    N-(5-Amino-9H-benzo[a]phenoxazin-9-ylidene)propan-1-aminium chlorides as antifungal agents and NIR fluorescence probes

    The search for benzo[a]phenoxazines, Nile Blue derivatives, with high antifungal activity and cell labelling capacity based on our previously published works in this type of compounds, led us to the design of compounds with specific substituents in the polycyclic system. Thus, in the present work, four new benzo[a]phenoxazinium chlorides, possessing at the 5-position amino or (3-aminopropyl) amino groups and at the 9-position propylamino or dipropylamino groups, were synthesized. Another analogue, with (3-aminopropyl) amino group at 5-position, ethyl amino group at 9-position and a methyl group at 10-position of the polycyclic system was also synthesized for comparison in the studies performed. Fundamental photophysics (absorption and fluorescent emission) was carried out in absolute ethanol, water, and other aqueous solutions of different pH values, relevant for the potential biological applications of these compounds. The antiproliferative activity of the synthesized benzo[a]phenoxazinium chlorides was determined using Saccharomyces cerevisiae PYCC 4072 and the microdilution method described for antifungal susceptibility tests in yeast. All compounds revealed antifungal activity, being the most active the one possessing an amino group at 5-position and an aminopropyl group at 9-position. The potential as fluorescent probes were evaluated by fluorescence microscopy, using S. cerevisae as a model system of eukaryotic cells, and it was found that the benzo[a]phenoxazinium chlorides stained the cells with preferential accumulation that seems to appear at the vacuolar membrane and/or the perinuclear membrane of the endoplasmatic reticulum.Thanks are due to Fundação para a Ciência e Tecnologia (FCT) and FEDER (European Fund for Regional Development)-COMPETE-QRENEU for financial support through the research centres CQ/UM (UID/QUI/0686/2019 and UIDB/00686/2020) and CBMA (PEst-OE/BIA/UI4050/2019 and UID/BIA/04050/2020), as well as a PhD grant to J.     The synthesis of four new Nile Blue derivatives with hydrogen, propyl and/or aminopropyl groups as substituents of the amines of 5- and 9-positions is described. Photophysical properties were evaluated in acidified ethanol and aqueous solution at physiological pH.     Organic–inorganic hybrid sol–gel material loaded with an heterocyclic aldehyde with potential application for Cu(II) detection

    Organic–inorganic hybrids (OIH) are a versatile type of material that joins organic and inorganic moieties in a polymeric matrix and can be easily synthesized by the sol–gel method. OIH can be doped with chemosensors to yield an ion-sensitive polymeric matrix that can be used in the design of a more complex sensory device. Heterocyclic aldehydes show promising optical properties for supramolecular interactions with different ions through molecular recognition and changes in the optical signal. Cu(II) is a cation with biological and environmental relevance, being an appealing target for the design of chemosensors. Considering the group’s work on OIH materials and heterocyclic chromophores/fluorophores, this study reports the synthesis and characterization of an OIH based on Jeffamine THF-170 and 3-isocyanatopropyltriethoxysilane (3-ICPTES), doped with a previously synthesized heterocyclic aldehyde that features a naked-eye detection of Cu(II). Additionally, the OIH material shows color changes in the presence of Cu(II). Therefore, the obtained material shows promising properties to be used in the development of an optical fiber sensor for Cu(II) detection

    New dinitrophenyl hydrazones as colorimetric probes for anions

    Anion sensing is a dynamic research field due to biological and environmental importance of some organic or inorganic anions. Hydrazones show promising properties in the design of anion chemosensors due to the presence of proton donor and acceptor sites in their structure. In this work, two novel dinitrophenyl hydrazones, functionalized with a quinoline moiety were synthesized and characterized by spectroscopic and spectrometric techniques. The interaction between the new compounds 3a-b with different organic and inorganic anions was assessed. The two compounds showed a change of color from light yellow to magenta in the presence of H2PO4-, CH3COO-, BzO-, CN-, and F-. The interactions were analyzed by spectrophotometric titrations and the stoichiometry of the interaction was assessed by the method of continuous variation. Compound 3b showed a remarkable sensitivity to CN- with a limit of detection of 0.35 µM.     Heterocyclic Aldehydes Based on Thieno[3,2-b]thiophene Core: Synthesis and Preliminary Studies as Ion Optical Chemosensors

    Heterocyclic aldehydes show a variety of optical properties and the versatility of their reactivity allows them to yield a wide range of more complex compounds, with application in areas such as medicinal, materials and supramolecular chemistry. The biological and environmental relevance of certain molecules and ions turns them into targets for the design of molecular recognition systems. Recently, heterocyclic aldehydes have been reported in the literature as ion chemosensors. Following the group's work on optical chemosensors, for the detection and quantification of ions and molecules with environmental and medicinal relevance, this work reports the synthesis and characterization of two heterocyclic aldehydes based on the thieno[3,2-b]thiophene core, by Suzuki coupling, as well as the synthesis of the corresponding precursors. Preliminary chemosensory studies for the synthesized heterocyclic aldehydes in the presence of selected cations were also performed, in solution, in order to determine their potential application as optical chemosensors

    PDMS based hybrid sol-gel materials for sensing applications in alkaline environments: synthesis and characterization

    Nowadays, concrete degradation is a major problem in the civil engineering field. Concrete carbonation, one of the main sources of structures’ degradation, causes concrete’s pH to decrease; hence, enabling the necessary conditions for corrosion reinforcement. An accurate, non-destructive sensor able to monitor the pH decrease resistant to concrete conditions is envisaged by many researchers. Optical fibre sensors (OFS) are generally used for concrete applications due to their high sensitivity and resistance to external interferences. Organic-inorganic hybrid (OIH) films, for potential functionalization of OFS to be applied in concrete structures, were developed. Polydimethylsiloxane (PDMS) based sol-gel materials were synthesized by the formation of an amino alcohol precursor followed by hydrolysis and condensation. Different ratios between PDMS and (3-aminopropyl)triethoxysilane (3-APTES) were studied. The synthesized OIH films were characterized by Fourier-transformed infrared spectroscopy (FTIR), UV–Vis spectroscopy, electrochemical impedance spectroscopy (EIS) and thermogravimetric analysis (TGA). The OIH films were doped with phenolphthalein (Phph), a pH indicator, and were characterized by UV–Vis and EIS. FTIR characterization showed that the reaction between both precursors, the hydrolysis and the condensation reactions occurred successfully. UV–Vis characterization confirmed the presence of Phph embedded in the OIH matrices.     Combined use of chitosan and olfactory mucosa mesenchymal stem/stromal cells to promote peripheral nerve regeneration in vivo

    Peripheral nerve injury remains a clinical challenge with severe physiological and functional consequences. Despite the existence of multiple possible therapeutic approaches, until now, there is no consensus regarding the advantages of each option or the best methodology in promoting nerve regeneration. Regenerative medicine is a promise to overcome this medical limitation, and in this work, chitosan nerve guide conduits and olfactory mucosa mesenchymal stem/stromal cells were applied in different therapeutic combinations to promote regeneration in sciatic nerves after neurotmesis injury. Over 20 weeks, the intervened animals were subjected to a regular functional assessment (determination of motor performance, nociception, and sciatic indexes), and after this period, they were evaluated kinematically and the sciatic nerves and cranial tibial muscles were evaluated stereologically and histomorphometrically, respectively. The results obtained allowed confirming the beneficial effects of using these therapeutic approaches. The use of chitosan NGCs and cells resulted in better motor performance, better sciatic indexes, and lower gait dysfunction after 20 weeks. The use of only NGGs demonstrated better nociceptive recoveries. The stereological evaluation of the sciatic nerve revealed identical values in the different parameters for all therapeutic groups. In the muscle histomorphometric evaluation, the groups treated with NGCs and cells showed results close to those of the group that received traditional sutures, the one with the best final values. The therapeutic combinations studied show promising outcomes and should be the target of new future works to overcome some irregularities found in the results and establish the combination of nerve guidance conduits and olfactory mucosa mesenchymal stem/stromal cells as viable options in the treatment of peripheral nerves after injury.info:eu-repo/semantics/publishedVersio

    Serum total and bone alkaline phosphatase levels and their correlation with serum minerals over the lifespan of sheep

    This study aimed to assess serum total alkaline phosphatase (ALP) and its bone isoform (BALP) levels during the ageing and in different physiologic states of sheep, in order to expand the knowledge about the variation of these biomarkers over the sheep lifespan. Ninety female sheep were divided into nine groups of various ages and physiological states (dry, lactation and pregnancy). Serum ALP, BALP and mineral levels were determined by commercial immunoassay, molecular absorbance spectrophotometry and chemical luminescence for BALP determination. Serum ALP and BALP decreased as sheep aged, and no statistically significant differences were obtained between ewes in different physiologic states. The continuous decline of serum BALP concentration along the sheep lifespan, namely in mature and old sheep, is a sign of decreasing bone turnover associated with ageing. Serum calcium concentrations increased slightly until 2 years of age and then showed a tenuous but statistically significant decrease in mature sheep, while serum phosphorus maintained an uninterrupted decrease as sheep matured. The knowledge of serum values of bone biomarkers throughout the sheep lifespan may be useful in preclinical orthopaedic research studies and for animal science studies using sheep.Cristina P.     Primary B-Cell Deficiencies Reveal a Link between Human IL-17-Producing CD4 T-Cell Homeostasis and B-Cell Differentiation

    IL-17 is a pro-inflammatory cytokine implicated in autoimmune and inflammatory conditions. The development/survival of IL-17-producing CD4 T cells (Th17) share critical cues with B-cell differentiation and the circulating follicular T helper subset was recently shown to be enriched in Th17 cells able to help B-cell differentiation. We investigated a putative link between Th17-cell homeostasis and B cells by studying the Th17-cell compartment in primary B-cell immunodeficiencies. Common Variable Immunodeficiency Disorders (CVID), defined by defects in B-cell differentiation into plasma and memory B cells, are frequently associated with autoimmune and inflammatory manifestations but we found no relationship between these and Th17-cell frequency. In fact, CVID patients showed a decrease in Th17-cell frequency in parallel with the expansion of activated non-differentiated B cells (CD21lowCD38low). Moreover, Congenital Agammaglobulinemia patients, lacking B cells due to impaired early B-cell development, had a severe reduction of circulating Th17 cells. Finally, we found a direct correlation in healthy individuals between circulating Th17-cell frequency and both switched-memory B cells and serum BAFF levels, a crucial cytokine for B-cell survival. Overall, our data support a relationship between Th17-cell homeostasis and B-cell maturation, with implications for the understanding of the pathogenesis of inflammatory/autoimmune diseases and the physiology of B-cell depleting therapies

    Bone turnover markers in sheep and goat: a review of the scientific literature

    Bone turnover markers (BTMs) are product of bone cell activity and are generally divided in bone formation and bone resorption markers. The purpose of this review was to structure the available information on the use of BTMs in studies on small ruminants, especially for monitoring their variations related to diet, exercise, gestation and metabolic lactation state, circadian and seasonal variations, and also during skeletal growth. Pre-clinical and translational studies using BTMs with sheep and goats as animal models in orthopaedic research studies to help in the evaluation of the fracture healing process and osteoporosis research are also described in this review. The available information from the reviewed studies was systematically organized in order to highlight the most promising BTMs in small ruminant research, as well as provide a wide view of the use of sheep and goat as animal models in orthopaedic research, type of markers and commercial assay kits with cross-reactivity in sheep and goat, method of sample and storage of serum and urine for bone turnover markers determination and the usefulness and limitations of bone turnover markers in the different studies, therefore an effective tool for researchers that seek answers to different questions while using BTMs in small ruminants.José Arthur de A.
