    Toxicity induced by Solanapyrone A in Chickpea shoots and its metabolism through Glutathione/Glutathione-S-Transferase system

    Solanapyrone A and C were isolated from a Pakistani isolate of Ascochyta rabiei, Pk-1. Two experiments were conducted to investigate the phytotoxic effects of the most potent toxin, solanapyrone A on chickpea cultivars and its subsequent detoxification through glutathion/glutathion-s-transferase(GST) system. When the shoots of cultivars were fed solanapyrone A, symptoms mimicking to Ascochyta blight appeared and extent of manifestation of symptoms varied with the cultivar. In the first experiment, the effect of three different plant ages of 2 cultivars with different levels of resistance to toxin was determined in terms of GST activity unit. GST activity in Balkasar-2000 (a resistant cultivar) increased 1.92 times, 1.72 and 1.65 times in two-week-old seedling, eight-week-old and adult plants (all treated) respectively as compared to their respective controls. In the highly susceptible cultivar, AUG-424, a slight increase (1.14 times) over control was noticed in GST activity at all the three ages. In the second experiment, where shoots of three cultivars were tested against 2 doses of the toxin, an increase in GST activity in Noor-91 (a moderately susceptible cultivar) and AUG-424 was significantly less than resistant cultivar, Balkasar-2000 showing direct relationship between resistance and activity of the enzyme. It may be concluded that it is a reason for difference in response of cultivars to the disease

    Environmental health and wildlife researchwith transnational education forebodings, applicability and approach

    Pakistan, the sixth most populous country (91.71 million)in the world, is battling to keep the balance between economic growth and environmental protection. At present the government is mainly focusing on meeting the energy and food needs of the country and environmental protection is lacking serious attention. Pollution (air, water, and soil), sanitation and loss of wildlife and biodiversity are growing environmental health issues in Pakistan. In order to deliver demand led solutions to these multifaceted challenges development of institutional and technical capacities in the countryis prerequisite. Transnational Education (TNE) provides institutional mobility in tertiary education and through British Council facilitated links we were successful in developing a much needed baseline courses work and underway the research for environmental health related issues. The Government of Pakistan will certainly use the data from the research and experts will also be available to concerned departments to tackle emerging environmental problems. Additionally the understanding and concerns of TNE in academic, economic, career and socio-cultural contexts could be imperative challenges. Through TNE it was learned that other nations / regions can be benefitted from such institutes and concerned people for a true flow of knowledge for reciprocated problems. The recommendations include general considerations for policy makers which are linked with environmental health and wildlife. Among the general considerations, or overarching advice, is that policies should promote community-wide action to manage wildlife, biodiversity, environmental health and that the safety of new emissions and exposure with technologies must be assessed rather than estimated

    Somatotrophs and lactotrophs: an immunohistochemical study of Gallus domesticus pituitary gland at different stages of induced moult

    The objective of this study was to determine the distribution of somatotrophs and lactotrophs and conduct a morphometrical analysis of immunoreactive somatotrophs and lactotrophs in the pituitary glands of White Leghorn Hens (Gallus domesticus) during the period of induced moult. We divided the periods of induced moulting into three phases viz. 7, 14 and 21 days. The labeled alkalinephsphatase method with anti-GH (growth hormone) and anti-PRL (prolactin) as a primary antibody was used to detect somatotrophs and lactotrophs, in the midsagital sections of chicken adenohypophysis. Immunohistochemistry showed that somatotrophs are not only confined to the cephalo-caudal axis but can also be found in the caudal lobe; while lactotrophs were distributed in both lobes of the anterior pituitary gland at all stages of moulting (7, 14 and 21 days). Lactotrophs were of different shapes but somatotrophs were oval to round in morphology. At the given stages of induced moulting, some hypertrophied lactotrophs were also present after 7 days of induced moult in the anterior pituitary gland. However, there were moulting-related changes: from 7 to 21 days of induced moulting the immunoreactive-PRL cell population decreased, while the mean lactotroph size was more than that of somatotrophs. Basic quantitative and morphological information relating to somatotrophs and lactotrophs during the period of induced moult in laying hens is reported here and the changes brought about by induced moulting are restricted to PRL positive cells rather than GH positive cells

    Evaluation of the genetic structure of the urban dwelling species of Bank Myna (Acridotheres ginginisnus) using random amplified polymorphic DNA (RAPD) analysis

    We used the random amplified polymorphic DNA (RAPD) technique to decipher the genetic structure of the Bank Myna (Acridotheres ginginisnus) in Pakistan. The samples were collected from four cities namely: Dera Ghazi Khan, Jahanian, Khanewal and Gujranwala. The analysis showed a high genetic diversity at species level (H = 0.318, S = 0.467) but low levels at population levels (H = 0.047, S = 0.069 to H = 0.195, S = 0.283). The four populations analyzed were genetically distant from each other and hinted the effect of urbanization role in isolating the urban dwelling species. The genetic distances between populations ranged from D = 0.2403 to D = 0.3419 and the similarity coefficient showed low range from 0.66 to 0.70. This study will help in future conservation plans and also help the understanding of the role of urbanization on fragmentation of species’ natural distribution and dispersal mechanism.Key words: Acridotheres ginginisnus, Bank Myna, random amplified polymorphic DNA (RAPD), genetic structure

    Prevalence and Exposure Assessment of Aflatoxins Through Black Tea Consumption in the Multan City of Pakistan and the Impact of Tea Making Process on Aflatoxins

    Aflatoxins are the highly toxic secondary metabolites of certain fungi, being mainly produced by Aspergillus flavus and Aspergillus parasiticus. Aflatoxins are classified as group 1 category carcinogens by the International Agency for Research on Cancer (IARC). A large number of food commodities are reported to be contaminated with aflatoxins. Tea is the world’s second most consumed beverage and the consumption of tea is increasing day by day. Besides being a source of several health promoting substances, tea leaves are also reported to be contaminated with aflatoxins. However, not a single study is reported from Pakistan regarding the level of aflatoxins in commercially available black tea samples. The current study aimed to quantify the level of aflatoxins in commercially available branded and non-branded black tea samples. The estimated daily intake (EDI) of aflatoxins through branded and non-branded black tea consumption and the health risk assessment based on margin of exposure (MOE) approach was assessed. Furthermore, the impact of local tea making processes on the concentration of aflatoxins in tea beverage (filtrate) was also investigated

    Population genetic structure of Rufous-Vented Prinia (Prinia burnesii) in Pakistan

    The objective of the study is to ascertain genetic variation within Rufous-vented Prinia, Prinia burnesii an endemic species, by DNA fingerprinting applying random amplified polymorphic DNA (RAPD) technique. Genetic material was obtained from three distant sites along western bank of River Indus. These sites include Chashma barrage (32°50’N, 71°20’E), in the north; Taunsa barrage (30°45’N, 70°45’E) and the southern population from Guddu barrage (28°26’N, 69°44’E). In total, 14 RAPD primers were selected to determine the genetic variability between three populations of the species. The Nei’s (1973) genetic diversity in three populations ranged from 0.190 to 0.320. The genetic distance between Populations ranged from 0.149 to 0.265. The polymorphism levels ranged from 48 to 83% in three populations and at species level, polymorphism were 93.07%.     Cloning and expression of Toxoplasma gondii tachyzoite P22 protein

    Delay in diagnosis of Toxoplasma gondii infection in pregnant women who have been infected during the first trimester of gestation can lead to death of her fetus. Serological tests based on recombinant proteins are the main diagnosis methods for the detection of anti Toxoplasma antibody in serum samples. The aim of this study was to clone and express a gene encoding a P22 protein of T. gondii tachyzoite as using antigen for ELISA serology method. DNA was extracted from T. gondii (RH-strain) tachyzoites and PCR reaction was done using corresponding primers. The PCR product was purified, ligated to PTZ57R plasmid via T/A cloning method and subcloned into SacI and BamHI digested pET32a expression vector. Recombinant plasmid was transformed in E. coli (Bl21 DE3) and induced by 1 mM IPTG and analyzed by 12% SDS-PAGE. Expressd protein was purified by affinity chromatography and confirmed by western blot analysis. We successfully cloned and expressed T. gondii P22 protein.Key words: Toxoplasma gondii, cloning, recombinant P22

    Induction and effector phase of allergic lung inflammation is independent of CCL21/CCL19 and LT-beta

    The chemokines CCL21 and CCL19, and cell bound TNF family ligand lymphotoxin beta (LTβ), have been associated with numerous chronic inflammatory diseases. A general role in chronic inflammatory diseases cannot be assumed however; in the case of allergic inflammatory disease, CCL21/CCL19 and LTβ have not been associated with the induction, recruitment, or effector function of Th2 cells nor dendritic cells to the lung. We have examined the induction of allergic inflammatory lung disease in mice deficient in CCL21/CCL19 or LTβ and found that both kinds of mice can develop allergic lung inflammation. To control for effects of priming differences in knockout mice, adoptive transfers of Th2 cells were also performed, and they showed that such effector cells had equivalent effects on airway hyper-responsiveness in both knockout background recipients. Moreover, class II positive antigen presenting cells (B cells and CD11c+ dendritic cells) showed normal recruitment to the peribronchial spaces along with CD4 T cells. Thus, the induction of allergic responses and recruitment of both effector Th2 cells and antigen presenting cells to lung peribronchial spaces can develop independently of CCL21/CCL19 and LTβ
