    Characterization of polychlorinated biphenyls in surface sediments of the North End Lake, Port Elizabeth, South Africa

    The distribution and concentrations of 6 indicator polychlorinated biphenyl (PCB) congeners, nos. 28, 52, 101, 138, 153 and 180, were determined in surface  sediments from the North End Lake in Port Elizabeth, South Africa. Forty-two  surficial sediment samples were collected from different locations covering the  region that receives the majority of the industrial waste, urban effluents and runoff  and thus expected to be contaminated with different degrees of contamination. The analysis was achieved by gas chromatography-mass spectrometry (GC/MS) using  the internal standard method. The total PCB concentrations in the samples ranged from 1.60 to 3.06 ng·g-1 dry weight (dw). The concentrations of congener profiles showed significant differences. Generally, the highest PCB concentrations were  associated with high organic matter contents and small grain size. The highly  chlorinated PCBs dominated with regards to the levels in sediments. PCB 138 was the major contributor to the total PCBs and was detected at 100% of sites. This study provided a snapshot of the PCB contamination status in the North End Lake  sediments, and allowed for a comparison between the investigated system and other systems worldwide.Keywords: PCBs, sediments, organic matter, particle size, North End Lak

    Distribution of polychlorinated biphenyl residues in several tissues of fish from the North End Lake, Port Elizabeth, South Africa

    The concentrations and distribution of 6 PCB indicator congeners (IUPAC nos. 28, 52, 101, 138, 153, and 180) were measured in 236 organ samples of fish (Cyprinus carpio and Oreochromis mossambicus) from the North End Lake in Port Elizabeth, South Africa. Polychlorinated biphenyls (PCBs) were extracted from the fish muscles, gills, gonads and livers using USEPA method 8082, followed by a clean-up using concentrated sulphuric acid and florisil column chromatography. Analysis was achieved by gas chromatography–mass spectrometry (GC/MS) using the internal standard method. The concentrations of total PCBs in the liver, gonads, gills and muscle were 95.69, 57.49, 44.63, 34.14 ng∙g-1 lipid weight (lw) in C. carpio and 119.73, 59.21, 49.78, 34.63 ng∙g-1 (lw) in O. mossambicus, respectively. These values were relatively low compared to those reported in the literature. PCB levels were predictably highest in the lipid-rich livers. Individual congeners were not distributed homogeneously within the investigated organs. PCBs 153 and 138 were present at higher concentrations than other PCB congeners for both species. PCB contaminants in fish act as indicators of pollution in aquatic ecosystems and are a potential threat to human health when consumed.Keywords: PCBs, tissues, fish, GC/MS, biomagnification, North End Lak

    Congener profiles of polychlorinated biphenyls and the effect on marine mussels at an outfall site, Port Elizabeth, South Africa

    This study investigated the effect of freshwater as a point source of PCBs contributing to a marine outfall region. Inflowing and outflowing water from the North End Lake, Port Elizabeth, was collected to determine the contamination levels of polychlorinated biphenyls (PCBs). Mussels at the outflow to the sea were also sampled. The samples were analysed by an internal standard method for 6 indicator PCB congeners using gas chromatography/mass spectrometry (GC/MS) in selected ion monitoring (SIM) mode. The total PCB concentrations in the water (dissolved plus particulate phases) ranged from 0.180 to 0.355 ng∙L-1 and from 20.84 to 31.34 ng∙g-1 wet weight (ww) in mussels. Lighter PCB congeners exhibited highest concentrations in the water samples while heavier PCBs were dominant in mussels. PCB-52 was the most abundant in the water samples while PCB-153 was abundant in mussels. To protect human health from the possible effects of eating fish and shellfish contaminated with PCBs, the Environmental Protection Agency regulates that the level of PCBs in water be no greater than 0.17 ng∙L-1 of water. The levels of PCBs in water from the North End Lake were found to be high (0.180 – 0.355 ng∙L-1) compared to the levels recommended by EPA.Keywords: dissolved phase, GC/MS, North End Lake, particulate phase, sedentary organism

    The Earliest Post-Paleozoic Freshwater Bivalves Preserved in Coprolites from the Karoo Basin, South Africa

    Background: Several clades of bivalve molluscs have invaded freshwaters at various times throughout Phanerozoic history. The most successful freshwater clade in the modern world is the Unionoida. Unionoids arose in the Triassic Period, sometime after the major extinction event at the End-Permian boundary and are now widely distributed across all continents except Antarctica. Until now, no freshwater bivalves of any kind were known to exist in the Early Triassic. Principal Findings: Here we report on a faunule of two small freshwater bivalve species preserved in vertebrate coprolites from the Olenekian (Lower Triassic) of the Burgersdorp Formation of the Karoo Basin, South Africa. Positive identification of these bivalves is not possible due to the limited material. Nevertheless they do show similarities with Unionoida although they fall below the size range of extant unionoids. Phylogenetic analysis is not possible with such limited material and consequently the assignment remains somewhat speculative. Conclusions: Bivalve molluscs re-invaded freshwaters soon after the End-Permian extinction event, during the earliest part of the recovery phase during the Olenekian Stage of the Early Triassic. If the specimens do represent unionoids then these Early Triassic examples may be an example of the Lilliput effect. Since the oldest incontrovertible freshwater unionoids are also from sub-Saharan Africa, it is possible that this subcontinent hosted the initial freshwater radiation of the Unionoida.     Genes left behind: Climate change threatens cryptic genetic diversity in the canopy-forming seaweed bifurcaria bifurcata

    The global redistribution of biodiversity will intensify in the coming decades of climate change, making projections of species range shifts and of associated genetic losses important components of conservation planning. Highly-structured marine species, notably brown seaweeds, often harbor unique genetic variation at warmer low-latitude rear edges and thus are of particular concern. Here, a combination of Ecological Niche Models (ENMs) and molecular data is used to forecast the potential near-future impacts of climate change for a warm-temperate, canopy forming seaweed, Bifurcaria bifurcata. ENMs for B. bifurcata were developed using marine and terrestrial climatic variables, and its range projected for 2040-50 and 2090-2100 under two greenhouse emission scenarios. Geographical patterns of genetic diversity were assessed by screening 18 populations spawning the entire distribution for two organelle genes and 6 microsatellite markers. The southern limit of B. bifurcata was predicted to shift northwards to central Morocco by the mid-century. By 2090-2100, depending on the emission scenario, it could either retreat further north to western Iberia or be relocated back to Western Sahara. At the opposing margin, B. bifurcata was predicted to expand its range to Scotland or even Norway. Microsatellite diversity and endemism were highest in Morocco, where a unique and very restricted lineage was also identified. Our results imply that B. bifurcata will maintain a relatively broad latitudinal distribution. Although its persistence is not threatened, the predicted extirpation of a unique southern lineage or even the entire Moroccan diversity hotspot will erase a rich evolutionary legacy and shrink global diversity to current (low) European levels.     The Osteology of the Basal Archosauromorph Tasmaniosaurus triassicus from the Lower Triassic of Tasmania, Australia

    Proterosuchidae are the most taxonomically diverse archosauromorph reptiles sampled in the immediate aftermath of the Permo-Triassic mass extinction and represent the earliest radiation of Archosauriformes (archosaurs and closely related species). Proterosuchids are potentially represented by approximately 15 nominal species collected from South Africa, China, Russia, Australia and India, but the taxonomic content of the group is currently in a state of flux because of the poor anatomic and systematic information available for several of its putative members. Here, the putative proterosuchid Tasmaniosaurus triassicus from the Lower Triassic of Hobart, Tasmania (Australia),is redescribed. The holotype and currently only known specimen includes cranial and postcranial remains and the revision of this material sheds new light on the anatomy of the animal, including new data on the cranial endocast. Several bones are re-identified or reinterpreted, contrasting with the descriptions of previous authors. The new information provided here shows that Tasmaniosaurus closely resembles the South African proterosuchid Proterosuchus, but it differed in the presence of, for example, a slightly downturned premaxilla, a shorter anterior process of maxilla, and a diamond-shaped anterior end of interclavicle. Previous claims for the presence of gut contents in the holotype of Tasmaniosaurus are considered ambiguous. The description of the cranial endocast of Tasmaniosaurus provides for the first time information about the anatomy of this region in proterosuchids. The cranial endocast preserves possibly part of the vomero-nasal (= Jacobson's) system laterally to the olfactory bulbs. Previous claims of the absence of the vomero-nasal organs in archosaurs, which is suggested by the extant phylogenetic bracket, are questioned because its absence in both clades of extant archosaurs seems to be directly related with the independent acquisition of a non-ground living mode of life

    The Sail-Backed Reptile Ctenosauriscus from the Latest Early Triassic of Germany and the Timing and Biogeography of the Early Archosaur Radiation

    Background Archosaurs (birds, crocodilians and their extinct relatives including dinosaurs) dominated Mesozoic continental ecosystems from the Late Triassic onwards, and still form a major component of modern ecosystems (>10,000 species). The earliest diverse archosaur faunal assemblages are known from the Middle Triassic (c. 244 Ma), implying that the archosaur radiation began in the Early Triassic (252.3–247.2 Ma). Understanding of this radiation is currently limited by the poor early fossil record of the group in terms of skeletal remains. Methodology/Principal Findings We redescribe the anatomy and stratigraphic position of the type specimen of Ctenosauriscus koeneni (Huene), a sail-backed reptile from the Early Triassic (late Olenekian) Solling Formation of northern Germany that potentially represents the oldest known archosaur. We critically discuss previous biomechanical work on the ‘sail’ of Ctenosauriscus, which is formed by a series of elongated neural spines. In addition, we describe Ctenosauriscus-like postcranial material from the earliest Middle Triassic (early Anisian) Röt Formation of Waldhaus, southwestern Germany. Finally, we review the spatial and temporal distribution of the earliest archosaur fossils and their implications for understanding the dynamics of the archosaur radiation. Conclusions/Significance Comprehensive numerical phylogenetic analyses demonstrate that both Ctenosauriscus and the Waldhaus taxon are members of a monophyletic grouping of poposauroid archosaurs, Ctenosauriscidae, characterised by greatly elongated neural spines in the posterior cervical to anterior caudal vertebrae. The earliest archosaurs, including Ctenosauriscus, appear in the body fossil record just prior to the Olenekian/Anisian boundary (c. 248 Ma), less than 5 million years after the Permian–Triassic mass extinction. These earliest archosaur assemblages are dominated by ctenosauriscids, which were broadly distributed across northern Pangea and which appear to have been the first global radiation of archosaurs

    Contrasting and congruent patterns of genetic structuring in two Microtus vole species using museum specimens

    The common vole (Microtus arvalis) and the field vole (Microtus agrestis) are morphologically similar species but are ecological distinctive and differ in the details of their evolutionary history as revealed by mitochondrial DNA (mtDNA). The aim of this study is to describe patterns of genetic variability using microsatellite markers in populations of the common and field vole in Poland using museum specimens, to assess the degree of congruence with mtDNA variation and thereby determine the factors that influence current patterns of gene flow. We genotyped 190 individuals of the common vole at 11 loci and 190 individuals of the field vole at 13 loci. Overall differentiation based on F ST was higher for the common vole than in the field vole. We detected a significant isolation by distance pattern for both species. Bayesian analysis in STRUCTURE identified Eastern and Western geographic groups in Poland based on microsatellites for both species. The location of river barriers is likely to be the main factor in these partitions. The eastern-western subdivision with microsatellites does not coincide with the distribution of mtDNA lineages for either species. Unlike previous studies in the common and field vole elsewhere in Europe, we found no evidence of reproductive isolation between the mtDNA lineages of these species at their contact zones in Poland.
