    Does 4-H Camp Influence Life Skill and Leadership Development?

    WV conducted a two-phase study involving over 2,000 campers to learn how 4-H camp affects life skills and leadership development. Camp is at the heart of many states\u27 4-H programs; however, there is limited research to document the impact. Fifteen counties with 28 individual camps participated in the study, which measured (1) camp experience, (2) targeted Life Skills, and (3) leadership skills. The study found that 4-H experiential learning activities at camp positively affect campers\u27 life skills and leadership skills. Results should be used to guide the future measurement of 4 H camp impact and to strengthen camping curriculums

    Finding AGN in Deep X-ray Flux States with Swift

    We report on our ongoing project of finding Active Galactic Nuclei (AGN) that go into deep X-ray flux states detected by Swift. Swift is performing an extensive study on the flux and spectral variability of AGN using Guest Investigator and team fill-in programs followed by triggering XMM_Newton for deeper follow-up observations. So far this program has been very successful and has led to a number of XMM-Newton follow up observations, including Mkn 335, PG 0844+349, and RX J2340.8-5329. Recent analysis of new Swift AGN observations reveal several AGN went into a very low X-ray flux state, particularly Narrow-Line Seyfert 1 galaxies.     Aging Reveals a Role for Nigral Tyrosine Hydroxylase ser31 Phosphorylation in Locomotor Activity Generation

    BACKGROUND:Tyrosine hydroxylase (TH) regulates dopamine (DA) bioavailability. Its product, L-DOPA, is an established treatment for Parkinson's disease (PD), suggesting that TH regulation influences locomotion. Site-specific phosphorylation of TH at ser31 and ser40 regulates activity. No direct evidence shows that ser40 phosphorylation is the dominating mechanism of regulating TH activity in vivo, and physiologically-relevant stimuli increase L-DOPA biosynthesis independent of ser40 phosphorylation. Significant loss of locomotor activity occurs in aging as in PD, despite less loss of striatal DA or TH in aging compared to the loss associated with symptomatic PD. However, in the substantia nigra (SN), there is equivalent loss of DA or TH in aging and at the onset of PD symptoms. Growth factors increase locomotor activity in both PD and aging models and increase DA bioavailability and ser31 TH phosphorylation in SN, suggesting that ser31 TH phosphorylation status in the SN, not striatum, regulates DA bioavailability necessary for locomotor activity. METHODOLOGY AND PRINCIPAL FINDINGS:We longitudinally characterized locomotor activity in young and older Brown-Norway Fischer 344 F(1) hybrid rats (18 months apart in age) at two time periods, eight months apart. The aged group served as an intact and pharmacologically-naïve source of deficient locomotor activity. Following locomotor testing, we analyzed DA tissue content, TH protein, and TH phosphorylation in striatum, SN, nucleus accumbens, and VTA. Levels of TH protein combined with ser31 phosphorylation alone reflected inherent differences in DA levels among the four regions. Measures strictly pertaining to locomotor activity initiation significantly correlated to DA content only in the SN. Nigral TH protein and ser31 phosphorylation together significantly correlated to test subject's maximum movement number, horizontal activity, and duration. CONCLUSIONS/SIGNIFICANCE:Together, these results show ser31 TH phosphorylation regulates DA bioavailability in intact neuropil, its status in the SN may regulate locomotor activity generation, and it may represent an accurate target for treating locomotor deficiency. They also show that neurotransmitter regulation in cell body regions can mediate behavioral outcomes and that ser31 TH phosphorylation plays a role in behaviors dependent upon catecholamines, such as dopamine

    Twitter: More than Tweets for Undergraduate Student Researchers

    During the COVID-19 pandemic, biology educators were forced to think of ways to communicate with their students, engaging them in science and with the scientific community. For educators using course-based undergraduate research experiences (CUREs), the challenge to have students perform real science, analyze their work, and present their results to a larger scientific audience was difficult as the world moved online. Many instructors were able to adapt CUREs utilizing online data analysis and virtual meeting software for class discussions and synchronous learning. However, interaction with the larger scientific community, an integral component of making science relevant for students and allowing them to network with other young scientists and experts in their fields, was still missing. Even before COVID-19, a subset of students would travel to regional or national meetings to present their work, but most did not have these opportunities. With over 300 million active users, Twitter provided a unique platform for students to present their work to a large and varied audience. The Cell Biology Education Consortium hosted an innovative scientific poster session entirely on Twitter to engage undergraduate researchers with one another and with the much broader community. The format for posting on this popular social media platform challenged students to simplify their science and make their points using only a few words and slides. Nineteen institutions and over one hundred students participated in this event. Even though these practices emerged as a necessity during the COVID-19 pandemic, the Twitter presentation strategy shared in this paper can be used widely

    From Genetics to Biotechnology: Synthetic Biology as a Flexible Course-embedded Research Experience

    The need for changing how science is taught and the expansion of undergraduate research experiences is essential to foster critical thinking in the Natural Sciences. Most faculty research programs only involve a small number of upper-level undergraduate students each semester. The course-based undergraduate research experience (CURE) model enables more students to take ownership over an independent project and experience authentic research. Further, by creating projects that fit into a curriculum\u27s learning goals and student-oriented outcomes, departments help strengthen critical thinking skills in the classroom. Here, we report on the incorporation of a synthetic biology CURE into a mid-level cellular biology course and two advanced level genetics/molecular biology courses. Synthetic biology involves systematic engineering of novel organisms, such as bacteria and plants, to work as functional devices to solve problems in medicine, agriculture, and manufacturing. The value of synthetic biology and its ultimate utility as a teaching tool relies on reusable, standard genetic parts that can be interchanged using common genetic engineering principles. This Synthetic biology CURE effectively achieves five essential goals: (1) a sense of project ownership; (2) self-efficacy: mastery of a manageable number of techniques; (3) increased tolerance for obstacles through challenging research; (4) increased communication skills; and (5) a sense of belonging in a larger scientific community. Based upon our student assessment data, we demonstrate that this course-based synthetic biology laboratory engages students directly in an authentic research experience and models important elements of collaboration, discovery, iteration, and critical thinking

    Primordial Black Hole Dark Matter Simulations Using PopSyCLE

    Primordial black holes (PBHs), theorized to have originated in the early universe, are speculated to be a viable form of dark matter. If they exist, they should be detectable through photometric and astrometric signals resulting from gravitational microlensing of stars in the Milky Way. Population Synthesis for Compact-object Lensing Events, or PopSyCLE, is a simulation code that enables users to simulate microlensing surveys, and is the first of its kind to include both photometric and astrometric microlensing effects, which are important for potential PBH detection and characterization. To estimate the number of observable PBH microlensing events we modify PopSyCLE to include a dark matter halo consisting of PBHs. We detail our PBH population model, and demonstrate our PopSyCLE + PBH results through simulations of the OGLE-IV and Roman microlensing surveys. We provide a proof-of-concept analysis for adding PBHs into PopSyCLE, and thus include many simplifying assumptions, such as fDMf_{\text{DM}}, the fraction of dark matter composed of PBHs, and mˉPBH\bar{m}_{\text{PBH}}, mean PBH mass. Assuming mˉPBH=30\bar{m}_{\text{PBH}}=30 M⊙M_{\odot}, we find ∼\sim 3.65fDMf_{\text{DM}} times as many PBH microlensing events than stellar evolved black hole events, a PBH average peak Einstein crossing time of ∼\sim 91.4 days, estimate on order of 102fDM10^2f_{\text{DM}} PBH events within the 8 year OGLE-IV results, and estimate Roman to detect on the order of 103fDM10^3f_{\text{DM}} PBH microlensing events throughout its planned microlensing survey

    Effect of Long-Duration Spaceflight on Postural Control During Self-Generated Perturbations

    This report is the first systematic evaluation of the effects of prolonged weightlessness on the bipedal postural control processes during self-generated perturbations produced by voluntary upper limb movements. Spaceflight impacts humans in a variety of ways, one of which is compromised postflight postural control. We examined the neuromuscular activation characteristics and center of pressure motion (COP) associated with arm movement of eight subjects who experienced long duration spaceflight (3-6 months) aboard the Mir space station. Surface electromyography (EMG), arm acceleration, and COP motion were collected while astronauts performed rapid unilateral shoulder flexions prior to and after spaceflight. Subjects displayed compromised postural control after flight as evidenced by modified peak-to-peak COP anterior-posterior and medio-lateral motion and COP pathlength relative to preflight values. These changes were associated with disrupted neuromuscular activation characteristics, particularly after the completion of arm acceleration (i.e. when subjects were attempting to maintain their upright posture). These findings suggest that although the subjects were able to assemble coordination modes that enabled them to generate rapid arm movements, the subtle control necessary to maintain bipedal equilibrium evident in their preflight performance is compromised after long duration spaceflight
