Insights into the origin of metazoan filopodia and microvilli.
Filopodia are fine actin-based cellular projections used for both environmental sensing and cell motility, and they are essential organelles for metazoan cells. In this study, we reconstruct the origin of metazoan filopodia and microvilli. We first report on the evolutionary assembly of the filopodial molecular toolkit and show that homologs of many metazoan filopodial components, including fascin and myosin X, were already present in the unicellular or colonial progenitors of metazoans. Furthermore, we find that the actin crosslinking protein fascin localizes to filopodia-like structures and microvilli in the choanoflagellate Salpingoeca rosetta. In addition, homologs of filopodial genes in the holozoan Capsaspora owczarzaki are upregulated in filopodia-bearing cells relative to those that lack them. Therefore, our findings suggest that proteins essential for metazoan filopodia and microvilli are functionally conserved in unicellular and colonial holozoans and that the last common ancestor of metazoans bore a complex and specific filopodial machinery
Role of delta-tubulin and the C-tubule in assembly of Paramecium basal bodies
BACKGROUND: A breakthrough in the understanding of centriole assembly was provided by the characterization of the UNI3 gene in Chlamydomonas. Deletion of this gene, found to encode a novel member of the tubulin superfamily, delta-tubulin, results in the loss of the C-tubule, in the nine microtubule triplets which are the hallmark of centrioles and basal bodies. Delta-tubulin homologs have been identified in the genomes of mammals and protozoa, but their phylogenetic relationships are unclear and their function is not yet known. RESULTS: Using the method of gene-specific silencing, we have inactivated the Paramecium delta-tubulin gene, which was recently identified. This inactivation leads to loss of the C-tubule in all basal bodies, without any effect on ciliogenesis. This deficiency does not directly affect basal body duplication, but perturbs the cortical cytoskeleton, progressively leading to mislocalization and loss of basal bodies and to altered cell size and shape. Furthermore, additional loss of B- and even A-tubules at one or more triplet sites are observed: around these incomplete cylinders, the remaining doublets are nevertheless positioned according to the native ninefold symmetry. CONCLUSIONS: The fact that in two distinct phyla, delta-tubulin plays a similar role provides a new basis for interpreting phylogenetic relationships among delta-tubulins. The role of delta-tubulin in C-tubule assembly reveals that tubulins contribute subtle specificities at microtubule nucleation sites. Our observations also demonstrate the existence of a prepattern for the ninefold symmetry of the organelle which is maintained even if less than 9 triplets develop
On The Spectral Energy Dependence of Gamma-Ray Burst Variability
The variable activity of a gamma-ray burst (GRB) source is thought to be
correlated with its absolute peak luminosity - a relation that, if confirmed,
can be used to derive an independent estimate of the redshift of a GRB. We find
that bursts with highly variable light curves have greater nu-F_nu spectral
peak energies, when we transform these energies to the cosmological rest frame
using the redshift estimates derived either from optical spectral features or
from the luminosity-variability distance indicator itself. This positive
correlation between peak energy and variability spans ~ 2 orders of magnitude
and appears to accommodate GRB 980425, lending credibility to the association
of this burst with SN 1998bw. The existence of such a correlation not only
provides an interesting clue to the nature of this luminosity indicator but
potentially reinforces the validity of the redshift estimates derived from this
method. It also implies that the rest frame GRB peak energy is correlated with
the intrinsic luminosity of the burst, as has been suggested in the past as an
explanation of the observed hardness-intensity correlation in GRBs.Comment: 10 pages, 3 figures; Accepted to Ap
Self‐Assembled Gels formed in Deep Eutectic Solvents : Supramolecular Eutectogels with High Ionic Conductivities
This paper reports the ability of 1,3:2,4‐dibenzylidene‐D‐ sorbitol (DBS), a simple, commercially‐relevant compound, to self‐assemble as a result of intermolecular non‐covalent interactions into supramolecular gels in deep eutectic solvents (DESs). The DESs are based on choline chloride combined with alcohols/ureas – DBS forms gels at a loading of 5% wt/vol. Rheology confirms the gel‐like nature of the materials, electron microscopy indicates underpinning nanofibrillar DBS networks and differential scanning calorimetry shows the deep eutectic solvent nature of the liquid‐like phase is retained. The ionic conductivities of the gels are similar to those of the unmodified DESs proving the deep eutectic nature of the ionic liquid‐like phase. Gelation is tolerant of ionic additives Li+, Mg2+ and Ca2+, with the resulting gels having similar conductivities to electrolyte dissolved in the native DES. The low‐molecular‐weight gelator DBS is a low‐cost additive – forming gels in DESs from readily‐available constituents, with conductivity levels suitable for practical applications. We suggest supramolecular eutectogels have potential uses ranging from energy technology to synthesis and catalysis

Induction and suppression of antiviral RNA silencing by Tomato spotted wilt virus
RNA silencing is an essential antiviral defense system in plants. Triggered by doublestranded RNA, silencing results in degradation or translational repression of target RNA. Viruses are inducers and targets of RNA silencing. To condition susceptibility, most plant viruses encode silencing suppressor proteins that interfere with RNA silencing. Tomato spotted wilt virus (TSWV) NSs protein is an RNA silencing suppressor. The mechanism of RNA silencing suppression by NSs and its role in virus infection and movement remain to be determined. We cloned NSs from the Hawaii isolate of TSWV. Using two independent assays, we show that NSs restored pathogenicity and supported the formation of local infection foci by suppressor-deficient Turnip mosaic virus (TuMV) and Turnip crinkle virus (TCV). Suppression of silencing directed against heterologous viruses establishes the foundation to determine the mechanism of antiviral RNA silencing suppression by NSs
