Selective targeting of neuroblastoma tumour-initiating cells by compounds identified in stem cell-based small molecule screens
Neuroblastoma (NB) is the most deadly extra-cranial solid tumour in children necessitating an urgent need for effective and less toxic treatments. One reason for the lack of efficacious treatments may be the inability of existing drugs to target the tumour-initiating or cancer stem cell population responsible for sustaining tumour growth, metastases and relapse. Here, we describe a strategy to identify compounds that selectively target patient-derived cancer stem cell-like tumour-initiating cells (TICs) while sparing normal paediatric stem cells (skin-derived precursors, SKPs) and characterize two therapeutic candidates. DECA-14 and rapamycin were identified as NB TIC-selective agents. Both compounds induced TIC death at nanomolar concentrations in vitro, significantly reduced NB xenograft tumour weight in vivo, and dramatically decreased self-renewal or tumour-initiation capacity in treated tumours. These results demonstrate that differential drug sensitivities between TICs and normal paediatric stem cells can be exploited to identify novel, patient-specific and potentially less toxic therapies
Chemotherapy-Induced Neuronal Maturation in Sinonasal Teratocarcinosarcomaâa Unique Observation
Sinonasal teratocarcinosarcoma (SNTCS) is a rare and highly malignant tumour with combined features of a teratoma and carcinosarcoma. We report the first case of a SNTCS in 23Â year old male treated with neo-adjuvant chemotherapy followed by cranio-facial resection. The resection specimen displayed cellular maturation in the neuroectodermal component. The patient presented with a short history of nasal obstruction, epistaxis and headache. On imaging, a bone destroying lesion of left paranasal sinuses and nasal cavity was identified. The diagnosis of SNTCS could be offered only on the third biopsy which showed heterogeneous admixture of primitive neuroectodermal, epithelial and mesenchymal elements. An adequate sampling with high index of suspicion is needed to catch hold this rare tumor. Tumor was excised after 4 cycles of neo-adjuvant chemotherapy. On microscopic examination, it showed similar epithelial and mesenchymal components as the pretreatment biopsies. However, the primitive neuroectodermal component displayed extensive neuronal maturation. The undifferentiated neuroectodermal cells were completely absent in the post chemotherapy specimen. This case throws light on the morphologic evidence of chemotherapy induced maturation in the neuroectodermal component within SNTCS, an event hitherto not reported in the literature in case of SNTCS

We herein present a patient with delayed-type allergic hypersensitivity against prilocaine leading to spreading eczematous dermatitis after subcutaneous injections for local anesthesia with prilocaine. Prilocaine allergy was proven by positive skin testing and subcutaneous provocation, whereas the evaluation of other local anesthetics - among them lidocaine, articaine and mepivacaine - did not exhibit any evidence for cross-reactivity

Eukaryotic Cells Producing Ribosomes Deficient in Rpl1 Are Hypersensitive to Defects in the Ubiquitin-Proteasome System
It has recently become clear that the misassembly of ribosomes in eukaryotic cells can have deleterious effects that go far beyond a simple shortage of ribosomes. In this work we find that cells deficient in ribosomal protein L1 (Rpl1; Rpl10a in mammals) produce ribosomes lacking Rpl1 that are exported to the cytoplasm and that can be incorporated into polyribosomes. The presence of such defective ribosomes leads to slow growth and appears to render the cells hypersensitive to lesions in the ubiquitin-proteasome system. Several genes that were reasonable candidates for degradation of 60S subunits lacking Rpl1 fail to do so, suggesting that key players in the surveillance of ribosomal subunits remain to be found. Interestingly, in spite of rendering the cells hypersensitive to the proteasome inhibitor MG132, shortage of Rpl1 partially suppresses the stress-invoked temporary repression of ribosome synthesis caused by MG132.United States. Reciprocal Prospective Relationships Between Loneliness and Weight Status in Late Childhood and Early Adolescence
Adolescents who do not conform to weight ideals are vulnerable to disapproval and victimization from peers in school. But, missing from the literature is a prospective examination of weight status and feelings of loneliness that might come from those experiences. Using data from the QuĂŠbec Longitudinal Study of Child Development, we filled that gap by examining the prospective associations between loneliness and weight status when the sample was aged 10 to 13 years. At ages 10, 12, and 13 years, 1042 youth (572 females; 92% from French speaking homes) reported on their loneliness and were weighed and measured. Family income sufficiency was included in our analyses given its relationship with weight status, but also its possible link with loneliness during early adolescence. The findings showed that (1) weight status and loneliness were not associated concurrently; (2) weight status predicted increases in loneliness from ages 12 to 13 years; and (3) loneliness predicted increases in weight from ages 12 to 13 years among female adolescents, but weight loss among male adolescents. The fact that loneliness was involved in weight gain for females suggests that interventions focused on reducing loneliness and increasing connection with peers during early adolescence could help in reducing obesity
Pervasiveness of Parasites in Pollinators
Many pollinator populations are declining, with large economic and ecological
implications. Parasites are known to be an important factor in the some of the
population declines of honey bees and bumblebees, but little is known about the
parasites afflicting most other pollinators, or the extent of interspecific
transmission or vectoring of parasites. Here we carry out a preliminary
screening of pollinators (honey bees, five species of bumblebee, three species
of wasp, four species of hoverfly and three genera of other bees) in the UK for
parasites. We used molecular methods to screen for six honey bee viruses,
Ascosphaera fungi, Microsporidia, and
Wolbachia intracellular bacteria. We aimed simply to detect
the presence of the parasites, encompassing vectoring as well as actual
infections. Many pollinators of all types were positive for
Ascosphaera fungi, while Microsporidia were rarer, being
most frequently found in bumblebees. We also detected that most pollinators were
positive for Wolbachia, most probably indicating infection with
this intracellular symbiont, and raising the possibility that it may be an
important factor in influencing host sex ratios or fitness in a diversity of
pollinators. Importantly, we found that about a third of bumblebees
(Bombus pascuorum and Bombus terrestris)
and a third of wasps (Vespula vulgaris), as well as all honey
bees, were positive for deformed wing virus, but that this virus was not present
in other pollinators. Deformed wing virus therefore does not appear to be a
general parasite of pollinators, but does interact significantly with at least
three species of bumblebee and wasp. Further work is needed to establish the
identity of some of the parasites, their spatiotemporal variation, and whether
they are infecting the various pollinator species or being vectored. However,
these results provide a first insight into the diversity, and potential
exchange, of parasites in pollinator communities
