    Uso do diluente água de coco em pó (ACP-103®) na conservação prolongada do sêmen do varrão: avaliação in vitro e in vivo

    O sêmen de oito reprodutores foi coletado e de cada ejaculado separou-se um total de 1,75x10(9)sptz, com concentração de 35x10(6)sptz/mL. Usou-se o Beltsville Thawing Solution (BTS) como controle para testar o diluente água de coco em pó (ACP). O dia de coleta foi o dia zero (D0), sendo o sêmen conservado durante cinco dias, com análises diárias no D0 e nos quatro dias seguintes (D1, D2, D3 e D4). A avaliação da qualidade espermática baseou-se nos resultados do vigor espermático, da porcentagem de células móveis, da morfologia espermática, da integridade da membrana plasmática e dos resultados de fertilidade. As avaliações do vigor espermático (4,1) e da porcentagem de espermatozoides móveis (91%) ficaram acima dos parâmetros mínimos exigidos (3,0 e 70%) para sua utilização em programas de inseminação artificial, pois não houve influência sobre as variáveis analisadas pelo protocolo experimental. O resultado médio da resistência osmótica foi de 71,3% de espermatozoides com cauda enrolada. Não houve diferenças entre os dois diluentes testados para a característica células com acrossoma intacto (BTS = 67,1%; ACP = 71,2%). O sêmen diluído em ACP apresentou maior número de células vivas (77,7 %) com membrana plasmática íntegra (74,2%) após a conservação. A escolha do diluente ACP é aconselhável para uso de rotina em laboratórios que trabalhem com conservação de sêmen suíno. Apesar dos bons resultados in vitro obtidos com o diluente ACP, o BTS apresentou os melhores resultados de fertilidade, 86,7% e 96,7%, respectivamente.The semen of eight boars was collected and a total of 1.75x10(9) spermatozoa were separated from each ejaculate, obtaining a concentration of 35x10(6)sptz/mL. The Beltsville Thawing Solution (BTS) was used as control, being tested the powder coconut water as extender (PCW). The day of collection was considered day zero (D0), and the semen was conserved for five days, with analyses on D0 and on four subsequent days (D1, D2, D3, and D4). The evaluation of spermatic quality was based on the results of spermatozoa vigour, cells motility, spermatic morphology, plasmatic membrane integrity, and fertility. The evaluations of the in natura semen presented mean values of spermatozoa vigour (4.1) and mobile spermatozoids percentage (91%) above the demanded minimum parameters (3.0 and 70%) for its use in artificial insemination programs without influencing on the analyzed variables. The mean results of the osmotic resistance were excellent, with 71.3% of spermatozoids with coiled tail. There were no differences between the two extenders tested for the characteristic cells with intact acrossome (BTS = 67.1%; PCW = 71.2%). The semen diluted in PCW extender presented a higher number of alive cells (77.7%) with a complete plasmatic membrane (74.2%) after conservation period. The choice of PCW extender is advisable for the routine use in laboratories that work with conservation of swine semen. In spite of the good results in vitro with the PCW extender, the BTS presented the best fertility results, 86.7% and 96.7%, respectively

    Comparação da qualidade seminal de caprinos das raças Canindé e Alpina Britânica no Nordeste brasileiro

    RESUMO O estudo objetivou avaliar a qualidade seminal de caprinos das raças Canindé (autóctone) e Alpina Britânica (exótica) no Nordeste brasileiro. O experimento foi realizado nos períodos de julho a setembro dos anos de 2015 e 2016. As coletas seminais foram realizadas com auxílio de vagina artificial, de machos das raças Canindé (n = 4) e Alpina Britânica (n = 7). Após a coleta, o sêmen foi avaliado quanto a: volume (ml), concentração (sptz/ml), motilidade (%) e vigor (1-5). Em seguida, diluído em ACP-101c e criopreservado em máquina TK3000TM. Posteriormente, foram analisados os parâmetros cinéticos, através do software SCA®, e a atividade mitocondrial. No sêmen fresco, as duas raças apresentaram valores de motilidade e concentração dentro do preconizado para a espécie. Entretanto, os valores de volume e circunferência escrotal foram superiores na raça Alpina Britânica (0,73 ml ± 0,32; 27,35 cm ± 2,09) do que na raça Canindé (0,36 ml ± 0,07; 23,25 cm ± 0,95) (p &lt; 0,05). Já no sêmen descongelado, os parâmetros motilidade total - MT (36,96% ± 11,16 vs. 20,50% ± 14,15), motilidade progressiva - MP (26,67% ± 11,13 vs. 11,29% ± 9,42), vigor (2,91 ± 0,90 vs. 1,88 ± 0,67), velocidade curvilinear - VCL (78,12 µm/s ± 12,41 vs. 59,28 µm/s ± 15,57), velocidade linear - VSL (49,23 µm/s ± 9,87 vs. 29,9 µm/s ± 9,48), velocidade média da trajetória - VAP (66,08 µm/s ± 12,77 vs. 45,70 µm/s ± 12,20), linearidade - LIN (62,97% ± 6,96 vs. 49,54%±9,50), retilinearidade - STR (74,63% ± 6,44 vs. 65,77% ± 8,92), e oscilação - WOB (84,32% ± 5,94 vs. 74,42% ± 7,31) foram superiores para a raça Canindé em detrimento à Alpina Britânica (p &lt; 0,05). Portanto, o sêmen fresco das duas raças podem ser utilizados em biotécnicas reprodutivas. Já o sêmen pós-descongelação da raça Canindé apresentou melhor qualidade, provavelmente pelos animais estarem mais adaptados às condições adversas da região Nordeste, sendo recomendado para programas de inseminação artificial.</jats:p

    Avaliação de espermatozoides caprinos congelados em meio à base de água de coco em pó (ACP-101®) ou TRIS

    Compararam-se as características cinéticas e morfológicas de espermatozoides caprinos congelados nos meios à base de ACP-101® e TRIS. Os diluentes utilizados foram: ACP-101® (+ 2,5% gema ovo + 7% glicerol) e TRIS (+ 20% gema ovo + 6,8% glicerol). Quarenta e oito ejaculados de quatro bodes foram coletados, avaliados, divididos em duas alíquotas e diluídos nos meios ACP-101® e TRIS, respectivamente, posteriormente congelados e, após 30 dias, descongelados. A avaliação da motilidade espermática por computador foi realizada aos 5, 60 e 120 minutos pós-descongelação. As características de motilidade espermática analisadas foram: motilidade total (MT) (%) e progressiva (MP) (%), velocidades média do trajeto do espermatozoide (VAP) (µm/s) e linear (VSL) (µm/s) e população de espermatozoides rápidos (ER) (%). As avaliações de morfologia espermática quantificaram a porcentagem de espermatozoides normais (N) e as alterações da cabeça (AC), da peça intermediária (API) e do flagelo (AF), aos cinco e 120 minutos pós-descongelação. O diluente TRIS apresentou resultados cinéticos mais elevados que o ACP-101® aos 60 e 120 minutos pós-descongelação. As AC aos 120 minutos pós-descongelação foram mais altas nos espermatozoides congelados em ACP-101®. Conclui-se que o diluente TRIS promoveu maior viabilidade in vitro dos espermatozoides caprinos pós-descongelação

    Comparison of the seminal quality of Canindé and British Alpine goats in the Brazilian Northeast

    ABSTRACT The objective of this study was to evaluate the sperm quality of Canindé (native) and British Alpine (exotic) goats in the Northeast of Brazil. The experiment was carried out from July to September of the years 2015 and 2016. Sperm collections of Canindé (n = 4) and British Alpine (n = 7) males were performed using artificial vagina. After collection, the sperm was evaluated for volume (ml), concentration (sptz/ml), motility (%), and vigor (1-5). Then diluted in ACP-101c and cryopreserved in TK3000TM machine. Subsequently, kinetic parameters were analyzed through SCA TM software and mitochondrial activity. In fresh sperm, the two races presented values of motility and concentration within the recommended for the specie. However, volume and scrotal circumference values were higher in the British Alpine breed (0.73 mL ± 0.32; 27.35 cm ± 2.09) than in the Canindé breed (0.36 mL ± 0.07; 23 , 25 cm ± 0.95) (p </p

    Behavioural and seminal characteristics of Santa Ines rams subjected to successive semen collections in the Amazonian region

    The objective of this study was to evaluate the reproductive performance of Santa Ines rams subjected to successive semen collections in an Amazonian climate. Four rams were subjected to successive ejaculations during a maximum period of three hours. This procedure was repeated three times at 15-day intervals. Sexual and behavioural (libido) and andrological (testicular and seminal) assessments were performed. A total of 81 ejaculates were collected. Libido and semen vigour, volume, appearance and concentration decreased as the ejaculation frequency increased (P<0.05) and sperm motility showed a decreasing trend (P=0.06). The seminal pH increased over the sequence of collections (P<0.05). The only significant differences observed between individual rams were the variable scrotal circumference and the percentages of live sperm and sperm abnormalities (P<0.05). All the parameters of the first ejaculation were within the normal range for this species, which suggests that the local climatic conditions (high temperature and humidity) did not affect the behavioural, testicular or seminal parameters of experimental rams. Our findings indicate that the reproductive performance of Santa Ines rams could be affected by the intensification of ejaculation frequency; however, individual male variation needs to be taken into consideration

    Insulin addition to swine semen diluted and cooled at 15 ºC

    The objective of this study was to evaluate the effect of adding different doses of insulin to swine semen processed and stored at 15 ºC. The experiment used sixteen ejaculates from four commercial breeding pigs, distributed in a randomized block design (ejaculate) with split plot along time (0, 24, 48 and 72 hours of storage) with four treatments (insulin levels - 0.0 4.0 8.0 and 12.0 IU per dose) and 16 repetitions. The experimental unit was made of two insemination doses of 100 mL each, with 3×10(9) spermatozoids. Insulin used was NPH-human, added at the time of processing the doses. The addition of insulin did not affect motility, sperm viability, the percentage of abnormal cells, the osmotic resistance or the degradation rate of motility in 120 minutes. There was a linear decrease in semen quality over storage time, regardless of insulin levels. The addition of insulin at the mentioned concentrations does not influence the quality of insemination dose in pigs

    USO DE DILUENTES E TEMPERATURAS ALTERNATIVAS NA CONSERVAÇÃO PROLONGADA DO SÊMEN DO VARRÃO

    The use of appropriate extenders is important for the success of an artificial insemination program. The objective of this study was to evaluate the efficiency of alternative extenders for swine semen at different temperatures (17 to 10 °C). The following extenders were used: Beltsville Thawing Solution (BTS), powdered coconut water (ACP-103®), and skimmed milk powder (LPD). The 50 ejaculates were analyzed daily, in natura and after dilution, during the 5-day period of semen preservation (D0 to D4), regarding spermatic vigor and motility. Acrosome integrity and sperm viability were evaluated on D0 and D4. Data were analyzed using Mann-Whitney, Students, Tukey and chi-square tests (p<0.05). The LPD extender at 10 °C presented higher motility and sperm vigor compared to BTS and ACP until D2, and to treatments stored at 17 °C. Acrosome vitality and integrity remained higher (p<0.001) with LPD at 10 °C on D0 and D4. LPD showed to be a good extender for the swine semen at lower temperature (10 °C).
