    An investigation into co2–brine–cement–reservoir rock interactions for wellbore integrity in co2 geological storage

    Geological storage of CO2 in saline aquifers and depleted oil and gas reservoirs can help mitigate CO2 emissions. However, CO2 leakage over a long storage period represents a potential concern. Therefore, it is critical to establish a good understanding of the interactions between CO2–brine and cement–caprock/reservoir rock to ascertain the potential for CO2 leakage. Accordingly, in this work, we prepared a unique set of composite samples to resemble the cement–reservoir rock inter-face. A series of experiments simulating deep wellbore environments were performed to investigate changes in chemical, physical, mechanical, and petrophysical properties of the composite samples. Here, we present the characterisation of composite core samples, including porosity, permeability, and mechanical properties, determined before and after long‐term exposure to CO2‐rich brine. Some of the composite samples were further analysed by X‐ray microcomputed tomography (X‐ray μ‐CT), X‐ray diffraction (XRD), and scanning electron microscopy–energy‐dispersive X‐ray (SEM–EDX). Moreover, the variation of ions concentration in brine at different timescales was studied by per-forming inductively coupled plasma (ICP) analysis. Although no significant changes were observed in the porosity, permeability of the treated composite samples increased by an order of magnitude, due mainly to an increase in the permeability of the sandstone component of the composite samples, rather than the cement or the cement/sandstone interface.     Addressing potential sources of variation in several non-destructive techniques for measuring firmness in apples

    Measurements of firmness have traditionally been carried out according to the Magness Taylor (MT) procedure; using a texture analyser or penetrometer in reference texture tests. Non-destructive tests like the acoustic impulse response of acoustic firmness sensors (AFSs), a low-mass impact firmness sensor Sinclair International (SIQ-FT) and impact test (Lateral Impact – UPM) have also been used to measure texture and firmness. The objectives of this study were to evaluate the influence of different sources of variation in these three non-destructive tests and to evaluate their respective capabilities of discriminating between fruit maturity at two different harvest dates, turgidity before and after dehydration treatment and ripening after different storage periods. According to our results, fruit studied an unexpected AFS trend with turgidity. Contact measurements (Lateral Impact – UPM and SIQ-FT) appeared highly sensitive to changes in turgidity, but were less able to follow changes in ripening caused by storage period. Contact measurements were suitable for detecting differences between fruits from different harvest dates and showed higher correlation coefficients with reference texture tests than acoustic measurements.     Learning Task-Specific Generalized Convolutions in the Permutohedral Lattice

    Dense prediction tasks typically employ encoder-decoder architectures, but the prevalent convolutions in the decoder are not image-adaptive and can lead to boundary artifacts. Different generalized convolution operations have been introduced to counteract this. We go beyond these by leveraging guidance data to redefine their inherent notion of proximity. Our proposed network layer builds on the permutohedral lattice, which performs sparse convolutions in a high-dimensional space allowing for powerful non-local operations despite small filters. Multiple features with different characteristics span this permutohedral space. In contrast to prior work, we learn these features in a task-specific manner by generalizing the basic permutohedral operations to learnt feature representations. As the resulting objective is complex, a carefully designed framework and learning procedure are introduced, yielding rich feature embeddings in practice. We demonstrate the general applicability of our approach in different joint upsampling tasks. When adding our network layer to state-of-the-art networks for optical flow and semantic segmentation, boundary artifacts are removed and the accuracy is improved.Comment: To appear at GCPR 201

    Selective suppression of α-Synuclein in monoaminergic neurons of mice by intranasal delivery of targeted small interfering RNA or antisense oligonucleotides: Potential therapy for Parkinson's disease

The brains of Parkinson patients typically contain insoluble intracellular protein inclusions called Lewy bodies. Increased neuronal α-Syn levels represent a major component of Lewy bodies and therefore, the suppression of α-Syn expression provides a valid therapeutic target for PD. The goal of this study was to assess the ability of various small interfering RNA (siRNA) and antisense oligonucleotide (ASO) sequences directed against α-Syn to downregulate endogenous or overexpressed α-Syn mRNA levels in BE-M17 neuroblastoma cells. Moreover, we evaluated the feasibility of reducing α-Syn expression selectively in PD-vulnerable brain areas including substantia nigra pars compacta (SNc), ventral tegmental area (VTA), locus coeruleus (LC) and dorsal raphe nucleus (DR) of mice after the internalization of conjugated siRNA/ASO molecules into monoamine neurons following intranasal administration. Conclusions: These results set the stage for the testing of these molecules as potential disease-modifying agents in neurotoxin-based and genetic models of PD linked to pathogenic increases in α-Syn.     Childhood craniopharyngioma: greater hypothalamic involvement before surgery is associated with higher homeostasis model insulin resistance index

    <p>Abstract</p> <p>Background</p> <p>Obesity seems to be linked to the hypothalamic involvement in craniopharyngioma. We evaluated the pre-surgery relationship between the degree of this involvement on magnetic resonance imaging and insulin resistance, as evaluated by the homeostasis model insulin resistance index (HOMA). As insulin-like growth factor 1, leptin, soluble leptin receptor (sOB-R) and ghrelin may also be involved, we compared their plasma concentrations and their link to weight change.</p> <p>Methods</p> <p>27 children with craniopharyngioma were classified as either grade 0 (n = 7, no hypothalamic involvement), grade 1 (n = 8, compression without involvement), or grade 2 (n = 12, severe involvement).</p> <p>Results</p> <p>Despite having similar body mass indexes (BMI), the grade 2 patients had higher glucose, insulin and HOMA before surgery than the grade 0 (P = 0.02, <0.05 and 0.02 respectively) and 1 patients (P < 0.02 and <0.03 for both insulin and HOMA). The grade 0 (5.8 ± 4.9) and 1 (7.2 ± 5.3) patients gained significantly less weight (kg) during the year after surgery than did the grade 2 (16.3 ± 7.4) patients. The pre-surgery HOMA was positively correlated with these weight changes (P < 0.03).</p> <p>The data for the whole population before and 6–18 months after surgery showed increases in BMI (P < 0.0001), insulin (P < 0.005), and leptin (P = 0.0005), and decreases in sOB-R (P < 0.04) and ghrelin (P < 0.03).</p> <p>Conclusion</p> <p>The hypothalamic involvement by the craniopharyngioma before surgery seems to determine the degree of insulin resistance, regardless of the BMI. The pre-surgery HOMA values were correlated with the post-surgery weight gain. This suggests that obesity should be prevented by reducing inn secretion in those cases with hypothalamic involvement.</p

    Machine learning-based approach unravels distinct pathological signatures induced by patient-derived α-synuclein seeds in monkeys. Dopaminergic neuronal cell death, associated with intracellular α-synuclein (α-syn)-rich protein aggregates [termed "Lewy bodies" (LBs)], is a well-established characteristic of Parkinson's disease (PD). Much evidence, accumulated from multiple experimental models, has suggested that α-syn plays a role in PD pathogenesis, not only as a trigger of pathology but also as a mediator of disease progression through pathological spreading. Here, we have used a machine learning-based approach to identify unique signatures of neurodegeneration in monkeys induced by distinct α-syn pathogenic structures derived from patients with PD. Unexpectedly, our results show that, in nonhuman primates, a small amount of singular α-syn aggregates is as toxic as larger amyloid fibrils present in the LBs, thus reinforcing the need for preclinical research in this species. Furthermore, our results provide evidence supporting the true multifactorial nature of PD, as multiple causes can induce a similar outcome regarding dopaminergic neurodegeneratio

    Border effects among Catalan dialects

    In this study, we investigate which factors influence the linguistic distance of Catalan dialectal pronunciations from standard Catalan. We use pronunciations from three regions where the northwestern variety of the Catalan language is spoken (Catalonia, Aragon and Andorra). In contrast to Aragon, Catalan has an official status in both Catalonia and Andorra, which likely influences standardization. Because we are interested in the potentially large range of differences that standardization might promote, we examine 357 words in Catalan varieties and in particular their pronunciation distances with respect to the standard. In order to be sensitive to differences among the words, we fitted a generalized additive mixed-effects regression model to this data. This allows us to examine simultaneously the general (i.e. aggregate) patterns in pronunciation distance and to detect those words that diverge substantially from the general pattern. The results revealed higher pronunciation distances from standard Catalan in Aragon than in the other regions. Furthermore, speakers in Catalonia and Andorra, but not in Aragon, showed a clear standardization pattern, with younger speakers having dialectal pronunciations closer to the standard than older speakers. This clearly indicates the presence of a border effect within a single country with respect to word pronunciation distances. Since a great deal of scholarship focuses on single segment changes, we compare our analysis to the analysis of three segment changes that have been discussed in the literature on Catalan. This comparison revealed that the pattern observed at the word pronunciation level was supported by two of the three cases examined. As not all individual cases conform to the general pattern,
