    A novel approach for classification of underactuated mechanism in myoelectric hand

    Concept of underactuation makes it possible to create such robotic hands that automatically adapt to the shape of the grasped object without using a complex control system or a large number of actuators. In underactuated hand prostheses, depending upon control strategy used, one can get more and more degree of freedoms (DOFs) to make its prosthetic device more versatile and easy to control. In spite of all the great work done by the researchers on underactuated prosthetic hands, no one has given any relationship between the number of motors (or actuators) and the number of degree of freedoms (DOFs). This novel concept named as “Coefficient of UAM” (CoUAM) is “the ratio between numbers of motors to the number of DOFs”. By reviewing various known studies, CoUAM of randomly selected prosthetic hands is calculated separately and analyzed merits/demerits, cost, weight, appearance, ease of controllability and their functionality to make this approach more objective and useful for the future researchers. Hence target of this paper is to assess a Coefficient of UAM (CoUAM) of different types of prostheses available and then classify them into red, green and yellow zones

    Characterization of wide cross derivatives in rice Oryza sativa L. using genomic in situ hybridization (GISH)

    Interspecific crosses provide a bridge by which the gene pool of rice can be increased. Introduction of alien genes requires hybridization followed by meiotic pairing and recombination between the chromosomes of cultivated and wild species. Attempts have been made to visualize the genomic constitution of wide-cross derivatives. Genomic in situ hybridization was used to detect Oryza australiensis chromosomes and introgressed segment from O. australiensis into the Oryza sativa background. Genomic DNA from O. australiensis was labeled with biotin-14-dATP and hybridized to the homologous chromosomes in hybrids, back cross progenies, monosomic alien addition line (MAAL) and introgression line. The probe hybridization fluoresced green and non-labeled O. sativa chromosomes appeared red due to counterstaining with propidium iodide (PI). This differential painting of chromosomes unequivocally detected the O. australiensis chromatin introgressed into the O. sativa genome. The probe produced uniform labeling pattern over the entire length of all the O. australiensis chromosomes. Genomic in situ hybridization (GISH) detected 12 O. australiensis chromosomes in the hybrid, O. sativa x O. australiensis in BC1 progenies, and a single chromosome in MAAL. Small segment of O. australiensis was localized on the chromosome 12 of the introgression line. However, results showed that GISH is a powerful technique to be used as an aid in selecting segregating progenies.Key words: Genomic in situ hybridization, wide hybrid, localizing introgression

    Localizing introgression on the chromosome of rice by genomic in situ hybridization (GISH)

    Genomic in situ hybridization was used to detect introgressed segment from Oryza australinesis onto the chromosomes of introgression line derived from the hybrid O. sativa x O. australinesis. Genomic DNA from Oryza australinesis was labeled with biotin and hybridized to the homologous sequences on the O. sativa chromosomes. The probe hybridization fluoresced green and non labeled O. sativa chromosomes appeared red or blue due to counterstaining with propidium iodide (PI) or 4,6-diamidino-2-phenylindole (DAPI). This differential painting of chromosomes unequivocally detected the introgressedsegment. Among the 200 cells analyzed, 6.5% of the cells showed  hybridization signal. Signal appeared on one chromosome in 5%, on two homologous chromosomes in 1% and on sister chromatids in 0.5%of the cells. Hybridization was seen on the short arm of the chromosome 12 of the introgression line

    An experimental investigation into the dimensional error of powder-binder three-dimensional printing

    This paper is an experimental investigation into the dimensional error of the rapid prototyping additive process of powder-binder three-dimensional printing. Ten replicates of a purpose-designed part were produced using a three-dimensional printer, and measurements of the internal and external features of all surfaces were made using a general purpose coordinate measuring machine. The results reveal that the bases of all replicates (nominally flat) have a concave curvature, producing a flatness error of the primary datum. This is in contrast to findings regarding other three-dimensional printing processes, widely reported in the literature, where a convex curvature was observed. All external surfaces investigated in this study showed positive deviation from nominal values, especially in the z-axis. The z-axis error consisted of a consistent positive cumulative error and a different constant error in different replicates. By compensating for datum surface error, the average total height error of the test parts can be reduced by 25.52 %. All the dimensional errors are hypothesised to be explained by expansion and the subsequent distortion caused by layer interaction during and after the printing process

    Improving additive manufacturing performance by build orientation optimization

    Additive manufacturing (AM) is an emerging type of production technology to create three-dimensional objects layer-by-layer directly from a 3D CAD model. AM is being extensively used in several areas by engineers and designers. Build orientation is a critical issue in AM since it is associated with the part accuracy, the number of supports required and the processing time to produce the object. This paper presents an optimization approach to solve the part build orientation problem taking into account some characteristics or measures that can affect the accuracy of the part, namely the volumetric error, the support area, the staircase effect, the build time, the surface roughness and the surface quality.     The Effect of the El Nino Southern Oscillation on Precipitation Extremes in the Hindu Kush Mountains Range

    The El Nino Southern Oscillation (ENSO) phenomenon is devastating as it negatively impacts global climatic conditions, which can cause extreme events, including floods and droughts, which are harmful to the region’s economy. Pakistan is also considered one of the climate change hotspot regions in the world. Therefore, the present study investigates the effect of the ENSO on extreme precipitation events across the Upper Indus Basin. We examined the connections between 11 extreme precipitation indices (EPIs) and two ENSO indicators, the Southern Oscillation Index (SOI) and the Oceanic Niño Index (ONI). This analysis covers both annual and seasonal scales and spans the period from 1971 to 2019. Statistical tests (i.e., Mann–Kendall (MK) and Innovative Trend Analysis (ITA)) were used to observe the variations in the EPIs. The results revealed that the number of Consecutive Dry Days (CDDs) is increasing more than Consecutive Wet Days (CWDs); overall, the EPIs exhibited increasing trends, except for the Rx1 (max. 1-day precipitation) and Rx5 (max. 5-day precipitation) indices. The ENSO indicator ONI is a temperature-related ENSO index. The results further showed that the CDD value has a significant positive correlation with the SOI for most of the UIB (Upper Indus Basin) region, whereas for the CWD value, high elevated stations gave a positive relationship. A significant negative relationship was observed for the lower portion of the UIB. The Rx1 and Rx5 indices were observed to have a negative relationship with the SOI, indicating that El Nino causes heavy rainfall. The R95p (very wet days) and R99p (extreme wet days) indices were observed to have significant negative trends in most of the UIB. In contrast, high elevated stations depicted a significant positive relationship that indicates they are affected by La Nina conditions. The PRCPTOT index exhibited a negative relationship with the SOI, revealing that the El Nino phase causes wet conditions in the UIB. The ONI gave a significant positive relationship for the UIB region, reinforcing the idea that both indices exhibit more precipitation during El Nino. The above observations imply that while policies are being developed to cope with climate change impacts, the effects of the ENSO should also be considered

    Diagnostic accuracy for different strategies of image-guided breast intervention in cases of nonpalpable breast lesions

    To find out whether ultrasound-guided fine-needle aspiration (FNA) and ultrasound and stereotactic-guided large core needle biopsy (LCNB) are reliable alternatives to needle-localised open breast biopsy (NLBB) in daily practice, we performed a retrospective study and evaluated the validity of these methods. In all, 718 women with 749 nonpalpable breast lesions from three Dutch Hospitals were included, and the validity of the various methods for diagnosis was assessed. This was carried out according to a method described by Burbank and Parker for evaluating the quality of an image-guided breast intervention. We compared our results with the outcome of the COBRA study. Overall, all diagnostic strategies (NLBB, FNA, LCNB ultrasound and stereotactic guided) show comparable agreement rates. However, the miss rates differ: 2% for NLBB, 3% for COBRA (LCNB in study setting), 5% for FNA and 8-12% for LCNB in practice. Fine-needle aspiration was nonconclusive in 29%, and shows an overestimation for DCIS in 9%. The DCIS underestimate rate in NLBB was 8%. For the assessment of lesions consisting of microcalcifications only and to exclude malignancy in all other lesions, a 14-gauge needle should be used. Ultrasound-guided intervention can be performed in a large percentage of nonpalpable lesions. Lesions consisting only of microcalcifications on mammography need special attention

    Expression of M. tuberculosis-induced suppressor of cytokine signaling (SOCS) 1, SOCS3, FoxP3 and secretion of IL-6 associates with differing clinical severity of tuberculosis

    Background Appropriate immune activation of T cells and macrophages is central for the control of Mycobacterium tuberculosis infections. IFN-γ stimulated responses are lowered in tuberculosis (TB), while expression of Suppressor of Cytokine Signaling (SOCS) molecules – 1 and 3 and CD4+CD25+FoxP3+T regulatory cells is increased. Here we investigated the association of these molecules in regard to clinical severity of TB. Methods Peripheral blood mononuclear cells (PBMCs) were isolated from patients with pulmonary TB (PTB, n = 33), extra-pulmonary TB (ETB, n = 33) and healthy endemic controls (EC, n = 15). Cases were classified as moderately advanced or far advanced PTB, and less severe or severe disseminated ETB. M. tuberculosis -stimulated IFN-γ, SOCS1, SOCS3 and FoxP3 gene expression and secretion of Th1 and Th2 cytokines was measured. Statistical analysis was performed using Mann–Whitney U, Wilcoxon Rank and Kruskal Wallis non-parametric tests. Results In un-stimulated PBMCs, IL-6 (p = 0.018) and IL-10 (p = 0.013) secretion levels were increased in PTB while IL-10 was also increased in ETB (p = 0.003), all in comparison with EC. M. tuberculosis-stimulated IL-6 (p = 0.003) was lowered in ETB as compared with EC. SOCS1 mRNA expression in M. tuberculosis stimulated PBMCs levels in moderately advanced PTB (p = 0.022), far advanced (p = 0.014) PTB, and severe ETB (p = 0.009) were raised as compared with EC. On the other hand, SOCS1 mRNA titers were reduced in less severe ETB, in comparison with severe ETB (p = 0.027) and far advanced PTB (p = 0.016). SOCS3 mRNA accumulation was reduced in far advanced PTB (p = 0.007) and FoxP3 mRNA expression was increased in less severe ETB as compared with EC (p = 0.017). Conclusions The lowered SOCS1 mRNA levels in patients with less severe extra-pulmonary TB as compared to those with more severe ETB and PTB may lead to elevated IFN-γ pathway gene expression in the latter group. As localized ETB has shown to be associated with more effective Th1 immunity and adaptive responses, this suggests a role for SOCS1 in determining disease outcome in extra-pulmonary TB
