    This submission is an analytical report that chronicles and evaluates the internship experience of Lisa Riley at the New York producing office of Elizabeth Ireland McCann (ElM). The internship period commenced August 4, 1986 and concluded December 2, 1986. As required by the University of New Orleans for the fulfillment for the degree of Master of Arts in Arts Administration, this report will describe the organizational structure of Elizabeth Ireland McCann Productions. It will then analyze management problems the intern discovered throughout the course of the internship and offer recommendations for the resolution of these managerial problems. Finally, a discussion will ensue to determine the actual long and short-term contributions the intern made towards the organization

    Does Channel Island Acmispon (Fabaceae) Form Cohesive Evolutionary Groups?

    The California Channel Islands are unique relative to other island chains due to their close proximity to the California mainland and the fact that individual islands, or groups of islands, vary in their distance to the mainland and other islands. This orientation raises questions about whether island taxa with widespread distributions form cohesive evolutionary units, or if they are actually composed of several distinct evolutionary entities, either derived from independent mainland-to-island colonization events or divergence due to prolonged allopatric isolation. The 4 northern islands are clustered in a line (6-8 km separation among islands), while the 4 southern islands are widely spaced (34-45 km separation among islands), which should impact the amount of gene flow and genetic connectivity among islands. We used nuclear microsatellite markers to examine the genetic structure and cohesion of 2 island shrubs, Acmispon dendroideus and A. argophyllus, which are widely distributed across the California Channel Islands. Both focal species contain varieties with multi-island distributions, with A. dendroideus exhibiting a greater distribution on the northern islands and A. argophyllus exhibiting a greater distribution on the southern islands. Substantial genetic divergence was observed for 2 single-island endemic varieties, A. dendroideus var. traskiae and A. agrophyllus var. niveus, confirming that allopatric isolation can lead to genetic divergence. The widespread Acmispon dendroideus var. dendroideus and single-island endemic A. dendroideus var. veatchii formed a cohesive evolutionary group that spans all 4 northern islands and 1 southern island, Santa Catalina, indicating that the northern and southern islands have been genetically linked in the past but do not display evidence of contemporary gene flow. In contrast, widespread A. argophyllus var. argenteus was composed of moderately distinct genetic groups on each of the 4 southern islands, with no evidence of recent gene flow among islands. These results demonstrate that isolation among islands has led to significant divergence among the southern islands, but that the commonly recognized split between northern and southern islands does not impact all taxa equally

    Bacteriophage-Based Tests for the Detection of \u3cem\u3eMycobacterium tuberculosis\u3c/em\u3e in Clinical Specimens: A Systematic Review and Meta-analysis

    Background: Sputum microscopy, the most important conventional test for tuberculosis, is specific in settings with high burden of tuberculosis and low prevalence of non tuberculous mycobacteria. However, the test lacks sensitivity. Although bacteriophage-based tests for tuberculosis have shown promising results, their overall accuracy has not been systematically evaluated. Methods: We did a systematic review and meta-analysis of published studies to evaluate the accuracy of phage-based tests for the direct detection of M. tuberculosis in clinical specimens. To identify studies, we searched Medline, EMBASE, Web of science and BIOSIS, and contacted authors, experts and test manufacturers. Thirteen studies, all based on phage amplification method, met our inclusion criteria. Overall accuracy was evaluated using forest plots, summary receiver operating (SROC) curves, and subgroup analyses. Results: The data suggest that phage-based assays have high specificity (range 0.83 to 1.00), but modest and variable sensitivity (range 0.21 to 0.88). The sensitivity ranged between 0.29 and 0.87 among smear-positive, and 0.13 to 0.78 among smear-negative specimens. The specificity ranged between 0.60 and 0.88 among smear-positive and 0.89 to 0.99 among smear-negative specimens. SROC analyses suggest that overall accuracy of phage-based assays is slightly higher than smear microscopy in direct head-to-head comparisons. Conclusion: Phage-based assays have high specificity but lower and variable sensitivity. Their performance characteristics are similar to sputum microscopy. Phage assays cannot replace conventional diagnostic tests such as microscopy and culture at this time. Further research is required to identify methods that can enhance the sensitivity of phage-based assays without compromising the high specificity

    A Spatial and Temporal Gradient of Fgf Differentially Regulates Distinct Stages of Neural Development in the Zebrafish Inner Ear

    Neuroblasts of the statoacoustic ganglion (SAG) initially form in the floor of the otic vesicle during a relatively brief developmental window. They soon delaminate and undergo a protracted phase of proliferation and migration (transit-amplification). Neuroblasts eventually differentiate and extend processes bi-directionally to synapse with hair cells in the inner ear and various targets in the hindbrain. Our studies in zebrafish have shown that Fgf signaling controls multiple phases of this complex developmental process. Moderate levels of Fgf in a gradient emanating from the nascent utricular macula specify SAG neuroblasts in laterally adjacent otic epithelium. At a later stage, differentiating SAG neurons express Fgf5, which serves two functions: First, as SAG neurons accumulate, increasing levels of Fgf exceed an upper threshold that terminates the initial phase of neuroblast specification. Second, elevated Fgf delays differentiation of transit-amplifying cells, balancing the rate of progenitor renewal with neuronal differentiation. Laser-ablation of mature SAG neurons abolishes feedback-inhibition and causes precocious neuronal differentiation. Similar effects are obtained by Fgf5-knockdown or global impairment of Fgf signaling, whereas Fgf misexpression has the opposite effect. Thus Fgf signaling renders SAG development self-regulating, ensuring steady production of an appropriate number of neurons as the larva grows

    Genetic and environmental drivers of colour and pattern in the Australian jacky dragon (Amphibolurus muricatus)

    The underlying drivers of variation in the colouration (colour and pattern) of animals can be genetic, non-genetic, or more likely, a combination of both. Understanding the role of heritable genetic elements, as well as non-genetic factors such as age, habitat or temperature, in shaping colouration can provide insight into the evolution and function of these traits, as well as the speed of response to changing environments. This project examined the genetic and non-genetic drivers of continuous variation in colouration in a lizard, the jacky dragon (Amphibolurus muricatus). We leveraged a large captive experiment that manipulated parental and offspring thermal environment to simultaneously estimate the genetic and non-genetic drivers of variation in colouration. We found that the overall brightness, the elongation of the longitudinal stripes on the dorsum and the contrast between light and dark patches of the pattern were all heritable. Colouration varied according to the age of the hatchling; however, the thermal environment of neither the parents nor offspring contributed significantly to colouration. It appears that developmental plasticity and maternal effects associated with temperature are not important drivers of variation in our measures of colouration

    Conducting Behavioural Research in the Zoo: A Guide to Ten Important Methods, Concepts and Theories

Robust methods that allow valid data collection and analysis constitute an evidence-based approach to animal care. Understanding behaviour is essential to improving animal management, and behavioural research is therefore popular, with a wide choice of behavioural methodologies and theories available. This review outlines ten methodological approaches, concepts or theories essential to zoo science that are based around behavioural observation. This list is not exhaustive but aims to define and describe key areas of consideration when planning and implementing a zoo-based behavioural project. We discuss the application of well-established methods (the construction of ethograms, use of time–activity patterns and measurement of space/enclosure use) as well as evaluating newer or less-widely applied analytical techniques, such as behavioural diversity indices, social networks analysis and Qualitative Behavioural Assessment. We also consider the importance of fundamental research methods, the application of pure science to understand and interpret zoo animal behaviour (with a review of a Tinbergian approach) and consideration of meta-analyses. The integration of observational techniques into experiments that aim to identify the cause and effect of behavioural performance is then explored, and we examine the assimilation of behavioural methods used in studies of environmental enrichment. By systematically studying animal behaviour, we can attempt to understand the welfare of individual animals in captivity, and here we present an example of our reviewed approaches to this area of zoo science. Combining multiple methodologies can lead to a greater understanding of behaviour and welfare, creating robust research, progressing husbandry and advancing conservation strategies. Collaborations between zoological collections and academic researchers (e.g., in Higher Education Institutions) can further refine and enhance the validity of research and husbandry practice alike

    Five ways to wellbeing at the zoo: improving human health and connection to nature

    Good mental and physical health go hand-in-hand when identifying factors that lead people to experience a better overall quality of life. A growing disconnect to the natural world is worsening the mental health of individuals in many societies. Numerous scientific publications have evidenced that being in nature and access to green and blue spaces positively impact upon humans’ physical and mental health. For many people, particularly those living in more urbanized areas, managed natural spaces and borrowed landscapes, such as those found in public parks, wildlife reserves and zoological gardens give the only opportunities for wider engagement with nature. Many zoos are designated green spaces and therefore human visitors can engage with native fauna and flora as well as exotic wild animals. This article reviews the UK Government’s “The Five Ways to Wellbeing” concept, applied to zoos and aquariums and thus suggests how zoos and aquariums can use this framework to promote positive nature-connectivity experiences for their visitors and promote good wellbeing. The Five Ways to Wellbeing are Connect, Be active, Take notice, Keep learning, and Give. We illustrate how zoos and aquariums could model their approaches to educational and engagement roles, as well as design initiatives to reach out to local communities via the Five Ways to Wellbeing concept. We show that many of the positive programs and works conducted by zoos and aquariums lend themselves to further engagement with the Five Ways to Wellbeing structure. By taking such a structured approach in the design, implementation and evaluation of their activities, zoos can expand their abilities in connecting humans with nature and further add value to their living collections of animals and plants. By including Wellbeing as a defined aim of the modern zoo, it will be clear to all of those involved in their work, visitors, workers, stakeholders, that zoos are working to promote, protect and preserve positive wellbeing outputs for humans and animals alike

    Effect of massage therapy on pain and quality of life in dogs: A cross sectional study

Quantitative efficacy data are lacking. This cross‐sectional study aimed to determine how dogs respond to canine massage therapy practiced by Canine Massage Guild UK practitioners. Methods: In 2018, case notes from a convenience sample of 527 dogs were shared, with permission from owners, by a self‐selected sample of 65 practitioners. Changes in number and severity of issues for five pain indicators (gait, posture, daily activity, behaviour, performance) and quality of life score, reported by owner and practitioners, were investigated. Results: Significant reductions in reported pain severity scores were recorded for all pain indicators over successive treatments (p < 0.001), with each treatment causing further significant reduction in pain severity. Number of pain indicators recorded over successive treatment sessions remained constant, in keeping with a cohort presenting with degenerative disease and chronic pain. All dogs and diagnostic variables responded similarly. Post‐treatment a dog was significantly more likely to have a ‘positive’ quality of life. Conclusions: This cross‐sectional study indicates canine massage therapy may effectively reduce myofascial and musculoskeletal pain severity reported by owners and practitioners associated with gait, posture, behavioural and performance issues and reduction in daily activities. Although this is not a double‐blind trial, and there is no control group, this study suggests massage therapy may be a valid treatment for myofascial and musculoskeletal pain typically derived from muscular injuries, arthritis/other orthopaedic conditions
