    Effect of Aspirin on Cell Growth of Human MG-63 Osteosarcoma Line

    Nonsteroidal anti-inflammatory drugs (NSAIDs) are commonly used in bone tissue repair treatment for their pharmacological action. The objective of this study was to determine the effect of aspirin, on osteoblast growth, using MG63 cell line as osteoblast model. MTT spectrophotometry results showed that 20, 100, and 1000 μM aspirin doses have an inhibitory effect on growth. Cell cycle analysis revealed that aspirin doses of 100 and 1000 μM arrest the cell cycle in phase GO/G1. Parallel apoptosis/necrosis studies showed no changes in comparison to control cells after treatment with 1 or 10 μM aspirin but a significantly increased percentage of cells in apoptosis at doses of 20, 100, and 1000 μM. We highlight that treatment of osteoblast-like cells with 1000 μM aspirin increased not only the percentage of cells in apoptosis but also the percentage of necrotic cells, which was not observed in aspirin treatments at lower doses

    Spread of the invasive alga Caulerpa racemosa var. cylindracea (Caulerpales, Chlorophyta) along the Mediterranean Coast of the Murcia region (SE Spain)

    The aim of this paper was to document the appearance and spread of the green alga Caulerpa racemosa along the coast of Murcia in south–eastern Spain. It was found for the first time in the area in 2005 and over the next two years the number of new sightings increased almost exponentially. In the period 2005–2007 the total surface area colonised by the alga in the region was estimated to be at least 265 ha. Benthic assemblages colonised by the alga were rocky bottoms with photophilic algae, dead P. oceanica rhizomes, infralittoral and circalittoral soft bottoms and maerl beds. No penetration of the alga was observed in P. oceanica meadows, except in one locality. Biometric analysis indicated high vegetative development in the established colonies in comparison to those described in other Mediterranean areas. Rapid spreading dynamics observed in the Murcia region is a potential threat for native benthic communities. El alga fue detectada por primera vez en el año 2005 y durante los dos años consecutivos se observó un crecimiento casi exponencial en el número de áreas colonizadas. La superficie total colonizada por el alga en Murcia durante el periodo 2005–2007 ha sido estimada en 265 ha., siendo las comunidades bentónicas afectadas algas fotófilas sobre sustrato rocoso, "mata muerta" de P. oceanica, fondos blandos infralitorales y circalitorales y fondos con comunidades de maërl. La presencia del alga dentro de praderas de P. oceanica solamente fue detectada en una localidad. Los estudios biométricos realizados muestran un elevado desarrollo vegetativo de las poblaciones de C. racemosa en Murcia en comparación con colonias de otras áreas del Mediterráneo, siendo esta rápida dinámica de expansión una amenaza potencial para las comunidades bentonicas nativas.     Changes in cannabinoid receptor binding and mRNA levels in several brain regions of aged rats

    AbstractWe have recently found that cannabinoid receptor binding and gene expression markedly decreased in extrapyramidal structures of aged rats. The present study was designed to analyze the possible existence of similar aging-induced changes in cannabinoid receptor binding and gene expression in brain regions other than extrapyramidal areas, but that also contain a significant population of cannabinoid receptors, such as the cerebellum, hippocampal structures, limbic and hypothalamic nuclei, the cerebral cortex and others. To this end, we analyzed cannabinoid receptor binding, using autoradiography, and cannabinoid receptor mRNA levels, using in situ hybridization, in slide-mounted brain sections obtained from young (3 month old) and aged (>2 year old) rats. Results were as follows. In the cerebellum, aged rats exhibited a marked decrease in cannabinoid receptor binding in the molecular layer (−33.3%), although accompanied by no changes in mRNA levels in the granular layer. In the cerebral cortex, a small, although statistically significant, decrease in binding was found in the deep layer (VI) (−18.3%) of aged rats, whereas no changes were found in the superficial layer (I). As in the case of the cerebellum, mRNA levels did not change in the cerebral cortex layers (II–III and V–VI). The different regions of the Ammon’s horn of the hippocampus exhibited similar cannabinoid receptor binding levels in aged and young rats. Interestingly, mRNA levels decreased in aged rats to a small, but statistically significant, extent (CA1: −26.1%; CA2: −21.6%; CA3: −14.4%). This was also seen in another hippocampal structure, the dentate gyrus (−14.6%), although in this region binding levels increased in aged rats (+28.4%). Two hypothalamic structures, the arcuate nucleus and the ventromedial hypothalamic nucleus, exhibited decreased cannabinoid receptor binding in aged rats (−31.1% and −30.3%, respectively), but this was not seen in the medial preoptic area. This was accompanied by no changes in mRNA levels in the ventromedial hypothalamic nucleus. In the limbic structures, aged rats exhibited similar binding levels to young rats. This was seen in the nucleus accumbens, septum nuclei and basolateral amygdaloid nucleus. However, mRNA levels slightly decreased in the basolateral amygdaloid nucleus (−13.4%), whereas they were not altered in the septum nuclei. Finally, other brain structures, such as the central gray substance and the brainstem, exhibited similar binding levels in aged and young rats. However, it is important to note that mRNA levels increased significantly (+211.2%) in the brainstem of aged rats, an area where the levels of binding and mRNA were very low in young rats. This marked increase may be related to an increase in the presence of glial elements in this region, as revealed by the increase in the immunoreactivity for glial fibrillary acidic protein observed in the brainstem of aged rats as compared to young animals. In summary, senescence was associated with changes in cannabinoid receptors in the cerebellum, the cerebral cortex, limbic and hypothalamic structures, the hippocampus and other brain regions. However, the changes observed (i) were not as marked and relevant as those early reported in extrapyramidal areas, and (ii) exhibited regional differences that might be attributed to the different roles played by these receptors in each region. Of particular relevance by their magnitude were the aging-induced decrease in binding found in the cerebellum and the hypothalamus, and the increase in mRNA levels observed in the brainstem.     Lateral impact sensor for measuring firmness of fruits in an experimental packing line.

    Different parameters are used to quantify the maturity of fruits at or near harvest (shape, color, flesh texture and internal composition). Flesh firmness is a critical handling parameter for fruits such as peach, pear and apple. Results of previous studies conducted by different researchers have shown that impact techniques can be used to evaluate firmness of fruits. A prototype impact system for firmness sorting of fruits was developed by Chen and Ruiz-Altisent (Chen et al, 1996). This sensor was mounted and tested successfully on a 3 m section of a commercial conveyor belt (Chen et al, 1998). This is a further development of the on-line impact system for firmness sorting of fruits. The design of the sensor has been improved and it has been mounted on a experimental fruit packing line (Ortiz-Cañavate et al 1999)

The boundary supersymmetric UU chain is solved by using the coordinate space Bethe ansatz technique and Bethe ansatz equations are derived.     Seasonal changes in feeding habits of Diplodus annularis (L., 1758) off southeast Iberian Peninsula

    The feeding habits of the sparid Diplodus annularis (L., 1758) from two Posidonia oceanica (L.) Delile, 1813 seagrass beds off Murcia (western Mediterranean), were analysed to determinate seasonal changes. Stomach contents of 289 specimens collected during experimental trawling from August 1993 to May 1994, were analysed. Diet varied seasonally: at Mazarron site, apendicularians and amphipods were more important in autumn, whereas during winter and spring amphipods were more important. At Cape Palos, Posidonia oceanica and algae were more important in summer, algae and hydrozoa in autumn, algae and briozoa in winter and harpacticoid copepods and gammarid amphipods in spring. The results indicate that Diplodus annularis feeds on a wide range of prey items, and could be considered a generalist feeder.Se estudiaron los hábitos alimenticios del espárido Diplodus annularis (L., 1758) de dos praderas de Posidonia oceanica (L.) Delile, 1813 de Murcia (Mediterráneo occidental) con el fin de detectar cambios estacionales en su dieta. Se analizaron los contenidos estomacales de 289 ejemplares capturados mediante arrastre experimental desde agosto de 1993 a mayo de 1994. La dieta varió estacionalmente; en Mazarrón, las presas más importantes en otoño fueron las apendicularias y los anfípodos, mientras que en invierno y primavera los anfípodos fueron las presas más importantes. En el cabo de Palos, Posidonia oceanica y algas fueron más importantes en verano, algas e hidrozoos en otoño, algas y briozoos en invierno, y copépodos harpacticoides y anfípodos gammáridos en primavera. Los resultados indican que Diplodus annularis se alimenta de una amplia variedad de tipos de presa, por lo que puede ser considerada una especie de alimentación generalista y poco selectiva.Instituto Español de Oceanografí

    Comparative study of the use of Doxycycline and Oxytetracycline to treat Anaplasmosis in fattening lambss

    Lamb icteric carcasses condemnation due to Anaplasma ovis is causing relevant economic losses. A comparative study was developed on the effects of different antibiotics to treat ovine anaplasmosis in fattening lambs. A total of 100 A. ovis naturally infected lambs were selected and randomly divided into four groups of 25 lambs: Group ID, treated with injectable doxycycline; Group OD, oral doxycycline; Group O, injectable oxytetracycline; and Group C, untreated animals for the control group. Clinical, haematological, and molecular analyses were performed before the treatment and 12 and 45 days after the beginning of the treatments, and carcass condemnation was followed after slaughter. The A. ovis bacterial load was high before the treatments in the four groups and decreased significantly 45 days after treatment in the ID and O Groups (p < 0.001). The parameters that were related to haemolysis showed similar results. At the abattoir, 15 out of the 47 examined carcasses were condemned; 7 of C Group, 6 of OD Group, 2 of O Group, and 0 of ID Group. It can be concluded that injectable doxycycline and oxytetracycline significantly reduce A. ovis bacterial load in blood and carcass condemnation at the abattoir. Further studies are needed in order to confirm these encouraging findings

    Discerning the ancestry of European Americans in genetic association studies

    European Americans are often treated as a homogeneous group, but in fact form a structured population due to historical immigration of diverse source populations. Discerning the ancestry of European Americans genotyped in association studies is important in order to prevent false-positive or false-negative associations due to population stratification and to identify genetic variants whose contribution to disease risk differs across European ancestries. Here, we investigate empirical patterns of population structure in European Americans, analyzing 4,198 samples from four genome-wide association studies to show that components roughly corresponding to northwest European, southeast European, and Ashkenazi Jewish ancestry are the main sources of European American population structure. Building on this insight, we constructed a panel of 300 validated markers that are highly informative for distinguishing these ancestries. We demonstrate that this panel of markers can be used to correct for stratification in association studies that do not generate dense genotype data

    Photo-acclimatory thresholds anticipate sudden shifts in seagrass ecosystem state under reduced light conditions

    Seagrass ecosystems usually respond in a nonlinear fashion to increasing pressures and environmental changes. Feedback mechanisms operating at the ecosystem level and involving multiple interactions among the seagrass meadow, its associated community and the physical environment are known to play a major role in such nonlinear responses. Phenotypic plasticity may also be important for buffering these ecological thresholds (i.e., regime shifts) as many physiological processes show nonlinear responses to gradual environmental changes, conferring the appearance of resistance before the effects at the organism and population levels are visible. However, the potential involvement of plant plasticity in driving catastrophic shifts in seagrass ecosystems has not yet been assessed. In this study, we conducted a manipulative 6-month light-gradient experiment in the field to capture nonlinearities of the physiological and population responses of the seagrass Cymodocea nodosa to gradual light reduction. The aim was to explore if and how the photo-acclimatory responses of shaded plants are translated to the population level and, hence, to the ecosystem level. Results showed that the seagrass population was rather stable under increasing shading levels through the activation of multilevel photo-acclimative responses, which are initiated with light reduction and modulated in proportion to shading intensity. The activation of photo-physiological and metabolic compensatory responses allowed shaded plants to sustain nearly constant plant productivity (metabolic carbon balance) along a range of shading levels before losing linearity and starting to decline. The species then activated plant- and meadow-scale photo-acclimative responses and drew on its energy reserves (rhizome carbohydrates) to confer additional population resilience. However, when the integration of all these buffering mechanisms failed to counterbalance the effects of extreme light limitation, the population collapsed, giving place to a phase shift from vegetated to bare sediments with catastrophic ecosystem outcomes. Our findings evidence that ecological thresholds in seagrass ecosystems under light limitation can be explained by the role of species’ compensatory responses in modulating population-level responses. The thresholds of these plastic responses anticipate the sudden loss of seagrass meadows with the potential to be used as early warning indicators signalling the imminent collapse of the ecosystem, which is of great value for the real-world management of seagrass ecosystems.En prens
