    Construction and evaluation of a gfp-tagged Salmonella Typhimurium strain for environmental applications

    Salmonella enterica ser. Typhimurium was isolated from freshwater sediments and chromosomally labelled with a stable variant of the green fluorescent protein (GFP). The pUT mini-Tn5 Km transposon was used to introduce the gfp gene onto the chromosome of the S. Typhimurium strain by tri-parental mating. Southern Blot hybridisation confirmed that the gene had integrated into the chromosome. The gfp gene was stably maintained and the labelled strain was not growth-rate impaired. The incorporation of the gfp gene did not convey any significant loss of phenotype which would affect the survival and behaviour of the tagged strains. The tagged S. Typhimurium strain was used to spike an established drinking water biofilm and was able to colonise and persist within the biofilm. The tagged strain was also successfully used to study the survival of S. Typhimurium in natural sediments under different temperatures. These tagged strains can therefore be used to study the fate and survival of different Salmonella strains in water environments.Keywords: biofilm, green fluorescent protein, Salmonella Typhimurium, surviva

    Addressing environmental and atmospheric challenges for capturing high-precision thermal infrared data in the field of astro-ecology

    Using thermal infrared detectors mounted on drones, and applying techniques from astrophysics, we hope to support the field of conservation ecology by creating an automated pipeline for the detection and identification of certain endangered species and poachers from thermal infrared data. We test part of our system by attempting to detect simulated poachers in the field. Whilst we find that we can detect humans hiding in the field in some types of terrain, we also find several environmental factors that prevent accurate detection, such as ambient heat from the ground, absorption of infrared emission by the atmosphere, obscuring vegetation and spurious sources from the terrain. We discuss the effect of these issues, and potential solutions which will be required for our future vision for a fully automated drone-based global conservation monitoring system

    Thermal-Drones as a Safe and Reliable Method for Detecting Subterranean Peat Fires

    Underground peat fires are a major hazard to health and livelihoods in Indonesia, and are a major contributor to carbon emissions globally. Being subterranean, these fires can be difficult to detect and track, especially during periods of thick haze and in areas with limited accessibility. Thermal infrared detectors mounted on drones present a potential solution to detecting and managing underground fires, as they allow large areas to be surveyed quickly from above and can detect the heat transferred to the surface above a fire. We present a pilot study in which we show that underground peat fires can indeed be detected in this way. We also show that a simple temperature thresholding algorithm can be used to automatically detect them. We investigate how different thermal cameras and drone flying strategies may be used to reliably detect underground fires and survey fire-prone areas. We conclude that thermal equipped drones are potentially a very powerful tool for surveying for fires and firefighting. However, more investigation is still needed into their use in real-life fire detection and firefighting scenarios

    High-Resolution Patterned Cellular Constructs by Droplet-Based 3D Printing.

    Bioprinting is an emerging technique for the fabrication of living tissues that allows cells to be arranged in predetermined three-dimensional (3D) architectures. However, to date, there are limited examples of bioprinted constructs containing multiple cell types patterned at high-resolution. Here we present a low-cost process that employs 3D printing of aqueous droplets containing mammalian cells to produce robust, patterned constructs in oil, which were reproducibly transferred to culture medium. Human embryonic kidney (HEK) cells and ovine mesenchymal stem cells (oMSCs) were printed at tissue-relevant densities (10(7) cells mL(-1)) and a high droplet resolution of 1 nL. High-resolution 3D geometries were printed with features of ≤200 μm; these included an arborised cell junction, a diagonal-plane junction and an osteochondral interface. The printed cells showed high viability (90% on average) and HEK cells within the printed structures were shown to proliferate under culture conditions. Significantly, a five-week tissue engineering study demonstrated that printed oMSCs could be differentiated down the chondrogenic lineage to generate cartilage-like structures containing type II collagen

    Safety and efficacy of an engineered hepatotropic AAV gene therapy for ornithine transcarbamylase deficiency in cynomolgus monkeys

    X-linked inherited ornithine transcarbamylase deficiency (OTCD) is the most common disorder affecting the liver-based urea cycle, a pathway enabling detoxification of nitrogen waste and endogenous arginine biosynthesis. Patients develop acute hyperammonemia leading to neurological sequelae or death despite the best-accepted therapy based on ammonia scavengers and protein-restricted diet. Liver transplantation is curative but associated with procedure-related complications and lifelong immunosuppression. Adeno-associated viral (AAV) vectors have demonstrated safety and clinical benefits in a rapidly growing number of clinical trials for inherited metabolic liver diseases. Engineered AAV capsids have shown promising enhanced liver tropism. Here, we conducted a good-laboratory practice-compliant investigational new drug-enabling study to assess the safety of intravenous liver-tropic AAVLK03 gene transfer of a human codon-optimized OTC gene. Juvenile cynomolgus monkeys received vehicle and a low and high dose of vector (2 × 1012 and 2 × 1013 vector genome (vg)/kg, respectively) and were monitored for 26 weeks for in-life safety with sequential liver biopsies at 1 and 13 weeks post-vector administration. Upon completion of monitoring, animals were euthanized to study vector biodistribution, immune responses, and histopathology. The product was well tolerated with no adverse clinical events, predominant hepatic biodistribution, and sustained supra-physiological OTC overexpression. This study supports the clinical deployment of intravenous AAVLK03 for severe OTCD

    Successful observation of orangutans in the wild with thermal-equipped drones

    We investigated the efficacy of a drone equipped with a thermal camera as a potential survey tool to detect wild Bornean orangutans (Pongo pygmaeus) and other tropical primates. Using the thermal camera we successfully detected 41 orangutans and a troop of proboscis monkeys, all of which were confirmed by ground observers. We discuss the potential advantages and limitations of thermal-equipped drones as a tool to complement other methods, and the potential of this technology for use as a future survey tool

    Low temperature scattering with the R-matrix method: the Morse potential

    Experiments are starting to probe collisions and chemical reactions between atoms and molecules at ultra-low temperatures. We have developed a new theoretical procedure for studying these collisions using the R-matrix method. Here this method is tested for the atom -- atom collisions described by a Morse potential. Analytic solutions for continuum states of the Morse potential are derived and compared with numerical results computed using an R-matrix method where the inner region wavefunctions are obtained using a standard nuclear motion algorithm. Results are given for eigenphases and scattering lengths. Excellent agreement is obtained in all cases.     REFERQUAL: A pilot study of a new service quality assessment instrument in the GP Exercise Referral scheme setting

    Background The development of an instrument accurately assessing service quality in the GP Exercise Referral Scheme (ERS) industry could potentially inform scheme organisers of the factors that affect adherence rates leading to the implementation of strategic interventions aimed at reducing client drop-out. Methods A modified version of the SERVQUAL instrument was designed for use in the ERS setting and subsequently piloted amongst 27 ERS clients. Results Test re-test correlations were calculated via Pearson's 'r' or Spearman's 'rho', depending on whether the variables were Normally Distributed, to show a significant (mean r = 0.957, SD = 0.02, p < 0.05; mean rho = 0.934, SD = 0.03, p < 0.05) relationship between all items within the questionnaire. In addition, satisfactory internal consistency was demonstrated via Cronbach's 'α'. Furthermore, clients responded favourably towards the usability, wording and applicability of the instrument's items. Conclusion REFERQUAL is considered to represent promise as a suitable tool for future evaluation of service quality within the ERS community. Future research should further assess the validity and reliability of this instrument through the use of a confirmatory factor analysis to scrutinise the proposed dimensional structure

    Carbon nanotube (CNT) has been considered as an ideal interconnect material for replacing copper for future nanoscale IC technology due to its outstanding current carrying capability, thermal conductivity, and mechanical robustness. In this paper, crosstalk problems for single-walled carbon nanotube (SWCNT) bundle interconnects are investigated; the interconnect parameters for SWCNT bundle are calculated first, and then the equivalent circuit has been developed to perform the crosstalk analysis. Based on the simulation results using SPICE simulator, the voltage of the crosstalk-induced glitch can be reduced by decreasing the line length, increasing the spacing between adjacent lines, or increasing the diameter of SWCNT
