    Sustainable Microalgal Harvesting Process Applying Opuntia cochenillifera: Process Parameters Optimization

    Microalgae harvesting by coagulation can use coagulant agents such as alum, synthetic polymers or biocoagulants. Biocoagulants have attracted the attention of researchers because they are natural, biodegradable, and promote high microalgal harvesting efficiencies. This study aims to optimize the harvesting of Chlorella vulgaris based on the dosage of the Opuntia cochenillifera extract and the choice of eluent for biopolymer extraction. The outdoor cultivation of C. vulgaris achieved a specific growth rate of 0.455 d(-1) and a maximum biomass concentration of 1.28 g(DW) L-1. In order to harvest the microalgal biomass, the polymer present in the mucilage of O. cochenillifera was extracted using NaOH and HCl. Coagulation and sedimentation assays were performed with different coagulant dosages: 3.5, 5.9, and 8.2 g L-1. The maximum harvesting efficiencies using the acid and alkaline extract coagulant solutions were 80.8% and 99.5%, respectively, with a dosage of 3.5 g L-1. According to the results, the C. vulgaris biomass can be harvested with the mucilage from O. cochenillifera in acid and alkaline eluents. The application of this biocoagulant constitutes a sustainable solution for microalgal harvesting

    Urbanisation and asthma in low-income and middle-income countries: a systematic review of the urban-rural differences in asthma prevalence.

    BACKGROUND: Urbanisation has been associated with temporal and geographical differences in asthma prevalence in low-income and middle-income countries (LMICs). However, little is known of the mechanisms by which urbanisation and asthma are associated, perhaps explained by the methodological approaches used to assess the urbanisation-asthma relationship. OBJECTIVE: This review evaluated how epidemiological studies have assessed the relationship between asthma and urbanisation in LMICs, and explored urban/rural differences in asthma prevalence. METHODS: Asthma studies comparing urban/rural areas, comparing cities and examining intraurban variation were assessed for eligibility. Included publications were evaluated for methodological quality and pooled OR were calculated to indicate the risk of asthma in urban over rural areas. RESULTS: Seventy articles were included in our analysis. Sixty-three compared asthma prevalence between urban and rural areas, five compared asthma prevalence between cities and two examined intraurban variation in asthma prevalence. Urban residence was associated with a higher prevalence of asthma, regardless of asthma definition: current-wheeze OR:1.46 (95% CI:1.22 to 1.74), doctor diagnosis OR:1.89 (95% CI:1.47 to 2.41), wheeze-ever OR:1.44 (95% CI:1.15 to 1.81), self-reported asthma OR:1.77 (95% CI:1.33 to 2.35), asthma questionnaire OR:1.52 (95% CI:1.06 to 2.16) and exercise challenge OR:1.96 (95% CI:1.32 to 2.91). CONCLUSIONS: Most evidence for the relationship between urbanisation and asthma in LMICs comes from studies comparing urban and rural areas. These studies tend to show a greater prevalence of asthma in urban compared to rural populations. However, these studies have been unable to identify which specific characteristics of the urbanisation process may be responsible. An approach to understand how different dimensions of urbanisation, using contextual household and individual indicators, is needed for a better understanding of how urbanisation affects asthma.     The bundles of algebraic and Dirac-Hestenes spinor fields

    Our main objective in this paper is to clarify the ontology of Dirac-Hestenes spinor fields (DHSF) and its relationship with even multivector fields, on a Riemann-Cartan spacetime (RCST) M=(M,g,del,tau(g),up arrow) admitting a spin structure, and to give a mathematically rigorous derivation of the so-called Dirac-Hestenes equation (DHE) in the case where M is a Lorentzian spacetime (the general case when M is a RCST will be discussed in another publication). To this aim we introduce the Clifford bundle of multivector fields (Cl(M,g)) and the left (Cl-Spin1,3(el)(M)) and right (Cl-Spin1,3(er)(M)) spin-Clifford bundles on the spin manifold (M,g). The relation between left ideal algebraic spinor fields (LIASF) and Dirac-Hestenes spinor fields (both fields are sections of Cl-Spin1,3(el)(M)) is clarified. We study in detail the theory of covariant derivatives of Clifford fields as well as that of left and right spin-Clifford fields. A consistent Dirac equation for a DHSF Psiis an element ofsec Cl-Spin1,3(el)(M) (denoted DECll) on a Lorentzian spacetime is found. We also obtain a representation of the DECll in the Clifford bundle Cl(M,g). It is such equation that we call the DHE and it is satisfied by Clifford fields psi(Xi)is an element ofsec Cl(M,g). This means that to each DHSF Psiis an element ofsec Cl-Spin1,3(el)(M) and spin frame Xiis an element ofsec P-Spin1,3(e)(M), there is a well-defined sum of even multivector fields psi(Xi)is an element ofsec Cl(M,g) (EMFS) associated with Psi. Such an EMFS is called a representative of the DHSF on the given spin frame. And, of course, such a EMFS (the representative of the DHSF) is not a spinor field. With this crucial distinction between a DHSF and its representatives on the Clifford bundle, we provide a consistent theory for the covariant derivatives of Clifford and spinor fields of all kinds. We emphasize that the DECll and the DHE, although related, are equations of different mathematical natures. We study also the local Lorentz invariance and the electromagnetic gauge invariance and show that only for the DHE such transformations are of the same mathematical nature, thus suggesting a possible link between them.     Production of monoclonal antibodies for detection of a secreted aspartyl proteinase from Candida spp. in biologic specimens

    Secreted acid proteinases (SAP) constitute an important group of virulence factors in Candida albicans. In the present work, an acid proteinase from C. albicans was sequentially purified from the supernatant of a yeast culture by precipitation with ammonium sulfate, ion exchange chromatography, and molecular exclusion chromatography, yielding a specific enzymatic activity of 204.1 IU/mg on bovine serum albumin (BSA). The molecular mass of the purified proteinase was estimated at 43 kd after exclusion chromatography and at 41 kd by nondenaturating sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The purified proteinase was able to degrade BSA at pH 2.5, but was not active on collagen, and it was significantly inhibited by pepstatin A. The immunization of BALB/c mice with the purified proteinase and later fusion of their spleen cells with myeloma cells resulted in 19 monoclonal antibody secreting hybridomas (MAbs) capable of detecting SAP in enzyme-linked immunosorbent assay ( ELISA) assays. All MAbs obtained are isotype IgG1 kappa (kappa) immunoglobulins and develop a 41 kd protein band by Western blot (WB) in samples of SAP obtained from C. albicans (12-A) and C. dubliniensis ( strain 778) crude extracts. The anti-SAP MAbs were used in capture ELISA and two combinations of these antibodies proved suitable for SAP detection, that is, MAP1 (1B1B3) or MAP2 (2D2C10) as coat antibodies, and biotinylated MAP3 (2A6E8) as detect antibody. Capture ELISA using these sets of MAbs detected over 32 ng/mL protein in purified SAP samples as well as in crude C. albicans and C. dubliniensis extracts. The results herein obtained allow for the prediction of how this set of antibodies can be useful for SAP detection in biologic specimens.26420120

    Effect of bilirubin on cytochrome c oxidase activity of mitochondria from mouse brain and liver

    <p>Abstract</p> <p>Background</p> <p>The unbound, free concentration (B<sub>f</sub>) of unconjugated bilirubin (UCB), and not the total UCB level, has been shown to correlate with bilirubin cytotoxicity, but the key molecular mechanisms accounting for the toxic effects of UCB are largely unknown.</p> <p>Findings</p> <p>Mouse liver mitochondria increase unbound UCB oxidation, consequently increasing the apparent rate constant for unbound UCB oxidation by HRP (Kp), higher than in control and mouse brain mitochondria, emphasizing the importance of determining Kp in complete systems containing the organelles being studied. The <it>in vitro </it>effects of UCB on cytochrome <it>c </it>oxidase activity in mitochondria isolated from mouse brain and liver were studied at B<sub>f </sub>ranging from 22 to 150 nM. The results show that UCB at B<sub>f </sub>up to 60 nM did not alter mitochondrial cytochrome <it>c </it>oxidase activity, while the higher concentrations significantly inhibited the enzyme activity by 20% in both liver and brain mitochondria.</p> <p>Conclusions</p> <p>We conclude that it is essential to include the organelles being studied in the medium used in measuring both Kp and B<sub>f</sub>. A moderately elevated, pathophysiologically-relevant B<sub>f </sub>impaired the cytochrome <it>c </it>oxidase activity modestly in mitochondria from mouse brain and liver.</p
