    Caracterización del Banco de La Concepción

    Se integra información hidrográfica, geomorfológica, sedimentológica, biológica, sobre hábitats marinos y pesquera, para establecer las bases ecológicas necesarias para la protección y conservación del Banco de La ConcepciónEl proyecto INDEMARES ha permitido utilizar amplios medios económicos y personales para estudiar en detalle y desde múltiples perspectivas la zona del Banco de La Concepción. Se han aplicado metodologías para el estudio de la hidrografía, caracterizando la región, describiendo sus principales masas de agua y la hidrodinámica de las corrientes. También se ha abordado la geología de la zona, incluyendo levantamientos batimétricos, perfiles sísmicos, muestreos de sedimento y petrológicos, obteniendo modelos digitales del terreno, mapas de tipos de fondo, geomorfológicos. Se han caracterizado las comunidades bentopelágicas, demersales, epibentónicas y endobentónicas, prestando especial atención a aquellas que conforman o estructuran los hábitats sensibles cuyo inventariado y cartografía era objeto principal del proyecto. Los trabajos de identificación de hábitats se han realizado con muestreadores directos, pero también con muestreadores visuales, que han permitido hacer un mayor esfuerzo de muestreo sin aumentar el impacto sobre los fondos de la zona. Por otro lado se ha estudiado la huella pesquera de la zona por medio del análisis de los datos VMS y los cuadernos de pesca proporcionados por la SGP, además de una ardua labor a pie de puerto de entrevistas y encuestas. Toda la información ha sido gestionada y analizada por diferentes grupos de investigación de diferentes centros (Universidad de La Laguna, Instituto Universitario de Ciencias Políticas y Sociales, Universidad de Las Palmas de Gran Canaria, Instituto de Ciencias del Mar de Barcelona, Universidad de Barcelona, Centro de Investigaciones Medioambientales del Atlántico, Centro Oceanográfico de Santander y Centro Oceanográfico de Málaga del Instituto Español de Oceanografía, …), aunque el grueso de esta labor, además de la coordinación, se ha llevado a cabo en el Centro Oceanográfico de Canarias del Instituto Español de Oceanografía. Los resultados permiten tener un conocimiento profundo y multidisciplinar de la zona de estudio comparable a pocos proyectos de investigación marina en la macaronesia. El estudio oceanográfico, el geológico, y el de las comunidades biológicas, ha permitido la caracterización de los hábitats de la zona, y su cartografiado mediante el intenso muestreo y la aplicación de análisis de idoneidad de hábitats.INDEMARES Project made possible to bring together economic and staff resources to study in detail, and from multiple perspectives, the area of Banco de La Concepción. Benthopelagic, demersal, epibenthic and endobenthic communities have been characterized, paying special attention to sensitive habitats which inventory and mapping was the project’s main aim. Habitat identification has been made with direct and visual samplers, the latter making a major sampling effort possible without an increase of bottom impact. Moreover, fisheries footprint has been identified by Vessel Monitoring System data, together with logbooks from Secretaría General de Pesca (Fisheries Ministry), as well as a hard interviewing and surveying task at landing points. Information has been managed and analysed by different research groups from different Centres (Universidad de La Laguna, Instituto Universitario de Ciencias Políticas y Sociales, Universidad de Las Palmas de Gran Canaria, Instituto de Ciencias del Mar de Barcelona, Universidad de Barcelona, Centro de Investigaciones Medioambientales del Atlántico, Centro Oceanográfico de Santander y Centro Oceanográfico de Málaga del Instituto Español de Oceanografía), although the main body of the work, plus coordination, has been made by the Canary Islands Oceanographic Centre, from the Spanish Institute of Oceanography. Results let us have a multidisciplinary profound knowledge of the study zone, comparable to few other marine research projects off Macaronesia.     Deepening our pelagic vision of the small pelagic fish in the Canary Islands (NW Africa, Spain)

    The small pelagic fish (SPF, i.e. Scomber colias, Trachurus spp, Sardina pilchardus and Sardinella spp) in the Canary Islands archipelago (Spain) are mainly targeted by artisanal purse-seiners, whose monitoring is included in the EU Data Collection Framework. The waters in the archipelago (located in the coastal transition zone of the Canary Current Eastern Boundary Upwelling System) are monitored since the late nineties by a hydrographic section (RAPROCAN) designed to study the temporal variability of the eastern subtropical gyre. Official fish landings, reported by the Government since 2007, are annually analyzed to assess the stock status of the SPF in the Fishery Committee for the Eastern Central Atlantic (CECAF) context. In this study we analyze the relationship between the SPF abundance assumed from official landings and the oceanographic parameters. Using the sea temperature in the 200-800 m layer, we obtained a statistically significant correlation with the SPF landings (ρ-Pearson=0.454, p<0.03) when we considered one-year time-lag. This layer is less affected by atmosphere variations and probably influence on the success of the SPF spawning and recruitment processes. In fact, recruitment to the fishery (i.e. individuals attain legal catchable sizes) approximately occurs for one-year-old individuals, and explaining the one-year time-lag. Keeping the monitoring systems is crucial to understand, foreseen and anticipate potential variations in the fishery resources and to aim the sustainable exploitation of the SPF populations, even more challenging in the current Climate Change scenario.FEM

    Dissecting the transcriptional response to elicitors in Vitis vinifera cells

    The high effectiveness of cyclic oligosaccharides like cyclodextrins in the production of trans-resveratrol in Vitis vinifera cell cultures is enhanced in the presence of methyl jasmonate. In order to dissect the basis of the interactions among the elicitation responses triggered by these two compounds, a transcriptional analysis of grapevine cell cultures treated with cyclodextrins and methyl jasmonate separately or in combination was carried out. The results showed that the activation of genes encoding enzymes from phenylpropanoid and stilbene biosynthesis induced by cyclodextrins alone was partially enhanced in the presence of methyl jasmonate, which correlated with their effects on trans-resveratrol production. In addition, protein translation and cell cycle regulation were more highly repressed in cells treated with cyclodextrins than in those treated with methyl jasmonate, and this response was enhanced in the combined treatment. Ethylene signalling was activated by all treatments, while jasmonate signalling and salicylic acid conjugation were activated only in the presence of methyl jasmonate and cyclodextrins, respectively. Moreover, the combined treatment resulted in a crosstalk between the signalling cascades activated by cyclodextrins and methyl jasmonate, which, in turn, provoked the activation of additional regulatory pathways involving the up-regulation of MYB15, NAC and WRKY transcription factors, protein kinases and calcium signal transducers. All these results suggest that both elicitors cause an activation of the secondary metabolism in detriment of basic cell processes like the primary metabolism or cell division. Crosstalk between cyclodextrins and methyl jasmonate-induced signalling provokes an intensification of these responses resulting in a greater trans-resveratrol production

    Monitoring the Oceanic Waters of the Canary Islands: the deep hydrographic section of the Canaries

    Due to the high density and heat capacity of water, the ocean modulates climate in scales much larger than the atmosphere, both spatially and temporally. In order to understand the mechanisms governing this internal variability of the ocean, and therefore climate, it is necessary to have long systematic observations. The Atlantic Meridional Overturning Circulation (AMOC), which is composed by the south- north circulation, transports 18Sv (1Sv=10E6m3/s) of water that carries more that 1.5PW (1PW=10E15W) of heat to the North Atlantic, and therefore plays a determining role in regulating the climate in Europe. An important component of the AMOC is the subtropical gyre, the largest oceanic structure of the North Atlantic. The Canary Islands are immersed in the eastern margin of the subtropical gyre, in the coastal transition zone of the Canary Current Upwelling System and therefore they are an ideal place for the study of the subtropical gyre variability. With this background, in 2006 the Spanish Institute of Oceanography began (IEO) the program deep hydrographic section around the Canary Islands (Raprocan), in order to establish the scales of variability in the range decadal/subdecadal in the subtropical gyre, specifically in its eastern margin. Based on previous results the observational strategy of Raprocan consists in hydrographic cruises in two seasons, with 50 hydrographic stations around the Canary archipelago. In each one of the stations velocity, temperature, salinity, pressure, oxygen, turbidity and fluorescence is continuously measured ( CTD ). In each station 24 samples are taken for calibration of the above variables as well as for determination of alkalinity, carbon content and chlorophyll. This program is carried out in collaboration with the University of Las Palmas de Gran Canaria (ULPGC) and with the Oceanic Platform of the Canary Islands (PLOCAN) that provides glider data since 2013. The warming of the upper 600 m continues at a rate of 0.14 ° C/ decade in the oceanic waters and 0.32 ° C / decade in the waters between Lanzarote and Africa under influence of upwelling off the African coast. At intermitted levels the warming continues at rate of 0.04 ° C/ decade in the oceanic waters and 0.08 ° C / in the waters between Lanzarote and Africa. At deeper levels, since 1997 there is not statistical significant trend. Regarding the oceanic circulation, the Canary Current presents a seasonal cycle, with the minimum transport occurring during fall, when only carries 3 Sv. During this season the transport is concentrated between the islands of Tenerife and Lanzarote and in the first 400 meters

    Enhanced extracellular production of trans-resveratrol in Vitis vinifera suspension cultured cells by using cyclodextrins and methyljasmonate

    In this work, the effect of different inducing factors on trans-resveratrol extracellular production in Monastrell grapevine suspension cultured cells is evaluated. A detailed analysis provides the optimal concentrations of cyclodextrins, methyljasmonate and UV irradiation dosage, optimal cell density, elicitation time and sucrose content in the culture media. The results indicate that trans-resveratrol production decreases as the initial cell density increases for a constant elicitor concentration in Monastrell suspension cultured cells treated with cyclodextrins individually or in combination with methyljasmonate, the decrease observed in cell cultures elicited with cyclodextrins alone being far more drastic than those observed in the combined treatment. trans-Resveratrol extracellular production observed by the joint use of cyclodextrins and methyljasmonate (1447.8 ± 60.4 μmol trans-resveratrol g-1 dry weight) is lower when these chemical compounds are combined with UV light short exposure (669.9 ± 45.2 μmol trans-resveratrol g-1 dry weight). Likewise, trans-resveratrol production is dependent on levels of sucrose in the elicitation medium being the maximal levels observed with 20 g l-1 sucrose and the joint action of cyclodextrins and 100 µM methyljasmonate. The sucrose concentration did not seem limiting the process although significantly affect the specific productivity since the lowest sucrose concentration is (10 g l-1), the highest productivity is reached (100.7 ± 5.8 μmol trans-resveratrol g-1 dry weight g-1 sucrose) using cyclodextrins and 25 µM methyljasmonate.Fil: Belchi-Navarro, Sarai.
