    Evidence for Fast Electron Transfer between the High-Spin Haems in Cytochrome bd-I from Escherichia coli

    Cytochrome bd-I is one of the three proton motive force-generating quinol oxidases in the O2-dependent respiratory chain of Escherichia coli. It contains one low-spin haem (b558) and the two high-spin haems (b595 and d) as the redox-active cofactors. In order to examine the flash-induced intraprotein reverse electron transfer (the so-called ''electron backflow''), CO was photolyzed from the ferrous haem d in one-electron reduced (b558 3+b595 3+d2+-CO) cytochrome bd-I, and the fully reduced (b5582+b5952+d2+-CO) oxidase as a control. In contrast to the fully reduced cytochrome bd-I, the transient spectrum of one-electron reduced oxidase at a delay time of 1.5 μs is clearly different from that at a delay time of 200 ns. The difference between the two spectra can be modeled as the electron transfer from haem d to haem b595 in 3–4% of the cytochrome bd-I population. Thus, the interhaem electron backflow reaction induced by photodissociation of CO from haem d in one-electron reduced cytochrome bd-I comprises two kinetically different phases: the previously unnoticed fast electron transfer from haem d to haem b595 within 0.2–1.5 μs and the slower well-defined electron equilibration with τ ~16 μs.     Theory of Chiral Modulations and Fluctuations in Smectic-A Liquid Crystals Under an Electric Field

    Chiral liquid crystals often exhibit periodic modulations in the molecular director; in particular, thin films of the smectic-C* phase show a chiral striped texture. Here, we investigate whether similar chiral modulations can occur in the induced molecular tilt of the smectic-A phase under an applied electric field. Using both continuum elastic theory and lattice simulations, we find that the state of uniform induced tilt can become unstable when the system approaches the smectic-A--smectic-C* transition, or when a high electric field is applied. Beyond that instability point, the system develops chiral stripes in the tilt, which induce corresponding ripples in the smectic layers. The modulation persists up to an upper critical electric field and then disappears. Furthermore, even in the uniform state, the system shows chiral fluctuations, including both incipient chiral stripes and localized chiral vortices.     Voiceless but empowered farmers in corporate supply chains: contradictory imagery and instrumental approach to empowerment

    There have been calls for a shift of focus towards the political and power-laden aspects of transitioning towards socially equitable global supply chains. This paper offers an empirically grounded response to these calls from a critical realist stance in the context of global food supply chains. We examine how an imaginary for sustainable farming structured around an instrumental construction of empowerment limits what is viewed as permissible, desirable and possible in global food supply chains. We adopt a multimodal Critical Discourse Analysis (CDA) to examine the sustainable farming imaginary for smallholder farmers constructed by one large organization, Unilever, in a series of videos produced and disseminated on YouTube. We expose the underlying mechanisms of power and marginalization at work within the sustainability imaginary and show how “empowerment” has the potential to create of new dependencies for these farmers. We recontextualize the representations to show that while the imaginary may be commercially feasible, it is less achievable in terms of empowering smallholder farmers

    A 2+1+1 quadruple star system containing the most eccentric, low-mass, short-period, eclipsing binary known

    We present an analysis of a newly discovered 2+1+1 quadruple system with TESS containing an unresolved eclipsing binary (EB) as part of TIC 121088960 and a close neighbor TIC 121088959. The EB consists of two very low-mass M dwarfs in a highly-eccentric (e = 0.709) short-period (P = 3.04358 d) orbit. Given the large pixel size of TESS and the small separation (3.{^{\prime\prime}_{.}}9) between TIC 121088959 and TIC 121088960 we used light centroid analysis of the difference image between in-eclipse and out-of-eclipse data to show that the EB likely resides in TIC 121088960, but contributes only ∼10% of its light. Radial velocity data were acquired with iSHELL at NASA’s Infrared Facility and the Coudé spectrograph at the McDonald 2.7-m telescope. For both images, the measured RVs showed no variation over the 11-day observational baseline, and the RV difference between the two images was 8 ± 0.3 km s−1. The similar distances and proper motions of the two images indicate that TIC 121088959 and TIC 121088960 are a gravitationally bound pair. Gaia’s large RUWE and astrometric_excess_noise parameters for TIC 121088960, further indicate that this image is the likely host of the unresolved EB and is itself a triple star. We carried out an SED analysis and calculated stellar masses for the four stars, all of which are in the M dwarf regime: 0.19 M⊙ and 0.14 M⊙ for the EB stars and 0.43 M⊙ and 0.39 M⊙ for the brighter visible stars, respectively. Lastly, numerical simulations show that the orbital period of the inner triple is likely the range 1 to 50 years.Accepted manuscrip

    New Insights into X-ray Binaries

    X-ray binaries are excellent laboratories to study collapsed objects. On the one hand, transient X-ray binaries contain the best examples of stellar-mass black holes while persistent X-ray binaries mostly harbour accreting neutron stars. The determination of stellar masses in persistent X-ray binaries is usually hampered by the overwhelming luminosity of the X-ray heated accretion disc. However, the discovery of high-excitation emission lines from the irradiated companion star has opened new routes in the study of compact objects.     The Pediatric Cell Atlas: defining the growth phase of human development at single-cell resolution

    Single-cell gene expression analyses of mammalian tissues have uncovered profound stage-specific molecular regulatory phenomena that have changed the understanding of unique cell types and signaling pathways critical for lineage determination, morphogenesis, and growth. We discuss here the case for a Pediatric Cell Atlas as part of the Human Cell Atlas consortium to provide single-cell profiles and spatial characterization of gene expression across human tissues and organs. Such data will complement adult and developmentally focused HCA projects to provide a rich cytogenomic framework for understanding not only pediatric health and disease but also environmental and genetic impacts across the human lifespan

    Mysterious Dust-emitting Object Orbiting TIC 400799224

    We report the discovery of a unique object of uncertain nature - but quite possibly a disintegrating asteroid or minor planet - orbiting one star of the widely separated binary TIC 400799224. We initially identified the system in data from TESS Sector 10 via an abnormally shaped fading event in the light curve (hereafter "dips"). Follow-up speckle imaging determined that TIC 400799224 is actually two stars of similar brightness at 0.″62 separation, forming a likely bound binary with projected separation of ∼300 au. We cannot yet determine which star in the binary is host to the dips in flux. ASAS-SN and Evryscope archival data show that there is a strong periodicity of the dips at ∼19.77 days, leading us to believe that an occulting object is orbiting the host star, though the duration, depth, and shape of the dips vary substantially. Statistical analysis of the ASAS-SN data shows that the dips only occur sporadically at a detectable threshold in approximately one out of every three to five transits, lending credence to the possibility that the occulter is a sporadically emitted dust cloud. The cloud is also fairly optically thick, blocking up to 37% or 75% of the light from the host star, depending on the true host. Further observations may allow for greater detail to be gleaned as to the origin and composition of the occulter, as well as to a determination of which of the two stars comprising TIC 400799224 is the true host star of the dips
