    The development of a 16S rRNA gene based PCR for the identification of Streptococcus pneumoniae and comparison with four other species specific PCR assays

    <p>Abstract</p> <p>Background</p> <p><it>Streptococcus pneumoniae </it>is one of the most frequently encountered pathogens in humans but its differentiation from closely related but less pathogenic streptococci remains a challenge.</p> <p>Methods</p> <p>This report describes a newly-developed PCR assay (Spne-PCR), amplifying a 217 bp product of the 16S rRNA gene of <it>S. pneumoniae</it>, and its performance compared to other genotypic and phenotypic tests.</p> <p>Results</p> <p>The new PCR assay designed in this study, proved to be specific at 57°C for <it>S. pneumoniae</it>, not amplifying <it>S. pseudopneumoniae </it>or any other streptococcal strain or any strains from other upper airway pathogenic species. PCR assays (psaA, LytA, ply, spn9802-PCR) were previously described for the specific amplification of <it>S. pneumoniae</it>, but <it>psaA</it>-PCR was the only one found not to cross-react with <it>S. pseudopneumoniae</it>.</p> <p>Conclusion</p> <p>Spne-PCR, developed for this study, and psaA-PCR were the only two assays which did not mis-identify <it>S. pseudopneumoniae </it>as <it>S. pneumoniae</it>. Four other PCR assays and the AccuProbe assay were unable to distinguish between these species.</p

    High-Fat Diet Induces Apoptosis of Hypothalamic Neurons

    Consumption of dietary fats is amongst the most important environmental factors leading to obesity. In rodents, the consumption of fat-rich diets blunts leptin and insulin anorexigenic signaling in the hypothalamus by a mechanism dependent on the in situ activation of inflammation. Since inflammatory signal transduction can lead to the activation of apoptotic signaling pathways, we evaluated the effect of high-fat feeding on the induction of apoptosis of hypothalamic cells. Here, we show that consumption of dietary fats induce apoptosis of neurons and a reduction of synaptic inputs in the arcuate nucleus and lateral hypothalamus. This effect is dependent upon diet composition, and not on caloric intake, since pair-feeding is not sufficient to reduce the expression of apoptotic markers. The presence of an intact TLR4 receptor, protects cells from further apoptotic signals. In diet-induced inflammation of the hypothalamus, TLR4 exerts a dual function, on one side activating pro-inflammatory pathways that play a central role in the development of resistance to leptin and insulin, and on the other side restraining further damage by controlling the apoptotic activity

    Adaptation to college and Ball State first year students of color : analysis of possible effects on adjustment resulting from the Multicultural Advisor Pilot Program

    The purpose of this study was to analyze possible effects of the 1997-1998 Multicultural Advisor Pilot Program which was designed to positively effect the adjustment of students of color to the Ball State University environment. The study utilizes the Student Adaptation to College Questionnaire which examines a student's full-scale adjustment, academic adjustment, social adjustment, personal-emotional adjustment, and attachment/commitment to their education and institution. Results of the study suggest various differences in adjustment to the Ball State University environment do exist between the first-year students of color affected by the program and the first-year students of color who were not involved with the Multicultural Advisor Pilot program.Thesis (M.A.)Department of Secondary, Higher, and Foundations of Educatio

    Prediction of Duration Limited Exposure for Participants Wearing Chemical Protective Clothing in the Cold

    The suitability of the IREQ (insulation required) index for predicting the thermal responses of 6 participants wearing chemical protective clothing was tested during exercise at -20 and -25 °C. IREQ was used to calculate duration limited exposure (DLE). Measured DLE correlated (r= .899, p < .001) with the predicted DLE. In exposures exceeding 40 min, however, the predicted DLE tended to be 10-20 min too short compared to the measured one. During short exposures the prediction was 5-20 min too long. The results show that IREQ overestimated the cold strain in participants wearing chemical protective clothing during cold exposures longer than 40 min. Nevertheless, predicted DLE never exceeded measured times and thus the prediction was always safe from the occupational point of view

    How useful is fluctuating asymmetry in conservation biology: asymmetry in rare and abundant Coenonympha butterflies

    It has been suggested that minor, fluctuating differences in size of bilateral traits could validly indicate individual differences in development stability. One plausible reason for instability to occur could be lowered population size, which has been suggested to increase fluctuating asymmetry due to inbreeding, for example. We measured seven wing asymmetries of three Coenonympha butterfly species in central Sweden. One species is abundant (C. pamphilus), one rather common (C. arcania), and one rare (C. hero). We expected that if fluctuating asymmetry is a reliable indicator of population quality and thus a useful tool for conservation purposes, the most abundant species should show lowest asymmetry and the most endangered, the highest. Contrary to our expectations, the highest wing asymmetry was found in the relatively common species C. arcania and the most abundant and rare species did not show significant differences in levels of wing asymmetry. Our results obtained from three Coenonympha species hence suggest that the use of fluctuating asymmetry as an indicator of population conservation status may be misleading. Possible increase in asymmetry of small and/or isolated populations of butterflies may be masked by local differences in environmental conditions that could have high impact on bilateral development as well

    On the forgetting of corporate irresponsibility

    Why are some serious cases of corporate irresponsibility collectively forgotten? Drawing on social memory studies, we examine how this collective forgetting process can occur. We propose that a major instance of corporate irresponsibility leads to the emergence of a stakeholder mnemonic community that shares a common recollection of the past incident. This community generates and then draws on mnemonic traces to sustain a collective memory of the past event over time. In addition to the natural entropic tendency to forget, collective memory is also undermined by instrumental "forgetting work," which we conceptualize in this article. Forgetting work involves manipulating short-term conditions of the event, silencing vocal "rememberers," and undermining collective mnemonic traces that sustain a version of the past. This process can result in a reconfigured collective memory and collective forgetting of corporate irresponsibility events. Collective forgetting can have positive and negative consequences for the firm, stakeholders, and society
