Ventricular assist devices implantation: surgical assessment and technical strategies
Along with the worldwide increase in continuous left ventricular assist device (LVAD) strategy adoption, more and more patients with demanding anatomical and clinical features are currently referred to heart failure (HF) departments for treatment. Thus surgeons have to deal, technically, with re-entry due to previous cardiac surgery procedures, porcelain aorta, peripheral vascular arterial disease, concomitant valvular or septal disease, biventricular failure. New surgical techniques and surgical tools have been developed to offer acceptable postoperative outcomes to all mechanical circulatory support recipients. Several less invasive and/or thoracotomic approaches for surgery combined with various LVAD inflow and outflow graft alternative anastomotic sites for system placement have been reported and described to solve complex clinical scenarios. Surgical techniques have been upgraded with further technical tips to preserve the native anatomy in case of re-entry for heart transplantation, myocardial recovery or device explant. The current continuous-flow miniaturized and intrapericardial devices provide versatility and technical advantages. However, the surgical planning requires a careful multidisciplinary evaluation which must be driven by a dedicated and well-trained Heart Failure team. Biventricular assist device (BVAD) implantation by adoption of the newer radial pumps might be a challenge. However, the results are encouraging thus remaining a valid option. Effects of asymmetric contacts on single molecule conductances of HS(CH2)nCOOH in nano-electrical junctions
A scanning tunnelling microscope has been used to determine the conductance of single molecular wires with the configuration X-bridge-X, X-bridge-Y and Y-bridge-Y (X = thiol terminus and Y = COOH). We find that for molecular wires with mixed functional groups (X-bridge-Y) the single molecule conductance decreases with respect to the comparable symmetric molecules. These differences are confirmed by theoretical computations based on a combination of density functional theory and the non-equilibrium Green functions formalism. This study demonstrates that the apparent contact resistance, as well as being highly sensitive to the type of the anchoring group is also strongly influenced by contact-asymmetry of the single molecular junction which in this case decreases the transmission. This highlights that contact asymmetry is a significant factor to be considered when evaluating nano-electrical junctions incorporating single molecules
Ultrasensititve system for the real time detection of H2O2 based on strong coupling in a bio-plasmonic system
We theoretically investigate the dependence of the different parameters of an optical biosensor for the detection of Hydrogen peroxide (H2O2) based on absorption enhancement of Cytochrome c molecules near gold nanoparticles. H2O2 is a major reactive oxygen species which is involved in signaling pathways and oxidative stress in cells. We use the Green's function approach as well as confirm the corresponding simulation results using the surface integral formulation. Further we show that this technique can be applied for detection of other small molecules, like oxygen and carbon monoxide

Biophotonic tool for sensing the dynamics of H2O2 extracellular release in stressed cells
Hydrogen peroxide (H2O2) is known to play a multifaceted role in cell physiology mechanisms involving oxidative stress and intracellular signal transduction. Therefore, the development of analytical tools providing information on the dynamics of H2O2 generation remains of utmost importance to achieve further insight in the complex physiological processes of living cells and their response to environmental stress(1). In the present work we developed a novel optic biosensor that provides continuous real-time quantification of the dynamics of the hydrogen peroxide release from cells under oxidative stress conditions. The biosensor is based on the ultra-sensitive dark field optical detection of cytochrome c (cyt c) that exhibits a narrow absorption peaks in its reduced state (Fe(II)) at lambda = 550 nm. In the presence of H2O2 the ferrous heme group Fe(II) is oxidised into Fe(III) providing the spectroscopic information exploited in this approach. Extremely low limit-of-detection for H2O2 down to the subnanomolar range is achieved by combining scattering substrates (eg. polystyrene beads) able to shelter cyt c and an inverted microscope in dark field configuration. The developed biosensor was able to perform real-time detection of H2O (2) extracellular release from human promyelocytic leukemia cells (HL-60) exposed to lipopolysaccaride (LPS) that elicits strong immune-response. This biosensing tool is currently being implemented to the real-time detection of superoxide anion (O-2(center dot-)) and offers the possibility to extend to further oxidative stress biomarkers such as glutathione. More generally, multianalyte and dynamic informations might bring new insights to understand complex cellular metabolisms involved in oxidative-stress-related diseases and cytotoxic responses
