Island tameness and the repeatability of flight initiation distance in a large herbivore

The final version is available from NRC Research Press via the DOI in this record.Antipredator behaviours can be lost relatively quickly in populations that are relieved of predation, as is known for several species inhabiting islands. Flight initiation distance (FID) is often studied in the context of island tameness; however, little is known about the factors that influence and maintain FID variation in predation-free populations. Here, we studied FID in foals of an isolated predator-free population of feral horses (Equus caballus L., 1758) on Sable Island, Canada, to determine if FID could be used for research on consistent individual differences in risk aversion and island tameness. In addition to testing for temporal, spatial, and sex effects on FID, we compared repeatability estimates at two temporal scales (within and among days). Similar FID for measurements obtained on the same day and for males and females indicated an absence of short-term desensitization and sex effects. In contrast, FID decreased for measurements made on subsequent days and from east to west, which could reflect habituation to human presence and (or) other temporal and spatial processes. Repeatability was high (0.42 ± 0.06), but tended to decrease with increasing time intervals. Efecto de la inclusión dietaria de glicerina sobre el desempeño productivo de conejos en crecimiento.
The possibility of using crude glycerine as a rabbit feed supplement is unknown in our country, therefore, this experiment was carried out in order to evaluate the effect of glycerin inclusion in the diet on growing rabbits performance. Thirty six White New Zealand weaned rabbits (18 male and 18 female) 35 d age (1008.61 ± 16.24 g initial BW) were assigned randomly to one of three treatments: A control diet and two levels of high purity glycerin (99%) inclusion (10 and 15%) in the diet; in a complete blocks experimental design with sub-sampling. No significant differences were observed in BW (P > 0.05) as a result of experimental diets. General means of daily weight gain were: 27.83, 26.71 and 23.38 g for T1, T2 and T3 treatments respectively, however differences between daily weight gains were not significant (P > 0.05). Calculated feed intake and feed conversion ratio were not affected by dietary treatments like the carcass yield (P > 0.05). Our results suggest that glycerin could be an appropiate energy source for growing rabbits diets at levels used in this experiment.La posibilidad de utilizar glicerina como fuente de energía en dietas para conejos en crecimiento no se ha evaluado en nuestro medio, por consiguiente se realizó un experimento para evaluar el efecto de la inclusión de glicerina en la ración sobre el desempeño productivo de conejos en crecimiento. Se utilizaron 36 conejos (18 hembras y 18 machos) destetos de raza Nueva Zelanda . Blanco, de 35 días de edad, con un peso inicial de 1008.61 ± 16.24 g, los cuales se sometieron a tres tratamientos: Una dieta control y dos tratamientos correspondientes a dos niveles de inclusión (10 y 15% en base seca) de glicerina de alta pureza (99%). Los animales fueron asignados aleatoriamente a los tratamientos en un diseño de bloques completos al azar con submuestreo. No se obtuvieron diferencias significativas (P > 0.05) en peso vivo por efecto de las dietas experimentales. El promedio general de ganancia diaria de peso fue de 26 g y no se observaron diferencias significativas entre tratamientos (P > 0.05). Tampoco se obtuvieron diferencias significativas (P > 0.05) por efecto de las dietas experimentales en el consumo estimado de alimento, conversión alimenticia estimada y rendimiento en canal (P > 0.05). Los resultados sugieren que la glicerina podría ser un ingrediente útil como fuente energética en raciones para conejos de engorde a los niveles de inclusión utilizados en el presente trabajo.
 
Deletion of parasite immune modulatory sequences combined with immune activating signals enhances vaccine mediated protection against filarial nematodes
<p>Background: Filarial nematodes are tissue-dwelling parasites that can be killed by Th2-driven immune effectors, but that have evolved to withstand immune attack and establish chronic infections by suppressing host immunity. As a consequence, the efficacy of a vaccine against filariasis may depend on its capacity to counter parasite-driven immunomodulation.</p>
<p>Methodology and Principal Findings: We immunised mice with DNA plasmids expressing functionally-inactivated forms of two immunomodulatory molecules expressed by the filarial parasite Litomosoides sigmodontis: the abundant larval transcript-1 (LsALT) and cysteine protease inhibitor-2 (LsCPI). The mutant proteins enhanced antibody and cytokine responses to live parasite challenge, and led to more leukocyte recruitment to the site of infection than their native forms. The immune response was further enhanced when the antigens were targeted to dendritic cells using a single chain Fv-αDEC205 antibody and co-administered with plasmids that enhance T helper 2 immunity (IL-4) and antigen-presenting cell recruitment (Flt3L, MIP-1α). Mice immunised simultaneously against the mutated forms of LsALT and LsCPI eliminated adult parasites faster and consistently reduced peripheral microfilaraemia. A multifactorial analysis of the immune response revealed that protection was strongly correlated with the production of parasite-specific IgG1 and with the numbers of leukocytes present at the site of infection.</p>
<p>Conclusions: We have developed a successful strategy for DNA vaccination against a nematode infection that specifically targets parasite-driven immunosuppression while simultaneously enhancing Th2 immune responses and parasite antigen presentation by dendritic cells.</p>
A single-electron transistor made from a cadmium selenide nanocrystal
The techniques of colloidal chemistry permit the routine creation of
semiconductor nanocrystals, whose dimensions are much smaller than those that
can be realized using lithographic techniques. The sizes of such nanocrystals
can be varied systematically to study quantum size effects or to make novel
electronic or optical materials with tailored properties. Preliminary studies
of both the electrical and optical properties of individual nanocrystals have
been performed recently. These studies show clearly that a single excess charge
on a nanocrystal can markedly influence its properties. Here we present
measurements of electrical transport in a single-electron transistor made from
a colloidal nanocrystal of cadmium selenide. This device structure enables the
number of charge carriers on the nanocrystal to be tuned directly, and so
permits the measurement of the energy required for adding successive charge
carriers. Such measurements are invaluable in understanding the energy-level
spectra of small electronic systems, as has been shown by similar studies of

Maternal corticotropin-releasing hormone is associated with LEP DNA methylation at birth and in childhood: an epigenome-wide study in Project Viva
BackgroundCorticotropin-releasing hormone (CRH) plays a central role in regulating the secretion of cortisol which controls a wide range of biological processes. Fetuses overexposed to cortisol have increased risks of disease in later life. DNA methylation may be the underlying association between prenatal cortisol exposure and health effects. We investigated associations between maternal CRH levels and epigenome-wide DNA methylation of cord blood in offsprings and evaluated whether these associations persisted into mid-childhood.MethodsWe investigated mother-child pairs enrolled in the prospective Project Viva pre-birth cohort. We measured DNA methylation in 257 umbilical cord blood samples using the HumanMethylation450 Bead Chip. We tested associations of maternal CRH concentration with cord blood cells DNA methylation, adjusting the model for maternal age at enrollment, education, maternal race/ethnicity, maternal smoking status, pre-pregnancy body mass index, parity, gestational age at delivery, child sex, and cell-type composition in cord blood. We further examined the persistence of associations between maternal CRH levels and DNA methylation in children's blood cells collected at mid-childhood (n = 239, age: 6.7-10.3 years) additionally adjusting for the children's age at blood drawn.ResultsMaternal CRH levels are associated with DNA methylation variability in cord blood cells at 96 individual CpG sites (False Discovery Rate <0.05). Among the 96 CpG sites, we identified 3 CpGs located near the LEP gene. Regional analyses confirmed the association between maternal CRH and DNA methylation near LEP. Moreover, higher maternal CRH levels were associated with higher blood-cell DNA methylation of the promoter region of LEP in mid-childhood (P < 0.05, β = 0.64, SE = 0.30).ConclusionIn our cohort, maternal CRH was associated with DNA methylation levels in newborns at multiple loci, notably in the LEP gene promoter. The association between maternal CRH and LEP DNA methylation levels persisted into mid-childhood
KCa2 channels activation prevents [Ca2+]i deregulation and reduces neuronal death following glutamate toxicity and cerebral ischemia
Exacerbated activation of glutamate receptor-coupled calcium channels and subsequent increase in intracellular calcium ([Ca2+]i) are established hallmarks of neuronal cell death in acute and chronic neurological diseases. Here we show that pathological [Ca2+]i deregulation occurring after glutamate receptor stimulation is effectively modulated by small conductance calcium-activated potassium (KCa2) channels. We found that neuronal excitotoxicity was associated with a rapid downregulation of KCa2.2 channels within 3 h after the onset of glutamate exposure. Activation of KCa2 channels preserved KCa2 expression and significantly reduced pathological increases in [Ca2+]i providing robust neuroprotection in vitro and in vivo. These data suggest a critical role for KCa2 channels in excitotoxic neuronal cell death and propose their activation as potential therapeutic strategy for the treatment of acute and chronic neurodegenerative disorders
Quantum Criticality in Heavy Fermion Metals
Quantum criticality describes the collective fluctuations of matter
undergoing a second-order phase transition at zero temperature. Heavy fermion
metals have in recent years emerged as prototypical systems to study quantum
critical points. There have been considerable efforts, both experimental and
theoretical, which use these magnetic systems to address problems that are
central to the broad understanding of strongly correlated quantum matter. Here,
we summarize some of the basic issues, including i) the extent to which the
quantum criticality in heavy fermion metals goes beyond the standard theory of
order-parameter fluctuations, ii) the nature of the Kondo effect in the quantum
critical regime, iii) the non-Fermi liquid phenomena that accompany quantum
criticality, and iv) the interplay between quantum criticality and

Caveolin 1 protein expression in renal cell carcinoma predicts survival
<p>Abstract</p> <p>Background</p> <p>Caveolae play a significant role in disease phenotypes such as cancer, diabetes, bladder dysfunction, and muscular dystrophy. The aim of this study was to elucidate the caveolin-1 <it>(</it>CAV1<it>) </it>protein expression in renal cell cancer (RCC) and to determine its potential prognostic relevance.</p> <p>Methods</p> <p>289 clear cell RCC tissue specimens were collected from patients undergoing surgery for renal tumors. Both cytoplasmic and membranous CAV1 expression were determined by immunohistochemistry and correlated with clinical variables. Survival analysis was carried out for 169 evaluable patients with a median follow up of 80.5 months (interquartile range (IQR), 24.5 - 131.7 months).</p> <p>Results</p> <p>A high CAV1 expression in the tumor cell cytoplasm was significantly associated with male sex (p = 0.04), a positive nodal status (p = 0.04), and poor tumor differentiation (p = 0.04). In contrast, a higher than average (i.e. > median) CAV1 expression in tumor cell membranes was only linked to male sex (p = 0.03). Kaplan-Meier analysis disclosed significant differences in 5-year overall (51.4 vs. 75.2%, p = 0.001) and tumor specific survival (55.3 vs. 80.1%, p = 0.001) for patients with higher and lower than average cytoplasmic CAV1 expression levels, respectively. Applying multivariable Cox regression analysis a high CAV1 protein expression level in the tumor cell cytoplasm could be identified as an independent poor prognostic marker of both overall (p = 0.02) and tumor specific survival (p = 0.03) in clear cell RCC patients.</p> <p>Conclusion</p> <p>Over expression of caveolin-1 in the tumour cell cytoplasm predicts a poor prognosis of patients with clear cell RCC. CAV1 is likely to be a useful prognostic marker and may play an important role in tumour progression. Therefore, our data encourage further investigations to enlighten the role of CAV1 and its function as diagnostic and prognostic marker in serum and/or urine of RCC patients.</p
Building an Aerial-Ground Robotics System for Precision Farming: An Adaptable Solution
The application of autonomous robots in agriculture is gaining increasing
popularity thanks to the high impact it may have on food security,
sustainability, resource use efficiency, reduction of chemical treatments, and
the optimization of human effort and yield. With this vision, the Flourish
research project aimed to develop an adaptable robotic solution for precision
farming that combines the aerial survey capabilities of small autonomous
unmanned aerial vehicles (UAVs) with targeted intervention performed by
multi-purpose unmanned ground vehicles (UGVs). This paper presents an overview
of the scientific and technological advances and outcomes obtained in the
project. We introduce multi-spectral perception algorithms and aerial and
ground-based systems developed for monitoring crop density, weed pressure, crop
nitrogen nutrition status, and to accurately classify and locate weeds. We then
introduce the navigation and mapping systems tailored to our robots in the
agricultural environment, as well as the modules for collaborative mapping. We
finally present the ground intervention hardware, software solutions, and
interfaces we implemented and tested in different field conditions and with
different crops. We describe a real use case in which a UAV collaborates with a
UGV to monitor the field and to perform selective spraying without human

Ferret badger rabies origin and its revisited importance as potential source of rabies transmission in Southeast China
<p>Abstract</p> <p>Background</p> <p>The frequent occurrence of ferret badger-associated human rabies cases in southeast China highlights the lack of laboratory-based surveillance and urges revisiting the potential importance of this animal in rabies transmission. To determine if the ferret badgers actually contribute to human and dog rabies cases, and the possible origin of the ferret badger-associated rabies in the region, an active rabies survey was conducted to determine the frequency of rabies infection and seroprevalence in dogs and ferret badgers.</p> <p>Methods</p> <p>A retrospective survey on rabies epidemics was performed in Zhejiang, Jiangxi and Anhui provinces in southeast China. The brain tissues from ferret badgers and dogs were assayed by fluorescent antibody test. Rabies virus was isolated and sequenced for phylogenetic analysis. The sera from ferret badgers and dogs were titrated using rabies virus neutralizing antibodies (VNA) test.</p> <p>Results</p> <p>The ferret badgers presented a higher percentage of rabies seroconversion than dogs did in the endemic region, reaching a maximum of 95% in the collected samples. Nine ferret badger-associated rabies viruses were isolated, sequenced, and were phylogenetically clustered as a separate group. Nucleotide sequence revealed 99.4-99.8% homology within the ferret badger isolates, and 83-89% homology to the dog isolates in the nucleoprotein and glycoprotein genes in the same rabies endemic regions.</p> <p>Conclusions</p> <p>Our data suggest ferret badger-associated rabies has likely formed as an independent enzootic originating from dogs during the long-term rabies infestation in southeast China. The eventual role of FB rabies in public health remains unclear. However, management of ferret badger bites, rabies awareness and control in the related regions should be an immediate need.</p
