    Fungal laccase: copper induction, semi-purification, immobilization, phenolic effluent treatment and electrochemical measurement

    The laccase activity induced by copper ions in Trametes versicolor by two inducers was studied and the aims were to show that it is possible to obtain a high laccase activity in almost 12 of the enzymesproduct using a basic culture medium but different copper concentration. Laccase was inducted by 2,5-xylidine and copper sulfate. Semi-purification of the crude laccase was carried out through precipitation and the column separation with NaCl gradient. In order to apply in an effluent treatment, laccase was immobilized on different vitroceramics supports, pyrolytic graphite and also on a carbon fiber electrode as biosensor. The maximum laccase activity was 40,774.0 U L-1 at the 12th day. The best support for immobilization was pyrolytic graphite (glutaraldehyde treated-94% efficiency). Total phenol removal in Kraft E1 effluent was 19% for immobilized pyrolytic graphite with the mediator present. The biosensor prepared with this material showed a good linear response to catechol. The optimization of laccaseactivity induction through the combination of 2,5-xylidine and cooper sulfate was obtained and led to its use in environmental remediation

    Progesterone Metabolism by Human and Rat Hepatic and Intestinal Tissue

    Following oral administration, the bioavailability of progesterone is low and highly variable. As a result, no clinically relevant, natural progesterone oral formulation is available. After oral delivery, first-pass metabolism initially occurs in the intestines; however, very little information on progesterone metabolism in this organ currently exists. The aim of this study is to investigate the contributions of liver and intestine to progesterone clearance. In the presence of NADPH, a rapid clearance of progesterone was observed in human and rat liver samples (t1/2 2.7 and 2.72 min, respectively). The rate of progesterone depletion in intestine was statistically similar between rat and human (t1/2 197.6 min in rat and 157.2 min in human). However, in the absence of NADPH, progesterone was depleted at a significantly lower rate in rat intestine compared to human. The roles of aldo keto reductases (AKR), xanthine oxidase (XAO) and aldehyde oxidase (AOX) in progesterone metabolism were also investigated. The rate of progesterone depletion was found to be significantly reduced by AKR1C, 1D1 and 1B1 in human liver and by AKR1B1 in human intestine. The inhibition of AOX also caused a significant reduction in progesterone degradation in human liver, whereas no change was observed in the presence of an XAO inhibitor. Understanding the kinetics of intestinal as well as liver metabolism is important for the future development of progesterone oral formulations. This novel information can inform decisions on the development of targeted formulations and help predict dosage regimens

    Cryptographic enforcement of information flow policies without public information via tree partitions

    We may enforce an information flow policy by encrypting a protected resource and ensuring that only users authorized by the policy are able to decrypt the resource. In most schemes in the literature that use symmetric cryptographic primitives, each user is assigned a single secret and derives decryption keys using this secret and publicly available information. Recent work has challenged this approach by developing schemes, based on a chain partition of the information flow policy, that do not require public information for key derivation, the trade-off being that a user may need to be assigned more than one secret. In general, many different chain partitions exist for the same policy and, until now, it was not known how to compute an appropriate one. In this paper, we introduce the notion of a tree partition, of which chain partitions are a special case. We show how a tree partition may be used to define a cryptographic enforcement scheme and prove that such schemes can be instantiated in such a way as to preserve the strongest security properties known for cryptographic enforcement schemes. We establish a number of results linking the amount of secret material that needs to be distributed to users with a weighted acyclic graph derived from the tree partition. These results enable us to develop efficient algorithms for deriving tree and chain partitions that minimize the amount of secret material that needs to be distributed.Comment: Extended version of conference papers from ACNS 2015 and DBSec 201

    Learning democracy in social work

    In this contribution, we discuss the role of social work in processes of democracy. A key question in this discussion concerns the meaning of ‘the social’ in social work. This question has often been answered in a self-referential way, referring to a methodological identity of social work. This defines the educational role of social work as socialisation (be it socialisation into obedience or into an empowered citizen). However, the idea of democracy as ‘ongoing experiment’ and ‘beyond order’ challenges this methodological identity of social work. From the perspective of democracy as an ‘ongoing experiment’, the social is to be regarded as a platform for dissensus, for ongoing discussions on the relation between private and public issues in the light of human rights and social justice. Hence, the identity of social work cannot be defined in a methodological way; social work is a complex of (institutionalized) welfare practices, to be studied on their underlying views on the ‘social’ as a political and educational concept, and on the way they influence the situation of children, young people and adults in society

    Action research and democracy

    This contribution explores the relationship between research and learning democracy. Action research is seen as being compatible with the orientation of educational and social work research towards social justice and democracy. Nevertheless, the history of action research is characterized by a tension between democracy and social engineering. In the social-engineering approach, action research is conceptualized as a process of innovation aimed at a specific Bildungsideal. In a democratic approach action research is seen as research based on cooperation between research and practice. However, the notion of democratic action research as opposed to social engineering action research needs to be theorized. So called democratic action research involving the implementation by the researcher of democracy as a model and as a preset goal, reduces cooperation and participation into instruments to reach this goal, and becomes a type of social engineering in itself. We argue that the relationship between action research and democracy is in the acknowledgment of the political dimension of participation: ‘a democratic relationship in which both sides exercise power and shared control over decision-making as well as interpretation’. This implies an open research design and methodology able to understand democracy as a learning process and an ongoing experiment

    Student responses to the introduction of case-based learning and practical activities into a theoretical obstetrics and gynaecology teaching programme

    BACKGROUND: The fourth-year Obstetrics and Gynaecology course at our institution had previously been taught using theory classes alone. A new teaching model was introduced to provide a better link with professional practice. We wished to evaluate the impact of the introduction of case discussions and other practical activities upon students' perceptions of the learning process. METHODS: Small-group discussions of cases and practical activities were introduced for the teaching of a fourth-year class in 2003 (Group II; 113 students). Comparisons were made with the fourth-year class of 2002 (Group I; 108 students), from before the new programme was introduced. Students were asked to rate their satisfaction with various elements of the teaching programme. Statistical differences in their ratings were analysed using the chi-square and Bonferroni tests. RESULTS: Group II gave higher ratings to the clarity of theory classes and lecturers' teaching abilities (p < 0.05) and lecturers' punctuality (p < 0.001) than did Group I. Group II had greater belief that the knowledge assessment tests were useful (p < 0.001) and that their understanding of the subject was good (p < 0.001) than did Group I. Group II gave a higher overall rating to the course (p < 0.05) than did Group I. However, there was no difference in the groups' assessments of the use made of the timetabled hours available for the subject or lecturers' concern for students' learning. CONCLUSIONS: Students were very receptive to the new teaching model

    Binary and Millisecond Pulsars at the New Millennium

    We review the properties and applications of binary and millisecond pulsars. Our knowledge of these exciting objects has greatly increased in recent years, mainly due to successful surveys which have brought the known pulsar population to over 1300. There are now 56 binary and millisecond pulsars in the Galactic disk and a further 47 in globular clusters. This review is concerned primarily with the results and spin-offs from these surveys which are of particular interest to the relativity community.Comment: 59 pages, 26 figures, 5 tables.
