    Evaluation of insect-mediated seed set among soybean lines segregating for male sterility at the ms6 locus

    Currently, there is no economical way to produce large quantities of F1 hybrid soybean seed in the USA. One of the fundamental requirements for hybrid seed production is the availability of a stable male-sterile, female-fertile system. However, the more challenging barrier is the efficient transfer of pollen from the male parent to the female parent. This could potentially be achieved through pollinator insects. Our observations suggested that seed set on male-sterile, female-fertile plants is a good indicator of insect attraction. The objective of this study was to evaluate seed set among male-sterile, female-fertile lines segregating for male-sterile, female-fertile ms6 allele by using Megachile rotundata as pollinator vector. Thirty-four pairs of near-isogenic lines, the ms6 w1w1 donor parent, and its two isogenic lines W1w1 and w1w1 segregating for male-sterile (ms6) allele were used. The W1 locus controls flower color and hypocotyle pigmentation. Seed set was evaluated on field-grown plants in 2001–2003 near Ames, IA. Although the observed seed set was not commercially acceptable, our results indicated significant differences in seed set among lines. This suggests that preferential attraction of pollinators occurred, and selection among male-sterile, female-fertile lines could be used to obtain female parents suitable to produce larger amounts of hybrid soybean seed. In addition, the effect of flower color on seed set was statistically significant. White-flowered lines (w1w1) produced more seed set compared to purple-flowered lines (W1W1). Lastly, the important effect of year suggested that the effect of environmental conditions on seed set among lines segregating for male sterility was of paramount importance to plant–pollinator interactions.     The first requirement to establish a successful hybrid soybean program is the availability of a stable male-sterile, female-fertile system. Male sterility has been an important tool in soybean breeding programs to improve traits such as yield, seed-protein and seed-oil content, and seed size. However, improvement of seed-set per se on male-sterile plants has not been an important breeding objective. The evaluation of the out-crossing potential of the available male-sterile, female-fertile soybean lines is crucial to determine the future of hybrid soybean. The objective of this study was to evaluate seed-set among 21 soybean lines segregating for male sterility at 10 different loci usingMegachile rotundata as insect pollinator. Seed-set was evaluated in field conditions in 2001, 2002, and 2003 near Ames, Iowa. Our results indicated significant differences in seed-set among male-sterile lines. The effects of maturity group, pubescence color, the locus and/or the allele, and the genetic background of the lines segregating for male sterility were significant. Differences for seed-set among lines with independent mutational events at the same locus, (i.e. different alleles), also were significant. These results imply that interaction effects with the genetic background of the lines for traits related to fertility/sterility and insect-pollinator attraction and reward are important. Flower color had little effect on seed-set in the evaluated lines. The effect of year was very important on the performance of the lines for seed-set. This suggested that environmental conditions that favor plant–pollinator relationships need to be determined in order to increase insect-mediated cross-pollination to develop an efficient hybrid soybean program

    Seed-set evaluation of four male-sterile, female-fertile soybean lines using alfalfa leafcutting bees and honey bees as pollinators

    Male-sterile, female-fertile plants were used to produce hybrid soybean seed. Manual cross-pollination using male-sterile plants to produce large quantities of hybrid seed is difficult and time-consuming because of the low success rate in cross-pollination. Insect pollinators may be suitable vectors to transfer pollen, but the most suitable vector for pollen transfer from the male parent to the female parent has not been identified for soybean. The objective of the present study was to evaluate seed-set on four male-sterile, female-fertile soybean lines by using alfalfa leafcutting bees (Megachile rotundata (F.)) and honey bees (Apis mellifera (L.)) as pollinators. Seed-set was evaluated in summers 2003 and 2005 near Ames, Iowa, USA and in summers 2003, 2004, and 2005 near Wooster, Ohio, USA. Neither the effect of pollinator species nor the interaction effect of pollinator species×location was significant for any year. Honey bees performed similarly to alfalfa leafcutting bees at both locations. The results indicated significant differences for seed-set among male-sterile lines, suggesting preferential pollination. Male-sterile lines, ms1(Urbana) and ms2 (Ames 2), had higher cross-pollinated seed-set compared to ms6 (Ames 1), and ms6 (Corsoy 79). At the Ames location, ms1ms1 (Urbana) plants had the highest seed-set (50·16 seeds per male-sterile plant in 2005). At the Wooster location, ms1ms1 (Urbana) plants also had the highest seed-set (92·04 seeds per male-sterile plant) in 2005. Costs and local conditions need to be addressed to support the choice of either pollinator species as a pollination vector to produce hybrid soybean seed

    Miocene Volcaniclastic Sequence Within the Xiyu Formation from Source to Sink: Implications for Drainage Development and Tectonic Evolution in Eastern Pamir, NW Tibetan Plateau

The formation of the Pamir salient and the Tashkorgan-Yarkand River is highly debated with the ages ranging from pre-Cenozoic to late Miocene. One approach to resolve these issues is to draw support from the sedimentary record in the surrounding basins. A volcaniclastic sequence, which divides into Lower and Upper Members, was identified in the southwestern Tarim Basin. The Lower Member was transported by dilute streamflows, which likely flowed during or soon after eruptions, while the Upper Member was formed by a syneruptive volcanic debris flow. Chronological, petrologic, and geochemical data consistently indicate that the sequence was derived from the Central Pamir at ~11 Ma. The ~11 Ma emplacement of the volcaniclastic sequence provides unique constraints for the evolution of the Tashkorgan-Yarkand River and the Pamir salient. Provenance data indicate a multistage evolutionary history of the Tashkorgan-Yarkand River. The paleo-Tashkorgan River was initially formed in the piedmont of the Pamir marginal range before ~15 Ma. This river cut back into the Tashkorgan region at ~15 Ma, after which it has eroded the Central Pamir by ~11 Ma. The N-S trending upper reaches of the Tashkorgan River and the Yarkand River were established after ~11 Ma. The emplacement of the volcanic debris flow, together with regional deformation evidence, indicates limited strike-slip motion between Pamir and the Tarim at least since ~11 Ma, which refutes hundreds of kilometers offset between the Pamir and the Tarim after this time and supports an earlier indention of the Pamir salient

    One hundred and five generic types of Pleosporales are described and illustrated. A brief introduction and detailed history with short notes on morphology, molecular phylogeny as well as a general conclusion of each genus are provided. For those genera where the type or a representative specimen is unavailable, a brief note is given. Altogether 174 genera of Pleosporales are treated. Phaeotrichaceae as well as Kriegeriella, Zeuctomorpha and Muroia are excluded from Pleosporales. Based on the multigene phylogenetic analysis, the suborder Massarineae is emended to accommodate five families, viz. Lentitheciaceae, Massarinaceae, Montagnulaceae, Morosphaeriaceae and Trematosphaeriaceae

    The evaluation of experimental data in fast range for n + 56

    Iron is one of the five materials selected for evaluation within the pilot international evaluation project CIELO. Analysis of experimental data for n+56Fe reaction is the basis for constraining theoretical calculations and eventual creation of the evaluated file. The detail analysis was performed for inelastic cross sections of neutron induced reactions with 56Fe in the fast range up to 20 MeV where there are significant differences among the main evaluated libraries, mainly caused by the different inelastic scattering cross section measurements. Gamma-ray production cross sections provide a way to gain experimental information about the inelastic cross section. Large discrepancies between experimental data for the 847-keV gamma ray produced in the 56Fe(n,n1'γ) reaction were analyzed. In addition, experimental data for elastic scattering cross section between 9.41∼11 MeV were used to deduce the inelastic cross section from the unitarity constrain

    Genome-Wide Profiling Identified a Set of miRNAs that Are Differentially Expressed in Glioblastoma Stem Cells and Normal Neural Stem Cells

    A major challenge in cancer research field is to define molecular features that distinguish cancer stem cells from normal stem cells. In this study, we compared microRNA (miRNA) expression profiles in human glioblastoma stem cells and normal neural stem cells using combined microarray and deep sequencing analyses. These studies allowed us to identify a set of 10 miRNAs that are considerably up-regulated or down-regulated in glioblastoma stem cells. Among them, 5 miRNAs were further confirmed to have altered expression in three independent lines of glioblastoma stem cells by real-time RT-PCR analysis. Moreover, two of the miRNAs with increased expression in glioblastoma stem cells also exhibited elevated expression in glioblastoma patient tissues examined, while two miRNAs with decreased expression in glioblastoma stem cells displayed reduced expression in tumor tissues. Furthermore, we identified two oncogenes, NRAS and PIM3, as downstream targets of miR-124, one of the down-regulated miRNAs; and a tumor suppressor, CSMD1, as a downstream target of miR-10a and miR-10b, two of the up-regulated miRNAs. In summary, this study led to the identification of a set of miRNAs that are differentially expressed in glioblastoma stem cells and normal neural stem cells. Characterizing the role of these miRNAs in glioblastoma stem cells may lead to the development of miRNA-based therapies that specifically target tumor stem cells, but spare normal stem cells

    Identification of a 4-microRNA Signature for Clear Cell Renal Cell Carcinoma Metastasis and Prognosis

    Renal cell carcinoma (RCC) metastasis portends a poor prognosis and cannot be reliably predicted. Early determination of the metastatic potential of RCC may help guide proper treatment. We analyzed microRNA (miRNA) expression in clear cell RCC (ccRCC) for the purpose of developing a miRNA expression signature to determine the risk of metastasis and prognosis. We used the microarray technology to profile miRNA expression of 78 benign kidney and ccRCC samples. Using 28 localized and metastatic ccRCC specimens as the training cohort and the univariate logistic regression and risk score methods, we developed a miRNA signature model in which the expression levels of miR-10b, miR-139-5p, miR-130b and miR-199b-5p were used to determine the status of ccRCC metastasis. We validated the signature in an independent 40-sample testing cohort of different stages of primary ccRCCs using the microarray data. Within the testing cohort patients who had at least 5 years follow-up if no metastasis developed, the signature showed a high sensitivity and specificity. The risk status was proven to be associated with the cancer-specific survival. Using the most stably expressed miRNA among benign and tumorous kidney tissue as the internal reference for normalization, we successfully converted his signature to be a quantitative PCR (qPCR)-based assay, which showed the same high sensitivity and specificity. The 4-miRNA is associated with ccRCC metastasis and prognosis. The signature is ready for and will benefit from further large clinical cohort validation and has the potential for clinical application

    The CIELO collaboration: Progress in international evaluations of neutron reactions on Oxygen, Iron, Uranium and Plutonium

    The CIELO collaboration has studied neutron cross sections on nuclides that significantly impact criticality in nuclear technologies – 16O, 56Fe, 235,8U and 239Pu – with the aim of improving the accuracy of the data and resolving previous discrepancies in our understanding. This multi-laboratory pilot project, coordinated via the OECD/NEA Working Party on Evaluation Cooperation (WPEC) Subgroup 40 with support also from the IAEA, has motivated experimental and theoretical work and led to suites of new evaluated libraries that accurately reflect measured data and also perform well in integral simulations of criticality
