    Exercise-induced fatigue in young people: advances and future perspectives

The final version is available from Springer Verlag via the DOI in this record.PURPOSE: In recent decades, the interest for exercise-induced fatigue in youth has substantially increased, and the effects of growth on the peripheral (muscular) and central (neural) mechanisms underpinning differences in neuromuscular fatigue between healthy children and adults have been described more extensively. The purpose of this review is to retrieve, report, and analyse the findings of studies comparing neuromuscular fatigue between children and adults. Objective measures of the evaluation of the physiological mechanisms are discussed. METHOD: Major databases (PubMed, Ovid, Scopus and Web of Science) were systematically searched and limited to English language from inception to September 2017. RESULT: Collectively, the analyzed studies indicate that children experience less muscular and potentially more neural fatigue than adults. However, there are still many unknown aspects of fatigue regarding neural (supraspinal and spinal) and peripheral mechanisms that should be more thoroughly examined in children. CONCLUSION: Suitable methods, such as transcranial magnetic stimulation, transcranial electrical stimulation, functional magnetic resonance imaging, near-infrared spectroscopy, tendon vibration, H-reflex, and ultrasound are recommended in the research field of fatigue in youth. By designing studies that test the fatigue effects in movements that replicate daily activities, new knowledge will be acquired. The linkage and interaction between physiological, cognitive, and psychological aspects of human performance remain to be resolved in young people. This can only be successful if research is based on a foundation of basic research focused on the mechanisms of fatigue while measuring all three above aspects

    Effects of sports drinks on the maintenance of physical performance during 3 tennis matches: A randomized controlled study

    Background: Tennis tournaments often involve playing several consecutive matches interspersed with short periods of recovery. Objective: The objective of this study was firstly to assess the impact of several successive tennis matches on the physical performance of competitive players and secondly to evaluate the potential of sports drinks to minimize the fatigue induced by repeated matches. Methods: This was a crossover, randomized controlled study. Eight male regionally-ranked tennis players participated in this study. Players underwent a series of physical tests to assess their strength, speed, power and endurance following the completion of three tennis matches each of two hours duration played over three consecutive half-days (1.5 day period for each condition). In the first condition the players consumed a sports drink before, during and after each match; in the second, they drank an identical volume of placebo water. The results obtained were compared with the third 'rest' condition in which the subjects did not play any tennis. Main outcomes measured were maximal isometric strength and fatigability of knee and elbow extensors, 20-m sprint speed, jumping height, specific repeated sprint ability test and hand grip strength. Results: The physical test results for the lower limbs showed no significant differences between the three conditions. Conversely, on the upper limbs the EMG data showed greater fatigue of the triceps brachii in the placebo condition compared to the rest condition, while the ingestion of sports drinks attenuated this fatigue. Conclusions: This study has demonstrated for the first time that, when tennis players are adequately hydrated and ingest balanced meals between matches, then no large drop in physical performance is observed even during consecutive competitive matches

    Utilizing dietary probiotics can boost amberjack (Seriola dumerili) lysozyme activity, antioxidant capacity, and gut microbiota

    In aquaculture, natural and eco-friendly resources are prioritized over synthetic inputs (e.g., antibiotics), and optimizing sustainable production methods is necessary. Probiotics have been well known for various potential functions in animal performance and health when supplemented as a single or mix of strain (s) in the diet. Consuming the right probiotic blend (synergistic types) is more advantageous than using each kind individually due to combining each species’ functions. The present trial aimed to examine the potential impacts of two probiotic mixtures on the performance and well-being of amberjack juveniles (5.63 ± 0.04 g; Number = 300) for a 60-day feed trial. Three experimental diets (43.5 ± 0.50% total protein and 12.55 ± 0.17% total lipid) were formulated with no probiotics supplement for the control (CD1) or with probiotics mixture supplement. Probiotic mixtures were supplemented at 2 g/kg diet for groups D2 [Bacillus amyloliquefaciens TOA5001 (BA) + Streptococcus faecalis T-110 (SF)] and D3 [Bacillus amyloliquefaciens, Streptococcus faecalis T-110, Lactobacillus plantarum TO-A, Bacillus mesentericus TO-A] respectively. The analysis revealed no difference in all means of growth parameters, feed utilization, survival rate, biometric indices, whole-body composition, and blood parameters between control and probiotic-supplemented diet groups. Liver lysozyme activity, superoxide dismutase, peroxidase activity, and biological antioxidant (BAP) were significantly improved in fish fed D2 compared to CD1 (P < 0.05). The higher (P < 0.05) content of intestinal lactic acid bacteria was noted in fish group fed diet D2, with an improved intestinal histological structure such as increased villi length, cryptal depth, and goblet cells compared to those provided with CD1 and D3 diets. In conclusion, incorporating a mix of probiotic bacteria [BA+SF] at a 2 g/kg diet has remarkable effects on intestinal health, immune responses, and oxidative status of amberjack, S. dumerili, suggesting a potential probiotic candidate for this species

    Exocyclic Carbons Adjacent to the N6 of Adenine are Targets for Oxidation by the Escherichia coli Adaptive Response Protein AlkB

    The DNA and RNA repair protein AlkB removes alkyl groups from nucleic acids by a unique iron- and α-ketoglutarate-dependent oxidation strategy. When alkylated adenines are used as AlkB targets, earlier work suggests that the initial target of oxidation can be the alkyl carbon adjacent to N1. Such may be the case with ethano-adenine (EA), a DNA adduct formed by an important anticancer drug, BCNU, whereby an initial oxidation would occur at the carbon adjacent to N1. In a previous study, several intermediates were observed suggesting a pathway involving adduct restructuring to a form that would not hinder replication, which would match biological data showing that AlkB almost completely reverses EA toxicity in vivo. The present study uses more sensitive spectroscopic methodology to reveal the complete conversion of EA to adenine; the nature of observed additional putative intermediates indicates that AlkB conducts a second oxidation event in order to release the two-carbon unit completely. The second oxidation event occurs at the exocyclic carbon adjacent to the N[superscript 6] atom of adenine. The observation of oxidation of a carbon at N[superscript 6] in EA prompted us to evaluate N[superscript 6]-methyladenine (m6A), an important epigenetic signal for DNA replication and many other cellular processes, as an AlkB substrate in DNA. Here we show that m6A is indeed a substrate for AlkB and that it is converted to adenine via its 6-hydroxymethyl derivative.     Match running performance and physical capacity profiles of U8 and U10 soccer players

    Aim This study aimed to characterize match running performance of very young soccer players and evaluate the relationship between these data and physical capacities and technical skills. Methods Distances covered at different speed thresholds were measured during 31 official matches using GPS technology in U10 (n = 12; age 10.1 ± 0.1 years) and U8 (n = 15; age 7.9 ± 0.1 years) national soccer players. Counter movement jump performance (CMJ), 20 m shuttle running (20 m-SR), linear sprint performance (10, 20, 30 m), shuttle (SHDT) and slalom dribble tests (SLDT) were performed to determine the players physical capacities and technical skills. Results Physical capacities and technical skills were higher in U10 versus U8 players [P 0.05, ES: 0.74). The U10 players covered more total (TD) and high-intensity running distance (HIRD) than their younger counterparts did (P 0.05, ES: 0.99). TD and HIRD covered across the three 15 min periods of match play did not decline (P > 0.05, ES: 0.02–0.55). Very large magnitude correlations were observed between the U8 and U10 players performances during the 20 m-SR versus TD (r = 0.79; P < 0.01) and HIRD (r = 0.82; P < 0.01) covered during match play. Conclusions Data demonstrate differences in match running performance and physical capacity between U8 and U10 players, and large magnitude relationships between match play measures and physical test performances. These findings could be useful to sports science staff working within the academies

    Quantitative In Vivo Magnetic Resonance Spectroscopy Using Synthetic Signal Injection

    Accurate conversion of magnetic resonance spectra to quantitative units of concentration generally requires compensation for differences in coil loading conditions, the gains of the various receiver amplifiers, and rescaling that occurs during post-processing manipulations. This can be efficiently achieved by injecting a precalibrated, artificial reference signal, or pseudo-signal into the data. We have previously demonstrated, using in vitro measurements, that robust pseudo-signal injection can be accomplished using a second coil, called the injector coil, properly designed and oriented so that it couples inductively with the receive coil used to acquire the data. In this work, we acquired nonlocalized phosphorous magnetic resonance spectroscopy measurements from resting human tibialis anterior muscles and used pseudo-signal injection to calculate the Pi, PCr, and ATP concentrations. We compared these results to parallel estimates of concentrations obtained using the more established phantom replacement method. Our results demonstrate that pseudo-signal injection using inductive coupling provides a robust calibration factor that is immune to coil loading conditions and suitable for use in human measurements. Having benefits in terms of ease of use and quantitative accuracy, this method is feasible for clinical use. The protocol we describe could be readily translated for use in patients with mitochondrial disease, where sensitive assessment of metabolite content could improve diagnosis and treatment

    High-Intensity Interval Training Interventions in Children and Adolescents: A Systematic Review

    BackgroundWhilst there is increasing interest in the efficacy of high-intensity interval training in children and adolescents as a time-effective method of eliciting health benefits, there remains little consensus within the literature regarding the most effective means for delivering a high-intensity interval training intervention. Given the global health issues surrounding childhood obesity and associated health implications, the identification of effective intervention strategies is imperative.ObjectivesThe aim of this review was to examine high-intensity interval training as a means of influencing key health parameters and to elucidate the most effective high-intensity interval training protocol.MethodsStudies were included if they: (1) studied healthy children and/or adolescents (aged 5–18 years); (2) prescribed an intervention that was deemed high intensity; and (3) reported health-related outcome measures.ResultsA total of 2092 studies were initially retrieved from four databases. Studies that were deemed to meet the criteria were downloaded in their entirety and independently assessed for relevance by two authors using the pre-determined criteria. From this, 13 studies were deemed suitable. This review found that high-intensity interval training in children and adolescents is a time-effective method of improving cardiovascular disease biomarkers, but evidence regarding other health-related measures is more equivocal. Running-based sessions, at an intensity of >90% heart rate maximum/100–130% maximal aerobic velocity, two to three times a week and with a minimum intervention duration of 7 weeks, elicit the greatest improvements in participant health.ConclusionWhile high-intensity interval training improves cardiovascular disease biomarkers, and the evidence supports the effectiveness of running-based sessions, as outlined above, further recommendations as to optimal exercise duration and rest intervals remain ambiguous owing to the paucity of literature and the methodological limitations of studies presently available

    DNA methylation on N6-adenine in mammalian embryonic stem cells

    It has been widely accepted that 5-methylcytosine is the only form of DNA methylation in mammalian genomes. Here we identify N6-methyladenine as another form of DNA modification in mouse embryonic stem cells. Alkbh1 encodes a demethylase for N6-methyladenine. An increase of N6-methyladenine levels in Alkbh1-deficient cells leads to transcriptional silencing. N6-methyladenine deposition is inversely correlated with the evolutionary age of LINE-1 transposons; its deposition is strongly enriched at young (6 million years old) L1 elements. The deposition of N6-methyladenine correlates with epigenetic silencing of such LINE-1 transposons, together with their neighbouring enhancers and genes, thereby resisting the gene activation signals during embryonic stem cell differentiation. As young full-length LINE-1 transposons are strongly enriched on the X chromosome, genes located on the X chromosome are also silenced. Thus, N6-methyladenine developed a new role in epigenetic silencing in mammalian evolution distinct from its role in gene activation in other organisms. Our results demonstrate that N6-methyladenine constitutes a crucial component of the epigenetic regulation repertoire in mammalian genomes

    Can physical education and physical activity outcomes be developed simultaneously using a game-centered approach?

    The primary objective of this study was to evaluate the efficacy of a pilot intervention using a gamecentered approach for improvement of physical activity (PA) and physical education (PE) outcomes simultaneously, and if this had an impact on enjoyment of PE. A group-randomized controlled trial with a 7-week wait-list control group was conducted in one primary school in the Hunter Region, NSW, Australia. Participants (n = 107 students; mean age = 10.7 years, SD 0.87) were randomized by class group into the Professional Learning for Understanding Games Education (PLUNGE) pilot intervention (n = 52 students) or the control (n = 55) conditions. PLUNGE involved 6 x 60 min PE lessons based on game-centered curriculum delivered via an in-class teacher mentoring program. Students were assessed at baseline and 7-week follow-up for fundamental movement skills (FMS) of throw and catch, game play abilities of decision making, support and skill performance; in-class PA; and enjoyment of PA. Linear mixed models revealed significant group-by-time intervention effects (p < 0.05) for throw (effect size: d = 0.9) and catch (d = 0.4) FMS, decision making (d = 0.7) and support (d = 0.9) during game play, and in-class PA (d = 1.6). No significant intervention effects (p > 0.05) were observed for skills outcome during game play (d = -0.2) or student enjoyment (d = 0.1). Game-centered pedagogy delivered via a teacher professional learning program was efficacious in simultaneously improving students' FMS skills, in-class PA and their decision making and support skills in game play
