Selection and geographic isolation influence hummingbird speciation: genetic, acoustic and morphological divergence in the wedge-tailed sabrewing (Campylopterus curvipennis)
<p>Abstract</p> <p>Background</p> <p>Mesoamerica is one of the most threatened biodiversity hotspots in the world, yet we are far from understanding the geologic history and the processes driving population divergence and speciation for most endemic taxa. In species with highly differentiated populations selective and/or neutral factors can induce rapid changes to traits involved in mate choice, promoting reproductive isolation between allopatric populations that can eventually lead to speciation. We present the results of genetic differentiation, and explore drift and selection effects in promoting acoustic and morphological divergence among populations of <it>Campylopterus curvipennis</it>, a lekking hummingbird with an extraordinary vocal variability across Mesoamerica.</p> <p>Results</p> <p>Analyses of two mitochondrial genes and ten microsatellite loci genotyped for 160 individuals revealed the presence of three lineages with no contemporary gene flow: <it>C. c. curvipennis, C. c. excellens</it>, and <it>C. c. pampa </it>disjunctly distributed in the Sierra Madre Oriental, the Tuxtlas region and the Yucatan Peninsula, respectively. Sequence mtDNA and microsatellite data were congruent with two diversification events: an old vicariance event at the Isthmus of Tehuantepec (<it>c</it>. 1.4 Ma), and a more recent Pleistocene split, isolating populations in the Tuxtlas region. Hummingbirds of the <it>excellens </it>group were larger, and those of the <it>pampa </it>group had shorter bills, and lineages that have been isolated the longest shared fewer syllables and differed in spectral and temporal traits of a shared syllable. Coalescent simulations showed that fixation of song types has occurred faster than expected under neutrality but the null hypothesis that morphological divergence resulted from drift was not rejected.</p> <p>Conclusions</p> <p>Our phylogeographic analyses uncovered the presence of three Mesoamerican wedge-tailed sabrewing lineages, which diverged at different time scales. These results highlight the importance of the Isthmus of Tehuantepec and more recent Pleistocene climatic events in driving isolation and population divergence. Coalescent analyses of the evolution of phenotypic traits suggest that selection is driving song evolution in wedge-tailed sabrewings but drift could not be rejected as a possibility for morphological divergence.</p
Finite element approximation of nematic liquid crystal flows using a saddle-point structure
In this work, we propose finite element schemes for the numerical approximation of nematic liquid crystal flows, based on a saddle-point formulation of the director vector sub-problem. It introduces a Lagrange
multiplier that allows to enforce the sphere condition. In this setting, we can consider the limit problem (without penalty) and the penalized problem (using a Ginzburg-Landau penalty function) in a unified way. Further, the
resulting schemes have an stable behavior with respect to the value of the penalty parameter, a key difference with respect to the existing schemes. Two different methods have been considered for the time integration. First, we have considered an implicit algorithm that is unconditionally stable and energy preserving. The linearization of the problem at every time step value can be performed using a quasi-Newton method that allows to decouple fluid
velocity and director vector computations for every tangent problem. Then, we have designed a linear semi-implicit algorithm (i.e. it does not involve nonlinear iterations) and proved that it is unconditionally stable, verifying a discrete energy inequality. Finally, some numerical simulations are provided
An overview on numerical analyses of nematic liquid crystal flows
The purpose of this work is to provide an overview of the most recent numerical developments in the field of nematic liquid crystals. The Ericksen-Leslie equations govern the motion of a nematic liquid crystal. This system, in its simplest form, consists of the Navier-Stokes equations coupled with an extra anisotropic stress tensor, which represents the effect of the nematic liquid crystal on the fluid, and a convective harmonic map equation. The
sphere constraint must be enforced almost everywhere in order to obtain an energy estimate. Since an almost everywhere satisfaction of this restriction is not appropriate at a numerical level, two alternative approaches have been introduced: a penalty method and a saddle-point method. These approaches are suitable for their numerical approximation by finite elements,
since a discrete version of the restriction is enough to prove the desired energy estimate. The Ginzburg-Landau penalty function is usually used to enforce the sphere constraint. Finite element methods of mixed type will play an important role when designing numerical approximations for the penalty method in order to preserve the intrinsic energy estimate. The inf-sup condition that makes the saddle-point method well-posed is not clear yet.
The only inf-sup condition for the Lagrange multiplier is obtained in the dual space of H1(Ω). But such an inf-sup condition requires more regularity for the director vector than the one provided by the energy estimate. Herein, we will present an alternative inf-sup condition whose proof for its discrete counterpart with finite elements is still open.

GWAS and meta-analysis identifies 49 genetic variants underlying critical COVID-19
Critical illness in COVID-19 is an extreme and clinically homogeneous disease
phenotype that we have previously shown1 to be highly efficient for discovery of
genetic associations2. Despite the advanced stage of illness at presentation, we have
shown that host genetics in patients who are critically ill with COVID-19 can identify
immunomodulatory therapies with strong beneficial effects in this group3. Here we
analyse 24,202 cases of COVID-19 with critical illness comprising a combination of
microarray genotype and whole-genome sequencing data from cases of critical illness
in the international GenOMICC (11,440 cases) study, combined with other studies
recruiting hospitalized patients with a strong focus on severe and critical disease:
ISARIC4C (676 cases) and the SCOURGE consortium (5,934 cases). To put these results
in the context of existing work, we conduct a meta-analysis of the new GenOMICC
genome-wide association study (GWAS) results with previously published data. We
find 49 genome-wide significant associations, of which 16 have not been reported
previously. To investigate the therapeutic implications of these findings, we infer the
structural consequences of protein-coding variants, and combine our GWAS results
with gene expression data using a monocyte transcriptome-wide association study
(TWAS) model, as well as gene and protein expression using Mendelian randomization.
We identify potentially druggable targets in multiple systems, including inflammatory
signalling ( JAK1), monocyte–macrophage activation and endothelial permeability
(PDE4A), immunometabolism (SLC2A5 and AK5), and host factors required for viral
entry and replication (TMPRSS2 and RAB2A)
Participatory diagnosis for the development of municipal housing plans: The cases of Bormujos and Bollullos de la Mitación (Andalusía)
The bursting of the Spanish real estate bubble in 2008 revealed a serious problem of housing access for a growing sector of the population that cannot afford market prices, while part of the existing housing stock remains vacant. In a context of lack of resources, the Andalusian Housing and Land Plan (2016-2020) proposes that municipalities develop Municipal Housing and Land Plans (known as PMVS in Spanish) to explore innovative solutions that contribute to bringing these two extremes closer together based on knowledge of the specific situation in each locality and the participation of the social actors involved. This article sets out to create a participatory diagnostic of the situation of unresolved demand and vacant housing stock on a local scale in order to situate the challenges that a PMVS should address. To this end, under a methodological framework of participatory action research, the study analyses the participatory process implemented in two municipalities in the Seville metropolitan area, Bormujos and Bollullos de la Mitación, as case studies representative of the urban processes that took place during the years of the real estate bubble. Therefore, the results were used to create a profile of the existing needs and the available resources that could be allocated to meet them, in this case empty housing and vacant land. Likewise, through this experience it was possible to conclude how the participation of technicians, politicians and citizens in public policy development contributes to building critical awareness, a sense of appropriation and the commitment of local agents to the challenge faced, which facilitates their subsequent involvement in the implementation of collaborative actions
