Interferon-stimulated gene (ISG)-expression screening reveals the specific antibunyaviral activity of ISG20
Bunyaviruses pose a significant threat to human health, prosperity and food security. In response to viral infections, interferons (IFNs) upregulate the expression of hundreds of interferon stimulated genes (ISGs) whose cumulative action can potently inhibit the replication of bunyaviruses. We used a flow cytometry-based method to screen the ability of ∼500 unique ISGs from humans and rhesus macaques to inhibit the replication of Bunyamwera orthobunyavirus (BUNV), the prototype of both the Peribunyaviridae family and Bunyavirales order. Candidates possessing antibunyaviral activity were further examined using a panel of divergent bunyaviruses. Interestingly, one candidate, ISG20, exhibited potent antibunyaviral activity against most viruses examined from the Peribunyaviridae, Hantaviridae and Nairoviridae families, whereas phleboviruses (Phenuiviridae) largely escaped inhibition. Similar to other viruses known to be targeted by ISG20, the antibunyaviral activity of ISG20 is dependent upon its functional ribonuclease activity. Through use of an infectious VLP assay (based on the BUNV minigenome system), we confirmed that gene expression from all 3 viral segments is strongly inhibited by ISG20. Using in vitro evolution, we generated a substantially ISG20-resistant BUNV and mapped the determinants of ISG20 sensitivity/resistance. Taken together, we report that ISG20 is a broad and potent antibunyaviral factor yet some bunyaviruses are remarkably ISG20 resistant. Thus, ISG20 sensitivity/resistance could influence the pathogenesis of bunyaviruses, many of which are emerging viruses of clinical or veterinary significance
Why do some coronaviruses become pandemic threats when others do not?
Despite multiple spillover events and short chains of transmission on at least 4 continents, Middle East Respiratory Syndrome Coronavirus (MERS-CoV) has never triggered a pandemic. By contrast, its relative, Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) has, despite apparently little, if any, previous circulation in humans. Resolving the unsolved mystery of the failure of MERS-CoV to trigger a pandemic could help inform how we understand the pandemic potential of pathogens, and probing it underscores a need for a more holistic understanding of the ways in which viral genetic changes scale up to population-level transmission
Photocathode Behavior During High Current Running in the Cornell ERL Photoinjector
The Cornell University Energy Recovery Linac (ERL) photoinjector has recently
demonstrated operation at 20 mA for approximately 8 hours, utilizing a
multialkali photocathode deposited on a Si substrate. We describe the recipe
for photocathode deposition, and will detail the parameters of the run.
Post-run analysis of the photocathode indicates the presence of significant
damage to the substrate, perhaps due to ion back-bombardment from the residual
beamline gas. While the exact cause of the substrate damage remains unknown, we
describe multiple surface characterization techniques (X-ray fluorescence
spectroscopy, X-ray diffraction, atomic force and scanning electron microscopy)
used to study the interesting morphological and crystallographic features of
the photocathode surface after its use for high current beam production.
Finally, we present a simple model of crystal damage due to ion
back-bombardment, which agrees qualitatively with the distribution of damage on

Physico-chemical spectroscopic mapping of the planetary nebula NGC 40 and the 2D_NEB, a new 2D algorithm to study ionised nebulae
In this paper we present an analysis of the physical and chemical conditions
of the planetary nebula NGC 40 through spatially-resolved spectroscopic maps.
We also introduce a new algorithm --2D_NEB-- based on the well-established IRAF
nebular package, which was developed to enable the use of the spectroscopic
maps to easily estimate the astrophysical quantities of ionised nebulae. The
2D_NEB was benchmarked, and we clearly show that it works properly, since it
compares nicely with the IRAF nebular software.
Using this software, we derive the maps of several physical parameters of NGC
40. From these maps, we conclude that Te[NII] shows only a slight temperature
variation from region to region, with its values constrained between ~8,000 K
and ~9,500 K. Electron densities, on the other hand, have a much more prominent
spatial variation, as Ne[SII] values vary from ~1,000 cm^(-3) to ~3,000
cm^(-3). Maps of the chemical abundances also show significant variations. From
the big picture of our work, we strongly suggest that analysis with spatial
resolution be mandatory for more complete study of the physical and chemical

Genomic and physiological footprint of the Deepwater Horizon oil spill on resident marsh fishes
The biological consequences of the Deepwater Horizon oil spill are unknown, especially for resident organisms. Here, we report results from a field study tracking the effects of contaminating oil across space and time in resident killifish during the first 4 mo of the spill event. Remote sensing and analytical chemistry identified exposures, which were linked to effects in fish characterized by genome expression and associated gill immunohistochemistry, despite very low concentrations of hydrocarbons remaining in water and tissues. Divergence in genome expression coincides with contaminating oil and is consistent with genome responses that are predictive of exposure to hydrocarbon-like chemicals and indicative of physiological and reproductive impairment. Oil-contaminated waters are also associated with aberrant protein expression in gill tissues of larval and adult fish. These data suggest that heavily weathered crude oil from the spill imparts significant biological impacts in sensitive Louisiana marshes, some of which remain for over 2 mo following initial exposures

First Plant Cell Atlas symposium report
The Plant Cell Atlas (PCA) community hosted a virtual symposium on December 9 and 10, 2021 on single cell and spatial omics technologies. The conference gathered almost 500 academic, industry, and government leaders to identify the needs and directions of the PCA community and to explore how establishing a data synthesis center would address these needs and accelerate progress. This report details the presentations and discussions focused on the possibility of a data synthesis center for a PCA and the expected impacts of such a center on advancing science and technology globally. Community discussions focused on topics such as data analysis tools and annotation standards; computational expertise and cyber-infrastructure; modes of community organization and engagement; methods for ensuring a broad reach in the PCA community; recruitment, training, and nurturing of new talent; and the overall impact of the PCA initiative. These targeted discussions facilitated dialogue among the participants to gauge whether PCA might be a vehicle for formulating a data synthesis center. The conversations also explored how online tools can be leveraged to help broaden the reach of the PCA (i.e., online contests, virtual networking, and social media stakeholder engagement) and decrease costs of conducting research (e.g., virtual REU opportunities). Major recommendations for the future of the PCA included establishing standards, creating dashboards for easy and intuitive access to data, and engaging with a broad community of stakeholders. The discussions also identified the following as being essential to the PCA’s success: identifying homologous cell-type markers and their biocuration, publishing datasets and computational pipelines, utilizing online tools for communication (such as Slack), and user-friendly data visualization and data sharing. In conclusion, the development of a data synthesis center will help the PCA community achieve these goals by providing a centralized repository for existing and new data, a platform for sharing tools, and new analytical approaches through collaborative, multidisciplinary efforts.
