    Improving Interactivity in Instructional Design by Developing an ICT-based Social Studies Plan: Case Study of Smart Agriculture in the National Strategic Special Zone of Hyogo Prefecture’s Yabu City

    This research aimed at designing a lesson plan in elementary social studies that enhances interactivity by utilizing information and communications technology (ICT). Taking into consideration that lessons created along with students and ICT-based elementary social studies classes enhance students’ learning, and based on the features of iPad, applications, and Zoom, a lesson for the fifth grade—called “Agriculture in Japan-Smart Agriculture in Yabu City, National Strategic Special Zone”—was designed. To clarify the actual situation pertaining to ICT utilization in the classroom, the lesson plan was examined from the perspectives of “information gathering,” “information examination/thinking,” and “reflection.” Information gathering enabled searching the Web for supplementary/additional materials using learning content-related keywords, and also helped share information and enhance teaching materials. Information examination/thinking enabled the use of map-based applications to improve reading skills. Zoom also enabled interaction with persons mentioned in the learning and teaching materials. This way, students could apply their learning beyond the classroom to the real world. LoiLoNote, a class-support application, helped organize thoughts on cards. Like a portfolio, students were able to build a learning history on their own. All these made instant sharing of classmates’ learning possible. Thus, utilizing ICT contributed to upgrading teaching and learning style and designing elementary school social-studies lessons to enhance interactivity

    iSTFTNet2: Faster and More Lightweight iSTFT-Based Neural Vocoder Using 1D-2D CNN

    The inverse short-time Fourier transform network (iSTFTNet) has garnered attention owing to its fast, lightweight, and high-fidelity speech synthesis. It obtains these characteristics using a fast and lightweight 1D CNN as the backbone and replacing some neural processes with iSTFT. Owing to the difficulty of a 1D CNN to model high-dimensional spectrograms, the frequency dimension is reduced via temporal upsampling. However, this strategy compromises the potential to enhance the speed. Therefore, we propose iSTFTNet2, an improved variant of iSTFTNet with a 1D-2D CNN that employs 1D and 2D CNNs to model temporal and spectrogram structures, respectively. We designed a 2D CNN that performs frequency upsampling after conversion in a few-frequency space. This design facilitates the modeling of high-dimensional spectrograms without compromising the speed. The results demonstrated that iSTFTNet2 made iSTFTNet faster and more lightweight with comparable speech quality.     The nurse supporting robot system to prepare and hand surgical instruments to a surgeon is proposed to reduce work of nurse in a surgical operation. In this paper, the surgical instrument recognition system (SIRS) is developed to hand the surgical instruments to a surgeon by the robot. The characteristics ot the instruments are area of the instruments, ratio of minimum center-contour distance to maximum one and its outline contour, are recognized by using the image processing.     Periostin-expressing cell-specific transforming growth factor-β inhibition in pulmonary artery prevents pulmonary arterial hypertension

    Transforming growth factor beta (TGF-β) has been shown to play a critical role in pathogenesis of pulmonary arterial hypertension (PAH) although the precise role of TGF-β signaling remains uncertain. A recent report has shown that periostin (Pn) is one of the most upregulated proteins in human PAH lung compared with healthy lungs. We established type I TGF-β receptor knockout mice specifically with Pn expressing cell (Pn-Cre/Tgfb1fl/fl mice). Increases in PA pressure and pulmonary artery muscularization were induced by hypoxia of 10% oxygen for 4 weeks. Lung Pn expression was markedly induced by 4 week-hypoxia. Pn-Cre/Tgfb1fl/fl mice showed lower right ventricular pressure elevation, inhibition of PA medial thickening. Fluorescent co-immunostaining showed that Smad3 activation in Pn expressing cell is attenuated. These results suggest that TGF-β signaling in Pn expressing cell may have an important role in the pathogenesis of PAH by controlling medial thickening

    Background: Patients with essential thrombocythemia (ET) often experience bleeding associated with acquired von Willebrand syndrome (AVWS) when the platelet count is markedly increased. Objective: We investigated whether von Willebrand factor (VWF) degradation is enhanced in patients with ET. Methods: Seventy patients with ET underwent VWF multimer (VWFM) analysis and measurement of VWF-related parameters. We calculated the VWFM index, defined as the ratio of intensities of a patient's molecular weight-categorized VWFMs, and those of a healthy subject's, using densitometric analysis. VWF degradation product (DP) was measured via ELISA using a monoclonal antibody that specifically recognizes Y1605 at the C-terminal boundary, which is exposed following ADAMTS13-mediated cleavage of the Y1605-M1606 bond of the VWF A2 domain. Results: Patients with higher platelet counts had a significantly reduced high molecular weight (HMW)-VWFM index and an increased VWF-DP: VWF antigen (Ag) ratio compared to those with lower platelet counts. On multivariate analysis, the VWF-DP/ VWF:Ag ratio was an independent predictor of the HMW-VWFM index. Patients who underwent cytoreductive therapy had a significantly higher HMW-VWFM index and lower VWF-DP/ VWF:Ag ratio than those who did not. Among individual patients, there was also a significant increase in the HMW-VWFM index and a decrease in the VWF-DP/ VWF:Ag ratio after cytoreductive therapy compared to pre-therapy values. Conclusion: In patients with ET, an increased platelet count is associated with enhanced cleavage of VWF at the Y1605-M1606 bond, primarily by ADAMTS13, leading to AVWS. Cytoreductive therapy reduces the platelet count, prevents excessive VWF cleavage, and improves VWFM distributions.博士(医学)・甲第881号・令和5年3月15

    Effects of mild calorie restriction and high-intensity interval walking in middle-aged and older overweight Japanese

    We investigated whether a combination of mild calorie restriction (MCR) and high-intensity interval walking (HIW) improved physical fitness more than HIW alone in middle-aged and older overweight Japanese (40-69 years old, BMI >= 23.6 kg/m(2)). Forty-seven women and 16 men were divided into MCR + HIW and HIW groups. All subjects performed HIW: >= 5 sets of 3-min low-intensity walking (40% peak aerobic capacity for walking, VO2peak) and 3-min high-intensity walking (>= 70% VO2peak) per day, >= 4 days per week, for 16 weeks while energy expenditure was monitored with a tri-axial accelerometer. The MCR + HIW group consumed meal replacement formula (240 kcal): a mixture of low-carbohydrates and -fat and high-protein, for either lunch or dinner everyday and therefore, had similar to 87% of the energy intake of the HIW group during the intervention period. Although the HIW group showed improvements in BMI, blood pressure, and several blood chemicals, the MCR + HIW group had greater improvement. Moreover, the medical expenditure for the 6 months including the intervention period was 59% lower in the MCR + HIW group than in the HIW group.     Mutant PKC gamma in Spinocerebellar Ataxia Type 14 Disrupts Synapse Elimination and Long-Term Depression in Purkinje Cells In Vivo

    Cerebellar Purkinje cells (PCs) express a large amount of the gamma isoform of protein kinase C (PKC gamma) and a modest level of PKC alpha. The PKC gamma is involved in the pruning of climbing fiber (CF) synapses from developing PCs, and PKC alpha plays a critical role in long-term depression (LTD) at parallel fiber (PF)-PC synapses. Moreover, the PKC signaling in PCs negatively modulates the nonselective transient receptor potential cation channel type 3 (TRPC3), the opening of which elicits slow EPSCs at PF-PC synapses. Autosomal dominant spinocerebellar ataxia type 14 (SCA14) is caused by mutations in PKC gamma. To clarify the pathology of this disorder, mutant (S119P) PKC gamma tagged with GFP was lentivirally expressed in developing and mature mouse PCs in vivo, and the effects were assessed 3 weeks after the injection. Mutant PKC gamma-GFP aggregated in PCs without signs of degeneration. Electrophysiology results showed impaired pruning of CF synapses from developing PCs, failure of LTD expression, and increases in slow EPSC amplitude. We also found that mutant PKC gamma colocalized with wild-type PKC gamma, which suggests that mutant PKC gamma acts in a dominant-negative manner on wild-type PKC gamma. In contrast, PKC alpha did not colocalize with mutant PKC gamma. The membrane residence time of PKC alpha after depolarization-induced translocation, however, was significantly decreased when it was present with the mutant PKC gamma construct. These results suggest that mutant PKC gamma in PCs of SCA14 patients could differentially impair the membrane translocation kinetics of wild-type gamma and alpha PKCs, which would disrupt synapse pruning, synaptic plasticity, and synaptic transmission

    Complications Associated With Spine Surgery in Patients Aged 80 Years or Older: Japan Association of Spine Surgeons with Ambition (JASA) Multicenter Study

    Study Design:Retrospective study of registry data.Objectives:Aging of society and recent advances in surgical techniques and general anesthesia have increased the demand for spinal surgery in elderly patients. Many complications have been described in elderly patients, but a multicenter study of perioperative complications in spinal surgery in patients aged 80 years or older has not been reported. Therefore, the goal of the study was to analyze complications associated with spine surgery in patients aged 80 years or older with cervical, thoracic, or lumbar lesions.Methods:A multicenter study was performed in patients aged 80 years or older who underwent 262 spinal surgeries at 35 facilities. The frequency and severity of complications were examined for perioperative complications, including intraoperative and postoperative complications, and for major postoperative complications that were potentially life threatening, required reoperation in the perioperative period, or left a permanent injury.Results:Perioperative complications occurred in 75 of the 262 surgeries (29%) and 33 were major complications (13%). In multivariate logistic regression, age over 85 years (hazard ratio [HR] = 1.007, P = 0.025) and estimated blood loss ≥500 g (HR = 3.076, P = .004) were significantly associated with perioperative complications, and an operative time ≥180 min (HR = 2.78, P = .007) was significantly associated with major complications.Conclusions:Elderly patients aged 80 years or older with comorbidities are at higher risk for complications. Increased surgical invasion, and particularly a long operative time, can cause serious complications that may be life threatening. Therefore, careful decisions are required with regard to the surgical indication and procedure in elderly patients
