Iron abundances from optical Fe III absorption lines in B-type stellar spectra
The role of optical Fe III absorption lines in B-type stars as iron abundance
diagnostics is considered. To date, ultraviolet Fe lines have been widely used
in B-type stars, although line blending can severely hinder their diagnostic
power. Using optical spectra, covering a wavelength range ~ 3560 - 9200 A, a
sample of Galactic B-type main-sequence and supergiant stars of spectral types
B0.5 to B7 are investigated. A comparison of the observed Fe III spectra of
supergiants, and those predicted from the model atmosphere codes TLUSTY
(plane-parallel, non-LTE), with spectra generated using SYNSPEC (LTE), and
CMFGEN (spherical, non-LTE), reveal that non-LTE effects appear small. In
addition, a sample of main-sequence and supergiant objects, observed with
FEROS, reveal LTE abundance estimates consistent with the Galactic environment
and previous optical studies. Based on the present study, we list a number of
Fe III transitions which we recommend for estimating the iron abundance from

Identification of full length bovine TLR1 and functional characterization of lipopeptide recognition by bovine TLR2/1 heterodimer
Toll-like receptors (TLR) are highly conserved pattern recognition receptors of the innate immune system. Toll-like receptor 2 (TLR2) recognizes bacterial lipopeptides in a heterodimeric complex with TLR6 or TLR1, thereby discriminating between di- or triacylated lipopeptides, respectively. Previously, we found that HEK293 cells transfected with bovine TLR2 (boTLR2) were able to respond to diacylated lipopeptides but did not recognize triacylated lipopeptides, even after cotransfection with the so far published sequence of boTLR1. In this study we now could show that primary bovine cells were in general able to detect triacylated lipopetides. A closer investigation of the boTLR1 gene locus revealed an additional ATG 195 base pairs upstream from the published start codon. Its transcription would result in an N-terminus with high identity to human and murine TLR1 (huTLR1, muTLR1). Cloning and cotransfection of this longer boTLR1 with boTLR2 now resulted in the recognition of triacylated lipopeptides by HEK293 cells, thereby resembling the ex vivo observation. Analysis of the structure-activity relationship showed that the ester-bound acid chains of these lipopeptides need to consist of at least 12 carbon atoms to activate the bovine heterodimer showing similarity to the recognition by huTLR2/huTLR1. In contrast, HEK293 cell cotransfected with muTLR2 and muTLR1 could already be activated by lipopeptides with shorter fatty acids of only 6 carbon atoms. Thus, our data indicate that the additional N-terminal nucleotides belong to the full length and functionally active boTLR1 (boTLR1-fl) which participates in a species-specific recognition of bacterial lipopeptides
Characterization of a Legionella pneumophila gene encoding a lipoprotein antigen
A prominent 19kDa surface antigen of Legionella pneumophila , cloned in Escherichia coli , was found to be intimately associated with peptidoglycan. The DNA region encoding this antigen was mapped on an 11.9kb plasmid by means of deletion analysis and transposon mutagenesis. PhoA + gene fusions, generated by Tn phoA insertions into this region, confirmed the presence of a gene encoding a secreted protein. PhoA + transposon insertions were also associated with loss of the 19 kDa antigen in immunoassay s using a monoclonal antibody (mAb1E9) and the replacement of the 19kDa antigen with larger fusion proteins in immunoblots using Legionella immune serum. A 1540bp PstI fragment carrying the gene was sequenced, and the open reading frame encoding the antigen was identified. The gene encodes a polypeptide 176 amino acid residues long and 18913Da in size. The presence of a signal sequence of 22 amino acids with a consensus sequence for cleavage by signal peptidase II indicates that the antigen is a lipoprotein, and striking similarity with peptidoglycan-associated lipoproteins (PALs) from E. coli (51% amino acid homology) and Haemophilus influenzae (55% homology) is noted. We conclude that the 19kDa antigen of L. pneumophila is the structural equivalent of the PAL found in other Gram-negative species and suggest that its post-translational acylation may explain its potency as an immunogen.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/75712/1/j.1365-2958.1991.tb00824.x.pd
A-type Supergiant Abundances in the SMC: Probes of Evolution
New abundances of N, O, Na, Mg, Si, Ca, Sc, Ti, Cr, Fe, Sr, Zr, and Ba are
presented for 10 A-type supergiants in the SMC, plus upper limits for C. In
interpreting the CNO results for constraints on stellar evolution theories,
careful attention has been paid to the comparison abundances, i.e., the present
day abundances of SMC nebulae and B-dwarf stars. These new results are also
compared to published results from F-K supergiant analyses, and found to be in
good agreement when both sets of data are carefully examined as differential
(SMC minus Galactic standard) abundances.
With the exception of nitrogen, very small star-to-star abundance variations
are found for all elements in this analysis. The N variations are not predicted
by standard stellar evolution models. Instead, the results support the new
predictions reported from rotating stellar models, where the range in nitrogen
is the result of partial mixing of CN-cycled gas from the stellar interior due
to main-sequence rotation at different rates (c.f., Langer & Heger 1998). The
overall overabundance of nitrogen in the sampled stars also implies these stars
have undergone the first dredge-up in addition to having been mixed while on
the main-sequence.
The alpha-elements (O, Mg, Si, Ca, Ti) have similar underabundances to Fe,
which is not the same as seen in metal-poor stars in the solar neighborhood of
the Galaxy. In addition, certain light s-process elements (Zr, Ba) are slightly
more underabundant than Fe, which is predicted by the bursting chemical
evolution model presented by Pagel & Tautvaisiene (1998) for the SMC.Comment: Accepted for publication in the Astrophysical Journal, Manuscript

Tato diplomová práce se zabývá procesem digitalizace konkrétního produktu. Teoretická část popisuje nejprve postavení zákazníka na trhu a jeho vztah s podnikem, následně se soustředí na vymezení pojmů produkt a distribuce a jejich zasazení do kontextu této práce. Dále je rozebíráno téma životního pojištění se svými aktuálními trendy. Praktická část se soustředí kolem projektu digitalizace produktu, jeho ovlivňujících faktorů a názorů vybraných respondentů, následně je schematicky naznačen proces takového prodejního kanálu. Závěr práce obsahuje závěrečné zhodnocení přínosu projektu a souhrn daných doporučení.This diploma thesis deals with the digitization process of a concrete product. Theoreti-cal part describes the position of a customer in the market and his connection to the company. Then it focuses on defining the terms of product and distribution and their placement in the context of this thesis. The topic of life insurance with its current trends is also discussed. The practical part targets the process of digitization of a prod-uct, its influential factors and the opinions based on answers by selected respondents. Then the process of this kind of selling channel is schematically represented and de-scribed. The conclusion of the thesis contains a final evaluation of the project's contri-bution and summary of given recommendations

Teoretická část popisuje nejprve obecně systém odměňování a mzdové formy, posléze se soustřeďuje na definici motivace a motivační teorie. V praktické části je zkoumán systém odměňování konkrétní organizace, následné dotazníkové šetření je zaměřeno na zjištění jeho nedostatků. Závěr práce obsahuje návrhy vedoucí k možnému zvýšení motivace zaměstnanců.This Bachelor thesis deals with relationship between compensation and motivation. Theoretical part describes at first system of compensation and payroll forms in general and later focuses on definition of motivation and motivational theories. In practical part there is a specific system of compensation researched and following questionnaire investigation pays attention to finding the deficits. The final part contains suggestions which will eventually lead to increase of employee's motivation
