    A modified expression of the major hydrolase activator in Hypocrea jecorina (Trichoderma reesei) changes enzymatic catalysis of biopolymer degradation

    AbstractHypocrea jecorina (anamorph Trichoderma reesei) is a saprophytic fungus that produces hydrolases, which are applied in different types of industries and used for the production of biofuel. A recombinant Hypocrea strain, which constantly expresses the main transcription activator of hydrolases (Xylanase regulator 1), was found to grow faster on xylan and its monomeric backbone molecule d-xylose. This strain also showed improved ability of clearing xylan medium on plates. Furthermore, this strain has a changed transcription profile concerning genes encoding for hydrolases and enzymes associated with degradation of (hemi)celluloses. We demonstrated that enzymes of this strain from a xylan cultivation favoured break down of hemicelluloses to the monomer d-xylose compared to the parental strain, while the enzymes of the latter one formed more xylobiose. Applying supernatants from cultivation on carboxymethylcellulose in enzymatic conversion of hemicelluloses, the enzymes of the recombinant strain were clearly producing more of both, d-xylose and xylobiose, compared to the parental strain. Altogether, these results point to a changed hydrolase expression profile, an enhanced capability to form the xylan-monomer d-xylose and the assumption that there is a disordered induction pattern if the Xylanase regulator 1 is de-regulated in Hypocrea

    A 29-year-old man complained of a 2-day history of frontal headache and new-onset fever but no other symptoms. Two months prior to admission, he underwent his third kidney transplantation. Clinical and laboratory examinations were unremarkable. Brain MRI showed a meningeal irritation consistent with viral meningitis. A diagnosis of cryptococcal meningitis and fungaemia was made after detection of a remarkably high and visible load of Cryptococcus neoformans in the cerebrospinal fluid

    A Coupled Snow Operations-Skier Demand Model for the Ontario (Canada) Ski Region

    The multi-billion dollar global ski industry is one of the tourism subsectors most directly impacted by climate variability and change. In the decades ahead, the scholarly literature consistently projects decreased reliability of natural snow cover, shortened and more variable ski seasons, as well as increased reliance on snowmaking with associated increases in operational costs. In order to develop the coupled snow, ski operations and demand model for the Ontario ski region (which represents approximately 18% of Canada's ski market), the research utilized multiple methods, including: a in situ survey of over 2400 skiers, daily operations data from ski resorts over the last 10 years, climate station data (1981-2013), climate change scenario ensemble (AR5 - RCP 8.5), an updated SkiSim model (building on Scott et al. 2003; Steiger 2010), and an agent-based model (building on Pons et al. 2014). Daily snow and ski operations for all ski areas in southern Ontario were modeled with the updated SkiSim model, which utilized current differential snowmaking capacity of individual resorts, as determined from daily ski area operations data. Snowmaking capacities and decision rules were informed by interviews with ski area managers and daily operations data. Model outputs were validated with local climate station and ski operations data. The coupled SkiSim-ABM model was run with historical weather data for seasons representative of an average winter for the 1981-2010 period, as well as an anomalously cold winter (2012-13) and the record warm winter in the region (2011-12). The impact on total skier visits and revenues, and the geographic and temporal distribution of skier visits were compared. The implications of further climate adaptation (i.e., improving the snowmaking capacity of all ski areas to the level of leading resorts in the region) were also explored. This research advances system modelling, especially improving the integration of snow and ski operations models with demand and socioeconomic implications. This innovative integrated systems model approach can be exported to other major ski tourism markets (e.g., Canada, USA, Western and Eastern Europe, Australia, Japan) to facilitate global comparative assessments of ski tourism vulnerability to climate change, establishing the standard for ski tourism vulnerability assessments and advancing scholarly work on sustainable tourism and climate-compatible development in mountain communities

    Assessing the Factorial Validity of the Attitudes and Belief Scale 2-Abbreviated Version: A Call for the Development a Gold Standard Method of Measuring Rational and Irrational Beliefs

    Rational Emotive Behaviour Therapy (REBT) does not possess a measure of rational and irrational beliefs that meets internationally recognised standards for acceptable psychometric properties. Without such a measure the theory/practice of REBT cannot be rigorously evaluated, thus undermining its scientific veracity. The current study investigates the validity and reliability of a recently developed measure of rational and irrational beliefs: the Attitudes and Belief Scale 2-Abbreviated Version (ABS-2-AV). University students from three countries completed the ABS-2-AV (N = 397). An alternative models framework using confirmatory factor analysis indicated that a theoretically consistent eight-factor model of the ABS-2-AV provided the best fit of the data. A number of post-hoc modifications were required in order to achieve acceptable model fit results, and these modifications revealed important methodological limitations with the ABS-2-AV. Results indicated that the validity of the ABS-2-AV was undermined due to items measuring both the psychological process of interest (rational and irrational beliefs) and the context in which these beliefs processes are presented. This is a serious methodological limitation of the ABS-2 and all questionnaires derived from it, including the ABS-2-AV. This methodological limitation resulted in the ABS-2-AV possessing poor internal reliability. These limitations are discussed in relation to the broader REBT literature and the impact such problems have on research and practice. A call is made for REBT researchers to come together to develop a “gold standard” method of assessing rational and irrational beliefs that meets international standard for psychometric excellence

    Effects of Manganese Porphyrins on Cellular Sulfur Metabolism

    Manganese porphyrins (MnPs), MnTE-2-PyP5+, MnTnHex-2-PyP5+ and MnTnBuOE-2-PyP5+, are superoxide dismutase (SOD) mimetics and form a redox cycle between O2 and reductants, including ascorbic acid, ultimately producing hydrogen peroxide (H2O2). We previously found that MnPs oxidize hydrogen sulfide (H2S) to polysulfides (PS; H2Sn, n = 2–6) in buffer. Here, we examine the effects of MnPs for 24 h on H2S metabolism and PS production in HEK293, A549, HT29 and bone marrow derived stem cells (BMDSC) using H2S (AzMC, MeRho-AZ) and PS (SSP4) fluorophores. All MnPs decreased intracellular H2S production and increased intracellular PS. H2S metabolism and PS production were unaffected by cellular O2 (5% versus 21% O2), H2O2 or ascorbic acid. We observed with confocal microscopy that mitochondria are a major site of H2S production in HEK293 cells and that MnPs decrease mitochondrial H2S production and increase PS in what appeared to be nucleoli and cytosolic fibrillary elements. This supports a role for MnPs in the metabolism of H2S to PS, the latter serving as both short- and long-term antioxidants, and suggests that some of the biological effects of MnPs may be attributable to sulfur metabolism
