    Threat of Foreign Arthropod-Borne Pathogens to Livestock in the United States

    There are many exotic animal pathogens throughout the world that, if introduced into the United States, could have a significant detrimental impact on the health of livestock, agricultural economy, the environment, and public health. Many of these pathogens are arthropod-borne and potential vectors are readily available in the United States. A number of these arthropod-borne pathogens are discussed here as examples that illustrate the potential risk and the consequences of inadvertent introductions. Several International agencies have a role in global surveillance and in controlling animal diseases should they begin to expand their range. The risk to the United States is considerable. We propose that the United States invest in the improved infrastructure needed to reduce the risk of foreign arthropod-borne pathogens. Current U.S. programs focus on the exclusion of pathogens through regulation of animal movements and products, surveillance, especially trained animal disease diagnosticians, research support, international cooperation and, should pathogens enter our country, the resources for their prompt eradication. We suggest that the United States needs to develop a comprehensive, updated strategic plan to assess all aspects of current and future requirements, objectives, and resources needed to protect its national interests

    Honokiol Blocks Store Operated Calcium Entry in CHO Cells Expressing the M3 Muscarinic Receptor: Honokiol and Muscarinic Signaling

    Background: Honokiol, a cell-permeable phenolic compound derived from the bark of magnolia trees and present in Asian herbal teas, has a unique array of pharmacological actions, including the inhibition of multiple autonomic responses. We determined the effects of honokiol on calcium signaling underlying transmission mediated by human M3 muscarinic receptors expressed in Chinese hamster ovary (CHO) cells. Receptor binding was determined in radiolabelled ligand binding assays; changes in intracellular calcium concentrations were determined using a fura-2 ratiometric imaging protocol; cytotoxicity was determined using a dye reduction assay. Results: Honokiol had a potent (EC50 ≈ 5 μmol/l) inhibitory effect on store operated calcium entry (SOCE) that was induced by activation of the M3 receptors. This effect was specific, rapid and partially reversible, and was seen at concentrations not associated with cytotoxicity, inhibition of IP3 receptor-mediated calcium release, depletion of ER calcium stores, or disruption of M3 receptor binding. Conclusions: It is likely that an inhibition of SOCE contributes to honokiol disruption of parasympathetic motor functions, as well as many of its beneficial pharmacological properties

    The Evolution of Quasar CIV and SiIV Broad Absorption Lines Over Multi-Year Time Scales

    We investigate the variability of CIV 1549A broad absorption line (BAL) troughs over rest-frame time scales of up to ~7 yr in 14 quasars at redshifts z>2.1. For 9 sources at sufficiently high redshift, we also compare CIV and SiIV 1400A absorption variation. We compare shorter- and longer-term variability using spectra from up to four different epochs per source and find complex patterns of variation in the sample overall. The scatter in the change of absorption equivalent width (EW), Delta EW, increases with the time between observations. BALs do not, in general, strengthen or weaken monotonically, and variation observed over shorter (<months) time scales is not predictive of multi-year variation. We find no evidence for asymmetry in the distribution of Delta EW that would indicate that BALs form and decay on different time scales, and we constrain the typical BAL lifetime to be >~30 yr. The BAL absorption for one source, LBQS 0022+0150, has weakened and may now be classified as a mini-BAL. Another source, 1235+1453, shows evidence of variable, blue continuum emission that is relatively unabsorbed by the BAL outflow. CIV and SiIV BAL shape changes are related in at least some sources. Given their high velocities, BAL outflows apparently traverse large spatial regions and may interact with parsec-scale structures such as an obscuring torus. Assuming BAL outflows are launched from a rotating accretion disk, notable azimuthal symmetry is required in the outflow to explain the relatively small changes observed in velocity structure over times up to 7 yr

    The X-Ray Properties of the Optically Brightest Mini-BAL Quasars from the Sloan Digital Sky Survey

    We have compiled a sample of 14 of the optically brightest radio-quiet quasars (mim_{i}~\le~17.5 and zz~\ge~1.9) in the Sloan Digital Sky Survey Data Release 5 quasar catalog that have C IV mini-BALs present in their spectra. X-ray data for 12 of the objects were obtained via a Chandra snapshot survey using ACIS-S, while data for the other two quasars were obtained from archival XMM-Newton observations. Joint X-ray spectral analysis shows the mini-BAL quasars have a similar average power-law photon index (Γ1.9\Gamma\approx1.9) and level of intrinsic absorption (NH8×1021 cm2N_H \lesssim 8\times 10^{21} \ {\rm cm}^{-2}) as non-BMB (neither BAL nor mini-BAL) quasars. Mini-BAL quasars are more similar to non-BMB quasars than to BAL quasars in their distribution of relative X-ray brightness (assessed with Δαox\Delta\alpha_{\rm ox}). Relative colors indicate mild dust reddening in the optical spectra of mini-BAL quasars. Significant correlations between Δαox\Delta\alpha_{\rm ox} and UV absorption properties are confirmed for a sample of 56 sources combining mini-BAL and BAL quasars with high signal-to-noise ratio rest-frame UV spectra, which generally supports models in which X-ray absorption is important in enabling driving of the UV absorption-line wind. We also propose alternative parametrizations of the UV absorption properties of mini-BAL and BAL quasars, which may better describe the broad absorption troughs in some respects.Comment: ApJ accepted; 21 pages, 11 figures, and 9 table

    Oxidative Stress Disruption of Receptor-Mediated Calcium Signaling Mechanisms

    Background: Oxidative stress increases the cytosolic content of calcium in the cytoplasm through a combination of effects on calcium pumps, exchangers, channels and binding proteins. In this study, oxidative stress was produced by exposure to tert-butyl hydroperoxide (tBHP); cell viability was assessed using a dye reduction assay; receptor binding was characterized using [3H]N-methylscopolamine ([3H]MS); and cytosolic and luminal endoplasmic reticulum (ER) calcium concentrations ([Ca2+]i and [Ca2+]L, respectively) were measured by fluorescent imaging. Results: Activation of M3 muscarinic receptors induced a biphasic increase in [Ca2+]i: an initial, inositol trisphosphate (IP3)-mediated release of Ca2+ from endoplasmic reticulum (ER) stores followed by a sustained phase of Ca2+ entry (i.e., store-operated calcium entry; SOCE). Under non-cytotoxic conditions, tBHP increased resting [Ca2+]i; a 90 minute exposure to tBHP (0.5-10 mM) increased [Ca2+]i from 26 to up to 127 nM and decreased [Ca2+]L by 55%. The initial response to 10 μM carbamylcholine was depressed by tBHP in the absence, but not the presence, of extracellular calcium. SOCE, however, was depressed in both the presence and absence of extracellular calcium. Acute exposure to tBHP did not block calcium influx through open SOCE channels. Activation of SOCE following thapsigargin-induced depletion of ER calcium was depressed by tBHP exposure. In calcium-free media, tBHP depressed both SOCE and the extent of thapsigargin-induced release of Ca2+ from the ER. M3 receptor binding parameters (ligand affinity, guanine nucleotide sensitivity, allosteric modulation) were not affected by exposure to tBHP. Conclusions: Oxidative stress induced by tBHP affected several aspects of M3 receptor signaling pathway in CHO cells, including resting [Ca2+]i, [Ca2+]L, IP3 receptor mediated release of calcium from the ER, and calcium entry through the SOCE. tBHP had little effect on M3 receptor binding or G protein coupling. Thus, oxidative stress affects multiple aspects of calcium homeostasis and calcium dependent signaling

    Prophylactic methylprednisolone to reduce inflammation and improve outcomes from one lung ventilation in children: a randomized clinical trial.

    BACKGROUND: One lung ventilation (OLV) results in inflammatory and mechanical injury, leading to intraoperative and postoperative complications in children. No interventions have been studied in children to minimize such injury. OBJECTIVE: We hypothesized that a single 2-mg·kg(-1) dose of methylprednisolone given 45-60 min prior to lung collapse would minimize injury from OLV and improve physiological stability. METHODS: Twenty-eight children scheduled to undergo OLV were randomly assigned to receive 2 mg·kg(-1) methylprednisolone (MP) or normal saline (placebo group) prior to OLV. Anesthetic management was standardized, and data were collected for physiological stability (bronchospasm, respiratory resistance, and compliance). Plasma was assayed for inflammatory markers related to lung injury at timed intervals related to administration of methylprednisolone. RESULTS: Three children in the placebo group experienced clinically significant intraoperative and postoperative respiratory complications. Respiratory resistance was lower (P = 0.04) in the methylprednisolone group. Pro-inflammatory cytokine IL-6 was lower (P = 0.01), and anti-inflammatory cytokine IL-10 was higher (P = 0.001) in the methylprednisolone group. Tryptase, measured before and after OLV, was lower (P = 0.03) in the methylprednisolone group while increased levels of tryptase were seen in placebo group after OLV (did not achieve significance). There were no side effects observed that could be attributed to methylprednisolone in this study. CONCLUSIONS: Methylprednisolone at 2 mg·kg(-1) given as a single dose prior to OLV provides physiological stability to children undergoing OLV. In addition, methylprednisolone results in lower pro-inflammatory markers and higher anti-inflammatory markers in the children\u27s plasma

    On the variability of quasars: a link between Eddington ratio and optical variability?

    Repeat scans by the Sloan Digital Sky Survey (SDSS) of a 278 square degree stripe along the Celestial equator have yielded an average of over 10 observations each for nearly 8,000 spectroscopically confirmed quasars. Over 2500 of these quasars are in the redshift range such that the CIV emission line is visible in the SDSS spectrum. Utilising the width of these CIV lines and the luminosity of the nearby continuum, we estimate black hole masses for these objects. In an effort to isolate the effects of black hole mass and luminosity on the photometric variability of our dataset, we create several subsamples by binning in these two physical parameters. By comparing the ensemble structure functions of the quasars in these bins, we are able to reproduce the well-known anticorrelation between luminosity and variability, now showing that this anticorrelation is independent of the black hole mass. In addition, we find a correlation between variability and the mass of the central black hole. By combining these two relations, we identify the Eddington ratio as a possible driver of quasar variability, most likely due to differences in accretion efficiency.Comment: 13 pages, 5 figures, Accepted for publication in MNRA
