    Non-alcoholic fatty liver disease (NAFLD) is the most common liver disease worldwide, and its prevalence is reaching epidemic characteristics both in adults and in children. The increase of NAFLD prevalence parallels that of obesity, now representing the major cause of liver inflammation, increasing the risk of cirrhosis and hepatocarcinoma. Furthermore, NAFLD is a risk factor for cardiovascular diseases and type 2 diabetes, two of the major leading causes of morbidity and mortality in western countries. Thus a significant amount of studies have dealt with the evaluation of the possible molecular mechanisms leading to NAFLD and its inflammatory consequences within the liver, the non-alcoholic steatohepatitis, and cirrhosis. The inflammasome is a key player in the inflammation and fibrogenic responses in many different tissues, including the liver. The activation of the NLRP3 inflammasome requires the activation by extracellular adenosine tri-phosphate (ATP) of a specific purinergic receptor named P2X7 located in the target cells, although other pathways have been described. To this regard, extracellular ATP acts as an internal danger signal coming from damaged cells participating in the activation of the inflammatory process, a signaling pathway common to many different tissues. Here, we briefly review the involvement of the P2X7 receptor/inflammasome NLRP3 axis in the pathophysiological events leading to NAFLD and its inflammatory and fibrotic evolutions, reporting the possible therapeutical strategies targeting the P2X7 receptor/NLRP3 inflammasome

    Supercritical Extraction from Vinification Residues: Fatty Acids, α-Tocopherol, and Phenolic Compounds in the Oil Seeds from Different Varieties of Grape

    Supercritical fluid extraction has been widely employed in the extraction of high purity substances. In this study, we used the technology to obtain oil from seeds from a variety of grapes, from vinification residues generated in the Southern region of the state of Rio Grande do Sul, Brazil. This work encompasses three varieties of Vitis vinifera (Moscato Giallo, Merlot, and Cabernet Sauvignon) and two of Vitis labrusca (Bordô e Isabel), harvested in 2005 and 2006. We obtained the highest oil content from Bordô (15.40%) in 2005 and from Merlot (14.66%), 2006. The biggest concentration of palmitic, stearic, and linoleic acids was observed in Bordô, 2005, and in Bordô, Merlot, and Moscato Giallo, 2006. Bordô showed the highest concentration of oleic acid and α-tocopherol in both seasons too. For the equivalent of procyanidins, we did not notice significant difference among the varieties from the 2005 harvest. In 2006, both varieties Isabel and Cabernet Sauvignon showed a value slightly lower than the other varieties. The concentration of total phenolics was higher in Bordô and Cabernet Sauvignon. The presence of these substances is related to several important pharmacological properties and might be an alternative to conventional processes to obtain these bioactives

    Stability Analysis of a Landslide Scarp by Means of Virtual Outcrops: The Mt. Peron Niche Area (Masiere di Vedana Rock Avalanche, Eastern Southern Alps)

    We investigated the Mt. Peron niche area of the Masiere di Vedana rock avalanche (BL), one of the major mass movements that affected the Eastern Southern Alps in historical times. So far, a geomechanical characterization and a stability analysis of the niche area, where potential rockfall sources are present, are lacking. The Mt. Peron niche area is a rocky cliff almost inaccessible to field-based measurements. In order to overcome this issue, we performed a geo-structural characterization of a sector of the cliff by means of a UAV-based photogrammetric survey. From the virtual outcrop, we extracted the orientation of 159 fractures that were divided into sets based on a K-means clustering algorithm and field-checked with some measurements collected along a rappelling descent route down to the cliff. Finally, with the aim of evaluating the stability of the volume under investigation, we performed a stability analysis of three rock pillars included in our survey by means of a distinct element numerical simulation. Our results indicate that two out of the three pillars are characterized by a stable state, under the simulation assumptions, whereas the third is close to failure, and for this reason, its condition needs further investigation

    Formas de fósforo no tecido de plantas anuais de inverno cultivadas em Latossolo Vermelho sob diferentes sistemas de manejo de solo

    As plantas de cobertura propiciam a proteção e a reciclagem de nutrientes do solo, como o fósforo (P). Contudo, o teor e as formas de acúmulo de P podem variar entre espécies e sistemas de manejo de solo. O objetivo do trabalho foi avaliar as formas de acúmulo de P no tecido vegetal de plantas anuais de inverno, manejadas sob Sistema Plantio Direto (SPD) e Cultivo Convencional (SCC). O experimento foi instalado em 1986 no Instituto Agronômico do Paraná, em Pato Branco-PR, sobre um Latossolo Vermelho aluminoférrico. Ele se baseia no uso de seis plantas anuais de inverno (trigo, aveia branca, centeio, ervilhaca, nabo forrageiro e tremoço azul) que foram manejadas sob SPD e SCC. Em maio de 2011 as espécies foram semeadas, sem o uso de fertilizantes, e em setembro de 2011 efetuou-se a coleta da parte aérea das plantas. No Laboratório de Química e Fertilidade do Solo da UFSM realizou-se o fracionamento de P do tecido vegetal, além da determinação dos teores de carbono total, nitrogênio total e P disponível do solo. A partir dos dados obtidos, conclui-se que as formas de acúmulo de P no tecido das plantas anuais de inverno variam em função da espécie e da disponibilidade de P no ambiente; para todas as plantas anuais de inverno o P inorgânico solúvel é a principal forma de acúmulo de P no tecido vegetal, principalmente quando são cultivadas em solos com maior disponibilidade de P, como sob SPD; Em ambientes com baixa disponibilidade de P, plantas anuais de inverno tendem a diminuir as formas solúveis de P e, ao mesmo tempo, acumular formas orgânicas estáveis, principalmente na forma de Prna e Plip

    O sistema de manejo pode afetar a disponibilidade de nutrientes para as plantas, em decorrência do sistema de rotação de culturas e da quantidade de resíduo vegetal presente na superfície do solo. O objetivo deste trabalho foi avaliar a quantidade de nitrogênio (N) e fósforo (P) acumulados no tecido vegetal de aveia preta, trigo, centeio, ervilhaca, nabo e tremoço azul, além de verificar seu estado nutricional de N e P. O experimento esta instalado no Instituto Agronômico do Paraná (IAPAR) em Pato Branco, Paraná, em um Latossolo Vermelho aluminoférrico. Em maio de 2011 os tratamentos foram semeados, sendo seis plantas de cobertura: trigo, aveia preta, centeio, ervilhaca, nabo forrageiro e tremoço azul cultivados sob Sistema Plantio Direto (SPD) e Sistema de Cultivo Convencional (SCC). Em setembro de 2011efetuou-se a coleta do material verde da parte aérea, o qual foi seco a 65ºC e posteriormente digerido com uma mistura de H2SO4 e H2O2, onde foram determinados os teores de nitrogênio total (Ntotal) e fósforo total (Ptotal), também foi quantificado o teor de carbono das amostras, e por fim calculou-se o índice de nutrição de fósforo (P índex) e de nitrogênio (N índex). Realizou-se a comparação das médias das plantas anuais de inverno e dos sistemas de manejo de solo por meio do teste de SCOTT-KNOTT. O sistema de cultivo influencia no acúmulo de N e P, consequentemente no índice de nutrição dos mesmos, sendo maior no SPD. A ervilhaca e centeio possuem grande capacidade de reciclar P

    Reproducibility Analysis Of The Stability And Treatment Of Vertebral Metastatic Lesions

    Objectives: To investigate the reproducibility among spine surgeons in defining the treatment of vertebral metastatic lesions, taking into account the mechanical stability of injuries. Methods: Twenty cases of isolated vertebral metastatic lesions were presented to ten experts. Their opinion was then asked about the stability of the lesion, as well as their treatment option. Results: The interobserver Kappa coefficient obtained both for stability analysis as to the decision of the treatment was poor (0.334 and 0.248, respectively).     Nicotine modulates the long-lasting storage of fear memory

    Late post-training activation of the ventral tegmental area (VTA)–hippocampus dopaminergic loop controls the entry of information into long-term memory (LTM). Nicotinic acetylcholine receptors (nAChR) modulate VTA function, but their involvement in LTM storage is unknown. Using pharmacological and behavioral tools, we found that α7-nAChR-mediated cholinergic interactions between the pedunculopontine tegmental nucleus and the medial prefrontal cortex modulate the duration of fear-motivated memories, maybe by regulating the activation state of VTA–hippocampus dopamine connections.Fil: Lima, Ramón H..
