En ledande frÄga - Om ledarskap och positivt klassrumsklimat
Sammanfattning
Pedagogens yrkesroll Àr i ett förÀnderligt skede, numera krÀvs en stor yrkeskompetens för att hantera
komplexiteten i att undervisa. Skolan har ett uppdrag att skapa en lÀrandemiljö som gynnar elevernas
utveckling. Detta kan enbart ske nÀr alla som Àr verksamma i skolan funderar över och reflekterar kring sin roll
i elevernas lÀrande och utveckling.
VÄrt syfte Àr att undersöka hur pedagoger talar kring och gestaltar sin roll i skapandet av klassrumsklimat.
För att uppfylla detta syfte valde vi att utgÄ frÄn tre frÄgestÀllningar: Vilka faktorer pÄverkar klassrumklimatet
enligt pedagogerna? Hur pÄverkar pedagogens förhÄllningssÀtt klassrumsklimatet? Och vad kan pedagogen göra
för att skapa studiero och ett positivt klassrumsklimat?
Vi har utgÄtt frÄn ledarskapsteorier, för att stÀlla dessa mot resultaten. Detta för att undersöka hur
betydelsefulla teorierna Àr i pedagogens yrkesroll.
I insamlingen av material har vi valt att anvÀnda oss av en tredagars fokuserad observation och
kvalitativ/halvstrukturerad intervju med tre informanter för att för att fÄ en inblick i frÄgestÀllningarna.
I resultatet fann vi svÄrigheten i att översÀtta teori till praktik och komplexiteten i att skapa ett positivt
klassrumsklimat. Ăven om vi i vĂ„r undersökning utgick frĂ„n pedagogens roll i skapandet av klassrumsklimat, sĂ„
har vi sett att ocksÄ en mÀngd andra faktorer Àr med och samspelar. MÄnga av dessa Àr svÄra för att förÀndra.
Ăven om pedagogens förhĂ„llningssĂ€tt har inflytande pĂ„ klassrumsklimatet, sĂ„ Ă€r det sĂ„ledes inte tillrĂ€ckligt
för att beskriva komplexiteten i skapandet av klassrumsklimat. Vi har kommit fram till att pedagogens
förhÄllningssÀtt pÄverkar och att pedagogen behöver reflektera för att bli medveten om sin delaktighet i och om
de faktorer som ligger utanför dennes möjlighet att förÀndra. Vi har genom arbetet fÄtt en större insikt i
betydelsen av reflektion för att utveckla verksamheten och för att skapa en gynnsam lÀrandemiljö.
Microscopic description of translationally invariant core+N+N overlap functions
We derive expressions for core+N+N overlap integrals starting from microscopic wave functions obtained in the ab initio no-core shell model. These overlap integrals correspond to three-body channel form factors and can be used to investigate the clustering of many-body systems into a core plus two nucleons. We consider the case when the composite system and the core are described in Slater determinant, harmonic oscillator bases, and we show how to remove spurious center-of-mass components exactly in order to derive translationally invariant overlap integrals. We study in particular the Borromean 6He nucleus using realistic chiral nuclear interactions, and we demonstrate that the observed clusterization in this system is a Pauli focusing effect. The inclusion of three-body forces has a small effect on this structure. In addition, we discuss the issue of absolute normalization for spectroscopic factors, which we show is larger than one. As part of this study we also perform extrapolations of ground-state observables and investigate the dependence of these results on the resolution scale of the interaction
Large-scale exact diagonalizations reveal low-momentum scales of nuclei
Ab initio methods aim to solve the nuclear many-body problem with controlled
approximations. Virtually exact numerical solutions for realistic interactions
can only be obtained for certain special cases such as few-nucleon systems.
Here we extend the reach of exact diagonalization methods to handle model
spaces with dimension exceeding on a single compute node. This allows
us to perform no-core shell model (NCSM) calculations for 6Li in model spaces
up to and to reveal the 4He+d halo structure of this
nucleus. Still, the use of a finite harmonic-oscillator basis implies
truncations in both infrared (IR) and ultraviolet (UV) length scales. These
truncations impose finite-size corrections on observables computed in this
basis. We perform IR extrapolations of energies and radii computed in the NCSM
and with the coupled-cluster method at several fixed UV cutoffs. It is shown
that this strategy enables information gain also from data that is not fully UV
converged. IR extrapolations improve the accuracy of relevant bound-state
observables for a range of UV cutoffs, thus making them profitable tools. We
relate the momentum scale that governs the exponential IR convergence to the
threshold energy for the first open decay channel. Using large-scale NCSM
calculations we numerically verify this small-momentum scale of finite nuclei.Comment: Minor revisions.Accepted for publication in Physical Review
Förvaltning av offentliga skogsfastigheter
I Sverige finns det 290 kommuner varav majoriteten Àger skogsfastigheter. 60 procent av den urbana skogen Àgs av kommuner, vilket betyder att strategin för förvaltning av de offentliga skogsfastigheterna fÄr betydelse för sÀkerstÀllande av bland annat frilufts- och rekreationsomrÄden men Àven som markreserv för framtida exploatering. Enligt kommunallagen Àr det upp till varje kommun att formulera strategier för hur förvaltningsarbetet ska utformas och genomföras. Eftersom kommunernas uppgift Àr bred med ett konglomerat av uppdrag och tjÀnster konkurrerar ofta olika krafter inom organisationerna med varandra, vilket kan orsaka implementeringsproblem. För en del kommuner kan det innebÀra att förvaltningen av offentliga skogsfastigheter ÄsidosÀtts. DÀrför behövs det en ökad lokal och nationell uppmÀrksamhet dÄ de offentliga skogsfastigheterna Àr invÄnarnas samfÀllda egendom.
Studiens övergripande syfte Àr att identifiera och beskriva strategier och handlingsplaner hos olika kommuner för förvaltning av offentliga fastigheter. Vidare Àr syftet att se hur kommunala strategier för offentliga skogsfastigheter ser ut, i vilken grad beslutsprocesser Àr metodiskt genomförda, vilka mÄl och mÄlsÀttningar som finns, vilken skoglig kunskap som finns hos kommunerna samt hur förvaltningsarbete kan förbÀttras och utvecklas. Studiens undersökningsobjekt Àr VÀxjö, Uddevalla, Sollentuna, Sandvikens och PiteÄ kommun.
Eftersom forskningsomrÄdet Àr relativt outforskat och det inte finns sÄ mycket fÀrdig teoribildning har den kvalitativa metoden grundad teori anvÀnts. Genom intervjuer med personer pÄ olika hierarkiska nivÄer hos de fem olika kommunerna har material samlats in. Den grundade teorin utgÄr frÄn empirin och kan genom datamaterial generera ny teori. En viktig ansats i metodarbetet Àr att det inledande arbetet, i största möjliga mÄn, ska ske förutsÀttningslöst. För att undvika att styra arbetet mot redan etablerade teorier och begrepp tillkommer en litteraturstudie i avsnittet för analys och diskussion. Etablerade teorier och begrepp anvÀnds för att styrka den genererade teorin. Metoden skiljer sig frÄn andra metoder dÀr man vanligtvis utgÄr frÄn etablerad teori som sedan testas i empirin.
Resultatet visar att utformningen av kommunernas förvaltning av skogsinnehavet skiljer sig Ät och att det finns brister inom strategiarbetet hos flera kommuner. MÄlformulering, strategisk utveckling, mÄluppfyllelse, uppföljning och sÀkerstÀllande av utförda ÄtgÀrder, funktioner i organisationen för resursutnyttjande Àr kÀrnkategorier som tagits fram för att identifiera och beskriva strategier hos kommunerna.
Resultatet visar att ÀndamÄlet för de offentliga skogsfastigheterna skiljer sig Ät mellan förvaltare, högre tjÀnstemÀn och politiker. TvÄ av kommunerna har formellt antagna skogspolicys dÀr det finns övergripande strategier för de offentliga skogsfastigheterna. Högre tjÀnstemÀn och politiker kÀnde dock inte till antagna policys som ligger till grund för operativa beslut som fattas i skogsförvaltningens dagliga arbete. Hos en kommun finns en mycket kort övergripande plan men det Àr oklart om den har antagits i nÄgon nÀmnd. Den Àr inte tillrÀckligt utförlig för att kunna anvÀndas som policy till kommunens strategiarbete. Hos en kommun finns ingen formell antagen policy. Förvaltaren har arbetat under flera Är med att ta fram en policy men den Àr fortfarande under arbete. Arbetet sker enligt en naturvÄrdsplan som förvaltaren tagit fram. Hos den femte kommunen finns ingen skogspolicy utan arbetet sker utifrÄn en skogsbruksplan. Operativa och strategiska beslut fattas av tjÀnstemÀn inom skogsförvaltningarna. Resultatet visar att kommunikationen och Äterkopplingen mellan förvaltningen av de offentliga skogsfastigheterna och den övriga kommunala verksamheten Àr lÄg. Det Àr förvaltarna sjÀlva som formulerar mÄl och bedriver arbetet inom de offentliga skogsfastigheterna.
Studien visar att det behövs gemensamma och tydliga riktlinjer för mĂ„l och strategier för innehavet av de offentliga skogsfastigheterna. För att förbĂ€ttra och utveckla flera steg inom strategiarbetet behövs samordnade exkursioner i fĂ€lt för förvaltare, högre tjĂ€nstemĂ€n och politiker. NĂ€tverkande mellan olika kommuner rekommenderas för att bland annat kunna ge samhörighet, lĂ€rande och kunskap i organisationerna.Sweden has 290 municipalities where most of them own forestland. 60 percent of the urban forests are owned by municipalities, which mean that the strategy for management of the forests will be important to ensure recreation areas as well as land reserved for future development. According to the Swedish Local Government act, each municipality has to form their own strategies for how to design and implement the management of forests. Since the municipalities has a wide commission with a conglomerate of duties and services many of the forces within the organizations often compete with each other, which can cause implementation problems. For some municipalities, it may mean that the management of municipal forests is undermined. Therefore there is a need for an increased local and national attention due to the residentâs collective possession.
The study's overall purpose is to identify and describe strategies for management of municipal forests. An additional purpose is to understand how work is organized, which the goals are and how strategies can be developed or improved. The study has been made in the following municipalities: VÀxjö, Uddevalla, Sollentuna, Sandviken and PiteÄ.
Since the literature background is considered low the qualitative method of grounded theory is used. Data has been collected through interviews with people at different hierarchical levels in the five different municipalities. The theory is based on empirical data, which is used to generate new theory. An important approach to the method is that the initial work, as far as possible, should be unbiased. Therefore the literature review and established theories are placed in the latter section of the study. The grounded theory differs from other methods, which are usually based on established theory and then empirically tested.
The results show that the strategy of municipal organizations differs and there are shortcomings in the strategy of several municipalities. Objectives, strategic development, objective compliance, monitoring and management, functions of the organization are the core categories that were developed for identifying and describing the strategies of the municipalities.
The results show that the objectives of municipal forest differ between managers, senior officials and politicians. Two of the municipalities have formally enrolled forest policies where there are comprehensive approaches to municipal forests. Senior officials and politicians were not aware of the forest policies that form the basis for operational decisions made in the daily work of forest management. Two of the municipalities have forest policies, but they were not formally enrolled. One of the municipalities did not have any long-term and overall goals and strategies. Officials in the forestry administrations make the operational and strategic decisions. The result shows that communication and feedback between the management of municipal forests and other municipal areas is low. It is the managers of the municipal forests that formulate objectives and conduct the work.
The study indicates that there is a need for common and clear guidance on goals and strategies for the maintenance of municipal forests. To improve and develop parts within the strategy coordinated field trips for managers, senior officials and politicians are needed. Networking between various municipalities is recommended for instance, to give unity, learning and knowledge in organizations
Att fÄ sÀtta spÄr
Every place is a stage where people interact. The nature of the stage and its scenography to some extent shapes the interaction.
Considered as a place, the school ground gives rise to many varying expectations. Lower school pupils often have clear ideas of how they are expected to behave and how they are expected to use the place. But as the pupils age, the purpose of the school ground grows less obvious. This presents the planner with a challenge as well as creative freedom. Obviously, the basic functions of the school grounds remain the same whether children or young adults are using it; windows should offer a pleasant view; pupils should have ready access to a place for gathering during breaks. The grounds should offer pupils a chance to relax from the demands of the classroom and fill their lungs with fresh air and their heads with new thoughts. In the best of worlds, the school ground can become an extra class room and a pedagogical resource for the school.
But whereas school grounds present younger children with ample room for games and play, an activity void appears as pupils age. The identity and usage of place is no longer determined by games and play. Something else must fill the void: the school ground can become a place where you can set tracks.
Since adults create school grounds, they express an adult perspective. They are by and large shaped by adult values, adult's readiness to delegate responsibility to children, and adult's faith in children's judgement. Our personalities are partly formed the physical environment in which we develop. People shape places that in turn shape people. The school ground is then a space that not only influences the physical health of sub-adults, but also their personality. Consequently, the best of school grounds embraces the tracks set by the pupils By allowing someone to shape their own environment, you allow them to shape their own self. The school ground then becomes a place that belongs to the pupils, a place to which they can belong.
There are two ways to set tracks. One is to effectively change the physical situation. The other is to instruct someone who is involved in the planning process. In this work I have examined various ways of designing school grounds in collaboration with its users. It has been my intention to use the visions that students of the upper secondary school have of their outdoor environment. The work has allowed me as a landscape architect to examine how I can create external places based in part on students' inner worlds. The work also includes a proposal to renovate the school ground at Alströmergymnasiet, an upper secondary school in the south of Sweden. My aim has been to rely on user participitation both for creating an inventory of the grounds and as a method for realising the proposal.
The occupational role of the landscape architect offers an array of choices. We can choose to use those roles most appropriate for the project in question. One of the more inspiring roles during this work has been to allow the actual end-users to paint me a picture of their dream-places.SkolgÄrden Àr en plats som rymmer mÄnga

vara en plats att trÀffa kompisar pÄ, för ett
avbrott i lektionstempot och en möjlighet
att fylla lungor och hjÀrna med frisk luft.
Den kan vara ett av skolans pedagogiska
rum och en resurs i undervisningen. Men
dÀr leken och kreativiteten inte lÀngre har
nÄgot givet utrymme mÄste nÄgot annat till.
En skolgÄrd kan vara en plats att fÄ sÀtta
avtryck pÄ.
Det finns platser som Àr fÀrdiga,
iordningstÀllda och regisserade. Platser
dÀr rollerna Àr givna och dÀr alla vet hur
man ska bete sig. SÄ kan en skolgÄrd
vara, inte minst för de lÀgre Ärskurserna
dÀr skolgÄrden Àr till för lek och spring i
benen.
Det finns ytor som vÀntar pÄ att nÄgot ska
hÀnda, pÄ att nÄgot ska ta plats pÄ dem.
MÄnga gymnasieskolgÄrdar kan upplevas
just sÄ. Leken Àr inte lÀngre en socialt
godkÀnd syssla och sportlekar (eller
tjuvrökning) passar inte alla.
Det finns ocksÄ platser som söker ett
utrymme att hÀnda pÄ. Inom varje
mÀnniska ligger tusen sÄdana platser och
vÀntar pÄ att fÄ komma fram. Inom varje
elev, lÀrare och rektor finns dessa platser.
Detta examensarbete har som mÄl att
undersöka hur gymnasieskolgÄrden kan
bli det utrymme dÀr de inre platserna hos
eleverna kan fÄ finna ett fysiskt uttryck.
SkolgÄrden Àr en plats dit eleverna Àr
hÀnvisade, den Àr gjord just för dem. Den
Àr skapad av vuxna och blir dÀrmed ett
uttryck för hur vuxenvÀrlden ser pÄ och
vÀrderar ungdomarna, vilket ansvar de ger
dem, vad man betror dem med. SkolgÄrden
Àr pÄ sÄ sÀtt en plats som inte bara pÄverkar
ungdomarnas hÀlsa, utan Àven formar
deras personlighet. I sjÀlva skapandet, i
formandet av sin omgivning, formas ocksÄ
den egna personen. MĂ€nniskan formar
dÀrför platsen som formar mÀnniskan.
Den skolgÄrd som ger utrymme för
elevernas medbestÀmmande skulle dÀrmed
pÄ ett effektivt sÀtt kunna bidra till att
att skapa nya aktiva medborgare. Den
bÀsta skolgÄrden Àr den som tillÄter och
omfamnar de spÄr som eleverna sÀtter, en
plats som de kan göra till sin egen, en plats
som de kan göra till en del av sig.
Det finns tvÄ sÀtt att sÀtta spÄr. Det ena Àr
att sjÀlv gÄ ut och delta i arbetet med att
fysiskt förÀndra. Det andra Àr att fÄ berÀtta
för nÄgon annan hur de ska göra det, att fÄ
göra sin röst hörd i planeringsprocessen.
I detta arbete har jag prövat olika former
av brukarinflytande för att undersöka
hur man som landskapsarkitekt kan dra
nytta av gymnasieelevers funderingar och
önskemÄl om sin egen skolgÄrd. Som en
del av detta arbete har det ocksÄ ingÄtt
att ta fram ett förslag för upprustning
av Alströmergymnasiet i AlingsÄs
skolgÄrd. Mitt mÄl med det fÀrdiga
förslaget har varit att förverkligandet av
det i sin tur ska rymma medverkan frÄn
eleverna i sÄvÀl detaljutformning som i
byggandet. Brukarinflytandet har anvÀnts i
analysarbetet och samtidigt varit ett sÀtt att
pÄbörja den interna förankringen pÄ skolan.
De metoder som kan anvÀndas för
att engagera och integrera brukarna
av en plats i en planeringsprocess Àr
bÄde mÄnga och skiftande. DÀr finns
de allmÀnt anvÀnda, som ex stormöte
och samrÄdsmöte som frÀmst handlar
om förankring av en redan pÄbörjad
process. Men det finns ocksÄ en uppsjö
av mer processinriktade som ex. charet,
framtidsverkstad m.fl. De metoder jag har
valt att arbeta med Àr gÄtur tillsammans
med elev- och personalrepresentanter och
en framtidsverkstad med en elevgrupp.
Dessa valdes dels för att de ger ett stort
utbyte pÄ kort tid, dels dÀrför att de
angriper problemet frÄn tvÄ olika synsÀtt.
GÄtur grundar sig pÄ att en expert kommer
in och vÀljer de omrÄden som Àr viktiga
att diskutera. Brukarna ska ge svaren till
experten. Framtidsverkstaden Ă„ andra sidan
ser brukarna som experterna som ska hitta
lösningarna.
Arbetet har varit ett sÀtt att undersöka pÄ
vilket sÀtt jag som landskapsarkitekt kan
anvÀnda mig av brukarnas inre platser, pÄ
vilket sÀtt jag kan ge dessa en fysisk form.
Under arbetets gÄng har denna roll flera
gÄnger förÀndrats. Att anvÀnda de faktiska
brukarnas tankar och upplevelser som en
del i analysen har ocksÄ inneburit att jag
har varit tvungen att ta stÀllning till min
egen roll i situationer dÀr deras och mina
bilder behöver jÀmkas. En av slutsatserna
av detta arbete Àr att vi ofta sÄg problem
pÄ olika strukturnivÄer och förslagen
kompletterade dÀrför varandra. Att lÄta
elever och personal pÄ skolan vara med och
pÄverka utformningen av sin gemensamma
miljö Àr bÄde berikande och utmanande för
den som ska planera en gÄrd. De problem
som uppstÄtt pÄ vÀgen och de lösningar
som jag dÀrigenom varit tvungen att söka,
kan ses som ett sÀtt att utveckla den egna
yrkesrollen. Landskapsarkitektens yrkesroll
Àr pÄ gott och ont öppen och rymmer
mÄnga valmöjligheter. Det innebÀr att vi
kan vÀlja att anvÀnda de roller som passar
bÀst för just det projekt som gÀller. En av
de mer inspirerande rollerna under detta
arbete har för mig varit att ge de faktiska
brukarna möjlighet att kunna visa vilken
plats de drömmer om
Membrane topology of the 60 kDa Oxa1p-homologue from Escherichia coli
We have characterized the membrane topology of a 60-kDa inner membrane protein from Escherichia coli that is homologous to the recently identified Oxa1p protein in Saccharomyces cerevisiae mitochondria implicated in the assembly of mitochondrial inner membrane proteins. Hydrophobicity and alkaline phosphatase fusion analyses suggest a membrane topology with six transmembrane segments, including an N-terminal signal-anchor sequence not present in mitochondrial Oxa1p. In contrast to partial N-terminal fusion protein constructs, the full-length protein folds into a protease-resistant conformation, suggesting that important folding determinants are present in the C-terminal part of the molecule
MicroRNAs: Novel Regulators Involved in the Pathogenesis of Psoriasis?
MicroRNAs are a recently discovered class of posttranscriptional regulators of gene expression with critical functions in health and disease. Psoriasis is the most prevalent chronic inflammatory skin disease in adults, with a substantial negative impact on the patients' quality of life. Here we show for the first time that psoriasis-affected skin has a specific microRNA expression profile when compared with healthy human skin or with another chronic inflammatory skin disease, atopic eczema. Among the psoriasis-specific microRNAs, we identified leukocyte-derived microRNAs and one keratinocyte-derived microRNA, miR-203. In a panel of 21 different human organs and tissues, miR-203 showed a highly skin-specific expression profile. Among the cellular constituents of the skin, it was exclusively expressed by keratinocytes. The up-regulation of miR-203 in psoriatic plaques was concurrent with the down-regulation of an evolutionary conserved target of miR-203, suppressor of cytokine signaling 3 (SOCS-3), which is involved in inflammatory responses and keratinocyte functions. Our results suggest that microRNA deregulation is involved in the pathogenesis of psoriasis and contributes to the dysfunction of the cross talk between resident and infiltrating cells. Taken together, a new layer of regulatory mechanisms is involved in the pathogenesis of chronic inflammatory skin diseases
Promoter polymorphism -119C/G in MYG1 (C12orf10) gene is related to vitiligo susceptibility and Arg4Gln affects mitochondrial entrance of Myg1
<p>Abstract</p> <p>Background</p> <p><it>MYG1 </it>(<it>Melanocyte proliferating gene 1</it>, also C12orf10 in human) is a ubiquitous nucleo-mitochondrial protein, involved in early developmental processes and in adult stress/illness conditions. We recently showed that <it>MYG1 </it>mRNA expression is elevated in the skin of vitiligo patients. Our aim was to examine nine known polymorphisms in the <it>MYG1 </it>gene, to investigate their functionality, and to study their association with vitiligo susceptibility.</p> <p>Methods</p> <p>Nine single nucleotide polymorphisms (SNPs) in the <it>MYG1 </it>locus were investigated by SNPlex assay and/or sequencing in vitiligo patients (n = 124) and controls (n = 325). <it>MYG1 </it>expression in skin biopsies was detected by quantitative-real time PCR (Q-RT-PCR) and polymorphisms were further analysed using luciferase and YFP reporters in the cell culture.</p> <p>Results</p> <p>Control subjects with -119G promoter allele (rs1465073) exhibited significantly higher <it>MYG1 </it>mRNA levels than controls with -119C allele (<it>P </it>= 0.01). Higher activity of -119G promoter was confirmed by luciferase assay. Single marker association analysis showed that the -119G allele was more frequent in vitiligo patients (47.1%) compared to controls (39.3%, <it>P </it>< 0.05, OR 1.37, 95%CI 1.02-1.85). Analysis based on the stage of progression of the vitiligo revealed that the increased frequency of -119G allele occurred prevalently in the group of patients with active vitiligo (n = 86) compared to the control group (48.2% <it>versus </it>39.3%, <it>P </it>< 0.05; OR 1.44, 95%CI 1.02-2.03). Additionally, we showed that glutamine in the fourth position (in Arg4Gln polymorphism) completely eliminated mitochondrial entrance of YFP-tagged Myg1 protein in cell culture. The analysis of available EST, cDNA and genomic DNA sequences revealed that Myg1 4Gln allele is remarkably present in human populations but is never detected in homozygous state according to the HapMap database.</p> <p>Conclusions</p> <p>Our study demonstrated that both <it>MYG1 </it>promoter polymorphism -119C/G and Arg4Gln polymorphism in the mitochondrial signal of Myg1 have a functional impact on the regulation of the <it>MYG1 </it>gene and promoter polymorphism (-119C/G) is related with suspectibility for actively progressing vitiligo.</p
Filaggrin Genotype Determines Functional and Molecular Alterations in Skin of Patients with Atopic Dermatitis and Ichthyosis Vulgaris
BACKGROUND: Several common genetic and environmental disease mechanisms are important for the pathophysiology behind atopic dermatitis (AD). Filaggrin (FLG) loss-of-function is of great significance for barrier impairment in AD and ichthyosis vulgaris (IV), which is commonly associated with AD. The molecular background is, however, complex and various clusters of genes are altered, including inflammatory and epidermal-differentiation genes. OBJECTIVE: The objective was to study whether the functional and molecular alterations in AD and IV skin depend directly on FLG loss-of-function, and whether FLG genotype determines the type of downstream molecular pathway affected. METHODS AND FINDINGS: Patients with AD/IV (nâ=â43) and controls (nâ=â15) were recruited from two Swedish outpatient clinics and a Swedish AD family material with known FLG genotype. They were clinically examined and their medical history recorded using a standardized questionnaire. Blood samples and punch biopsies were taken and trans-epidermal water loss (TEWL) and skin pH was assessed with standard techniques. In addition to FLG genotyping, the STS gene was analyzed to exclude X-linked recessive ichthyosis (XLI). Microarrays and quantitative real-time PCR were used to compare differences in gene expression depending on FLG genotype. Several different signalling pathways were altered depending on FLG genotype in patients suffering from AD or AD/IV. Disease severity, TEWL and pH follow FLG deficiency in the skin; and the number of altered genes and pathways are correlated to FLG mRNA expression. CONCLUSIONS: We emphasize further the role of FLG in skin-barrier integrity and the complex compensatory activation of signalling pathways. This involves inflammation, epidermal differentiation, lipid metabolism, cell signalling and adhesion in response to FLG-dependent skin-barrier dysfunction
