    Study of the performance of disinfection with sodium hypochlorite on a full-scale sewage treatment plant

    A full-scale sewage treatment plant was investigated to assess the performance of the disinfection stage. Sodium hypochlorite was used as a disinfectant agent and the process efficiency was evaluated by E.coli removal. The research took place over a period of two years in order to evaluate the effect of retention time (t) and residual chlorine (Cr) under different seasonal conditions. The effectiveness of E.coli removal with sodium hypochlorite proved to be strictly dependent on the factor CR t (product of residual chlorine with the contact time). The regression line of the experimental points was, on the whole, well comparable with the model proposed by Collins, especially in the field of CRt lower than 30 mg L-1 min

    IEC 61508: Effect of test policy on the probability of failure on demand of safety instrumented systems

    Standard IEC 61508 provides probabilistic equations for determining the Average Probability of Failure on Demand (PFDavg) and the Average Probability of Failure per Hour (PFHavg) for some architectures of Safety Instrumented Systems (SIS) under the hypothesis of equal redundant components, taking into account Common Cause Failures (CCF), Detection Coverage (DC) and Proof Test Coverage (PTC) parameters. Surprisingly, IEC standard does not mention the testing policy aspects of SIS redundant components. However, from a close examination of the probabilistic equations, it is possible to recognize that the simultaneous/sequential testing policy has been implicitly assumed. This paper describes the conditions under which the staggered testing policy - which is better than all the others in case of independent tested components - can be advantageously applied to reduce PFDavg when CCF, DC and PTC parameters are taken into account

    The sensitivity of a specific denitrification rate under the dissolved oxygen pressure

    The biological denitrification process is extensively discussed in scientific literature. The process requires anoxic conditions, but the influence of residual dissolved oxygen (DO) on the efficiency is not yet adequately documented. The present research aims to fill this gap by highlighting the effects of DO on the specific denitrification rate (SDNR) and consequently on the efficiency of the process. SDNR at a temperature of 20â—¦C (SDNR20â—¦C) is the parameter normally used for the sizing of the denitrification reactor in biological-activated sludge processes. A sensitivity analysis of SNDR20â—¦C to DO variations is developed. For this purpose, two of the main empirical models illustrated in the scientific literature are taken into consideration, with the addition of a deterministic third model proposed by the authors and validated by recent experimentations on several full-scale plants. In the first two models, SDNR20â—¦C is expressed as a function of the only variable food:microrganism ratio in denitrification (F:MDEN), while in the third one, the dependence on DO is made explicit. The sensitivity analysis highlights all the significant dependence of SDNR20â—¦C on DO characterized by a logarithmic decrease with a very pronounced gradient in correspondence with low DO concentrations. Moreover, the analysis demonstrates the relatively small influence of F:MDEN on the SDNR20â—¦C and on the correlation between SDNR20â—¦C and DO. The results confirm the great importance of minimizing DO and limiting, as much as possible, the transport of oxygen in the denitrification reactor through the incoming flows and mainly the mixed liquor recycle. Solutions to achieve this result in full-scale plants are reported

    Salicylic Acid Peel Incorporating Triethyl Citrate and Ethyl Linoleate in the Treatment of Moderate Acne: A New Therapeutic Approach.

    BACKGROUND: Acne affects many adolescents. Conventional therapy often results in side effects and poor adherence, and the treatment does not consider the psychological effect of acne on patients, which is comparable with that of disabling diseases. OBJECTIVES: To evaluate the efficacy and tolerability of a peel (30% salicylic acid, triethyl citrate and ethyl linoleate) combined with a home therapy with three topical agents (triethyl citrate, ethyl linoleate and salicylic acid 0.5% cream, lotion) in moderate acne of the face. DESIGN: Prospective, observational, multicenter, open-label, postmarketing, phase IV study. METHODS: Patients were assessed by comparing Global Acne Grading System (GAGS) score and total lesion count from 15 days before the first peel (T-15 ), after four salicylic peels (every 10 ± 2 days (T0 , T10 , T20 , T30 ), and 20 days after of the end of the study (T50 ). This treatment was associated to a home therapy. RESULTS: Fifty-three patients completed the study. The average GAGS score fell 49% between T-15 and T50 (p < .001). No patient withdrew for adverse events. CONCLUSIONS: This therapy was effective and well-tolerated in all cases. Chemo-exfoliation sessions ensured the continuous monitoring of clinical results and improved patient quality of life

    Infecção respiratória aguda por adenovirus: comparação dos métodos de PCR e imunofluorescência indireta para o seu diagnóstico e dados clínicos dos pacientes infectados

    Infecções respiratórias por Adenovírus (ADV) são geralmente descritas associadas com alta mortalidade. O diagnóstico laboratorial é essencial para o estabelecimento da terapêutica adequada e para orientar a implantação de medidas preventivas evitando a propagação da infecção. Com o objetivo de analisar a sensibilidade e a especificidade dos métodos de avaliação de diagnóstico laboratorial, foi comparada a detecção de antígeno por imunofluorescência indireta (IF) com a reação em cadeia da polimerase específica (PCR) para detectar AdV em amostras respiratórias coletadas de pacientes internados com doença respiratória aguda. As amostras com resultados positivos foram inoculadas em cultura celular. Foram analisadas 381 amostras da secreção nasofaríngea coletadas durante o ano de 2008, das quais 2,6% foram positivas pela IF e 10% pela PCR, isolamento positivo foi obtido em 40% e 26% dos casos positivos pelos testes anteriores, respectivamente. A maioria dos pacientes infectados eram crianças com menos de seis meses de idade, e apesar do fato de que um número significativo de pacientes necessitou de cuidados intensivos, a taxa de mortalidade foi baixa (5%). Em conclusão, os métodos moleculares são úteis para o diagnóstico rápido de infecções por adenovírus com maior sensibilidade do que a detecção do antígeno, a sua introdução na rotina permitiu um aumento significativo no diagnóstico de infecções por adenovírus.Adenovirus (AdV) respiratory infections are usually described as being associated with high mortality rates. Laboratory diagnosis is essential for the establishment of the appropriate therapy, and for guiding the implementation of preventive measures in order to prevent the spread of the infection. Aiming to analyze the sensitivity and specificity of the laboratorial diagnosis methods available, we compared antigen detection by indirect immunofluorescence assay (IF), and a specific nested polymerase chain reaction (PCR), to detect AdV in respiratory samples collected from patients admitted to hospital with acute respiratory disease. Positive samples were inoculated into a cell culture to confirm the results. We analyzed 381 samples from the nasopharyngeal aspirates collected during the year 2008; of these, 2.6% tested were positive for adenovirus through IF and 10% through PCR; positive isolation was obtained in 40% and 26% of these cases, respectively. Most infected patients were children under six months of age, and despite of the fact that a significant number of patients required intensive care, the mortality rate was low (5%). In conclusion, molecular methods were found to be useful for rapid diagnosis of adenovirus infections with higher sensitivity than antigen detection; their introduction permitted a significant increase in diagnoses of adenovirus infections

This virus is carried by 2 rodent species, Oligoryzomys nigripes and Oxymycterus nasutus.     Potential geographic distribution of Hantavirus reservoirs in Brazil

    Hantavirus cardiopulmonary syndrome is an emerging zoonosis in Brazil. Human infections occur via inhalation of aerosolized viral particles from excreta of infected wild rodents. Necromys lasiurus and Oligoryzomys nigripes appear to be the main reservoirs of hantavirus in the Atlantic Forest and Cerrado biomes. We estimated and compared ecological niches of the two rodent species, and analyzed environmental factors influencing their occurrence, to understand the geography of hantavirus transmission. N. lasiurus showed a wide potential distribution in Brazil, in the Cerrado, Caatinga, and Atlantic Forest biomes. Highest climate suitability for O. nigripes was observed along the Brazilian Atlantic coast. Maximum temperature in the warmest months and annual precipitation were the variables that most influence the distributions of N. lasiurus and O. nigripes, respectively. Models based on occurrences of infected rodents estimated a broader area of risk for hantavirus transmission in southeastern and southern Brazil, coinciding with the distribution of human cases of hantavirus cardiopulmonary syndrome. We found no demonstrable environmental differences among occurrence sites for the rodents and for human cases of hantavirus. However, areas of northern and northeastern Brazil are also apparently suitable for the two species, without broad coincidence with human cases. Modeling of niches and distributions of rodent reservoirs indicates potential for transmission of hantavirus across virtually all of Brazil outside the Amazon Basin

    Phylogeographic Analysis of HIV-1 Subtype C Dissemination in Southern Brazil

    The HIV-1 subtype C has spread efficiently in the southern states of Brazil (Rio Grande do Sul, Santa Catarina and Paraná). Phylogeographic studies indicate that the subtype C epidemic in southern Brazil was initiated by the introduction of a single founder virus population at some time point between 1960 and 1980, but little is known about the spatial dynamics of viral spread. A total of 135 Brazilian HIV-1 subtype C pol sequences collected from 1992 to 2009 at the three southern state capitals (Porto Alegre, Florianópolis and Curitiba) were analyzed. Maximum-likelihood and Bayesian methods were used to explore the degree of phylogenetic mixing of subtype C sequences from different cities and to reconstruct the geographical pattern of viral spread in this country region. Phylogeographic analyses supported the monophyletic origin of the HIV-1 subtype C clade circulating in southern Brazil and placed the root of that clade in Curitiba (Paraná state). This analysis further suggested that Florianópolis (Santa Catarina state) is an important staging post in the subtype C dissemination displaying high viral migration rates from and to the other cities, while viral flux between Curitiba and Porto Alegre (Rio Grande do Sul state) is very low. We found a positive correlation (r2 = 0.64) between routine travel and viral migration rates among localities. Despite the intense viral movement, phylogenetic intermixing of subtype C sequences from different Brazilian cities is lower than expected by chance. Notably, a high proportion (67%) of subtype C sequences from Porto Alegre branched within a single local monophyletic sub-cluster. These results suggest that the HIV-1 subtype C epidemic in southern Brazil has been shaped by both frequent viral migration among states and in situ dissemination of local clades

    The Role of Oligomerization and Cooperative Regulation in Protein Function: The Case of Tryptophan Synthase

    The oligomerization/co-localization of protein complexes and their cooperative regulation in protein function is a key feature in many biological systems. The synergistic regulation in different subunits often enhances the functional properties of the multi-enzyme complex. The present study used molecular dynamics and Brownian dynamics simulations to study the effects of allostery, oligomerization and intermediate channeling on enhancing the protein function of tryptophan synthase (TRPS). TRPS uses a set of α/β–dimeric units to catalyze the last two steps of L-tryptophan biosynthesis, and the rate is remarkably slower in the isolated monomers. Our work shows that without their binding partner, the isolated monomers are stable and more rigid. The substrates can form fairly stable interactions with the protein in both forms when the protein reaches the final ligand–bound conformations. Our simulations also revealed that the α/β–dimeric unit stabilizes the substrate–protein conformation in the ligand binding process, which lowers the conformation transition barrier and helps the protein conformations shift from an open/inactive form to a closed/active form. Brownian dynamics simulations with a coarse-grained model illustrate how protein conformations affect substrate channeling. The results highlight the complex roles of protein oligomerization and the fine balance between rigidity and dynamics in protein function
