    Plasmodium falciparum OTU‐like cysteine protease (PfOTU) is essential for apicoplast homeostasis and associates with noncanonical role of Atg8

    The metabolic pathways associated with the mitochondrion and the apicoplast in Plasmodium, 2 parasite organelles of prokaryotic origin, are considered as suitable drug targets. In the present study, we have identified functional role of a novel ovarian tumour unit (OTU) domain-containing cysteine protease of Plasmodium falciparum (PfOTU). A C-terminal regulatable fluorescent affinity tag on native protein was utilised for its localization and functional characterization. Detailed studies showed vesicular localization of PfOTU and its association with the apicoplast. Degradation-tag mediated knockdown of PfOTU resulted in abnormal apicoplast development and blocked development of parasites beyond early-schizont stages in subsequent cell cycle; downregulation of PfOTU hindered apicoplast protein import. Further, the isoprenoid precursor-mediated parasite growth-rescue experiments confirmed that PfOTU knockdown specifically effect development of functional apicoplast. We also provide evidence for a possible biological function of PfOTU in membrane deconjugation of Atg8, which may be linked with the apicoplast protein import. Overall, our results show that the PfOTU is involved in apicoplast homeostasis and associates with the noncanonical function of Atg8 in maintenance of parasite apicoplast

    Effectiveness of oral health education on knowledge, attitude, practices and oral hygiene status among 12–15-year-old schoolchildren of fishermen of Kutch district, Gujarat, India

    Background: To assess the effectiveness of oral health education on oral health knowledge, attitude, practices and oral hygiene status among 12–15-year-old school children of fishermen of Kutch district, Gujarat, India.Materials and methods: A before-and-after experimental study was conducted among all (n = 205) the 12–15-year-old children from two schools of Bhadreshwar village of Mundra taluka of Kutch district, Gujarat, India from January 2013 to December 2013. At baseline, children were assessed for oral health knowledge, attitude and practices using a self-administered structured questionnaire and oral hygiene was assessed using Oral Hygiene Index-Simplified (OHI-S). Oral health education was provided after baseline assessment, at 3 months and at 6 months. Follow up study was done after 1 year from baseline. Statistical tests applied were Independent t test, paired t test and McNemar test. Level of significance and confidence level were set at 5% and 95%, respectively.Results: Mean OHI-S scores reduced significantly at 1 year follow up interval. All the questions showed statistically significant improvement in knowledge, attitude and practices except the frequency of change of tooth brush which showed no improvement.Conclusions: The results of the study reflects the accomplishment of upgrading oral health knowledge, attitude, practices and oral hygiene status of fishermen children through school oral health education programme. Organizing oral health education in high school children of fishermen community could lead to improvement in students’ oral hygiene to ultimately enhance their oral health

    The Relationship Between Multiple Intelligences with Preferred Science Teaching and Science Process Skills

    This study was undertaken to identify the relationship between multiple intelligences with preferred science teaching and science process skills. The design of the study is a survey using three questionnaires reported in the literature: Multiple Intelligences Questionnaire, Preferred Science Teaching Questionnaire and Science Process Skills Questionnaire. The study selected 300 primary school students from five (5) primary schools in Penang, Malaysia. The findings showed a relationship between kinesthetic, logical-mathematical, visual-spatial and naturalistic intelligences with the preferred science teaching. In addition there was a correlation between kinesthetic and visual-spatial intelligences with science process skills, implying that multiple intelligences are related to science learning

    : This aim of this research is to know (1) the differences of students\u27 motivation and achievement after using tapioca waste as learning resource in tenth grade, MAN 2 Pati in Archaebacteria and Eubacteria materials: (2) the effects of using tapioka waste as learning resources on students\u27 motivation and achievement in tenth grade, MAN 2 Pati. This research is quasi-experimental design with randomized control group, pre-post test. The study population was all students of Class X MAN 2 Pati, while the research sample is Class X-2 as the control group and Class X-3 as the experimental group. Data collection techniques were test and non-test techniques. The analyses used the t-test to determine differences in motivation and achievement. The results show that there is difference in learning motivation after using tapioca waste as a source of learning, indicated by the result of paired samples t-test, value of sig.(2-tailed)>α. There is no difference in learning achievement after using tapioca waste as a source of learning, which is indicated by paired sample t-test, the value of sig.(2-tailed)<α. Learning motivation and achievement of students who use tapioca waste as learning resources is higher than that of students who use the conventional learning resources

    Effects of hospital facilities on patient outcomes after cancer surgery: an international, prospective, observational study

    Background Early death after cancer surgery is higher in low-income and middle-income countries (LMICs) compared with in high-income countries, yet the impact of facility characteristics on early postoperative outcomes is unknown. The aim of this study was to examine the association between hospital infrastructure, resource availability, and processes on early outcomes after cancer surgery worldwide.Methods A multimethods analysis was performed as part of the GlobalSurg 3 study-a multicentre, international, prospective cohort study of patients who had surgery for breast, colorectal, or gastric cancer. The primary outcomes were 30-day mortality and 30-day major complication rates. Potentially beneficial hospital facilities were identified by variable selection to select those associated with 30-day mortality. Adjusted outcomes were determined using generalised estimating equations to account for patient characteristics and country-income group, with population stratification by hospital.Findings Between April 1, 2018, and April 23, 2019, facility-level data were collected for 9685 patients across 238 hospitals in 66 countries (91 hospitals in 20 high-income countries; 57 hospitals in 19 upper-middle-income countries; and 90 hospitals in 27 low-income to lower-middle-income countries). The availability of five hospital facilities was inversely associated with mortality: ultrasound, CT scanner, critical care unit, opioid analgesia, and oncologist. After adjustment for case-mix and country income group, hospitals with three or fewer of these facilities (62 hospitals, 1294 patients) had higher mortality compared with those with four or five (adjusted odds ratio [OR] 3.85 [95% CI 2.58-5.75]; p&lt;0.0001), with excess mortality predominantly explained by a limited capacity to rescue following the development of major complications (63.0% vs 82.7%; OR 0.35 [0.23-0.53]; p&lt;0.0001). Across LMICs, improvements in hospital facilities would prevent one to three deaths for every 100 patients undergoing surgery for cancer.Interpretation Hospitals with higher levels of infrastructure and resources have better outcomes after cancer surgery, independent of country income. Without urgent strengthening of hospital infrastructure and resources, the reductions in cancer-associated mortality associated with improved access will not be realised
