Dietary exposure to parabens and body mass index in an adolescent Spanish population
This work was carried out in line with 'GP/EFSA/ENCO/380 2018/03/G04: OBEMIRISK: Knowledge platform for assessing the risk of Bisphenols on gut microbiota and its role in obesogenic phenotype: looking for biomarkers'. This research was also funded by Plan Estatal de I + D + I 2013-2016 and co-funded by FEDER-ISCIII PI17/01758, FEDER-Consejeria de Salud y Familias, Junta de Andalucia PE-0250-2019, FEDER/Junta de Andalucia-Consejeria de Transformacion Economica, Industria, Conocimiento y Universidades/Proyecto P18-RT-4247. All individuals participating in this research signed an informed consent form and the study protocol was approved by the Ethics Com-mittee of the University of Granada.Parabens are alkyl esters of p-hydroxybenzoic acid which are extensively used in cosmetics, pharmaceuticals and
foodstuffs due to their antimicrobial properties. The most commonly used parabens are methyl-(MeP), ethyl-
(EtP), propyl-(PrP) and butyl-(BuP) paraben. Most human exposure to parabens is achieved through the consumption
of food or pharmaceutical products and the use of personal care products. However, studies on dietary
parabens exposure and the associated factors are very scarce. The main aim of the present study was to explore
factors associated with dietary exposure to parabens in Spanish adolescents according to gender. Dietary data
and anthropometric measures were collected from 585 adolescents (53.4% boys) aged 12–16 years. Parabens
exposure through diet was assessed using a food frequency questionnaire with food products providing more
than 95% of energy and macronutrient intake being included in analysis. Stepwise regression was used to
identify the foods that most contributed to parabens intake. Logistic regression was used to evaluate factors
predicting higher dietary exposure to parabens. The main contributors to dietary MeP, EtP, PrP and BuP exposure
in adolescent boys were eggs (41.9%), canned tuna (46.4%), bakery and baked goods products (57.3%) and
pineapple (61.1%). In adolescent girls, the main contributors were apples and pears (35.3%), canned tuna
(42.1%), bakery and baked goods products (55.1%) and olives (62.1%). Overweight/obese girls were more likely
to belong to the highest tertile of overall parabens intake (odds ratio [OR]: 3.32; 95% confidence interval [95%
CI]: 1.21–9.15) and MeP (OR: 3.05; 95% CI: 1.14–8.12) than those with a body mass index lower than 25 kg/m2.
These findings suggest a positive association between dietary exposure to parabens and overweight/obesity in
adolescent girls.Plan Estatal de I + D + I 2013-2016FEDER-ISCIII PI17/01758FEDER-Consejeria de Salud y FamiliasJunta de Andalucia PE-0250-2019FEDER/Junta de Andalucia-Consejeria de Transformacion Economica, Industria, Conocimiento y Universidades/Proyecto P18-RT-424
Novel Horizons in Postbiotics: Lactobacillaceae Extracellular Vesicles and Their Applications in Health and Disease
Lactobacillus probiotics contained in dietary supplements or functional foods are wellknown
for their beneficial properties exerted on host health and diverse pathological situations. Their
capacity to improve inflammatory bowel disease (IBD) and regulate the immune system is especially
remarkable. Although bacteria–host interactions have been thought to occur directly, the key role that
extracellular vesicles (EVs) derived from probiotics play on this point is being unveiled. EVs are lipid
bilayer-enclosed particles that carry a wide range of cargo compounds and act in different signalling
pathways. Notably, these EVs have been recently proposed as a safe alternative to the utilisation of
live bacteria since they can avoid the possible risks that probiotics may entail in vulnerable cases
such as immunocompromised patients. Therefore, this review aims to give an updated overview of
the existing knowledge about EVs from different Lactobacillus strains, their mechanisms and effects in
host health and different pathological conditions. All of the information collected suggests that EVs
could be considered as potential tools for the development of future novel therapeutic approaches.Junta de Andalucia CTS 164

Vanadium Decreases Hepcidin mRNA Gene Expression in STZ-Induced Diabetic Rats, Improving the Anemic State
We are grateful for the support received from the Consejeria de Innovacion, Ciencia y Empresa, Andalusian Regional Government (Project P06-CTS-01435), and from the Spanish Ministry of Economy and Competitivity (SAF2011-29648); the CIBERehd is funded by the Instituto de Salud Carlos III.This paper forms part of the Doctoral Program in Nutrition and Food Science of
the University of Granada.Diabetes is a disease with an inflammatory component that courses with an anemic state. Vanadium (V) is an antidiabetic agent that acts by stimulating insulin signaling. Hepcidin blocks the intestinal absorption of iron and the release of iron from its deposits. We aim to investigate the effect of V on hepcidin mRNA expression and its consequences on the hematological parameters in streptozotocin-induced diabetic Wistar rats. Control healthy rats, diabetic rats, and diabetic rats treated with 1 mgV/day were examined for five weeks. The mineral levels were measured in diet and serum samples. Hepcidin expression was quantified in liver samples. Inflammatory and hematological parameters were determined in serum or whole blood samples. The inflammatory status was higher in diabetic than in control rats, whereas the hematological parameters were lower in the diabetic rats than in the control rats. Hepcidin mRNA expression was significantly lower in the V-treated diabetic rats than in control and untreated diabetic rats. The inflammatory status remained at a similar level as the untreated diabetic group. However, the hematological profile improved after the V-treatment, reaching similar levels to those found in the control group. Serum iron level was higher in V-treated than in untreated diabetic rats. We conclude that V reduces gene expression of hepcidin in diabetic rats, improving the anemic state caused by diabetes.Consejeria de Innovacion, Ciencia y Empresa, Andalusian Regional Government

Comparative Study of the Antioxidant and Anti-Inflammatory E ects of Leaf Extracts from Four Di erent Morus alba Genotypes in High Fat Diet-Induced Obesity in Mice
The authors declare no conflict of interest. The funders had no role in the design of the study;

the results.Increased levels of reactive oxygen species (ROS) and a low-grade chronic inflammation in
multiple organs have been demonstrated in obesity. Morus alba leaves extracts (MAEs) have been used
in traditional medicine as anti-inflammatory agents. In this work, the bioactive compounds of di erent
genotypes of M. alba L. (Filipina, Valenciana Temprana, Kokuso, and Italia) were analyzed not only by
reverse phase high performance liquid chromatography–electrospray ionization-time of flight-mass
spectrometry (RP-HPLC-ESI-TOF-MS) and hydrophilic interaction chromatography–electrospray
ionization-time of flight-mass spectrometry (HILIC-ESI-TOF-MS), but also screened for in vitro and
in vivo antioxidant activity by means of DPPH radical scavenging assay and Caenorhabditis elegans
model. These MAEs were administered daily in a model of diet-induced obesity in mice. Filipina
and Italia genotypes significantly reduced weight gain, the glycemic levels in high fat diet, as well
as, levels of LDL-cholesterol and triglycerides. Filipina and Italia MAEs also reduced the expression
of proinflammatory mediators such as Tnf- , Il-1 , Il-6 and increased the levels of adiponectin and
AMPK, which exert anti-inflammatory e ects. Moreover, Italia genotype ameliorated the intestinal
barrier function. In conclusion, Filipina and Italia methanolic extracts show the highest antioxidant
and anti-inflammatory e ect, due to the presence of compounds such as protocatechuic acid or
quercetin-3-glucoside, and they could be developed as a complementary treatment for obesity and
metabolic disorders.Junta de Andalucia

Beneficial Effects of Limosilactobacillus fermentum in the DCA Experimental Model of Irritable Bowel Syndrome in Rats
Limosilactobacillus fermentum CECT5716, a probiotic strain isolated from human milk, has
reported beneficial effects on different gastrointestinal disorders. Moreover, it has shown its ability to
restore altered immune responses, in association with microbiome modulation in different pathological
conditions. Therefore, our aim was to assess the effects of a Limosilacbacillus fermentum CECT5716
in a rat experimental model of irritable bowel syndrome (IBS) that resembles human IBS. The experimental
IBS was induced by deoxycholic acid (DCA) in rats and then, Limosilactobacillus fermentum
CECT5716 (109 CFU/day/rat) was administered. Behavioral studies, hyperalgesia and intestinal
hypersensitivity determinations were performed and the impact of the probiotic on the inflammatory
and intestinal barrier integrity was evaluated. Additionally, the gut microbiota composition was
analyzed. Limosilactobacillus fermentum CECT5716 attenuated the anxiety-like behavior as well as the
visceral hypersensitivity and referred pain. Moreover, this probiotic ameliorated the gut inflammatory
status, re-establishing the altered intestinal permeability, reducing the mast cell degranulation and
re-establishing the gut dysbiosis in experimental IBS. Therefore, our results suggest a potential use of
Limosilactobacillus fermentum CECT5716 in clinical practice for the management of IBS patients.Junta de Andalucia A-CTS-447-UGR18

The melatonergic agonist agomelatine ameliorates high fat diet-induced obesity in mice through the modulation of the gut microbiome
Background and purpose: Melatonin has shown beneficial effects on obesity, both in humans and experimental
models, via regulating the altered circadian rhythm and thus ameliorating the gut dysbiosis associated with this
metabolic condition. However, its clinical use is limited, mostly due to its short half-life. Agomelatine is an
agonist of the melatonin receptors that could be used to manage obesity and offer a better profile than melatonin.
Experimental approach: Male C57BL/6 mice were fed a high fat diet and orally treated for five weeks with agomelatine,
or melatonin or metformin, used as control drugs. Metabolic profile, inflammatory status, vascular
dysfunction and intestinal microbiota composition were assessed.
Key results: Agomelatine lessened body weight gain, enhanced glucose and lipid metabolisms, and improved
insulin resistance. It also reduced the obesity-associated inflammatory status and endothelial dysfunction,
probably linked to its effect on gut dysbiosis, consisting of the restoration of bacterial populations with key
functions, such as short chain fatty acid production.
Conclusions and implications: Agomelatine can be considered as a novel therapeutic tool for the management of
human obesity and its associated comorbidities.
