    Histomorphological and Immunophenotypic Characterization of Feline Injection Site-Associated Sarcoma

    Background: Feline Injection Site-Associated Sarcoma (FISS) is a mesenchymal neoplasia of aggressive behavior that develops in sites where vaccine or drugs were administered. FISS is clinically characterized by the appearance of a solitary firm nodule or a diffuse mass, adhered to tissues, in regions associated to vaccine or drug applications. Despite low prevalence, tumor recurrence rates can reach 80%. FISS present more aggressive histological characteristics when compared to sarcomas not associated to injection sites. The aim of this paper is to contribute towards the understanding of the biological behavior of FISS.Materials, Methods & Results: Sixteen samples of FISS were analyzed. Fibrosarcomas were the most frequent histological subtype (62.5%). Malignant peripheral nerve sheath tumor was diagnosed in 18.75% cases. Ten (62.5%) FISS were classified as grade II; 4/16 (25%) grade I, and 2/16 (12.5%) grade III. Cox-2 overexpression occurred in 3/16 (18.75%) samples, with positive correlation between Cox-2 expression and cellularity (r = 0.696, P = 0.003). Mitotic index lower than 9 events was found in 11/16 (68.7%) samples and between 10 and 19 mitotic events in 5/16 (31.3%) cases. Mean Ki-67 expression was 2.39 ± 2.48%. FISS characterized as fibrosarcomas presented longer overall survival (median 545 days) than other histological subtypes (median 130.5 days) [P = 0.01].Discussion: Patients with FISS generally present with larger nodules than those with sarcomas not associated to injections, suggesting a challenge for pet owners to note subcutaneous tumors in the interscapular region, in addition to the aggressive biological behavior of FISS. The influence of size on prognosis remains controversial. An association between histologic grade and the development of metastasis has been observed, with patients with grade III FISS associated with an increase in the metastatic rate. The present study did not find a correlation between overall survival and histologic grade. A positive correlation between the presence of giant multinucleated cells and tumor grade has been observed.Despite the absence of such correlation in the present study, possibly due to a small sample, a trend for higher frequency of giant cells in advanced histologic grade was observed. Cox-2 expression in 81.75% and overexpression in 18.75% of our samples contrasts with the 64% Cox-2 expression and the absence of Cox-2 expression found by other authors. A positive moderate correlation between cellularity and Cox-2 expression was also observed, while another study did not find a correlation of Cox-2 expression with tumor grade, recurrence rates or overall survival. Cox metabolites such as prostaglandins can enhance cellular proliferation, inhibit apoptosis, induce angiogenesis, alter cellular adherence to facilitate metastatic development and inhibit immune surveillance. In the present study, no correlation was found between Cox-2 and angiogenesis in FISS. Our findings demonstrated low immunolabeling for Ki-67. A previous study analyzed 52 samples of FISS, 51% of them considered grade III, with a mean Ki-67 labeling of 14%. The lower Ki-67 staining in the samples of the present study may be related to the lower number of samples of grade III FISS or to a difference in the studied population. Fibrosarcomas are associated with better prognosis than other histological subtypes. Furthermore, malignant peripheral nerve sheath tumors were diagnosed as a possible histological subtype of FISS

    Echocardiographic indices in normal German shepherd dogs

    Sixty clinically normal German shepherd dogs, 31 males and 29 females, ranging in age from 1 and 5 years and with a body weight ranging from 22 to 37.2 kg, were examined by the two-dimensional mode, M-mode, and Doppler echocardiography. In Doppler mode, the mitral valve flows were obtained, where the aim was to determine the velocity peaks and ratios of the E and A waves and the mitral E wave deceleration time. The velocity peaks were obtained for the tricuspid, pulmonary and aortic valves. On the left ventricular outflow tract flow, the time velocity integral and aortic cross-sectional area was used to calculate the cardiac output. A statistically significant correlation with the body weight was found for the systolic left atrial and diastolic aortic diameter on two-dimensional mode. On M-mode, there was a significant correlation between the body weight and the systolic left atrium and diastolic aortic dimension, systolic and diastolic left ventricular, septal and posterior wall dimensions. Doppler echocardiography showed that there was no significant correlation between the body weight and the mitral, tricuspid, pulmonary and aortic valves flows. These results demonstrate that it is important to know the normal echocardiographic values for German shepherd dogs because there are some characteristics peculiar to this particular breed. The data obtained is expected to be helpful for studies on small animal cardiology

    Evaluation and comparison between methods of measurement of systolic blood pressure in healthy conscious cats

    Após validação de dois diferentes métodos de mensuração de pressão arterial sistólica (PAS) não invasivos ou indiretos (oscilométrico e Doppler vascular) com o invasivo ou direto (cateterização arterial padrão ouro) em 12 gatos hígidos anestesiados de mesma faixa etária, os métodos não invasivos foram avaliados e comparados em 24 gatos hígidos e conscientes, divididos em quatro grupos de idade. Em cada animal, procedeu-se à mensuração da pressão pelos métodos Doppler e oscilométrico. O método oscilométrico foi o primeiro a ser realizado, e, logo em seguida, o método Doppler. Os valores considerados foram obtidos das médias de sete medidas consecutivas para cada método, com intervalo de 30 segundos entre elas. A primeira medida e as medidas discrepantes (variação ˃ 20% PAS), obtidas com sinais óbvios de estresse e/ou com movimentação do animal, foram descartadas. Os métodos de mensuração oscilométrico e Doppler, embora tenham apresentado médias estatisticamente diferentes em todos os grupos, quando comparados em cada grupo, apresentaram uma correlação significativa, alta e positiva. Ou seja, sempre que for obtido um valor elevado por um método, o mesmo fato ocorrerá com o outro método e vice-versa. Clinicamente, a diferença observada não é relevante, uma vez que a diferença média dos valores obtida pelos dois métodos, em cada grupo, foi menor que 1,1%.After two different validation systolic blood pressure measurement methods (PAS), not invasive or indirect (oscillometric and doppler vascular) with invasive or direct (arterial catheterization - Gold Standard) in 12 anesthetized healthy cats of the same age group, not invasive methods were evaluated and compared in 24 healthy conscious cats, divided into four age groups. In each animal, we proceeded to measure the pressure by Doppler and oscillometric methods. The oscillometric method was the first to be performed, and, shortly thereafter, the doppler method. The values considered were obtained from averages from seven consecutive measurements for each method with an interval of thirty seconds between them. The first measurement and disparate measurements (range ˃ 20% PAS) obtained with obvious signs of stress and / or animal movement were discarded. Methods of oscillometric and doppler measurement, although statistically different averages were shown in all groups when compared, each group showed a significant high positive correlation, meaning that when a high value is obtained by a method, the same occurs with the other method, and vice versa. Clinically, the observed difference is not relevant since the mean difference values obtained by the two methods, in each group was lower than 1.1%

    ABSTRACT: Targeted therapy of neoplasms is an emergent approach in human and veterinary medicine. Cyclooxygenase (COX) is a class of catalytic enzymes related to the formation of inflammatory mediators. COX-2 is expressed constitutively in a few body tissues, but it may be induced in specific pathophysiologic conditions, such as cancer. COX-2 expression in neoplams may be considered a potential predictive factor, due to the possible association of selective COX-2 inhibitors in adjuvant treatments. This scientific communication has the objective to report COX-2 expression in seven neoplasms of dogs and the usage of adjuvant treatment with COX-2 selective inhibitors as an effective and feasible option in cancer treatment

    A imuno-histoquímica pode ser aplicada na oncologia veterinária para a definição do imunofenótipo neoplásico, com possibilidades ilimitadas de utilização. A técnica baseia-se na identificação de antígenos "in situ", que podem apresentar valor prognóstico e terapêutico. A expressão de receptores de estrógeno e progesterona está diretamente relacionada ao melhor prognóstico para os neoplasmas mamários, enquanto a super-expressão do receptor KIT (CD117), proteínas Ki-67 (MIB-1), VEGF, CD31 e COX-2 podem ser associados à progressão tumoral para vários tipos histológicos. Essa associação possui valor preditivo em potencial, tendo em vista a possibilidade da utilização de bloqueadores específicos. Esta revisão tem como objetivo apresentar as possibilidades da utilização da imuno-histoquímica nos tumores de cães, visando à definição mais precisa do prognóstico e a indicação de tratamentos específicos para cada paciente.Immunohistochemistry can be applied in veterinary oncology to define the immunophenotype of neoplastic cells with unlimited possibilities of application. The technique is based on the identification of antigens "in situ", which may have prognostic and therapeutic value. Expression of estrogen and progesterone is directly related to better prognosis for breast cancer, while over-expression of receptor KIT (CD117) proteins Ki-67 (MIB-1), COX-2, VEGF and CD31 can be associated with tumor progression for several histological types. This association has a potential predictive value, due to the possibility of using specific blockers. This revision aims at presenting the possibilities of immunohistochemical's application in tumors of dogs, searching for a more precise definition of the prognosis and indication of specific treatments for each patient

    Este estudo teve como objetivo estabelecer os índices ecocardiográficos modo-M normais de cães da raça Pastor Alemão do canil da Polícia Militar do Estado de Minas Gerais. Sessenta cães clinicamente sadios foram submetidos ao exame ecocardiográfico. O diâmetro interno do ventrículo direito na diástole e do ventrículo esquerdo na diástole e na sístole, massa ventricular esquerda, dimensão do arco aórtico e átrio esquerdo, espessura do septo interventricular e parede posterior do ventrículo esquerdo na diástole e sístole tiveram correlações significativas com o peso corporal. A relação espessura do septo interventricular e parede posterior, no final da diástole, relação diâmetro do átrio esquerdo e aorta, porcentagem de espessamento sistólico do septo interventricular, porcentagem de espessamento sistólico da parede posterior de ventrículo esquerdo e porcentagem de encurtamento sistólico do diâmetro ventricular esquerdo não se correlacionaram com o peso corporal, sexo ou idade.The normal M-mode echocardiographic parameters were determined in German Shepherd dogs from Minas Gerais State Police kennel. Sixty clinically normal dogs were submitted to the echocardiographic examination. The right ventricular internal dimension at end-diastole, the left ventricular internal dimension at end-diastole and end-systole, the left ventricular mass, the aortic root dimension, left atrial systolic dimension, end-diastolic interventricular septal thickness and end-diastolic and end-systolic left ventricular posterior wall thickness had significant correlation with body weight. The ratio of septal to left ventricular posterior wall thickness at end-distole, the left atrial to aortic root ratio, the thickening fraction of the interventricular septum, the thickening fraction of the left ventricular posterior wall, and left ventricular shortening fraction showed no linear correlations with body weight, sex or age

    <div><p>ABSTRACT: Extramedullary plasmacytomas (EPs) are responsible for 2.5% of neoplasms in dogs. They are solitary, smooth, elevated, pink or red nodules, of 1 to 2cm in diameter. Cutaneous and oral extramedullary plasmacytomas in dogs are usually benign tumors, treated with local therapies. Prognosis is generally good. Recurrence and metastatic rates are low. Electrochemotherapy is a local treatment that combines chemotherapy and electroporation and shows objective responses of 70% to 94% with few local and systemic side effects. This scientific communication has the objective to report treatment of three canine patients with oral extramedullary plasmacytoma. Nodules were located on the tongue and patients were submitted to one or two electrochemotherapy sessions, which preserved the tongue without mutilation and cured the patients.</p></div
