Diseño y caracterización de péptidos específicamente dirigidos al bloqueo de la formación del apresorio en el hongo fitopatógeno Magnaporthe oryzae
La piriculariosis del arroz, causada por el hongo Magnaporthe oryzae, es la enfermedad que más pérdidas genera devastando miles de hectáreas de cultivo anualmente. Los tratamientos antifúngicos frente a esta enfermedad suelen ser de poca utilidad y se vuelven ineficaces en poco tiempo debido a la alta tasa de variación del hongo. Además, dentro del contexto legislativo actual a nivel europeo, se está restringiendo cada vez más la utilización de compuestos antifúngicos de amplio espectro debido a los posibles daños que pueden ocasionar sobre el ecosistema y la salud humana. Una alternativa prometedora al uso de los antifúngicos convencionales son los péptidos antimicrobianos, y más concretamente los péptidos dirigidos capaces de bloquear un proceso esencial para la infección del patógeno. Un paso fundamental en el proceso infectivo del hongo M. oryzae es la formación del apresorio, cuya finalidad es romper la superficie foliar y así penetrar en el mesófilo de la hoja. El desarrollo de compuestos capaces de bloquear específicamente este proceso sin afectar al ciclo vital del hongo constituiría una estrategia ventajosa en el control de la piriculariosis, a priori no tóxica para otros organismos incluido el ser humano. En este trabajo, hemos abordado este objetivo mediante dos aproximaciones, el cribado de una biblioteca combinatoria de péptidos y la modificación de secuencias de AMPs naturales mediante diseño racional. Gracias al conocimiento que se tiene sobre la secuencia y el modo de acción de los péptidos antimicrobianos, hemos diseñado una serie de péptidos capaces de controlar el desarrollo de la piriculariosis mediante inhibición específica de la formación del apresorio en M. oryzae, pero sin afectar a la viabilidad celular. Nuestros resultados indican que péptidos pequeños, como el heptapéptido PAF104, son capaces de reprimir la expresión de los genes MoMSB2 y MoMST11, implicados en la ruta de señalización de la MAPK Pmk1 imprescindible para la formación del apresorio en M. oryzae. Además, demostramos un comportamiento diferencial frente a distintas estructuras infectivas (i.e., apresorio, estructuras similares al apresorio e hifopodios), lo que apoya la idea propuesta por otros investigadores de que los mecanismos moleculares que regulan estás estructuras no son exactamente iguales. Mediante microscopía confocal mostramos que el tratamiento del hongo con MgAPI16 da lugar a apresorios aberrantes y no funcionales. En conclusión la principal aportación de esta investigación es el diseño de una serie de péptidos sintéticos, los heptapéptidos diseñados de novo PAF104, MgAPI24, MgAPI40 y MgAPI47, y el péptido MgAPI16 derivado del péptido antimicrobiano natural CecA, capaces de controlar el desarrollo de la piriculariosis en arroz mediante la inhibición específica de la formación del apresorio. Este trabajo contribuye por tanto al diseño de estrategias de control en protección vegetal basadas en péptidos dirigidos, las cuales deberían ser sostenibles y respetuosas con el medio ambiente.Rice blast caused by the fungus Magnaporthe oryzae is the most devastating rice disease, causing losses and ravaging thousands of hectares every year. Antifungal treatments are often ineffective and become useless soon due to the high fungus variation rate. Furthermore, within the current legislative framework at European level, it is increasingly restricting the use of broad-spectrum antifungal compounds because of the potential damage that can cause to the ecosystem and human health. One promising alternative to conventional antifungal compounds are the use of antimicrobial peptides, and in particular the target-orientated peptides able to block an essential infective process. A basic step on the development of rice blast diseases is the formation of appressoria, necessary to break the leaf surface and penetrate the mesophyll. To develop novel compounds able to block this process without affecting the life cycle of the fungus could be a promising alternative to control rice blast diseases, a priori no toxic to other organisms including humans. In this work, we board this objective by performing two approaches, the screening of a peptide combinatorial library and the rational design of natural known antimicrobial peptides. We have design a set of peptides able to control the infection caused by M. oryzae by specific inhibition of appressorium formation, but without a lytic effect. Our results show that small peptides, such as the heptapeptide PAF104, represses the gene expression of MoMSB2 and MoMST11, involved in the Pmk1 MAPK pathway essential to M. oryzae appressorium formation. Moreover, the designed peptides showed a differential effect on some infective structures (i.e., appressoria, appressorium-like structures and hyphopodia), supporting that the molecular regulation of these structures is different, an idea previously proposed by other researchers. By confocal microscopy we showed that the appressoria treated with MgAPI16 present aberrant physical and not functional structures. As a conclusion, the main contribution of this work is the design of a set of synthetic peptides, de novo designed heptapeptides PAF104, MgAPI24, MgAPI40 y MgAPI47 and the peptide MgAPI16 derived from the natural antimicrobial peptide Cecropin A, which are able to control rice blast development through the specific inhibition of the appressorium formation in M. oryzae. Screening of a synthetic peptide combinatorial library to identify inhibitors of the appressorium formation in Magnaporthe oryzae
The rice blast disease caused by Magnaporthe oryzae is one of the most devastating diseases of cultivated rice. One of the most important stages in the infective cycle of M. oryzae is the formation of the dome-shaped structure called appressorium. The purpose of the present study was to identify novel peptides to control the rice blast disease by blocking the appressorium formation through screening of a synthetic peptide combinatorial library. As result of the screening, a set of 29 putative bioactive peptides were identified, synthesized and assayed in comparison with the previously identified peptide PAF104. The peptides MgAPI24, MgAPI40 and MgAPI47 showed improved inhibitory activity on the M. oryzae appressorium formation. Our data show that these peptides have a differential effect on two developmental structures: appressoria and appressorium-like structures. Antimicrobial assays against M. oryzae and other non-target microorganisms showed a weak or no toxicity of these peptides, demonstrating their specific activity blocking the appressorium formation. Background: The Tobacco Products Directive
(2014/40/EU) obliges manufacturers and importers of
tobacco products and related to notify Member States
the commercialization of these products through the EU
Common Entry Portal (EU-CEG). The objective of this
study is to describe the main data available in EU-CEG
in Spain and analyze its implications for smoking control
policies.
Methods: Cross-sectional analysis of the data obtained
in EU-CEG for Spain. The access is done through
.xml or .pdf files, downloaded from the portal or consulted
through its interface. It describes the distribution of products,
manufacturers, presence of priority ingredients, Slim
cigarettes and concentration of nicotine in liquids.
Results: In the EU-CEG portal for Spain there are
currently 42,736 records. 83.7% corresponds to the devices
susceptible to nicotine release (67% of refill liquids,
mainly with concentrations of 3, 6, 12 and 18 mg). In tobacco
products, cigarettes (5%), cigars (5.9%) and water
pipe tobacco (2.8%) stand out. Herbal products for smoking
account for 0.1%. 5 of the priority ingredients for
surveillance are present in more than 75% of cigarettes
and rolling tobacco. There are 44 records of Slim cigarettes
and 9 novel tobacco products.
Conclusions: The EU-CEG portal has become a
powerful tool in tobacco control. We highlight challenges
that require a joint and comprehensive approach in aspects
such as labeling and ingredients, Slim cigarettes and new
forms of consumption.Fundamentos: La Directiva de Productos del
Tabaco (2014/40/EU) obliga a los fabricantes e importadores
de productos de tabaco y relacionados a notificar a
los Estados miembros la comercialización de estos productos,
a través del Portal de Entrada Común de la UE
(EU-CEG). El objetivo del estudio fue describir los principales
datos disponibles en el EU-CEG en España, así
como analizar sus implicaciones para las políticas de control
del tabaquismo.
Material y métodos: Se realizó un análisis transversal
de los datos obtenidos en el EU-CEG para España.
El acceso se realizó a través de archivos en formato xml
o pdf, descargados del portal o consultados mediante su
interfaz. Se describió la distribución de productos, de fabricantes,
la presencia de aditivos prioritarios, y la concentración
de nicotina en los líquidos.
Resultados: En el portal EU-CEG para España existen
actualmente 42.736 registros. El 83,7% correspondía a
los dispositivos susceptibles de liberación de nicotina (67%
de líquidos de recarga, principalmente con concentraciones
de 3, 6, 12 y 18 mg). En productos de tabaco destacaron los
cigarrillos (5%), los puros (5,9%) y el tabaco para pipa de
agua (2,8%). Los productos de hierbas para fumar supusieron
el 0,1%. Cinco de los aditivos prioritarios para su vigilancia
estaban presentes en más del 75% de los cigarrillos y
del tabaco de liar. Existían 44 registros de cigarrillos Slim y
9 de productos novedosos de tabaco.
Conclusiones: El portal EU-CEG se ha convertido
en una potente herramienta en el control del tabaco.
Destacan los retos que requieren un abordaje conjunto e
integral en aspectos como el etiquetado y los ingredientes,
los cigarrillos Slim y las nuevas formas de consumo
