    Investigation of the hydrophobic sites of DREAM

    DREAM (downstream regulatory element antagonist modulator) also known as calsenilin or KChIP-3 is a calcium binding protein that has been associated with memory retention, pain sensing, calcium homeostasis and Alzheimer’s disease. It has two EF hands that bind Ca2+ and change the conformation of the protein. DREAM has two hydrophobic cavities in both the N- and C- terminus. These hydrophobic sites act as important binding sites with Kv channels, presenilin and other small molecules. Previously our group has showed that hydrophobic molecules such as NS5806 and 1,8- ANS can bind to the hydrophobic cavities in the C-terminus. The aim of this investigation was to characterize interactions of other hydrophobic molecules with DREAM with a long-term goal of finding a potential inhibitor for DREAM interacting with presenilin. In silico and in vitro approaches were used to determine a mechanism of binding of DREAM with hydrophobic molecules NS5806, NS3623, bezafibrate, trifluoperazine, 5-biphenyl-2-yl-1H-tetrazole, N-[(4-phenylphenyl)methyl]- 2H-tetrazol-5-amine and cholesterol. Three binding sites were identified at the C-terminus and Nterminus. The most favored binding site was in the C-terminus in the hydrophobic pocket of the EF- 4. All the hydrophobic molecules tested showed strong binding in this region. The major contributing residues were HIS 214, TYR 174, MET 187 and PHE 218. Other binding sites found resided in the hydrophobic pocket in the N-terminus but binding was shown to be weaker with many unfavorable residue interactions. Additional binding site was identified in between EF-3 and EF-4 but had many unfavorable interactions. An inhibitor with a high affinity for DREAM binding to presenilin would be beneficial in the blocking of gamma secretase complex which may lead to a better understanding of the physiology and treatment of Alzheimer disease

Curs 2021/2022 (A distància)La ecuación Ginzburg-Landau compleja en su forma cúbica describe una amplia gama de fenómenos para muchos sistemas de la física y presenta muchas estructuras coherentes estables entre sus soluciones. De ahí que se haya estudiado intensamente a lo largo de los años y que sea un laboratorio ideal para aprender sobre los esquemas de integración temporal. Desde el punto de vista numérico se pueden utilizar métodos pseudoespectrales para resolver la discretización espacial, hay que resolverlo con variable compleja y se pueden utilizar métodos específicos para la integración temporal como la Exponential Time Differencing o Integrating Factor Methods gracias a la presencia de términos lineales y no lineales integrables. El objetivo de esta tesis es comparar algunos de estos métodos analizando su orden y eficiencia. Para ello, se han programado los diferentes esquemas en Fortran y se han validado sus resultados mediante una pila de casos de prueba. A continuación, utilizando uno de ellos como referencia y calculando la solución de una prueba con un paso de tiempo muy pequeño, se han comparado los demás. Entre otros resultados, se ha constatado la buena convergencia de los métodos de orden superior o la mejor velocidad de los métodos que integran analíticamente parte de la ecuación o que necesitan menos transformadas de Fourier

Esa imagen colectiva se adquiere poco a poco de la información que se extrae de aspectos cotidianos como el cine, la literatura, la publicidad y de otras muchas facetas que nos rodean en nuestro día a día y que nada tienen que ver con una idea pedagógica o divulgativa pero que acaban distorsionando este periodo de la historia. En este trabajo analizamos brevemente algunas de estas facetas cotidianas que generan una visión irreal de la Prehistoria.The vision that society have in general on Prehistory, most of the times do not come from exhibitions or publications developed by experts. The collective image of the Prehistory most of the times come from the Cinema, the Literature, the commercial spots and other aspects that surrounds us in our quatidianity and nothing has to do with a pedagogic or divulgative idea, but the end in the distortion of this History period.     MinkSORT: A 3D deep feature extractor using sparse convolutions to improve 3D multi-object tracking in greenhouse tomato plants

    The agro-food industry is turning to robots to address the challenge of labour shortage. However, agro-food environments pose difficulties for robots due to high variation and occlusions. In the presence of these challenges, accurate world models, with information about object location, shape, and properties, are crucial for robots to perform tasks accurately. Building such models is challenging due to the complex and unique nature of agro-food environments, and errors in the model can lead to task execution issues. In this paper, we propose MinkSORT, a novel method for generating tracking features using a 3D sparse convolutional network in a deepSORT-like approach to improve the accuracy of world models in agro-food environments. We evaluated our feature extractor network using real-world data collected in a tomato greenhouse, which significantly improved the performance of our baseline model that tracks tomato positions in 3D using a Kalman filter and Mahalanobis distance. Our deep learning feature extractor improved the HOTA from 42.8% to 44.77%, the association accuracy from 32.55% to 35.55%, and the MOTA from 57.63% to 58.81%. We also evaluated different contrastive loss functions for training our deep learning feature extractor and demonstrated that our approach leads to improved performance in terms of three separate precision and recall detection outcomes. Our method improves world model accuracy, enabling robots to perform tasks such as harvesting and plant maintenance with greater efficiency and accuracy, which is essential for meeting the growing demand for food in a sustainable manner

    Development and evaluation of automated localization and reconstruction of all fruits on tomato plants in a greenhouse based on multi-view perception and 3D multi-object tracking

    Accurate representation and localization of relevant objects is important for robots to perform tasks. Building a generic representation that can be used across different environments and tasks is not easy, as the relevant objects vary depending on the environment and the task. Furthermore, another challenge arises in agro-food environments due to their complexity, and high levels of clutter and occlusions. In this paper, we present a method to build generic representations in highly occluded agro-food environments using multi-view perception and 3D multi-object tracking. Our representation is built upon a detection algorithm that generates a partial point cloud for each detected object. The detected objects are then passed to a 3D multi-object tracking algorithm that creates and updates the representation over time. The whole process is performed at a rate of 10 Hz. We evaluated the accuracy of the representation on a real-world agro-food environment, where it was able to successfully represent and locate tomatoes in tomato plants despite a high level of occlusion. We were able to estimate the total count of tomatoes with a maximum error of 5.08% and to track tomatoes with a tracking accuracy up to 71.47%. Additionally, we showed that an evaluation using tracking metrics gives more insight in the errors in localizing and representing the fruits.Comment: Pre-print, article submitted and in review proces

    Comunicación corporativa del Real Valladolid y el Real Betis Balompié: Análisis y comparación de su ejecución en plataformas 2.0

    Desde el nacimiento de las redes sociales ha habido un cambio en la vida de las personas y de las empresas. Las primeras han cambiado su forma de consumir información: antes lo hacían comprando el periódico diariamente, pero ahora la población, al tener smartphone, lo tienen todo a mano, consumen información de lo que les interesa a través de aplicaciones como Twitter, Instagram, YouTube y Facebook, entre otras. Este cambio a la hora de obtener información llevó a las empresas, incluidos los clubes de fútbol profesional, a adaptarse a esta nueva situación. Primero, tuvieron que tener claro las ideas en el interior del club, una filosofía, y segundo darse a conocer en el exterior, saber lo que se quiere transmitir a los aficionados. Por último, con el paso de los años, aumentar la presencia en las redes sociales para potenciar la visibilidad de su propia marca e incrementar el número de seguidores y aficionados e interactuar con ellos. Partiendo del contexto anterior, en este Trabajo de Fin de Grado se ha tomado de objeto de estudio la comunicación del Real Valladolid Club de Fútbol y el Real Betis Balompié, equipos de la Liga Santander (Primera División de España) para explorar la estrategia comunicativa mediante el análisis de sus publicaciones y entrevistas para detectar las claves de la comunicación corporativa 2.0 de ambos clubes. Se puso la atención en el tipo de contenidos multimedia que emplean en cada publicación, que son sobre todo las imágenes; las interacciones de sus aficionados, que en la entidad verdiblanca destaca bastante estos aspectos y, por último, las diferencias entre los dos modelos de comunicación, que son bastantes, sobre todo en su estrategia comunicativa en redes sociales. Para realizar la investigación hemos hecho entrevistas en profundidad y un análisis de contenido de las principales redes sociales: Twitter, Instagram, YouTube y Facebook, de los dos clubes, para al final, visualizar los resultados obtenidos.With the birth of social networks, the lives of people and companies have experienced a change. The former have changed the way of acquiring information, which previously was limited to reading the daily news in the newspaper, whereas nowadays they have access to all the information through the apps on smartphones, such as Twitter, Instagram, YouTube or Facebook. This transformation led companies, in particular Professional Football Clubs, to adapt to this new situation. Firstly, the staff of the Football Club had to follow a clear philosophy. Secondly, the Club needed to communicate effectively with their fans so as to make itself known among the population. Lastly, the presence in social networks had to be improved in order to promote the brand and raise the number of followers and interact with them. Based on the previous context, the subject matter of this Final Project is the Corporate Communication of Real Valladolid Club de Fútbol and Real Betis Balompié, Clubs in the Santander League from Spain's First Division.The Communication Strategies from both Clubs will be invesitagted through a content analysis of publications and interviews, in order to identify the key elements of their Corporate Communication in 2.0 platforms. This research focuses on the type of media content used in the publications, mainly images; the interactions with fans, which really stand out in Real Betis social networks; and the differences between the communication methods from both Football Clubs, which are numerous within their communication strategies in social networks. In order to conduct this research, we will follow two steps: in-depth interviews with professionals and a content analysis of the main social networks from the aforementioned football clubs, which are Twitter, Instagram, YouTube and Facebook.     El objeto de estudio de este trabajo es el mito de la mandrágora en la literatura alemana del Romanticismo. La elección de este tema deriva de nuestro gran interés por la mitología y la lietaruta alemana, especialmente la perteneciente a dicho movimiento. También queríamos elaborar un trabajo cuyo objeto de estudio fuera diferente y un tanto misterioso, puesto que presentamos gran fascinación por lo oscuro y lo sobrenatural.
