    I can see CRISPR now, even when phage are gone: a view on alternative CRISPR-Cas functions from the prokaryotic envelope

    Purpose of review: CRISPR-Cas systems are prokaryotic immune systems against invading nucleic acids that adapt as new environmental threats arise. There are emerging examples of CRISPR-Cas functions in bacterial physiology beyond their role in adaptive immunity. This highlights the poorly understood, but potentially common, moonlighting functions of these abundant systems. We propose that these noncanonical CRISPR-Cas activities have evolved to respond to stresses at the cell envelope. Recent findings: Here, we discuss recent literature describing the impact of the extracellular environment on the regulation of CRISPR-Cas systems, and the influence of CRISPR-Cas activity on bacterial physiology. These described noncanonical CRISPR-Cas functions allow the bacterial cell to respond to the extracellular environment, primarily through changes in envelope physiology. Summary: This review discusses the expanding noncanonical functions of CRISPR-Cas systems, including their roles in virulence, focusing mainly on their relationship to the cell envelope. We first examine the effects of the extracellular environment on regulation of CRISPR-Cas components, and then discuss the impact of CRISPR-Cas systems on bacterial physiology, concentrating on their roles in influencing interactions with the environment including host organisms

    Does it matter to a member of the military whether the military campaign in which he is taking part is lawful or not? Despite the observation that the crime of aggression (post Kampala 2010) constitutes a ‘leadership crime par excellence,’ which limits any (future) criminal responsibility accordingly, the legality or illegality of any military action under international law can create moral implications for the common foot soldier and mid-level officer and also have a tangible impact on the national legal frameworks under which these forces operate. This short article uses the example of Operation Iraqi Freedom (2003) to discuss the repercussions of a—most likely—illegal military campaign for individual members of democratic armed forces before the background of the present discussion of NATO led action in Libya

    Ion and polymer dynamics in polymer electrolytes PPO-LiClO4: II. 2H and 7Li NMR stimulated-echo experiment

    We use 2H NMR stimulated-echo spectroscopy to measure two-time correlation functions characterizing the polymer segmental motion in polymer electrolytes PPO-LiClO4 near the glass transition temperature Tg. To investigate effects of the salt on the polymer dynamics, we compare results for different ether oxygen to lithium ratios, namely, 6:1, 15:1, 30:1 and infinity. For all compositions, we find nonexponential correlation functions, which can be described by a Kohlrausch function. The mean correlation times show quantitatively that an increase of the salt concentration results in a strong slowing down of the segmental motion. Consistently, for the high 6:1 salt concentration, a high apparent activation energy E_a=4.1eV characterizes the temperature dependence of the mean correlation times at Tg < T< 1.1T_g, while smaller values E_a=2.5eV are observed for moderate salt contents. The correlation functions are most nonexponential for 15:1 PPO-LiClO4, whereas the stretching is reduced for higher and lower salt concentrations. A similar dependence of the correlation functions on the evolution time in the presence and in the absence of ions indicates that addition of salt hardly affects the reorientational mechanism. For all compositions, mean jump angles of about 15 degree characterize the segmental reorientation. In addition, comparison of results from 2H and 7Li NMR stimulated-echo experiments suggests a coupling of ion and polymer dynamics in 15:1 PPO-LiClO4.Comment: 14 pages, 12 figure

    Methods & proposal for metadata guiding principles for scholarly communications

    This article describes an international community-based effort to create metadata guiding principles for adopting and using richer metadata and advancing its application in scholarly communications. These principles can facilitate the dissemination, discoverability and use/reuse of many types of research and scholarly outputs. While much work remains to be done, these principles serve as a starting point for the evolution of processes that span communities including publishers, researchers, scholars, authors and other creators, librarians, curators, custodians, and consumers of scholarly works. These aspirational Metadata 2020 Principles are designed to encompass the needs of our entire community while ensuring thoughtful, purposeful, and reusable metadata resources. They provide a framework for all of us to be good metadata citizens. They also provide a foundation for considering related work from Metadata 2020 and must be interpreted within the legal and practical context in which we operate. They are intended to guide the broadest possible cross-section of our community in improving research communications, publishing, and discoverability

    Effects Of Length, Complexity, And Grammatical Correctness On Stuttering In Spanish-Speaking Preschool Children

    Purpose: To explore the effects of utterance length, syntactic complexity, and grammatical correctness on stuttering in the spontaneous speech of young, monolingual Spanish-speaking children. Method: Spontaneous speech samples of 11 monolingual Spanish-speaking children who stuttered, ages 35 to 70 months, were examined. Mean number of syllables, total number of clauses, utterance complexity (i.e., containing no clauses, simple clauses, or subordinate and/or conjoined clauses), and grammatical correctness (i.e., the presence or absence of morphological and syntactical errors) in stuttered and fluent utterances were compared. Results: Findings revealed that stuttered utterances in Spanish tended to be longer and more often grammatically incorrect, and contain more clauses, including more subordinate and/or conjoined clauses. However, when controlling for the interrelatedness of syllable number and clause number and complexity, only utterance length and grammatical incorrectness were significant predictors of stuttering in the spontaneous speech of these Spanish-speaking children. Use of complex utterances did not appear to contribute to the prediction of stuttering when controlling for utterance length. Conclusions: Results from the present study were consistent with many earlier reports of English-speaking children. Both length and grammatical factors appear to affect stuttering in Spanish-speaking children. Grammatical errors, however, served as the greatest predictor of stuttering.Communication Sciences and Disorder

    Equidistribution Rates, Closed String Amplitudes, and the Riemann Hypothesis

    We study asymptotic relations connecting unipotent averages of Sp(2g,Z)Sp(2g,\mathbb{Z}) automorphic forms to their integrals over the moduli space of principally polarized abelian varieties. We obtain reformulations of the Riemann hypothesis as a class of problems concerning the computation of the equidistribution convergence rate in those asymptotic relations. We discuss applications of our results to closed string amplitudes. Remarkably, the Riemann hypothesis can be rephrased in terms of ultraviolet relations occurring in perturbative closed string theory.Comment: 15 page

    Black Sea coastal forecasting system

    The Black Sea coastal nowcasting and forecasting system was built within the framework of EU FP6 ECOOP (European COastalshelf sea OPerational observing and forecasting system) project for five regions: the south-western basin along the coasts of Bulgaria and Turkey, the north-western shelf along the Romanian and Ukrainian coasts, coastal zone around of the Crimea peninsula, the north-eastern Russian coastal zone and the coastal zone of Georgia. The system operates in the real-time mode during the ECOOP project and afterwards. The forecasts include temperature, salinity and current velocity fields. Ecosystem model operates in the off-line mode near the Crimea coast

The definitive version was published in Oncogene 32 (2013): 1135–1143, doi:10.1038/onc.2012.135.Neurofibromatosis type 2 patients develop schwannomas, meningiomas and ependymomas resulting from mutations in the tumor suppressor gene, NF2, encoding a membrane-cytoskeleton adapter protein called merlin. Merlin regulates contact inhibition of growth and controls the availability of growth factor receptors at the cell surface. We tested if microtubule-based vesicular trafficking might be a mechanism by which merlin acts. We found that schwannoma cells, containing merlin mutations and constitutive activation of the Rho/Rac family of GTPases, had decreased intracellular vesicular trafficking relative to normal human Schwann cells. In Nf2−/− mouse Schwann (SC4) cells, re-expression of merlin as well as inhibition of Rac or its effector kinases, MLK and p38SAPK, each increased the velocity of Rab6 positive exocytic vesicles. Conversely, an activated Rac mutant decreased Rab6 vesicle velocity. Vesicle motility assays in isolated squid axoplasm further demonstrated that both mutant merlin and active Rac specifically reduce anterograde microtubule-based transport of vesicles dependent upon the activity of p38SAPK kinase. Taken together, our data suggest loss of merlin results in the Rac-dependent decrease of anterograde trafficking of exocytic vesicles, representing a possible mechanism controlling the concentration of growth factor receptors at the cell surface.This work was supported by NIH R01 CA118032 (to NR), and MBL research fellowships (to NR and GM), NIH R01 NS23868 (to STB)

    Phase transitions for suspension flows

    This paper is devoted to study thermodynamic formalism for suspension flows defined over countable alphabets. We are mostly interested in the regularity properties of the pressure function. We establish conditions for the pressure function to be real analytic or to exhibit a phase transition.
