Comparing carbon isotope composition of bulk wood and holocellulose fromQuercus cerris,Fraxinus ornusandPinus radiatatree rings
Tree-ring \u3b413C is widely employed in ecophysiological studies, because it represents an integrated proxy of the ratio between photosynthesis (A) and stomatal conductance (g), which expresses the intrinsic water use efficiency (iWUE), strongly affected by the environmental conditions experienced by the plant during its life span. Tree-ring \u3b413C also reflects long term variations of atmospheric CO2 concentration and of its carbon isotope composition, partly due to increasing anthropogenic emissions. Carbon isotope abundances in tree rings can be assessed on bulk wood as well as on wood biochemical components, wich show different \u3b413C values because of secondary discrimination during biosynthesis. We present the results of a comparison between \u3b413C values of bulk wood and holocellulose samples obtained from the last three (1999, 2000 and 2001) annual growth rings of two hardwood (Quercus cerris L. and Fraxinus ornus L. and one conifer (Pinus radiata D. Don, species. We found that 13C values differed significantly among tree species, both in the case of holocellulose and bulk wood, but only in the case of P. radiata bulk wood samples tend to provide more negative \u3b413C values than holocellulose, as reported in the literature. We suggest that, at least for the two hardwood species studied, bulk wood is a suitable material to work with for \u3b413C assessment, whilst in P. radiata holocellulose could provide a more stable and reliable index, when studying plant ecophysiological responses to changing environmental conditions

Role of glucose as a modulator of anabolic and catabolic gene expression in normal and osteoarthritic human chondrocytes
Cartilage matrix homeostasis involves a dynamic balance between numerous signals that modulate chondrocyte functions. This study aimed at elucidating the role of the extracellular glucose concentration in modulating anabolic and catabolic gene expression in normal and osteoarthritic (OA) human chondrocytes and its ability to modify the gene expression responses induced by pro-anabolic stimuli, namely Transforming Growth Factor-β (TGF). For this, we analyzed by real time RT-PCR the expression of articular cartilage matrix-specific and non-specific genes, namely collagen types II and I, respectively. The expression of the matrix metalloproteinases (MMPs)-1 and -13, which plays a major role in cartilage degradation in arthritic conditions, and of their tissue inhibitors (TIMP) was also measured. The results showed that exposure to high glucose (30 mM) increased the mRNA levels of both MMPs in OA chondrocytes, whereas in normal ones only MMP-1 increased. Collagen II mRNA was similarly increased in normal and OA chondrocytes, but the increase lasted longer in the later. Exposure to high glucose for 24 h prevented TGF-induced downregulation of MMP-13 gene expression in normal and OA chondrocytes, while the inhibitory effect of TGF on MMP-1 expression was only partially reduced. Other responses were not significantly modified. In conclusion, exposure of human chondrocytes to high glucose, as occurs in vivo in diabetes mellitus patients and in vitro for the production of engineered cartilage, favors the chondrocyte catabolic program. This may promote articular cartilage degradation, facilitating OA development and/or progression, as well as compromise the quality and consequent in vivo efficacy of tissue engineered cartilage
Differential prefrontal-like deficit in children after cerebellar astrocytoma and medulloblastoma tumor
Background: This study was realized thanks to the collaboration of children and adolescents who had been resected from cerebellar tumors. The medulloblastoma group (CE+, n = 7) in addition to surgery received radiation and chemotherapy. The astrocytoma group (CE, n = 13) did not receive additional treatments. Each clinical group was compared in their executive functioning with a paired control group (n = 12). The performances of the clinical groups with respect to controls were compared considering the tumor's localization (vermis or hemisphere) and the affectation (or not) of the dentate nucleus. Executive variables were correlated with the age at surgery, the time between surgery-evaluation and the resected volume.
Methods: The executive functioning was assessed by means of WCST, Complex Rey Figure, Controlled Oral Word Association Test (letter and animal categories), Digits span (WISC-R verbal scale) and Stroop test. These tests are very sensitive to dorsolateral PFC and/or to medial frontal cortex functions. The scores for the non-verbal Raven IQ were also obtained. Direct scores were corrected by age and transformed in standard scores using normative data. The neuropsychological evaluation was made at 3.25 (SD = 2.74) years from surgery in CE group and at 6.47 (SD = 2.77) in CE+ group.
Results: The Medulloblastoma group showed severe executive deficit (</= 1.5 SD below normal mean) in all assessed tests, the most severe occurring in vermal patients. The Astrocytoma group also showed executive deficits in digits span, semantic fluency (animal category) and moderate to slight deficit in Stroop (word and colour) tests. In the astrocytoma group, the tumor's localization and dentate affectation showed different profile and level of impairment: moderate to slight for vermal and hemispheric patients respectively. The resected volume, age at surgery and the time between surgery-evaluation correlated with some neuropsychological executive variables.
Conclusion: Results suggest a differential prefrontal-like deficit due to cerebellar lesions and/or cerebellar-frontal diaschisis, as indicate the results in astrocytoma group (without treatments), that also can be generated and/or increased by treatments in the medulloblastoma group. The need for differential rehabilitation strategies for specific clinical groups is remarked. The results are also discussed in the context of the Cerebellar Cognitive Affective Syndrome.4:18; pags: 1-16

Recovery of electronic wastes as fillers for electromagnetic shielding in building components: an LCA study
The present study reports the development of sandwich panels for building walls having electromagnetic interference (EMI) shielding abilities. Conductive polymer composites (CPCs) have started being employed as EMI shielding materials. In this paper we propose the use of a conductive polymer composite flat sheet made of high-density polyethylene (HDPE) recovered from municipal solid wastes (MSW) used as polymeric matrix, “doped” with dispersed metal fillers recycled from e-wastes. Test results proved that the recycled metal fillers enhance the electrical conductivity and enable EMI shielding. Different sandwich panels were discussed in the context of building applications, using identical HDPE/metal-filler EMI sheets, but different thermal insulation material (polystyrene and glass wool). The life cycle assessment (LCA) methodology was applied to evaluate the environmental impact generated during the following steps: a) recycling of thermoplastic materials from MSW; b) recovering of metallic components from waste PCB; c) re-use of the recovered components into sandwich panels with electromagnetic shielding properties for buildings. The goal of the LCA was to perform a comparative analysis of the composite sandwich structures manufactured to be used as EMI shielding in buildings applications in order to assist the materials selection and eco-design. By means of the LCA results it was possible to manufacture a building component with good EMI shielding properties and reduced environmental impact
Rabies virus in a pregnant naturally infected southern yellow bat (Lasiurus ega)
Current knowledge on bat lyssavirus infections in their native hosts is limited and little is known about the virulence, virus dissemination and transmission among free-living insectivorous bats. The present study is a brief description of rabies virus (RABV) dissemination in tissues of a naturally infected pregnant southern yellow bat (Lasiurus ega) and its fetuses, obtained by reverse-transcriptase polymerase chain reaction (RT-PCR). The RT-PCR was positive in samples from the brain, salivary gland, tongue, lungs, heart, kidneys and liver. On the other hand, the placenta, three fetuses, spleen, intestine and brown fat tissue tested negative. This research demonstrated the absence of rabies virus in the fetuses, thus, in this specific case, the transplacentary transmission was not observed
Comparative descriptions of eggs from three species of Rhodnius (Hemiptera: Reduviidae: Triatominae)
The authors describe and compare the morphological and ultrastructural characteristics of eggs from the three most recent described species of the genus Rhodnius Stål, 1859, which have not previously been studied. These species are Rhodnius colombiensis (Mejia, Galvão & Jurberg 1999), Rhodnius milesi (Carcavallo, Rocha, Galvão & Jurberg 2001) and Rhodnius stali (Lent, Jurberg & Galvão 1993). The results revealed that there are similarities in the exochorial architecture of optical microscopy and scanning electron microscopy; these include the predominance of hexagonal cells that are common to all Rhodnius species and variable degrees of lateral flattening, which is common not only to species of this genus, but also to the Rhodniini tribe. Differences in overall colour, the presence of a collar in R. milesi, a longitudinal bevel in R. stali and the precise length of R. colombiensis can be useful distinguishing features. Effective silencing of ENaC by siRNA delivered with epithelial-targeted nanocomplexes in human cystic fibrosis cells and in mouse lung
Introduction Loss of the cystic fibrosis transmembrane conductance regulator in cystic fibrosis (CF) leads to hyperabsorption of sodium and fluid from the airway due to upregulation of the epithelial sodium channel (ENaC). Thickened mucus and depleted airway surface liquid (ASL) then lead to impaired mucociliary clearance. ENaC regulation is thus a promising target for CF therapy. Our aim was to develop siRNA nanocomplexes that mediate effective silencing of airway epithelial ENaC in vitro and in vivo with functional correction of epithelial ion and fluid transport.
Methods We investigated translocation of nanocomplexes through mucus and their transfection efficiency in primary CF epithelial cells grown at air–liquid interface (ALI).Short interfering RNA (SiRNA)-mediated silencing was examined by quantitative RT-PCR and western analysis of ENaC. Transepithelial potential (Vt), short circuit current (Isc), ASL depth and ciliary beat frequency (CBF) were measured for functional analysis. Inflammation was analysed by histological analysis of normal mouse lung tissue sections.
Results Nanocomplexes translocated more rapidly than siRNA alone through mucus. Transfections of primary CF epithelial cells with nanocomplexes targeting αENaC siRNA, reduced αENaC and βENaC mRNA by 30%. Transfections reduced Vt, the amiloride-sensitive Isc and mucus protein concentration while increasing ASL depth and CBF to normal levels. A single dose of siRNA in mouse lung silenced ENaC by approximately 30%, which persisted for at least 7 days. Three doses of siRNA increased silencing to approximately 50%.
Conclusion Nanoparticle-mediated delivery of ENaCsiRNA to ALI cultures corrected aspects of the mucociliary defect in human CF cells and offers effective delivery and silencing in vivo
