    Occurrence of phytoplasma associated with sugarcane yellow leaf syndrome in three regions of São Paulo State, Brazil

    Uma doença conhecida como síndrome do amarelecimento foliar da cana-de-açúcar ou amarelinho causou danos expressivos à cultura no estado de São Paulo, na década de noventa. A síndrome também foi registrada em outros países e investigações sobre a etiologia evidenciaram a associação da doença com vírus e fitoplasma, de forma isolada ou simultânea. A doença é potencialmente importante e ocorre endemicamente no estado paulista, por esta razão o presente estudo buscou demonstrar a ocorrência do fitoplasma em três tradicionais regiões produtoras de cana, localizadas no estado de São Paulo, através da detecção molecular deste agente em plantas naturalmente infectadas. Para isto, plantas sintomáticas das variedades SP71-6163, SP71-6180 e SP89-1115 foram amostradas em Piracicaba, Jaú e Ribeirão Preto e a partir do tecido foliar foi extraído o DNA total. PCR duplo foi processado com os "primers" P1/Tint-16F2n/R2 e os produtos foram analisados por eletroforese em gel de agarose. Amplificações de fragmentos de DNA de 1,2 kb evidenciaram a presença de fitoplasma em 36% das plantas sintomáticas e revelaram a sua ocorrência nas três regiões amostradas. Os resultados demonstraram a associação do fitoplasma com a doença e a importância de se manter uma atenta vigilância nos campos e viveiros, bem como buscar genótipos com bom nível de resistência nos programas de melhoramento.Sugarcane yellow leaf syndrome caused serious damage to crops in the São Paulo State, Brazil, in the 1990's. The syndrome was also reported in other countries and investigations into the etiology revealed an association between the disease and virus and/or phytoplasma. The disease is potentially important and occurs endemically in that State, and for this reason the present study was conducted in order to demonstrate the occurrence of phytoplasma in three traditional sugarcane-producing areas, in São Paulo State, through molecular detection from naturally infected plants. Symptomatic plants belonging to varieties SP71-6163, SP71-6180 and SP89-1115 were sampled from Piracicaba, Jaú and Ribeirão Preto, and total DNA was extracted from foliar tissues. Nested PCR was conducted with primer pairs P1/Tint-16F2n/R2, and the amplified products were analyzed by electrophoresis on agarose gels. Amplified DNA fragments of 1.2 kb evidenced the presence of phytoplasma in 36% of symptomatic plants and revealed its occurrence in all sampled regions. The results demonstrated that phytoplasma is associated with the disease and that it is important to keep a safe inspection of nurseries and monitoring plants in the field, as well as to select sugarcane genotypes with a good level of resistance in breeding programs.Coordenação de Aperfeiçoamento de Pessoal Nível Superior - CAPE

    Solidified floating organic drop microextraction (SFODME) for the simultaneous analysis of three non-steroidal anti-inflammatory drugs in aqueous samples by HPLC

    In this work, a liquid-liquid microextraction methodology using solidified floating organic drop (SFODME) was combined with liquid chromatography and UV/Vis detection to determine non-steroidal anti-inflammatory drugs (NSAIDs) naproxen (NPX), diclofenac (DCF), and mefenamic acid (MFN) in tap water, surface water, and seawater samples. Parameters that can influence the efficiency of the process were evaluated, such as the type and volume of the extractor and dispersive solvents, effect of pH, agitation type, and ionic strength. The optimized method showed low detection limits (0.09 to 0.25 μg L-1), satisfactory recovery rates (90 to 116%), and enrichment factors in the range between 149 and 199. SFODME showed simplicity, low cost, speed, and high concentration capacity of the analytes under study. Its use in real samples did not demonstrate a matrix effect that would compromise the effectiveness of the method, being possible to apply it successfully in water samples with different characteristics.publishe

    Comportamento do consumidor em resposta aos efeitos da pandemia da covid-19: um estudo sobre a relação entre a intenção de autoisolamento e compras incomuns

    Purpose: The aim of this study is to investigate consumer behavior in the context of the COVID-19 pandemic in Brazil to assess the relationship between the intention of self-isolation and to make unusual purchases. Methodology /Approach: Through an online survey with a sample of 181 individuals in Brazil, the proposed model and hypotheses were tested using Structural Equation Modeling (PLS-SEM.) Findings: The results demonstrate a link between perceived severity in the two behavioral responses measured, the intention to make unusual purchases and, more strongly, the intention to voluntary self-isolation. Originality/Value: The study discusses consumer behavior for unusual purchases (cyberchondria) in risky situations such as the COVID-19 pandemic in Brazil. Contributions and implications: We demonstrate how information overload leads to cyberchondria. In addition, the perceived severity leads the individual to make unusual purchases and self-isolation. In turn, exposure to online information sources leads to cyberchondria, which leads to behavior that increases the intention to make unusual purchases, and to self-isolation, which further increases exposure to online information. Furthermore, this study extends existing research (Laato et al., 2020) that suggests that research be carried out in different contexts.Objetivo: o propósito deste estudo é investigar o comportamento dos consumidores no contexto da pandemia de COVID-19 no Brasil para avaliar a relação entre a intenção de auto isolamento e eventuais compras incomuns. Método: Por meio de uma pesquisa on-line com uma amostra 181 indivíduos, o modelo proposto e as hipóteses foram testadas a partir da Modelagem de Equações Estruturais (PLS-SEM.)  Principais resultados: Os resultados demonstram ligação entre gravidade percebida nas duas respostas comportamentais medidas, a intenção de fazer comprar incomuns e, mais fortemente, a intenção de autoisolamento voluntário. Relevância/originalidade: O estudo traz para discussão o comportamento do consumidor para compras incomuns (cibercondria) em situações de risco como a pandemia da COVID-19 no Brasil. Contribuições e implicações: Demonstramos como a sobrecarga de informação leva à cibercondria. Além disso, que a gravidade percebida leva o indivíduo a fazer compras incomuns e ao autoisolamento. Por sua vez, a exposição a fontes de informação on-line leva à cibercondria, que leva a um comportamento que aumenta a intenção de fazer compras incomuns e ao autoisolamento o que aumenta ainda mais a exposição a informações on-line. Além disso, este estudo estende a pesquisa existente (Laato et al., 2020) que sugere que pesquisas sejam realizadas em contextos diferentes

    A extração do petróleo em áreas offshore é, em geral, acompanhada de água de formação, originária do reservatório. Esta água tem características oleosas com um alto teor de sais o que constitui um sério problema ambiental. O nível de concentração de amônia nesta água é superior ao limite máximo permitido para descarte. A principal dificuldade na remoção de amônia da água produzida é o alto nível de íons sódio e a presença de muitos outros íons interferentes encontrados nesta água.     Cereal bars functionalised with tempeh: nutritional composition, isoflavone content and consumer acceptance

    The aim of this study was to produce cereal bars (CB) added with tempeh flour (TF) and evaluate the nutritional composition, fatty acids profile, isoflavone content and sensory acceptance. Tempeh was produced from organic soya beans cotyledons fermented by Rhizophus oligosporus followed by freeze-drying and milling in order to obtain the TF. The addition of 10%, 15% and 20% TF to CB resulted in both protein and lipid content increase and a higher polyunsaturated/saturated fatty acids ratio. The content of isoflavone aglycones, the forms considered to have higher biological activity, was higher in TF than in soya bean, which was also reflected in the CB chemical composition. An acceptance test showed that CB added with TF (up to 15%) were sensory accepted. Overall results suggest that the addition of TF as an ingredient in cereal bars allows improving the chemical and functional characteristics of this type of ready-to-eat foods.info:eu-repo/semantics/publishedVersio

    Comprehensive analysis of The Cancer Genome Atlas reveals a unique gene and non-coding RNA signature of fibrolamellar carcinoma

    Fibrolamellar carcinoma (FLC) is a unique liver cancer primarily affecting young adults and characterized by a fusion event between DNAJB1 and PRKACA. By analyzing RNA-sequencing data from The Cancer Genome Atlas (TCGA) for >9,100 tumors across ~30 cancer types, we show that the DNAJB1-PRKACA fusion is specific to FLCs. We demonstrate that FLC tumors (n = 6) exhibit distinct messenger RNA (mRNA) and long intergenic non-coding RNA (lincRNA) profiles compared to hepatocellular carcinoma (n = 263) and cholangiocarcinoma (n = 36), the two most common liver cancers. We also identify a set of mRNAs (n = 16) and lincRNAs (n = 4), including LINC00473, that distinguish FLC from ~25 other liver and non-liver cancer types. We confirm this unique FLC signature by analysis of two independent FLC cohorts (n = 20 and 34). Lastly, we validate the overexpression of one specific gene in the FLC signature, carbonic anhydrase XII (CA12), at the protein level by western blot and immunohistochemistry. Both the mRNA and lincRNA signatures support a major role for protein kinase A (PKA) signaling in shaping the FLC gene expression landscape, and present novel candidate FLC oncogenes that merit further investigation
