    The importance and effectiveness of volunteer‐collected data in ecology and conservation

    Volunteers have been collecting ecological data for centuries. However, volunteercollected data are frequently challenged because they lack the precision and rigour of scientific studies. This thesis evaluates the advantages of volunteer‐collected data and the importance of such data for the study of ecology and conservation, and considers methods to verify data to avoid or reduce inaccuracies. Different case studies aimed to answer questions relating to species’ ecology, habitat selection, and behaviour. Charismatic mammals were selected in order to increase volunteer participation (Water voles Arvicola terrestris; dormice Muscardinus avellanarius; North American otters Lontra canadensis; hedgehogs Erinaceus europaeus). Simple, rapid data collection methods were used so that volunteers and citizen scientists could easily follow instructions. The findings show that simple methods such as scales and estimates can be an effective way of studying water vole habitat associations; however, inter‐observer variability was highly problematic when volunteers collected data based on subjective estimations. A volunteer‐collected long‐term dataset on dormouse nestbox occupancy provided excellent information on habitat selection despite some irregularities when the data were recorded. Untrained citizen scientists could not record activity budgets for captive otters despite simple instructions, whereas citizen scientists were able to record habitat variables within their gardens, but false absences were found to be an issue when they recorded hedgehog sightings. Overall, this thesis suggests that volunteer‐collected data can provide useful insights into various aspects of ecology, for example, for studying distributions and species‐habitat interactions. Encouraging volunteers to collect ecological data has additional benefits such as increasing the health and wellbeing of participants, and it also raises public awareness of conservation issues. Recommendations on how to increase participation rates while minimising sources of error and bias are given

    This thesis explores the position of female film directors working in Hollywood. It is intended to address an area in feminist film theory which has often been overlooked. Although it is incorrect to say there has been no feminist analysis of the "mainstream" woman director, most of the work which has been done concentrates either on finding the feminism or femininity of her films, or studies only a select few directors. This research widens the debate by validating the study of all women directors, and moves away from the search for definitive feminist meaning in the cinematic text. It employs a contextual and multi-theoretical approach to interrogate the multiplicity of meanings embodied by the phrase "woman director". The first chapter interrogates auteur theory because any discussion of female authorship must confront this critical perspective. The female director makes a problematic auteur since that figure is traditionally gendered as masculine. Chapter two is a "state of the industry" examination of the position of the woman director in Hollywood, with a special emphasis on mentoring. Chapter three examines the marketing of Mimi Leder's films The Peacemaker (1997) and Deep Impact (1999). Chapters four, five and six explore the construction of the woman director as "star", presenting in-depth case studies of Jodie Foster and Penny Marshall. Chapters seven and eight look at the reception of Blue Steel (1990) and Strange Days (1995) directed by Kathryn Bigelow, and Clueless (1995) directed by Amy Heckerling. Each chapter is designed to contextualise and historicise the woman director in order to better understand why her gender has prevented her from being seen as a "natural" director: that is, why directing has been viewed as a suitable job for a man but "no job for a lady"

    Breakfast and exercise contingently affect postprandial metabolism and energy balance in physically active males

    The present study examined the impact of breakfast and exercise on postprandial metabolism, appetite and macronutrient balance. A sample of twelve (blood variables n 11) physically active males completed four trials in a randomised, crossover design comprising a continued overnight fast followed by: (1) rest without breakfast (FR); (2) exercise without breakfast (FE); (3) breakfast consumption(1859 kJ) followed by rest (BR); (4) breakfast consumption followed by exercise (BE). Exercise was continuous, moderate-intensity running (expending approximately 2·9MJ of energy). The equivalent time was spent sitting during resting trials. A test drink (1500 kJ) was ingested on all trials followed 90 min later by an ad libitum lunch. The difference between the BR and FR trials in blood glucose time-averaged AUC following test drink consumption approached significance (BR: 4·33 (SEM 0·14) v. FR: 4·75 (SEM 0·16) mmol/l; P¼0·08); but it was not different between FR and FE (FE: 4·77 (SEM 0·14) mmol/l; P¼0·65); and was greater in BE (BE: 4·97 (SEM 0·13) mmol/l) v. BR(P¼0·012). Appetite following the test drink was reduced in BR v. FR (P¼0·006) and in BE v. FE (P¼0·029). Following lunch, the most positive energy balance was observed in BR and least positive in FE. Regardless of breakfast, acute exercise produced a less positive energy balance following ad libitum lunch consumption. Energy and fat balance is further reduced with breakfast omission. Breakfast improved the overall appetite responses to foods consumed later in the day, but abrogated the appetite suppressive effect of exercise

    Development of a critical appraisal tool to assess the quality of cross-sectional studies (AXIS)

    Objectives: The aim of this study was to develop a critical appraisal (CA) tool that addressed study design and reporting quality as well as the risk of bias in cross-sectional studies (CSSs). In addition, the aim was to produce a help document to guide the non-expert user through the tool. Design: An initial scoping review of the published literature and key epidemiological texts was undertaken prior to the formation of a Delphi panel to establish key components for a CA tool for CSSs. A consensus of 80% was required from the Delphi panel for any component to be included in the final tool. Results: An initial list of 39 components was identified through examination of existing resources. An international Delphi panel of 18 medical and veterinary experts was established. After 3 rounds of the Delphi process, the Appraisal tool for Cross-Sectional Studies (AXIS tool) was developed by consensus and consisted of 20 components. A detailed explanatory document was also developed with the tool, giving expanded explanation of each question and providing simple interpretations and examples of the epidemiological concepts being examined in each question to aid non-expert users. Conclusions: CA of the literature is a vital step in evidence synthesis and therefore evidence-based decision-making in a number of different disciplines. The AXIS tool is therefore unique and was developed in a way that it can be used across disciplines to aid the inclusion of CSSs in systematic reviews, guidelines and clinical decision-making

    Poly-ε-Lysine or Mel4 Antimicrobial Surface Modification on a Novel Peptide Hydrogel Bandage Contact Lens

    Microbial keratitis (MK) is a serious issue in many countries and is often caused by contact lens wear. Antimicrobial peptides (AMPs) are a potentially useful tool for creating antimicrobial surfaces in light of increasing antibiotic resistance. Poly-ε-lysine (pεK) is an AMP that has been used extensively as a food preservative and Mel4 has recently been synthesized and studied as an antimicrobial coating for contact lenses. A hydrogel synthesized of pεK cross-linked with biscarboxylic acids provides a potential lens material which has many surface free amines, that can be subsequently used to attach additional AMPs, creating an antimicrobial lens. The aim of this study is to investigate pεK hydrogels against a clinical strain of Pseudomonas aeruginosa (P. aeruginosa) for preventing or treating MK. Covalent attachment of AMPs is investigated and confirmed by fluorescently tagged peptides. Bound pεK effectively reduces the number of adherent P. aeruginosa in vitro (>3 log). In ex vivo studies positive antimicrobial activity is observed on bare pεK hydrogels and those with additionally bound pεK or Mel4; lenses allow the maintenance of the corneal epithelium. A pεK hydrogel contact lens with additional AMPs can be a therapeutic tool to reduce the incidence of MK

    The impact of axial length on retinal tamponade for gas, silicone oil, and heavy silicone oil, using an in vitro model

    Background: To investigate whether ocular axial length influences the tamponade efficacy of three commonly used agents: gas, silicone oil, and heavy silicone oil. Methods: A series of filling experiments was conducted using 19-mm and 25-mm surface-modified spherical model eye chambers to mimic the vitreous cavity. For each agent, tamponade efficacy was assessed across a range of percentage fills, and comparison was made between the two model eye chambers. The behavior of each tamponade agent was quantified by measuring (1) the maximum height of the tamponade bubble, and calculating (2) the arc of retinal contact subtended by the tamponade bubble. Results: Polynomial regression analysis found no statistically significant difference between the regression models for the different-sized model eye chambers for bubble height or arc of retinal contact subtended. This applied to all of the tamponade agents under investigation. Conclusions: Across the range of cavity sizes under investigation, no significant difference in tamponade efficacy (as reflected by the measured dimensions of the tamponade bubble) was identified. © 2010 The Author(s).published_or_final_versionSpringer Open Choice, 21 Feb 201

    In vitro and Computational Modelling of Drug Delivery across the Outer Blood-Retinal Barrier

    The ability to produce rapid, cost-effective and human-relevant data has the potential to accelerate development of new drug delivery systems. Intraocular drug delivery is an area undergoing rapid expansion due to the increase in sight-threatening diseases linked to increasing age and lifestyle factors. The outer bloodretinal barrier (OBRB) is important in this area of drug delivery, as it separates the eye from the systemic blood flow. This study reports the development of complementary in vitro and in silico models to study drug transport from silicone oil across the outer blood-retinal barrier. Monolayer cultures of a human retinal pigmented epithelium cell line, ARPE-19, were added to chambers and exposed to a controlled flow to simulate drug clearance across the OBRB. Movement of dextran molecules and release of ibuprofen from silicone oil in this model were measured. Corresponding simulations were developed using COMSOL Multiphysics computational fluid dynamics (CFD) software and validated using independent in vitro data sets. Computational simulations were able to predict dextran movement and ibuprofen release, with all of the features of the experimental release profiles being observed in the simulated data. Simulated values for peak concentrations of permeated dextran and ibuprofen released from silicone oil were within 18% of the in vitro results. This model could be used as a predictive tool of drug transport across this important tissue
