    Comiteco agave (Agave americana L.) is cultivated in the Comiteca Plateau, Chiapas, México, since the middle of the 19th Century as a small-scale economic activity. Currently it is considered an option for productive reconversion in some municipalities of the Plateau; therefore, geographic areas were evaluated to determine zones of higher productive potential in nine municipalities of the state of Chiapas, through determination of agroecologic requirements, the agroclimate characterization of the Comiteca Plateau and planning of the areas with agroclimate potential. A surface of 135,869.8 ha with agroclimate potential was identified, equivalent to 17 % of the total area of the Plateau to broaden the distribution of agaveplantations.El agave Comiteco (Agave americana L.) se cultiva en la Meseta Comiteca de Chiapas, México desdemediados del siglo XIX como una actividad económica en pequeña escala.     Radiographers supporting radiologists in the interpretation of screening mammography: a viable strategy to meet the shortage in the number of radiologists.

    BackgroundAn alternative approach to the traditional model of radiologists interpreting screening mammography is necessary due to the shortage of radiologists to interpret screening mammograms in many countries.MethodsWe evaluated the performance of 15 Mexican radiographers, also known as radiologic technologists, in the interpretation of screening mammography after a 6 months training period in a screening setting. Fifteen radiographers received 6 months standardized training with radiologists in the interpretation of screening mammography using the Breast Imaging Reporting and Data System (BI-RADS) system. A challenging test set of 110 cases developed by the Breast Cancer Surveillance Consortium was used to evaluate their performance. We estimated sensitivity, specificity, false positive rates, likelihood ratio of a positive test (LR+) and the area under the subject-specific Receiver Operating Characteristic (ROC) curve (AUC) for diagnostic accuracy. A mathematical model simulating the consequences in costs and performance of two hypothetical scenarios compared to the status quo in which a radiologist reads all screening mammograms was also performed.ResultsRadiographer's sensitivity was comparable to the sensitivity scores achieved by U.S. radiologists who took the test but their false-positive rate was higher. Median sensitivity was 73.3 % (Interquartile range, IQR: 46.7-86.7 %) and the median false positive rate was 49.5 % (IQR: 34.7-57.9 %). The median LR+ was 1.4 (IQR: 1.3-1.7 %) and the median AUC was 0.6 (IQR: 0.6-0.7). A scenario in which a radiographer reads all mammograms first, and a radiologist reads only those that were difficult for the radiographer, was more cost-effective than a scenario in which either the radiographer or radiologist reads all mammograms.ConclusionsGiven the comparable sensitivity achieved by Mexican radiographers and U.S. radiologists on a test set, screening mammography interpretation by radiographers appears to be a possible adjunct to radiologists in countries with shortages of radiologists. Further studies are required to assess the effectiveness of different training programs in order to obtain acceptable screening accuracy, as well as the best approaches for the use of non-physician readers to interpret screening mammography

    Genome Degradation in Brucella ovis Corresponds with Narrowing of Its Host Range and Tissue Tropism

    Brucella ovis is a veterinary pathogen associated with epididymitis in sheep. Despite its genetic similarity to the zoonotic pathogens B. abortus, B. melitensis and B. suis, B. ovis does not cause zoonotic disease. Genomic analysis of the type strain ATCC25840 revealed a high percentage of pseudogenes and increased numbers of transposable elements compared to the zoonotic Brucella species, suggesting that genome degradation has occurred concomitant with narrowing of the host range of B. ovis. The absence of genomic island 2, encoding functions required for lipopolysaccharide biosynthesis, as well as inactivation of genes encoding urease, nutrient uptake and utilization, and outer membrane proteins may be factors contributing to the avirulence of B. ovis for humans. A 26.5 kb region of B. ovis ATCC25840 Chromosome II was absent from all the sequenced human pathogenic Brucella genomes, but was present in all of 17 B. ovis isolates tested and in three B. ceti isolates, suggesting that this DNA region may be of use for differentiating B. ovis from other Brucella spp. This is the first genomic analysis of a non-zoonotic Brucella species. The results suggest that inactivation of genes involved in nutrient acquisition and utilization, cell envelope structure and urease may have played a role in narrowing of the tissue tropism and host range of B. ovis

    De Novo Missense Substitutions in the Gene Encoding CDK8, a Regulator of the Mediator Complex, Cause a Syndromic Developmental Disorder

    The Mediator is an evolutionarily conserved, multi-subunit complex that regulates multiple steps of transcription. Mediator activity is regulated by the reversible association of a four-subunit module comprising CDK8 or CDK19 kinases, together with cyclin C, MED12 or MED12L, and MED13 or MED13L. Mutations in MED12, MED13, and MED13L were previously identified in syndromic developmental disorders with overlapping phenotypes. Here, we report CDK8 mutations (located at 13q12.13) that cause a phenotypically related disorder. Using whole-exome or whole-genome sequencing, and by international collaboration, we identified eight different heterozygous missense CDK8 substitutions, including 10 shown to have arisen de novo, in 12 unrelated subjects; a recurrent mutation, c.185C>T (p.Ser62Leu), was present in five individuals. All predicted substitutions localize to the ATP-binding pocket of the kinase domain. Affected individuals have overlapping phenotypes characterized by hypotonia, mild to moderate intellectual disability, behavioral disorders, and variable facial dysmorphism. Congenital heart disease occurred in six subjects; additional features present in multiple individuals included agenesis of the corpus callosum, ano-rectal malformations, seizures, and hearing or visual impairments. To evaluate the functional impact of the mutations, we measured phosphorylation at STAT1-Ser727, a known CDK8 substrate, in a CDK8 and CDK19 CRISPR double-knockout cell line transfected with wild-type (WT) or mutant CDK8 constructs. These experiments demonstrated a reduction in STAT1 phosphorylation by all mutants, in most cases to a similar extent as in a kinase-dead control. We conclude that missense mutations in CDK8 cause a developmental disorder that has phenotypic similarity to syndromes associated with mutations in other subunits of the Mediator kinase module, indicating probable overlap in pathogenic mechanisms

    Con el propósito de determinar las especies de agave utilizadas para la elaboración de la bebida tradicional denominada “Comiteco” en el estado de Chiapas, se realizaron recorridos de verificación en campo en ocho localidades de los municipios de Las Rosas, Carranza y Comitán de Domínguez en el estado de Chiapas, donde se ubicaron algunas poblaciones naturales y plantaciones de Agave spp., recolectando material para herbario y plantas vivas, además de entrevistar habitantes que lo usan y poseen el recurso genético. Los resultados indicaron que las especies utilizadas para la elaboración de la bebida “Comiteco” son Agave americana L. y Agave salmiana Otto ex Salm-Dyck en Chiapas

    Optimal fertilization dose in castor bean (Ricinus communis L.) using partial budgets analysis

    Objective: To determine the economically adequate fertilization dose for a castor bean (Ricinus communis L.) crop that improves the producer's benefits using the partial budgeting technique. Design / methodology / approach: The experiment was established at a site located 500 m from the Centro de Chiapas experimental field, with an altitude of 800 m. Two production factors were studied: nitrogen (40, 60, and 80) and phosphorus (20, 40, and 60) with three levels each. The treatments had nine combinations in a randomized complete block experimental design and four replications. Phenological and morphological variables, seed yield, and variable costs economic components were evaluated as a response. Results: With the average yields the obtained net benefits were determined; an adjustment of the benefits was made, and its dominance was determined. Four experiments were dominated, and five showed favorable profits for the producer. The 80-60-00 formulation had a return rate (RR) of 23 %. Conclusions: The 40-20-00, 40-40-00, 40-60-00, 80-40-00 and 80-60-00 formulations were dominant. The 80-60-00 formulation reported the highest RR, an increase in variable cost of US9.64,likeotherdosesoflowercostsandbenefits.Forthisdose,thenetbenefitsincreasewasus9.64, like other doses of lower costs and benefits.
