Interleukin-12 bypasses common gamma-chain signalling in emergency natural killer cell lymphopoiesis
Differentiation and homeostasis of natural killer (NK) cells relies on common gamma-chain (γc)-dependent cytokines, in particular IL-15. Consequently, NK cells do not develop in mice with targeted γc deletion. Herein we identify an alternative pathway of NK-cell development driven by the proinflammatory cytokine IL-12, which can occur independently of γc-signalling. In response to viral infection or upon exogenous administration, IL-12 is sufficient to elicit the emergence of a population of CD122+CD49b+ cells by targeting NK-cell precursors (NKPs) in the bone marrow (BM). We confirm the NK-cell identity of these cells by transcriptome-wide analyses and their ability to eliminate tumour cells. Rather than using the conventional pathway of NK-cell development, IL-12-driven CD122+CD49b+ cells remain confined to a NK1.1lowNKp46low stage, but differentiate into NK1.1+NKp46+ cells in the presence of γc-cytokines. Buttressing staples with cholecyst-derived extracellular matrix (CEM) reinforces staple lines in an ex vivo peristaltic inflation model

Copyright @ Springer Science + Business Media, LLC 2008Background - Staple line leakage and bleeding are the most common problems associated with the use of surgical staplers for gastrointestinal resection and anastomotic procedures. These complications can be reduced by reinforcing the staple lines with buttressing materials. The current study reports the potential use of cholecyst-derived extracellular matrix (CEM) in non-crosslinked (NCEM) and crosslinked (XCEM) forms, and compares their mechanical performance with clinically available buttress materials [small intestinal submucosa (SIS) and bovine pericardium (BP)] in an ex vivo small intestine model.
Methods - Three crosslinked CEM variants (XCEM0005, XCEM001, and XCEM0033) with different degree of crosslinking were produced. An ex vivo peristaltic inflation model was established. Porcine small intestine segments were stapled on one end, using buttressed or non-buttressed surgical staplers. The opened, non-stapled ends were connected to a peristaltic pump and pressure transducer and sealed. The staple lines were then exposed to increased intraluminal pressure in a peristaltic manner. Both the leak and burst pressures of the test specimens were recorded.
Results - The leak pressures observed for non-crosslinked NCEM (137.8 ± 22.3 mmHg), crosslinked XCEM0005 (109.1 ± 14.1 mmHg), XCEM001 (150.1 ± 16.0 mmHg), XCEM0033 (98.8 ± 10.5 mmHg) reinforced staple lines were significantly higher when compared to non-buttressed control (28.3 ± 10.8 mmHg) and SIS (one and four layers) (62.6 ± 11.8 and 57.6 ± 12.3 mmHg, respectively) buttressed staple lines. NCEM and XCEM were comparable to that observed for BP buttressed staple lines (138.8 ± 3.6 mmHg). Only specimens with reinforced staple lines were able to achieve high intraluminal pressures (ruptured at the intestinal mesentery), indicating that buttress reinforcements were able to withstand pressure higher than that of natural tissue (physiological failure).
Conclusions - These findings suggest that the use of CEM and XCEM as buttressing materials is associated with reinforced staple lines and increased leak pressures when compared to non-buttressed staple lines. Ultraviolet asymptotics of scalar and pseudoscalar correlators in hot Yang-Mills theory
Inspired by recent lattice measurements, we determine the short-distance (a
> omega >> pi T) asymptotics
of scalar (trace anomaly) and pseudoscalar (topological charge density)
correlators at 2-loop order in hot Yang-Mills theory. The results are expressed
in the form of an Operator Product Expansion. We confirm and refine the
determination of a number of Wilson coefficients; however some discrepancies
with recent literature are detected as well, and employing the correct values
might help, on the qualitative level, to understand some of the features
observed in the lattice measurements. On the other hand, the Wilson
coefficients show slow convergence and it appears uncertain whether this
approach can lead to quantitative comparisons with lattice data. Nevertheless,
as we outline, our general results might serve as theoretical starting points
for a number of perhaps phenomenologically more successful lines of

Gauge invariant definition of the jet quenching parameter
In the framework of Soft-Collinear Effective Theory, the jet quenching
parameter, , has been evaluated by adding the effect of Glauber gluon
interactions to the propagation of a highly-energetic collinear parton in a
medium. The result, which holds in covariant gauges, has been expressed in
terms of the expectation value of two Wilson lines stretching along the
direction of the four-momentum of the parton. In this paper, we show how that
expression can be generalized to an arbitrary gauge by the addition of
transverse Wilson lines. The transverse Wilson lines are explicitly computed by
resumming interactions of the parton with Glauber gluons that appear only in
non-covariant gauges. As an application of our result, we discuss the
contribution to coming from transverse momenta of order in a

Multiscale methods for the solution of the Helmholtz and Laplace equations
This paper presents some numerical results about applications of multiscale techniques to boundary integral equations. The numerical schemes developed here are to some extent based on the results of the papers [6]—[10]. Section 2 deals with a short description of the theory of generalized Petrov-Galerkin methods for elliptic periodic pseudodifferential equations in covering classical Galerkin schemes, collocation, and other methods. A general setting of multiresolution analysis generated by periodized scaling functions as well as a general stability and convergence theory for such a framework is outlined. The key to the stability analysis is a local principle due to one of the authors. Its applicability relies here on a sufficiently general version of a so-called discrete commutator property of wavelet bases (see [6]). These results establish important prerequisites for developing and analysing methods for the fast solution of the resulting linear systems (Section 2.4). The crucial fact which is exploited by these methods is that the stiffness matrices relative to an appropriate wavelet basis can be approximated well by a sparse matrix while the solution to the perturbed problem still exhibits the same asymptotic accuracy as the solution to the full discrete problem. It can be shown (see [7]) that the amount of the overall computational work which is needed to realize a required accuracy is of the order , where is the number of unknowns and is some real number
Quantifying The Causes of Differences in Tropospheric OH within Global Models
The hydroxyl radical (OH) is the primary daytime oxidant in the troposphere and provides the main loss mechanism for many pollutants and greenhouse gases, including methane (CH4). Global mean tropospheric OH differs by as much as 80% among various global models, for reasons that are not well understood. We use neural networks (NNs), trained using archived output from eight chemical transport models (CTMs) that participated in the POLARCAT Model Intercomparison Project (POLMIP), to quantify the factors responsible for differences in tropospheric OH and resulting CH4 lifetime (τCH4) between these models. Annual average τCH4, for loss by OH only, ranges from 8.0–11.6 years for the eight POLMIP CTMs. The factors driving these differences were quantified by inputting 3-D chemical fields from one CTM into the trained NN of another CTM. Across all CTMs, the largest mean differences in τCH4 (ΔτCH4) result from variations in chemical mechanisms (ΔτCH4 = 0.46 years), the photolysis frequency (J) of O3→O(1D) (0.31 years), local O3 (0.30 years), and CO (0.23 years). The ΔτCH4 due to CTM differences in NOx (NO + NO2) is relatively low (0.17 years), though large regional variation in OH between the CTMs is attributed to NOx. Differences in isoprene and J(NO2) have negligible overall effect on globally averaged tropospheric OH, though the extent of OH variations due to each factor depends on the model being examined. ECOLOGICAL SERVICES OF AGROFORESTRY LANDSCAPES IN SELECTED WATERSHED AREAS IN THE PHILIPPINES AND INDONESIA
This article argues that the practice of agroforestry provides ecological contributions to the smallholder farmers cultivating in the watershed areas. Specifically, this farming system provides contribution to carbon sequestration potential of the woody perennials and the biodiversity conservation of the other components of the system.  This argument is based on the research conducted in Molawin-Dampalit Sub-Watershed, Mt. Makiling Forest Reserve in the Philippines and Way Betung Watershed in Indonesia. The research involved an interview session of 106 and 261 smallholder farmers and an assessment of 27 and 14 agroforesty plots for carbon stock assessment and biodiversity assessment, respectively. Results indicated that the total carbon found among the crop components was 52.32 MgC/ha in Molawin-Dampalit Sub-Watershed and 244.26 MgC/ha in Way Betung WatershedÂ, which suggested the high carbon sequestration potential of the woody perennials and understory crops in an agroforestry system. The farm lots being cultivated by the smallholder farmers were found to contribute to biodiversity conservation having a moderate biodiversity index of 2.59 and 2.53, respectively. With these findings, promotion of desired agroforestry systems in suitable portions of the watershed areas should be intensified and heightened to contribute to ecological balance across the landscape. Agroforestry should always be an integral part of all initiatives toward ecological restoration with the cultivators/smallholder farmers as potential partners. The agroforestry system should consider all the technical and socioeconomic considerations toward having diverse components and ensure food security among the smallholder farmers throughout the year
