Human CD4+ T lymphocytes consistently respond to the latent Epstein- Barr virus nuclear antigen EBNA1
The Epstein-Barr virus (EBV)-encoded nuclear antigen EBNA1 is critical for the persistence of the viral episome in replicating EBV-transformed human B cells. Therefore, all EBV-induced tumors express this foreign antigen. However, EBNA1 is invisible to CD8+ cytotoxic T lymphocytes because its Gly/Ala repeat domain prevents proteasome-dependent processing for presentation on major histocompatibility complex (MHC) class I. We now describe that CD4+ T cells from healthy adults are primed to EBNA1. In fact, among latent EBV antigens that stimulate CD4+ T cells, EBNA1 is preferentially recognized. We present evidence that the CD4+ response may provide a protective role, including interferon γ secretion and direct cytolysis after encounter of transformed B lymphocyte cell lines (B-LCLs). Dendritic cells (DCs) process EBNA1 from purified protein and from MHC class II-mismatched, EBNA1-expressing cells including B-LCLs. In contrast, B-LCLs and Burkitt\u27s lymphoma lines likely present EBNA1 after endogenous processing, as their capacity to cross-present from exogenous sources is weak or undetectable. By limiting dilution, there is a tight correlation between the capacity of CD4+ T cell lines to recognize autologous B-LCL-expressing EBNA1 and DCs that have captured EBNA1. Therefore, CD4+ T cells can respond to the EBNA1 protein that is crucial for EBV persistence. We suggest that this immune response is initiated in vivo by DCs that present EBV-infected B cells, and that EBNA1-specific CD4+ T cell immunity be enhanced to prevent and treat EBV-associated malignancies
Acoustic breathers in two-dimensional lattices
The existence of breathers (time-periodic and spatially localized lattice
vibrations) is well established for i) systems without acoustic phonon branches
and ii) systems with acoustic phonons, but also with additional symmetries
preventing the occurence of strains (dc terms) in the breather solution. The
case of coexistence of strains and acoustic phonon branches is solved (for
simple models) only for one-dimensional lattices.
We calculate breather solutions for a two-dimensional lattice with one
acoustic phonon branch. We start from the easy-to-handle case of a system with
homogeneous (anharmonic) interaction potentials. We then easily continue the
zero-strain breather solution into the model sector with additional quadratic
and cubic potential terms with the help of a generalized Newton method. The
lattice size is . The breather continues to exist, but is dressed
with a strain field. In contrast to the ac breather components, which decay
exponentially in space, the strain field (which has dipole symmetry) should
decay like . On our rather small lattice we find an exponent
1060 nm Single-Mode VCSEL and Single-Mode Fiber Links for Long-Reach Optical Interconnects
We investigate the use of a 1060 nm single-mode vertical-cavity surface-emitting laser (VCSEL) and a 1060 nm single-mode fiber as a competitive single-mode technology for cost-and power-efficient long-reach optical interconnects. Error-free transmission (bit error rate < 10-12) over 2 km is demonstrated at bitrates up to 40 Gb/s under on-off keying non-return-to-zero (OOK-NRZ) modulation, without equalization, forward-error correction, or other forms of digital signal processing. The VCSEL is extensively characterized with respect to its static and dynamic performances, including the power-voltage-current characteristics, spectral characteristics, beam divergence, modulation response, relative intensity noise, and frequency chirp. The measured dependence of power penalty on fiber length is consistent with an analysis of chirp-induced pulse compression and broadening along the negative chromatic dispersion fiber
Contrasting patterns of genetic diversity at three different genetic markers in a marine mammal metapopulation
Many studies use genetic markers to explore population structure and variability within species. However, only a minority use more than one type of marker and, despite increasing evidence of a link between heterozygosity and individual fitness, few ask whether diversity correlates with population trajectory. To address these issues, we analyzed data from the Steller’s sea lion, Eumetiopias jubatus, where three stocks are distributed over a vast geographical range and where both genetic samples and detailed demographic data have been collected from many diverse breeding colonies. To previously published mitochondrial DNA(mtDNA) and microsatellite data sets,we have added new data for amplified fragment length polymorphism (AFLP) markers, comprising 238 loci scored in 285 sea lions sampled from 23 natal rookeries. Genotypic diversity was low relative to most vertebrates, with only 37 loci (15.5%) being polymorphic. Moreover, contrasting geographical patterns of genetic diversity were found at the three markers, with Nei’s gene diversity tending to be higher for AFLPs and microsatellites in rookeries of the western and Asian stocks, while the highest mtDNA values were found in the eastern stock. Overall, and despite strongly contrasting demographic histories, after applying phylogenetic correction we found little correlation between genetic diversity and either colony size or demography. In contrast, we were able to show a highly significant positive relationship between AFLP diversity and current population size across a range of pinniped species, even though equivalent analyses did not reveal significant trends for either microsatellites or mtDNA
Theory of sound attenuation in glasses: The role of thermal vibrations
Sound attenuation and internal friction coefficients are calculated for a
realistic model of amorphous silicon. It is found that, contrary to previous
views, thermal vibrations can induce sound attenuation at ultrasonic and
hypersonic frequencies that is of the same order or even larger than in
crystals. The reason is the internal-strain induced anomalously large

Numerical study of anharmonic vibrational decay in amorphous and paracrystalline silicon
The anharmonic decay rates of atomic vibrations in amorphous silicon (a-Si)
and paracrystalline silicon (p-Si), containing small crystalline grains
embedded in a disordered matrix, are calculated using realistic structural
models. The models are 1000-atom four-coordinated networks relaxed to a local
minimum of the Stillinger-Weber interatomic potential. The vibrational decay
rates are calculated numerically by perturbation theory, taking into account
cubic anharmonicity as the perturbation. The vibrational lifetimes for a-Si are
found to be on picosecond time scales, in agreement with the previous
perturbative and classical molecular dynamics calculations on a 216-atom model.
The calculated decay rates for p-Si are similar to those of a-Si. No modes in
p-Si reside entirely on the crystalline cluster, decoupled from the amorphous
matrix. The localized modes with the largest (up to 59%) weight on the cluster
decay primarily to two diffusons. The numerical results are discussed in
relation to a recent suggestion by van der Voort et al. [Phys. Rev. B {\bf 62},
8072 (2000)] that long vibrational relaxation inferred experimentally may be
due to possible crystalline nanostructures in some types of a-Si.Comment: 9 two-column pages, 13 figure
Scholar-activists in an expanding European food sovereignty movement
This article analyzes the roles, relations, and positions of scholar-activists in the European food sovereignty movement. In doing so, we document, make visible and question the political dimensions of researchers' participation in the movement. We argue that scholar-activists are part of the movement, but are distinct from the affected constituencies, put in place to ensure adequate representation of key movement actors. This is because scholar-activists lack a collective identity, have no processes to formulate collective demands, and no mechanisms for inter-researcher and researchers-movement communication. We reflect on whether and how scholar-activists could organize, and discuss possible pathways for a more cohesive and stronger researcher engagement in the movement.</p
Guidelines for genetic data analysis
The IWC Scientific Committee recently adopted guidelines for quality control of DNA data. Once data have been collected, the next step is to analyse the data and make inferences that are useful for addressing practical problems in conservation and management of cetaceans. This is a complex exercise, as numerous analyses are possible and users have a wide range of choices of software programs for implementing the analyses. This paper reviews the underlying issues, illustrates application of different types of genetic data analysis to two complex management problems (involving common minke whales and humpback whales), and concludes with a number of recommendations for best practices in the analysis of population genetic data. An extensive Appendix provides a detailed review and critique of most types of analyses that are used with population genetic data for cetaceans.Publisher PDFPeer reviewe

Adeno-associated virus-8-Mediated intravenous transfer of myostatin propeptide leads to systemic functional improvements of slow but not fast muscle
Myostatin is a member of the transformating growth factor-_ (TGF-_) superfamily of proteins and is produced almost exclusively in skeletal muscle tissue, where it is secreted and circulates as a serum protein. Myostatin acts as a negative regulator of muscle mass through the canonical SMAD2/3/4 signaling pathway. Naturally occurring myostatin mutants exhibit a ‘double muscling’ phenotype in which muscle mass is dramatically increased as a result of both hypertrophy and hyperplasia. Myostatin is naturally inhibited by its own propeptide; therefore, we assessed the impact of adeno associated virus-8 (AAV8) myostatin propeptide vectors when systemically introduced in MF-1 mice. We noted a significant systemic increase in muscle mass in both slow and fast muscle phenotypes, with no evidence of hyperplasia; however, the nuclei-to- cytoplasm ratio in all myofiber types was significantly reduced. An increase in muscle mass in slow (soleus) muscle led to an increase in force output; however, an increase in fast (extensor digitorum longus [EDL]) muscle mass did not increase force output. These results suggest that the use of gene therapeutic regimens of myostatin inhibition for age-related or disease-related muscle loss may have muscle-specific effects
Population Genetics of Bowhead Whales (Baleana mysticetus) in the Western Arctic
Bowhead whales (Balaena mysticetus) in the Bering, Chukchi, and Beaufort seas experienced a severe reduction as a result of commercial whaling in the 19th century. Since the cessation of commercial whaling, the population has recovered to a size that is approaching pre-whaling estimates. Inupiat and Yupik communities in northern and western Alaska hunt these Western Arctic (WA) bowheads along their migratory path during spring and fall. This hunting is regulated by the International Whaling Commission. Recent but preliminary analysis of available genetic data (207 whales and 10 microsatellite markers) raised the question of the presence of multiple, genetically distinct populations within the WA bowheads. Here we re-examined this question on the basis of a study of 414 whales and 22 newly developed microsatellite loci. We identified widespread departures from Hardy-Weinberg equilibrium; however, we were unable to detect significant evidence of multiple genetic populations within the WA bowheads that could explain this Hardy-Weinberg disequilibrium, particularly when compared to the strength of evidence for differentiation between WA bowheads and other populations from distant regions such as the Okhotsk Sea and eastern Canada. There was conclusive evidence of genetic differentiation among the three regions. The statistical rejection of panmixia within the WA improves our understanding of bowhead whale biology, and the lack of evidence for multiple populations within the WA enables risk-averse management of aboriginal hunting of Western Arctic bowhead whales.La population de baleines boréales (Balaena mysticetus) des mers de Béring, de Tchoukotka et de Beaufort a enregistré un grave déclin en raison de la pêche commerciale à la baleine au XIXe siècle.
