    Negotiation is a method for people to settle differences and achieve solutions. While formal negotiations can take the forms of conflict resolutions or licensing agreement with vendor negotiations, there are many informal forms of negotiation that take place every day. For example, two co-workers discussing the respective work responsibilities in a project that they are working on is a form of negotiation; a librarian asking for the supervisor’s approval to attend a professional conference is also a form of negotiation; two friend deciding where to go to eat lunch is a form of negotiation. As the purpose of negotiation is to reach mutually beneficial solutions, it is a communication skillset that is highly valuable in the professional setting. Therefore, understanding library employees’ perception and habits of negotiation can potentially reveal any gaps or needs for professional develop in this area

    A Preliminary Investigation towards the Risk Stratification of Allogeneic Stem Cell Recipients with Respect to the Potential for Development of GVHD via Their Pre-Transplant Plasma Lipid and Metabolic Signature

    The clinical outcome of allogeneic hematopoietic stem cell transplantation (SCT) may be influenced by the metabolic status of the recipient following conditioning, which in turn may enable risk stratification with respect to the development of transplant-associated complications such as graft vs. host disease (GVHD). To better understand the impact of the metabolic profile of transplant recipients on post-transplant alloreactivity, we investigated the metabolic signature of 14 patients undergoing myeloablative conditioning followed by either human leukocyte antigen (HLA)-matched related or unrelated donor SCT, or autologous SCT. Blood samples were taken following conditioning and prior to transplant on day 0 and the plasma was comprehensively characterized with respect to its lipidome and metabolome via liquid chromatography/mass spectrometry (LCMS) and gas chromatography/mass spectrometry (GCMS). A pro-inflammatory metabolic profile was observed in patients who eventually developed GVHD. Five potential pre-transplant biomarkers, 2-aminobutyric acid, 1-monopalmitin, diacylglycerols (DG 38:5, DG 38:6), and fatty acid FA 20:1 demonstrated high sensitivity and specificity towards predicting post-transplant GVHD. The resulting predictive model demonstrated an estimated predictive accuracy of risk stratification of 100%, with area under the curve of the ROC of 0.995. The likelihood ratio of 1-monopalmitin (infinity), DG 38:5 (6.0), and DG 38:6 (6.0) also demonstrated that a patient with a positive test result for these biomarkers following conditioning and prior to transplant will be at risk of developing GVHD. Collectively, the data suggest the possibility that pre-transplant metabolic signature may be used for risk stratification of SCT recipients with respect to development of alloreactivity

    Layer specific and general requirements for ERK/MAPK signaling in the developing neocortex

    Aberrant signaling through the Raf/MEK/ERK (ERK/MAPK) pathway causes pathology in a family of neurodevelopmental disorders known as 'RASopathies' and is implicated in autism pathogenesis. Here, we have determined the functions of ERK/MAPK signaling in developing neocortical excitatory neurons. Our data reveal a critical requirement for ERK/MAPK signaling in the morphological development and survival of large Ctip2+ neurons in layer 5. Loss of Map2k1/2 (Mek1/2) led to deficits in corticospinal tract formation and subsequent corticospinal neuron apoptosis. ERK/MAPK hyperactivation also led to reduced corticospinal axon elongation, but was associated with enhanced arborization. ERK/MAPK signaling was dispensable for axonal outgrowth of layer 2/3 callosal neurons. However, Map2k1/2 deletion led to reduced expression of Arc and enhanced intrinsic excitability in both layers 2/3 and 5, in addition to imbalanced synaptic excitation and inhibition. These data demonstrate selective requirements for ERK/MAPK signaling in layer 5 circuit development and general effects on cortical pyramidal neuron excitability

    Miniature exoplanet radial velocity array I: design, commissioning, and early photometric results

    The MINiature Exoplanet Radial Velocity Array (MINERVA) is a US-based observational facility dedicated to the discovery and characterization of exoplanets around a nearby sample of bright stars. MINERVA employs a robotic array of four 0.7 m telescopes outfitted for both high-resolution spec- troscopy and photometry, and is designed for completely autonomous operation. The primary science program is a dedicated radial velocity survey and the secondary science objective is to obtain high precision transit light curves. The modular design of the facility and the flexibility of our hardware allows for both science programs to be pursued simultaneously, while the robotic control software provides a robust and efficient means to carry out nightly observations. In this article, we describe the design of MINERVA including major hardware components, software, and science goals. The telescopes and photometry cameras are characterized at our test facility on the Caltech campus in Pasadena, CA, and their on-sky performance is validated. New observations from our test facility demonstrate sub-mmag photometric precision of one of our radial velocity survey targets, and we present new transit observations and fits of WASP-52b—a known hot-Jupiter with an inflated radius and misaligned orbit. The process of relocating the MINERVA hardware to its final destination at the Fred Lawrence Whipple Observatory in southern Arizona has begun, and science operations are expected to commence within 2015

    Characterization of the Atmosphere of the Hot Jupiter HAT-P-32Ab and the M-dwarf Companion HAT-P-32B

We carried out adaptive optics imaging of the planet host star HAT-P-32A and its companion HAT-P-32B in the near-IR and the visible. We clearly resolve the two stars from each other and find a separation of 2.''923 ± 0.''004 and a position angle 110fdg64 ± 0fdg12. We measure the flux ratios of the binary in g'r'i'z' and H and KS bands, and determine T eff= 3565 ± 82 K for the companion star, corresponding to an M1.5 dwarf. We use PHOENIX stellar atmosphere models to correct the dilution of the secondary eclipse depths of the hot Jupiter due to the presence of the M1.5 companion. We also improve the secondary eclipse photometry by accounting for the non-classical, flux-dependent nonlinearity of the WIRC IR detector in the H band. We measure planet-to-star flux ratios of 0.090% ± 0.033%, 0.178% ± 0.057%, 0.364% ± 0.016%, and 0.438% ± 0.020% in the H, KS , 3.6 and 4.5 μm bands, respectively. We compare these with planetary atmospheric models, and find they prefer an atmosphere with a temperature inversion and inefficient heat redistribution. However, we also find that the data are equally well described by a blackbody model for the planet with T p = 2042 ± 50 K.     TIGIT Marks Exhausted T Cells, Correlates with Disease Progression, and Serves as a Target for Immune Restoration in HIV and SIV Infection.

    HIV infection induces phenotypic and functional changes to CD8+ T cells defined by the coordinated upregulation of a series of negative checkpoint receptors that eventually result in T cell exhaustion and failure to control viral replication. We report that effector CD8+ T cells during HIV infection in blood and SIV infection in lymphoid tissue exhibit higher levels of the negative checkpoint receptor TIGIT. Increased frequencies of TIGIT+ and TIGIT+ PD-1+ CD8+ T cells correlated with parameters of HIV and SIV disease progression. TIGIT remained elevated despite viral suppression in those with either pharmacological antiretroviral control or immunologically in elite controllers. HIV and SIV-specific CD8+ T cells were dysfunctional and expressed high levels of TIGIT and PD-1. Ex-vivo single or combinational antibody blockade of TIGIT and/or PD-L1 restored viral-specific CD8+ T cell effector responses. The frequency of TIGIT+ CD4+ T cells correlated with the CD4+ T cell total HIV DNA. These findings identify TIGIT as a novel marker of dysfunctional HIV-specific T cells and suggest TIGIT along with other checkpoint receptors may be novel curative HIV targets to reverse T cell exhaustion

    Design, motivation, and on-sky tests of an efficient fiber coupling unit for 1-meter class telescopes

    We present the science motivation, design, and on-sky test data of a high-throughput fiber coupling unit suitable for automated 1-meter class telescopes. The optical and mechanical design of the fiber coupling is detailed and we describe a flexible controller software designed specifically for this unit. The system performance is characterized with a set of numerical simulations, and we present on-sky results that validate the performance of the controller and the expected throughput of the fiber coupling. This unit was designed specifically for the MINERVA array, a robotic observatory consisting of multiple 0.7 m telescopes linked to a single high-resolution stabilized spectrograph for the purpose of exoplanet discovery using high-cadence radial velocimetry. However, this unit could easily be used for general astronomical purposes requiring fiber coupling or precise guiding
