    Ground and Water Source Heat Pump Performance and Design for Southern Climates

    Ground and water source heat pump systems have very attractive performance characteristics when properly designed and installed. These systems typically consist of a water-to-air or water-to-water heat pump linked to a closed loop vertical or horizontal ground-coupling, an open groundwater loop, or a surface water loop. This paper discusses system performance characteristics, component selection procedures presently being used, improvements currently being considered and future possibilities for improved efficiency and reliability. Optimum designs require proper matching of the heat pump unit to the water circulation system, the building space heating/cooling load and water heating requirements. General trends resulting from system and component choices will be discussed. Water heating methods with these heat pumps will be considered

    In vivo imaging of protease activity by Probody therapeutic activation.

    Probodyℱ therapeutics are recombinant, proteolytically-activated antibody prodrugs, engineered to remain inert until activated locally by tumor-associated proteases. Probody therapeutics exploit the fundamental dysregulation of extracellular protease activity that exists in tumors relative to healthy tissue. Leveraging the ability of a Probody therapeutic to bind its target at the site of disease after proteolytic cleavage, we developed a novel method for profiling protease activity in living animals. Using NIR optical imaging, we demonstrated that a non-labeled anti-EGFR Probody therapeutic can become activated and compete for binding to tumor cells in vivo with a labeled anti-EGFR monoclonal antibody. Furthermore, by inhibiting matriptase activity in vivo with a blocking-matriptase antibody, we show that the ability of the Probody therapeutic to bind EGFR in vivo was dependent on protease activity. These results demonstrate that in vivo imaging of Probody therapeutic activation can be used for screening and characterization of protease activity in living animals, and provide a method that avoids some of the limitations of prior methods. This approach can improve our understanding of the activity of proteases in disease models and help to develop efficient strategies for cancer diagnosis and treatment

    Isolation of Current Components and Partial Reaction Cycles in the Glial Glutamate Transporter EAAT2

    The kinetic properties of the excitatory amino acid transporter EAAT2 were studied using rapid applications of l-glutamate to outside-out patches excised from transfected human embryonic kidney 293 cells. In the presence of the highly permeant anion SCN−, pulses of glutamate rapidly activated transient anion channel currents mediated by the transporter. In the presence of the impermeant anion gluconate, glutamate pulses activated smaller currents predicted to result from stoichiometric flux of cotransported ions. Both anion and stoichiometric currents displayed similar kinetics, suggesting that anion channel gating and stoichiometric charge movements are linked to early transitions in the transport cycle. Transporter-mediated anion currents were recorded with ion and glutamate gradients favoring either unidirectional influx or exchange. Analysis of deactivation and recovery kinetics in these two conditions suggests that, after binding, translocation of substrate is more likely than unbinding under physiological conditions. The kinetic properties of EAAT2, the dominant glutamate transporter in brain astrocytes, distinguish it as an efficient sink for synaptically released glutamate

    Evaluating Common Measures 2.0 in 4-H: Intra- and Interpersonal Skills Predict Engaged Citizenship

    Research indicates that positive youth development (PYD) programming leads to improved developmental outcomes. Yet there continues to be a lack of clarity about how PYD programs are defined and measured. Therefore, Common Measures 2.0 was created for assessing 4-H youth development programs. We examined the universal skills (intrapersonal and interpersonal) and engaged citizenship scales of Common Measures 2.0 through a survey of 175 participants at a 4-H youth leadership conference. Intrapersonal and interpersonal skills were significantly positively associated with engaged citizenship. The universal skills and engaged citizenship scales showed acceptable factor loadings and Cronbach\u27s alpha values. Limitations and future directions are discussed

    Ovalization of Tubes Under Bending and Compression

    An empirical equation has been developed that gives the approximate amount of ovalization for tubes under bending loads. Tests were made on tubes in the d/t range from 6 to 14, the latter d/t ratio being in the normal landing gear range. Within the range of the series of tests conducted, the increase in ovalization due to a compression load in combination with a bending load was very small. The bending load, being the principal factor in producing the ovalization, is a rather complex function of the bending moment, d/t ratio, cantilever length, and distance between opposite bearing faces.     Dynamic Structural Flexibility of Fe-MOF-5 Evidenced by ⁔⁷Fe Mössbauer Spectroscopy

    Temperature-dependent ⁔⁷Fe Mössbauer spectra were collected on Fe[subscript x]Zn[subscript 4−x](1,4-benzenedicarboxylate)₃ (Fe-MOF-5). When measured under an Ar atmosphere, the data at higher temperatures reveal thermal population of the lowest-lying electronic excited state, as expected for low symmetry tetrahedral ferrous ions. In the presence of N₂, however, the temperature dependence becomes exaggerated and the spectra cannot be fitted to a single species. A fluctuating electric field gradient at the Fe nuclei best explains these data and suggests dynamic structural distortions induced by weak interactions with N₂. This direct evidence of dynamic behaviour at MOF open metal sites is relevant for the use of MOF SBUs in catalysis, gas separation, and other applications that invoke similar phenomena

    Clinical efficacy, radiographic and safety findings through 2 years of golimumab treatment in patients with active psoriatic arthritis: results from a long-term extension of the randomised, placebo-controlled GO-REVEAL study

    Objectives: To assess long-term golimumab efficacy/safety in patients with active psoriatic arthritis (PsA).<p></p> Methods Adult PsA patients (≥3 swollen, ≥3 tender joints, active psoriasis) were randomly assigned to subcutaneous injections of placebo, golimumab 50 mg or 100 mg every 4 weeks (q4wks) through week 20. All patients received golimumab 50 or 100 mg beginning week 24. Findings through 2 years are reported. Efficacy evaluations included ≥20% improvement in American College of Rheumatology (ACR20) response, good/moderate response in Disease Activity Scores incorporating 28 joints and C-reactive protein (DAS28-CRP), ≥75% improvement in Psoriasis Area and Severity Index (PASI75) and changes in PsA-modified Sharp/van der Heijde scores (SHS).<p></p> Results: Golimumab treatment through 2 years was effective in maintaining clinical response (response rates: ACR20 63%–70%, DAS28-CRP 77%–86%, PASI75 56%–72%) and inhibiting radiographic progression (mean change in PsA-modified SHS in golimumab-treated patients: −0.36), with no clear difference between doses. No new safety signals were identified through 2 years. With the study's tuberculosis screening and prophylactic measures, no patient developed active tuberculosis through 2 years.<p></p> Conclusions: Golimumab 50 and 100 mg for up to 2 years yielded sustained clinical and radiographic efficacy when administered to patients with active PsA. Increasing the golimumab dose from 50 to 100 mg q4wks added limited benefit. Golimumab safety through up to 2 years was consistent with other antitumour necrosis factor α agents used to treat PsA. Treatment of patients with latent tuberculosis identified at baseline appeared to be effective in inhibiting the development of active tuberculosis.<p></p&gt
