Lysosomal acidification dysfunction in microglia: an emerging pathogenic mechanism of neuroinflammation and neurodegeneration
Microglia are the resident innate immune cells in the brain with a major role in orchestrating immune responses. They also provide a frontline of host defense in the central nervous system (CNS) through their active phagocytic capability. Being a professional phagocyte, microglia participate in phagocytic and autophagic clearance of cellular waste and debris as well as toxic protein aggregates, which relies on optimal lysosomal acidification and function. Defective microglial lysosomal acidification leads to impaired phagocytic and autophagic functions which result in the perpetuation of neuroinflammation and progression of neurodegeneration. Reacidification of impaired lysosomes in microglia has been shown to reverse neurodegenerative pathology in Alzheimer's disease. In this review, we summarize key factors and mechanisms contributing to lysosomal acidification impairment and the associated phagocytic and autophagic dysfunction in microglia, and how these defects contribute to neuroinflammation and neurodegeneration. We further discuss techniques to monitor lysosomal pH and therapeutic agents that can reacidify impaired lysosomes in microglia under disease conditions. Finally, we propose future directions to investigate the role of microglial lysosomal acidification in lysosome-mitochondria crosstalk and in neuron-glia interaction for more comprehensive understanding of its broader CNS physiological and pathological implications
Prognostic potential of body composition indices in detecting risk of musculoskeletal injury in army officer cadet profiles.
OBJECTIVES: High values in most of the body composition indices have been related to musculoskeletal injuries, but limited data exists on the accuracy of these diagnoses when detecting musculoskeletal injuries in military populations. METHODS: The suitability of body fat percentage, body mass index, fat mass index and fat free mass index to identify injury risk was examined in a group of army officer recruits. All body composition diagnoses were measured in 268 male army officer recruits prior to the commencement of basic combat training. Musculoskeletal injury was identified using codes from the International Classification of Diseases. The area under the curve, in the receiver operating characteristic curve, was used to quantify the overall ability to discriminate between those who were injured and those who were not. RESULTS: The statistics indicated that all indices, apart from body mass index, had a significant possibility to detect musculoskeletal injury potential (p 22, for fat mass index >6.5 and for fat free mass index <16.5. CONCLUSION: Body mass index values can not similarly detect the possibility of occurrence of musculoskeletal injuries in army officer recruits, just as other body composition diagnoses related to fat mass or/and free fat mass. However, the cut off-points related to the overall diagnostic performance of each body composition index should be used with caution and in accordance with the aims of each experimental setting
Annually resolved North Atlantic marine climate over the last millennium

Available from Nature Publishing Group via the DOI in this record.Owing to the lack of absolutely dated oceanographic information before the modern instrumental period, there is currently significant debate as to the role played by North Atlantic Ocean dynamics in previous climate transitions (for example, Medieval Climate Anomaly-Little Ice Age, MCA-LIA). Here we present analyses of a millennial-length, annually resolved and absolutely dated marine ÎŽ(18)O archive. We interpret our record of oxygen isotope ratios from the shells of the long-lived marine bivalve Arctica islandica (ÎŽ(18)O-shell), from the North Icelandic shelf, in relation to seawater density variability and demonstrate that solar and volcanic forcing coupled with ocean circulation dynamics are key drivers of climate variability over the last millennium. During the pre-industrial period (AD 1000-1800) variability in the sub-polar North Atlantic leads changes in Northern Hemisphere surface air temperatures at multi-decadal timescales, indicating that North Atlantic Ocean dynamics played an active role in modulating the response of the atmosphere to solar and volcanic forcing.We thank the members of the RV Bjarni SĂŠmundsson (Cruise No. B05-2006). A CDCL-style calculus for solving non-linear constraints
In this paper we propose a novel approach for checking satisfiability of
non-linear constraints over the reals, called ksmt. The procedure is based on
conflict resolution in CDCL style calculus, using a composition of symbolical
and numerical methods. To deal with the non-linear components in case of
conflicts we use numerically constructed restricted linearisations. This
approach covers a large number of computable non-linear real functions such as
polynomials, rational or trigonometrical functions and beyond. A prototypical
implementation has been evaluated on several non-linear SMT-LIB examples and

A modern multicentennial record of radiocarbon variability from an exactly dated bivalve chronology at the Tree Nob site (Alaska coastal current)

Available from Cambridge University Press via the DOI in this record.âŻQuantifying the marine radiocarbon reservoir effect, offsets (ÎR), and ÎR variability over time is critical to improving dating estimates of marine samples while also providing a proxy of water mass dynamics. In the northeastern Pacific, where no high-resolution time series of ÎR has yet been established, we sampled radiocarbon (14C) from exactly dated growth increments in a multicentennial chronology of the long-lived bivalve, Pacific geoduck (Paneopea generosa) at the Tree Nob site, coastal British Columbia, Canada. Samples were taken at approximately decadal time intervals from 1725 CE to 1920 CE and indicate average ÎR values of 256 ± 22 years (1Ï) consistent with existing discrete estimates. Temporal variability in ÎR is small relative to analogous Atlantic records except for an unusually old-water event, 1802â1812. The correlation between ÎR and sea surface temperature (SST) reconstructed from geoduck increment width is weakly significant (r2 = .29, p = .03), indicating warm water is generally old, when the 1802â1812 interval is excluded. This interval contains the oldest (â2.1Ï) anomaly, and that is coincident with the coldest (â2.7Ï) anomalies of the temperature reconstruction. An additional 32 14C values spanning 1952â1980 were detrended using a northeastern Pacific bomb pulse curve. Significant positive correlations were identified between the detrended 14C data and annual El Niño Southern Oscillation (ENSO) and summer SST such that cooler conditions are associated with older water. Thus, 14C is generally relatively stable with weak, potentially inconsistent associations to climate variables, but capable of infrequent excursions as illustrated by the unusually cold, old-water 1802â1812 interval.National Science FoundationNational Science Foundatio

underutilised in physiotherapy research. This article aims to introduce interpretative phenomenological analysis
(IPA), a research methodology oriented toward exploring and understanding the experience of a particular
phenomenon (e.g., living with spinal cord injury or chronic pain, or being the carer of someone with a particular
health condition). Researchers using IPA try to find out how people make sense of their experiences and the
meanings they attach to them. The findings from IPA research are highly nuanced and offer a fine grained
understanding that can be used to contextualise existing quantitative research, to inform understanding of novel
or underresearched topics or, in their own right, to provoke a reappraisal of what is considered known about
a specified phenomenon. We advocate IPA as a useful and accessible approach to qualitative research that
can be used in the clinical setting to inform physiotherapy practice and the development of services from the

Attention modulates adaptive motor learning in the âbroken escalatorâ paradigm
The physical stumble caused by stepping onto a stationary (broken) escalator represents a locomotor aftereffect (LAE) that attests to a process of adaptive motor learning. Whether such learning is primarily explicit (requiring attention resources) or implicit (independent of attention) is unknown. To address this question, we diverted attention in the adaptation (MOVING) and aftereffect (AFTER) phases of the LAE by loading these phases with a secondary cognitive task (sequential naming of a vegetable, fruit and a colour). Thirty-six healthy adults were randomly assigned to 3 equally sized groups. They performed 5 trials stepping onto a stationary sled (BEFORE), 5 with the sled moving (MOVING) and 5 with the sled stationary again (AFTER). A 'Dual-Task-MOVING (DTM)' group performed the dual-task in the MOVING phase and the 'Dual-Task-AFTEREFFECT (DTAE)' group in the AFTER phase. The 'control' group performed no dual task. We recorded trunk displacement, gait velocity and gastrocnemius muscle EMG of the left (leading) leg. The DTM, but not the DTAE group, had larger trunk displacement during the MOVING phase, and a smaller trunk displacement aftereffect compared with controls. Gait velocity was unaffected by the secondary cognitive task in either group. Thus, adaptive locomotor learning involves explicit learning, whereas the expression of the aftereffect is automatic (implicit). During rehabilitation, patients should be actively encouraged to maintain maximal attention when learning new or challenging locomotor tasks
Critical evaluation of current data analysis strategies for psychophysiological measures of fear conditioning and extinction in humans
Fear conditioning and extinction is a construct integral to understanding trauma-, stress- and anxiety-related disorders. In the laboratory, associative learning paradigms that pair aversive with neutral stimuli are used as analogues to real-life fear learning. These studies use physiological indices, such as skin conductance, to sensitively measure rates and intensity of learning and extinction. In this review, we discuss some of the potential limitations in interpreting and analysing physiological data during the acquisition or extinction of conditioned fear. We argue that the utmost attention should be paid to the development of modelling approaches of physiological data in associative learning paradigms, by illustrating the lack of replicability and interpretability of results in current methods. We also show that statistical significance may be easily achieved in this paradigm without more stringent data and data analysis reporting requirements, leaving this particular field vulnerable to misleading conclusions. This review is written so that issues and potential solutions are accessible to researchers without mathematical training. We conclude the review with some suggestions that all laboratories should be able to implement, including visualising the full data set in publications and adopting modelling, or at least regression-based, approaches
Serologic evidence of human orthopoxvirus infections in Sierra Leone
<p>Abstract</p> <p>Background</p> <p>Orthopoxviruses, including variola virus, vaccinia virus, and monkeypox virus, have previously been documented in humans in West Africa, however, no cases of human orthopoxvirus infection have been reported in the region since 1986. We conducted a serosurvey to determine whether human exposure to orthopoxviruses continues to occur in eastern Sierra Leone.</p> <p>Findings</p> <p>To examine evidence of exposure to orthopoxviruses in the Kenema District of Sierra Leone, we collected and tested sera from 1596 persons by IgG ELISA and a subset of 313 by IgM capture ELISA. Eleven persons born after the cessation of smallpox vaccination had high orthopoxvirus-specific IgG values, and an additional 6 persons had positive IgM responses. No geographic clustering was noted.</p> <p>Conclusions</p> <p>These data suggest that orthopoxviruses continue to circulate in Sierra Leone. Studies aimed at obtaining orthopoxvirus isolates and/or genetic sequences from rodents and symptomatic humans in the area are indicated.</p
Testing the effectiveness of the forest integrity assessment: A fieldâbased tool for estimating the condition of tropical forest
1. Global targets to halt biodiversity losses and mitigate climate change will require protecting rainforest beyond current protected area networks, necessitating responsible forest stewardship from a diverse range of companies, communities and private individuals. Robust assessments of forest condition are critical for successful forest management, but many existing techniques are highly technical, time-consuming, expensive or require specialist knowledge.
2. To make assessment of tropical forests accessible to a wide range of actors, many of whom may be limited by resources or expertise, the High Conservation Value Resource Network (HCVRN), with the SE Asia Rainforest Research Partnership (SEARRP), developed a South East Asian version of the Forest Integrity Assessment (FIA) tool as a rapid (< 1 hour) method of assessing forest condition in the field, where non-experts respond to 50 questions about characteristics of the local environment while walking a site transect. Here, we examined the effectiveness of this survey tool by conducting ⌠1000 assessments of forest condition at 16 tropical rainforest sites with varying levels of disturbance in Sabah, Malaysian Borneo.
3. We found good agreement (R-squared range: 0.50â0.78) between FIA survey scores and independent measures of forest condition, including biodiversity, vegetation structure, aboveground carbon and other key metrics of ecosystem function, indicating that the tool performed well. Although there was variation among assessor responses when surveying the same forest sites, assessors were consistent in their ranking of those sites, and prior forest knowledge had a minimal effect on the FIA scores. Revisions or further training for questions where assessors disagree, for example, on the presence of fauna at a site, could improve consistency.
4. We conclude that the FIA survey tool is a robust method of assessing forest condition, providing a rapid and accessible means of forest conservation assessment. The FIA tool could be incorporated into management practices in a wide range of forest conservation schemes, from sustainability standards, to community forestry and restoration initiatives. The tool will enable more organizations and individuals to understand the conservation value of the forests they manage and to identify areas for targeted improvements
