    This paper describes the theoretical underpinnings of an ongoing research project that is examining the relationship between e-Democracy and the ‘digital divide’. The literature surrounding the Digital Divide is reviewed, and the importance of equitable physical access to ICTs in the drive to bridge the issue of social exclusion examined. It is argued that any discussion of the phenomenon of the digital divide must look beyond equitable physical access and take into consideration issues mentioned separately in the literature – ‘real access’, ‘reach’ and ‘socially responsible connectivity’

    Comparison of Convective Overshooting Models and Their Impact on Abundances from Integrated Light Spectroscopy of Young (<< 3 Gyr) Star Clusters

    As part of an ongoing program to measure detailed chemical abundances in nearby galaxies, we use a sample of young to intermediate age clusters in the Large Magellanic Cloud with ages of 10 Myr to 2 Gyr to evaluate the effect of isochrone parameters, specifically core convective overshooting, on Fe abundance results from high resolution, integrated light spectroscopy. In this work we also obtain fiducial Fe abundances from high resolution spectroscopy of the cluster individual member stars. We compare the Fe abundance results for the individual stars to the results from isochrones and integrated light spectroscopy to determine whether isochrones with convective overshooting should be used in our integrated light analysis of young to intermediate age (10 Myr -3 Gyr) star clusters. We find that when using the isochrones from the Teramo group, we obtain more accurate results for young and intermediate age clusters over the entire age range when using isochrones without convective overshooting. While convective overshooting is not the only uncertain aspect of stellar evolution, it is one of the most readily parametrized ingredients in stellar evolution models, and thus important to evaluate for the specific models used in our integrated light analysis. This work demonstrates that our method for integrated light spectroscopy of star clusters can provide unique tests for future constraints on stellar evolution models of young and intermediate age clusters.Comment: 16 pages, accepted for publication in Ap

    Using remote substituents to control solution structure and anion binding in lanthanide complexes.

    A study of the anion-binding properties of three structurally related lanthanide complexes, which all contain chemically identical anion-binding motifs, has revealed dramatic differences in their anion affinity. These arise as a consequence of changes in the substitution pattern on the periphery of the molecule, at a substantial distance from the binding pocket. Herein, we explore these remote substituent effects and explain the observed behaviour through discussion of the way in which remote substituents can influence and control the global structure of a molecule through their demands upon conformational space. Peripheral modifications to a binuclear lanthanide motif derived from α,α′-bis(DO3 Ayl)-m-xylene are shown to result in dramatic changes to the binding constant for isophthalate. In this system, the parent compound displays considerable conformational flexibility, yet can be assumed to bind to isophthalate through a well-defined conformer. Addition of steric bulk remote from the binding site restricts conformational mobility, giving rise to an increase in binding constant on entropic grounds as long as the ideal binding conformation is not excluded from the available range of conformers

    Perampanel add-on for drug-resistant focal epilepsy.

    BackgroundEpilepsy is one of the most common neurological disorders. Approximately 30% of people with epilepsy are considered to be drug-resistant, and usually need treatment with a combination of other antiepileptic drugs. Perampanel is a newer antiepileptic drug that has been investigated as add-on therapy for drug-resistant focal epilepsy.ObjectivesTo evaluate the benefits and harms of perampanel as add-on therapy for people with drug-resistant focal epilepsy.Search methodsWe used standard, extensive Cochrane search methods. The latest search date was 20 October 2022.Selection criteriaWe included randomised controlled trials comparing add-on perampanel with placebo.Data collection and analysisWe used standard Cochrane methods. Our primary outcome was 1. 50% or greater reduction in seizure frequency. Our secondary outcomes were 2. seizure freedom, 3. treatment withdrawal due to any reason, 4. treatment withdrawal due to adverse effects, and 5.Adverse effectsWe used an intention-to-treat population for all primary analyses. We presented the results as risk ratios (RR) with 95% confidence intervals (CIs), except for individual adverse effects, which we reported with 99% CIs to compensate for multiple testing. We used GRADE to assess certainty of evidence for each outcome.Main resultsWe included seven trials involving 2524 participants, all aged over 12 years. The trials were double-blind, randomised, placebo-controlled trials with treatment duration of 12 to 19 weeks. We assessed four trials at overall low risk of bias, and three trials at overall unclear risk of bias, due to risk of detection, reporting, and other biases. Compared with placebo, participants receiving perampanel were more likely to achieve a 50% or greater reduction in seizure frequency (RR 1.67, 95% CI 1.43 to 1.95; 7 trials, 2524 participants; high-certainty evidence). Compared to placebo, perampanel increased seizure freedom (RR 2.50, 95% CI 1.38 to 4.54; 5 trials, 2323 participants; low-certainty evidence) and treatment withdrawal (RR 1.30, 95% CI 1.03 to 1.63; 7 trials, 2524 participants; low-certainty evidence). Participants treated with perampanel were more likely to withdraw from treatment due to adverse effects compared to those receiving placebo (RR 2.36, 95% CI 1.59 to 3.51; 7 trials, 2524 participants; low-certainty evidence). A higher proportion of participants receiving perampanel reported one or more adverse effects when compared to participants who received placebo (RR 1.17, 95% CI 1.10 to 1.24; 7 trials, 2524 participants; high-certainty evidence). Compared with placebo, participants receiving perampanel were more likely to experience ataxia (RR 14.32, 99% CI 1.09 to 188.31; 2 trials, 1098 participants; low-certainty evidence), dizziness (RR 2.87, 99% CI 1.45 to 5.70; 7 trials, 2524 participants; low-certainty evidence), and somnolence (RR 1.76, 99% CI 1.02 to 3.04; 7 trials, 2524 participants). Subgroup analysis indicated that a larger proportion of participants who received perampanel at a dose of 4 mg/day (RR 1.38, 95% CI 1.05 to 1.83; 2 trials, 710 participants), 8 mg/day (RR 1.83, 95% CI 1.51 to 2.22; 4 trials, 1227 participants), or 12 mg/day (RR 2.38, 95% CI 1.86 to 3.04; 3 trials, 869 participants) achieved a 50% or greater reduction in seizure frequency compared to placebo; however, treatment with perampanel 12 mg/day also increased treatment withdrawal (RR 1.77, 95% CI 1.31 to 2.40; 3 trials, 869 participants).Authors' conclusionsAdd-on perampanel is effective at reducing seizure frequency and may be effective at maintaining seizure freedom for people with drug-resistant focal epilepsy. Although perampanel was well-tolerated, there was a higher proportion of treatment withdrawals with perampanel compared with placebo. Subgroup analysis suggested that 8 mg/day and 12 mg/day are the most efficacious perampanel doses; however, the use of 12 mg/day would likely increase the number of treatment withdrawals. Future research should focus on investigating the efficacy and tolerability of perampanel with longer-term follow-up, as well as exploring an optimal dose

    Robust analysis of prokaryotic pangenome gene gain and loss rates with Panstripe

    Horizontal gene transfer (HGT) plays a critical role in the evolution and diversification of many microbial species. The resulting dynamics of gene gain and loss can have important implications for the development of antibiotic resistance and the design of vaccine and drug interventions. Methods for the analysis of gene presence/absence patterns typically do not account for errors introduced in the automated annotation and clustering of gene sequences. In particular, methods adapted from ecological studies, including the pangenome gene accumulation curve, can be misleading as they may reflect the underlying diversity in the temporal sampling of genomes rather than a difference in the dynamics of HGT. Here, we introduce Panstripe, a method based on generalized linear regression that is robust to population structure, sampling bias, and errors in the predicted presence/absence of genes. We show using simulations that Panstripe can effectively identify differences in the rate and number of genes involved in HGT events, and illustrate its capability by analyzing several diverse bacterial genome data sets representing major human pathogens.Peer reviewe

    Globular Cluster Abundances from High-Resolution, Integrated-Light Spectroscopy. III. The Large Magellanic Cloud: Fe and Ages

    In this paper we refine our method for the abundance analysis of high resolution spectroscopy of the integrated light of unresolved globular clusters (GCs). This method was previously demonstrated for the analysis of old (>>10 Gyr) Milky Way GCs. Here we extend the technique to young clusters using a training set of 9 GCs in the Large Magellanic Cloud (LMC). Depending on the signal-to-noise ratio of the data, we use 20-100 Fe lines per cluster to successfully constrain the ages of old clusters to within a ∼\sim5 Gyr range, the ages of ∼\sim2 Gyr clusters to a 1-2 Gyr range, and the ages of the youngest clusters (0.05-1 Gyr) to a ∼\sim200 Myr range. We also demonstrate that we can measure [Fe/H] in clusters with any age less than 12 Gyrs with similar or only slightly larger uncertainties (0.1-0.25 dex) than those obtained for old Milky Way GCs (0.1 dex); the slightly larger uncertainties are due to the rapid evolution in stellar populations at these ages. In this paper, we present only Fe abundances and ages. In the next paper in this series, we present our complete analysis of the ∼20\sim 20 elements for which we are able to measure abundances. For several of the clusters in this sample, there are no high resolution abundances in the literature from individual member stars; our results are the first detailed chemical abundances available.
