    Epidemiological situation of bovine brucellosis in the State of Sergipe, Brazil

    Realizou-se um estudo para caracterizar a situação epidemiológica da brucelose bovina no Estado de Sergipe. O Estado foi estratificado em dois circuitos produtores. Em cada circuito produtor foram amostradas aleatoriamente cerca de 300 propriedades e, dentro dessas foi escolhido de forma aleatória um número pré-estabelecido de animais, dos quais foi obtida uma amostra de sangue. No total foram amostrados 4.757 animais, provenientes de 590 propriedades. Em cada propriedade amostrada foi aplicado um questionário epidemiológico para verificar o tipo de exploração da propriedade e as práticas zootécnicas e sanitárias que poderiam estar associadas ao risco de infecção pela doença. O protocolo de testes utilizado foi o da triagem com o teste do antígeno acidificado tamponado e a confirmação dos positivos com o teste do 2-mercaptoetanol. O rebanho foi considerado positivo, se pelo menos um animal foi reagente às duas provas sorológicas. A prevalência de focos e a de animais foram: 12,6% [9,2-16,0%] e 3,4% [2,3-4,4%], respectivamente. As prevalências de focos e de animais infectados para os circuitos pecuários foram: circuito 1, 11,1% [7,9-15,0%] e 2,6% [1,6-3,5%]; circuito 2, 12,9% [9,1-17,6%] e 6,2% [3,0-9,5%]. Os fatores de risco (odds ratio, OR) associados à condição de foco foram: assistência veterinária (OR= 2,89 [1,15-7,23]), tamanho do rebanho ≥30 fêmeas adultas (OR= 1,88 [1,07-3,28]) e uso de inseminação artificial (OR= 1,92 [0,84-4,38]). _______________________________________________________________________________________________________ ABSTRACTA study to characterize the epidemiological situation of bovine brucellosis in the State of Sergipe was carried out. The State was divided in two regions. Around three hundred herds were randomly sampled in each region and a pre-established number of animals were sampled in each of these herds. A total of 4,757 serum samples from 590 herds were collected. In each herd, it was applied an epidemiological questionnaire focused on herd traits as well as husbandry and sanitary practices that could be associated with the risk of infection. The serum samples were screened by the Rose-Bengal Test (RBT) and positive sera were re-tested by the 2-mercaptoethanol test (2-ME). The herd was considered positive if at least one animal was positive on both RBT and 2-ME tests. The prevalences of infected herds and animals in the State were, respectively: 12.6% [9.2-16.0%] and 3.4% [2.3-4.4%]. The prevalences of infected herds and animals in the regions were, respectively: region 1, 11.1% [7.9-15.0%] and 2.6% [1.6-3.5%]; and region 2, 12.9% [9.1-17.6%] and 6.2% [3.0-9.5%]. The risk factors associated with the presence of the infection were: veterinary assistance (OR= 2.89 [1.15-7.23]), herd size ≥30 adult females (OR= 1.88 [1.07-3.28]), and use of artificial insemination (OR=1.92 [0.84-4.38])

    Alteplase for Massive Pulmonary Embolism after Complicated Pericardiocentesis

    Background: The occurrence of a high-risk pulmonary embolism (PE) within 48 hours of a complicated pericardiocentesis to remove a haemorrhagic pericardial effusion, is an uncommon clinical challenge. Case summary: The authors report the case of a 75-year-old woman who presented with signs of imminent cardiac tamponade due to recurring idiopathic pericardial effusion. The patient underwent pericardiocentesis that was complicated by the loss of 1.5 litres of blood. Within 48 hours, the patient had collapsed with clear signs of obstructive shock. This was a life-threating situation so alteplase was administered after cardiac tamponade and hypertensive pneumothorax had been excluded. CT chest angiography later confirmed bilateral PE. The patient achieved haemodynamic stability less than an hour after receiving the alteplase. However, due to the high risk of bleeding, the medical team suspended the thrombolysis protocol and switched to unfractionated heparin within the hour. The cause of the PE was not identified despite extensive study, but after 1 year of follow-up the patient remained asymptomatic. Discussion: Despite the presence of a contraindication, the use of thrombolytic therapy in obstructive shock after exclusion of hypertensive pneumothorax can be life-saving, and low-dose thrombolytic therapy may be a valid option in such cases

    In vitro development of bovine preantral follicles in monolayer of ovarian cells

    O presente trabalho teve como objetivos determinar a influência de células ovarianas no desenvolvimento in vitro de folículos pré-antrais, avaliar a viabilidade das células ovarianas em monocamada e a influência do soro na manutenção de folículos pré-antrais in vitro. Folículos pré-antrais (FPs) e células ovarianas foram isolados de ovários de fetos bovinos, com idade entre 6 e 8 meses de gestação, oriundos de matadouro. Células ovarianas em monocamada foram cultivadas em meio TCM-199, e a viabilidade celular, após o cultivo na presença ou ausência de FSH, foi determinada com o corante vital azul de tripan. FPs foram distribuidos em quatro tratamentos e cultivados em TCM- 199 modificado, contendo soro de novilho castrado (SNC), SNC em monocamada de células ovarianas (MCO), MCO com FSH ou meio definido com álcool polivinílico (PVA) como macromolécula. A viabilidade celular não foi afetada em conseqüência da presença ou ausência de FSH. No entanto, houve um incremento significativo no tamanho dos FPs cultivados na presença de SNC, MCO e FSH (P<0,05), o que não foi observado nos demais tratamentos. A adição de FSH ao meio de cultivo não influencia a viabilidade de células ovarianas ao longo de 20 dias. Folículos pré-antrais, com área superficial entre 4000 e 8000µm2, mas não folículos com área superficial superiores a 8000µm2, crescem in vitro na presença de células ovarianas, independentemente da presença de FSH. _________________________________________________________________________________ ABSTRACTThe aim of the present work was to determine the influence of ovarian cells in the in vitro development of preantral follicles (PF). The viability of monolayer ovarian cells and the effect of the serum in the survive of in vitro PF was also investigated. Ovarian cells and PF were isolated from ovaries of bovine fetus between 6 and 8 months of pregnancy, obtained in a slaughterhouse. Monolayer of ovarian cells were cultured in a modified TCM-199 in the presence and absence of FSH and its viability after incubation was determined with Trypan Blue. PFs were divided in four different treatments, cultured in modified TCM-199, containing serum of castrated steer (SCS), SCS in monolayer of ovarian cells (MOC), MOC with FSH or a defined medium with polyvinyl alcohol (PVA) as macromolecule. The cellular viability was not affected by the presence or absence of FSH. However, PFs had a significant development (P<0.05) when cultivated in the presence of SNC, MCO and FSH, which was not observed in the other treatments. With these results, it was possible to conclude that FSH does not influence the viability of the ovarian cells when cultured in a monolayer for 20 days. Preantral follicles, measuring between 4000 and 8000µm2 but not with size greater than 8000µm2, grow on ovarian cells in vitro, independently of FSH

    Clinicopathologic data of individuals with oral lichen planus : a Brazilian case series

    The aim of the present series was to analyze the sociodemographic characteristics, clinicopathologic features, and oral health-related quality of life of 41 individuals with oral lichen planus (OLP). In a retrospective analysis (1998-2018), individuals with a clinical diagnosis of OLP from a referral service of Oral Medicine of Brazil were invited for follow-up. The individuals were assessed using the Oral Health Impact Profile-14 (OHIP-14) form. Histopathological data were reviewed according to the latest criteria proposed by the American Academy of Oral and Maxillofacial Pathology (AAOMP/2016). This series mainly consisted of females (70.7%) in their forties (31.7%). The buccal mucosa (68.2%) was the most commonly affected site. Reticular (56.1%) and erosive (34.3%) appearances were the most frequent. According to OHIP-14, individuals with OLP at multiple sites in the oral cavity showed worse values in the handicap domain and those who did not respond to corticosteroids showed a higher score on the psychological discomfort domain. The findings of the present study, using the AAOMP/2016 criteria, agree with case series and retrospective studies reported in the literature. Besides, OLP in its more severe clinical forms had an influence on patient quality of life

    First inventory of the sandfly fauna (Diptera: Psychodidae, Phlebotominae) in the municipality of Juiz de Fora, State of Minas Gerais, Brazil

    Introduction: This study aimed to inventory the phlebotomine sandfl y fauna present in the urban area of Juiz de Fora, with an emphasis on the genus Lutzomyia. Methods: Capture was performed from March to September 2012, using HP light traps placed at peridomestic sites, in a municipal kennel and a forest biome. Results: A total of 133 specimens were captured, representing eight species of the genus Lutzomyia. Lutzomyia pascalei was the most prevalent species. Conclusions: This research provides an inventory and description of the spatial locations of the phlebotomine sandfl y fauna of Juiz de Fora.

    Pesquisa malacológica de Biomphalaria em municípios da Estrada Real, situados no sudeste do Estado de Minas Gerais

    INTRODUCTION: The increasing practice of ecotourism and rural tourism in the State of Minas Gerais, Brazil, highlights the importance of studies concerning the occurrence of potential intermediate hosts of Schistosoma mansoni. This study aimed to identify species of Biomphalaria snails in municipalities along the Estrada Real, an important Brazilian tourism project. METHODS: The specimens were collected in different water collections of 36 municipalities along the Estrada Real in the southeast of the State of Minas Gerais. Biomphalaria species were characterized using both morphological and molecular approaches. The research was conducted between August 2005 and September 2009 and all the sites visited were georeferenced using GPS. RESULTS: Six Biomphalaria species were found in 30 of the 36 municipalities studied: glabrata, tenagophila, straminea, peregrina, occidentalis and schrammi. The first three species of Biomphalaria, recognized as intermediate hosts of S. mansoni, were present in 33.3%, 47.2% and 8.3% of the municipalities studied, respectively. The mollusks were found in different types of water collections and no infection by S. mansoni was detected. The highest occurrence of Biomphalaria concentration was verified in the area covered by the Caminho Novo route (Diamantina/MG to Rio de Janeiro/RJ). CONCLUSIONS: Considering the occurrence of schistosomiasis in the State of Minas Gerais and the socioeconomic repercussions involved in the Estrada Real Project, this work focuses on the vulnerability of water collections due to the presence of Biomphalaria mollusks and emphasizes the need for epidemiological surveillance and sanitary and educational measures integrated with the local community and tourism sectors.INTRODUÇÃO: O aumento das práticas de ecoturismo e turismo rural, em Minas Gerais, Brasil, evidencia a importância de se realizarem estudos sobre a ocorrência de hospedeiros intermediários do Schistosoma mansoni, no estado. O presente trabalho objetivou a busca e identificação das espécies de caramujos Biomphalaria encontrados em municípios mineiros pertencentes à Estrada Real, um importante projeto de turismo brasileiro. MÉTODOS: Os moluscos foram coletados em 36 municípios da Estrada Real, no sudeste de Minas Gerais. A pesquisa foi realizada de agosto de 2005 a setembro de 2009 e todos os locais visitados foram georreferenciados com o uso de GPS. RESULTADOS: Dos 36 municípios estudados, 30 apresentaram a ocorrência de pelo menos uma entre as seis espécies de Biomphalaria: glabrata, tenagophila, straminea, peregrina, occidentalis e schrammi. As três primeiras espécies citadas, reconhecidas como hospedeiras intermediárias do S. mansoni, estavam presentes em 33,3%, 47,2% e 8,3% dos municípios estudados, respectivamente. Os moluscos foram encontrados em diferentes tipos de coleções hídricas e em nenhum deles foi detectada infecção pelo S. mansoni. Houve maior ocorrência de Biomphalaria na área referente ao Caminho Novo (Diamantina/MG ao Rio de Janeiro/RJ). CONCLUSÕES: Considerando-se a ocorrência da esquistossomose, no Estado de Minas Gerais, e as repercussões socioeconômicas que envolvem o projeto Estrada Real, este trabalho aponta para a vulnerabilidade das coleções hídricas devido à presença de moluscos Biomphalaria e enfatiza a necessidade de vigilância epidemiológica e medidas educativas e sanitárias integradas com a comunidade local e setores de turismo

    Chemical characterization and bioactivity of phytochemicals from Iberian endemic Santolina semidentata and strategies for ex situ propagation

    Asteraceae family members are well-known for their medicinal potential, comprising several properties that make them unique among plants. Here we focus on Santolina semidentata, an endemic plant from the Iberian Peninsula, not yet described for its medicinal properties. Phytochemical characterization of S. semidentata was performed, concerning total phenol content, flavonoid content, antioxidant capacity, HPLC-DAD profile, acetylcholinesterase inhibitory capacity, cytotoxicity and neuroprotective effect in a human neurodegeneration cell model. Moreover, essential oil composition and antifungal activity were also analised. This oil might be useful for therapeutical purposes, particularly in the treatment of dermatophytosis. S. semidentata potential for neuroprotection was revealed by acetylcholinesterase inhibitory capacity and also by an effective protective effect in human neuronal cells. Furthermore, different seed conservation protocols, as well as successful in vitro propagation were established which may be useful when integrated in a broad strategy for the conservation of these endemic plants and their sustainable use for potential biotechnological applications. The results presented here greatly contribute to value this species regarding its potential as a source of phytochemicals with prospective neuroprotective health benefits, either as alternative neuroprotective drugs or as leads for synthetizing more effective molecules.The authors wish to thank to “Fundo EDP para a Biodiversidade” for financial support.
