    Boundaries of Semantic Distraction: Dominance and Lexicality Act at Retrieval

    Three experiments investigated memory for semantic information with the goal of determining boundary conditions for the manifestation of semantic auditory distraction. Irrelevant speech disrupted the free recall of semantic category-exemplars to an equal degree regardless of whether the speech coincided with presentation or test phases of the task (Experiment 1) and occurred regardless of whether it comprised random words or coherent sentences (Experiment 2). The effects of background speech were greater when the irrelevant speech was semantically related to the to-be-remembered material, but only when the irrelevant words were high in output dominance (Experiment 3). The implications of these findings in relation to the processing of task material and the processing of background speech is discussed

    Working memory capacity modulates habituation rate: Evidence from a cross-modal auditory distraction paradigm

    Habituation of the orienting response is a pivotal part of selective attention, and previous research has related working memory capacity (WMC) to attention control. Against this background, the purpose of this study was to investigate whether individual differences in WMC contribute to habituation rate. The participants categorized visual targets across six blocks of trials. Each target was preceded either by a standard sound or, on rare trials, by a deviant. The magnitude of the deviation effect (i.e., prolonged response time when the deviant was presented) was relatively large in the beginning but attenuated toward the end. There was no relationship between WMC and the deviation effect at the beginning, but there was at the end, and greater WMC was associated with greater habituation. These results indicate that high memory ability increases habituation rate, and they support theories proposing a role for cognitive control in habituation and in some forms of auditory distraction

    Targeted resequencing of candidate genes using selector probes

    Targeted genome enrichment is a powerful tool for making use of the massive throughput of novel DNA-sequencing instruments. We herein present a simple and scalable protocol for multiplex amplification of target regions based on the Selector technique. The updated version exhibits improved coverage and compatibility with next-generation-sequencing (NGS) library-construction procedures for shotgun sequencing with NGS platforms. To demonstrate the performance of the technique, all 501 exons from 28 genes frequently involved in cancer were enriched for and sequenced in specimens derived from cell lines and tumor biopsies. DNA from both fresh frozen and formalin-fixed paraffin-embedded biopsies were analyzed and 94% specificity and 98% coverage of the targeted region was achieved. Reproducibility between replicates was high (R2 = 0, 98) and readily enabled detection of copy-number variations. The procedure can be carried out in <24 h and does not require any dedicated instrumentation

    Functional loss of IκBε leads to NF-κB deregulation in aggressive chronic lymphocytic leukemia

    NF-κB is constitutively activated in chronic lymphocytic leukemia (CLL); however, the implicated molecular mechanisms remain largely unknown. Thus, we performed targeted deep sequencing of 18 core complex genes within the NF-κB pathway in a discovery and validation CLL cohort totaling 315 cases. The most frequently mutated gene was NFKBIE (21/315 cases; 7%), which encodes IκBε, a negative regulator of NF-κB in normal B cells. Strikingly, 13 of these cases carried an identical 4-bp frameshift deletion, resulting in a truncated protein. Screening of an additional 377 CLL cases revealed that NFKBIE aberrations predominated in poor-prognostic patients and were associated with inferior outcome. Minor subclones and/or clonal evolution were also observed, thus potentially linking this recurrent event to disease progression. Compared with wild-type patients, NFKBIE-deleted cases showed reduced IκBε protein levels and decreased p65 inhibition, along with increased phosphorylation and nuclear translocation of p65. Considering the central role of B cell receptor (BcR) signaling in CLL pathobiology, it is notable that IκBε loss was enriched in aggressive cases with distinctive stereotyped BcR, likely contributing to their poor prognosis, and leading to an altered response to BcR inhibitors. Because NFKBIE deletions were observed in several other B cell lymphomas, our findings suggest a novel common mechanism of NF-κB deregulation during lymphomagenesis

    Distraction of mental arithmetic by background speech: Further evidence for the habitual-response priming view of auditory distraction

    When solving mental arithmetic problems, one can easily be distracted by someone speaking in the background and this distraction is greater if the speech comprises numbers. We explored the basis of this disruption by asking participants to solve mental addition problems (e.g., "45 + 17 = ?") in three different conditions: background speech comprising numbers in ascending order (e.g., "61, 62, 63, 64, 65"), background speech comprising numbers in descending order (e.g., "65, 64, 63, 62, 61"), and quiet. Performance was best in quiet, worse in the descending numbers condition, and poorest in the ascending numbers condition. In view of these findings, we suggest that disruption arises as a by-product of preventing the primed, but inaccurate, candidate responses from assuming the control of action. Alternative explanations are also discussed

    Children's recall of words spoken in their first and second language : Effects of Signal-to-Noise Ratio and Reverberation Time

    Speech perception runs smoothly and automatically when there is silence in the background, but when the speech signal is degraded by background noise or by reverberation, effortful cognitive processing is needed to compensate for the signal distortion. Previous research has typically investigated the effects of signal-to-noise ratio (SNR) and reverberation time in isolation, whilst few have looked at their interaction. In this study, we probed how reverberation time and SNR influence recall of words presented in participants' first- (L1) and second-language (L2). A total of 72 children (10 years old) participated in this study. The to-be-recalled wordlists were played back with two different reverberation times (0.3 and 1.2 s) crossed with two different SNRs (+3 dBA and +12 dBA). Children recalled fewer words when the spoken words were presented in L2 in comparison with recall of spoken words presented in L1. Words that were presented with a high SNR (+12 dBA) improved recall compared to a low SNR (+3 dBA). Reverberation time interacted with SNR to the effect that at +12 dB the shorter reverberation time improved recall, but at +3 dB it impaired recall.
