    Measuring vertebrate telomeres: applications and limitations

    Telomeres are short tandem repeated sequences of DNA found at the ends of eukaryotic chromosomes that function in stabilizing chromosomal end integrity. In vivo studies of somatic tissue of mammals and birds have shown a correlation between telomere length and organismal age within species, and correlations between telomere shortening rate and lifespan among species. This result presents the tantalizing possibility that telomere length could be used to provide much needed information on age, ageing and survival in natural populations where longitudinal studies are lacking. Here we review methods available for measuring telomere length and discuss the potential uses and limitations of telomeres as age and ageing estimators in the fields of vertebrate ecology, evolution and conservation

    Rapid circulation of warm subtropical waters in a major glacial fjord in East Greenland

One possible cause is increased melting at the ice/ocean interface driven by the synchronous warming of subtropical waters offshore of Greenland. This hypothesis is largely untested, however, because of the lack of observations from Greenland’s glacial fjords and our limited understanding of their dynamics. Here, we present new ship-based and moored oceanographic data, collected in Sermilik Fjord, a large glacial fjord in East Greenland, showing that subtropical waters are present throughout the fjord and are continuously replenished via a wind-driven exchange with the shelf, where they occur year-round. The temperature and rapid renewal of these waters suggest that, at present, they drive enhanced submarine melting at the terminus. Key controls on the melting rate are the volume and properties of subtropical waters on the shelf and the patterns of the along-shore winds, suggesting the glaciers’ acceleration was triggered by a combination of atmospheric and oceanic changes.     Rapid response of Helheim Glacier in Greenland to climate variability over the past century

The definitive version was published in Nature Geoscience 5 (2012): 37-41, doi:10.1038/ngeo1349.During the early 2000s the Greenland Ice Sheet experienced the largest ice mass loss observed on the instrumental record1, largely as a result of the acceleration, thinning and retreat of major outlet glaciers in West and Southeast Greenland2-5. The quasi-simultaneous change in the glaciers suggests a common climate forcing and increasing air6 and ocean7-8 temperatures have been indicated as potential triggers. Here, we present a new record of calving activity of Helheim Glacier, East Greenland, extending back to c. 1890 AD. This record was obtained by analysing sedimentary deposits from Sermilik Fjord, where Helheim Glacier terminates, and uses the annual deposition of sand grains as a proxy for iceberg discharge. The 120 year long record reveals large fluctuations in calving rates, but that the present high rate was reproduced only in the 1930s. A comparison with climate indices indicates that high calving activity coincides with increased Atlantic Water and decreased Polar Water influence on the shelf, warm summers and a negative phase of the North Atlantic Oscillation. Our analysis provides evidence that Helheim Glacier responds to short-term (3-10 years) large-scale oceanic and atmospheric fluctuations.This study has been supported by Geocenter Denmark in financial support to the SEDIMICE project.     Unconventional Transcriptional Response to Environmental Enrichment in a Mouse Model of Rett Syndrome

    Background: Rett syndrome (RTT) is an X-linked postnatal neurodevelopmental disorder caused by mutations in the gene encoding methyl-CpG binding protein 2 (MeCP2) and one of the leading causes of mental retardation in females. RTT is characterized by psychomotor retardation, purposeless hand movements, autistic-like behavior and abnormal gait. We studied the effects of environmental enrichment (EE) on the phenotypic manifestations of a RTT mouse model that lacks MeCP2 (Mecp2 2/y). Principal Findings: We found that EE delayed and attenuated some neurological alterations presented by Mecp2 2/y mice and prevented the development of motor discoordination and anxiety-related abnormalities. To define the molecular correlate of this beneficial effect of EE, we analyzed the expression of several synaptic marker genes whose expression is increased by EE in several mouse models.     Age before beauty? Relationships between fertilization success and age-dependent ornaments in barn swallows

    When males become more ornamented and reproduce more successfully as they grow older, phenotypic correlations between ornament exaggeration and reproductive success can be confounded with age effects in cross-sectional studies, and thus say relatively little about sexual selection on these traits. This is exemplified here in a correlative study of male fertilization success in a large colony of American barn swallows (Hirundo rustica erythrogaster). Previous studies of this species have indicated that two sexually dimorphic traits, tail length and ventral plumage coloration, are positively correlated with male fertilization success, and a mechanism of sexual selection by female choice has been invoked. However, these studies did not control for potential age-related variation in trait expression. Here, we show that male fertilization success was positively correlated with male tail length but not with plumage coloration. We also show that 1-year-old males had shorter tails and lower fertilization success than older males. This age effect accounted for much of the covariance between tail length and fertilization success. Still, there was a positive relationship between tail length and fertilization success among older males. But as this group consisted of males from different age classes, an age effect may be hidden in this relationship as well. Our data also revealed a longitudinal increase in both tail length and fertilization success for individual males.     Density-Independent Mortality and Increasing Plant Diversity Are Associated with Differentiation of Taraxacum officinale into r- and K-Strategists

    Background: Differential selection between clones of apomictic species may result in ecological differentiation without mutation and recombination, thus offering a simple system to study adaptation and life-history evolution in plants. Methodology/Principal Findings: We caused density-independent mortality by weeding to colonizer populations of the largely apomictic Taraxacum officinale (Asteraceae) over a 5-year period in a grassland biodiversity experiment (Jena Experiment). We compared the offspring of colonizer populations with resident populations deliberately sown into similar communities. Plants raised from cuttings and seeds of colonizer and resident populations were grown under uniform conditions. Offspring from colonizer populations had higher reproductive output, which was in general agreement with predictions of r-selection theory. Offspring from resident populations had higher root and leaf biomass, fewer flower heads and higher individual seed mass as predicted under K-selection. Plants grown from cuttings and seeds differed to some degree in the strength, but not in the direction, of their response to the r- vs. K-selection regime. More diverse communities appeared to exert stronger K-selection on resident populations in plants grown from cuttings, while we did not find significant effects of increasing species richness on plants grown from seeds. Conclusions/Significance: Differentiation into r- and K-strategists suggests that clones with characteristics of r-strategists were selected in regularly weeded plots through rapid colonization, while increasing plant diversity favoured the selection of clones with characteristics of K-strategists in resident populations. Our results show that different selection pressures may result in a rapid genetic differentiation within a largely apomictic species. Even under the assumption that colonizer and resident populations, respectively, happened to be r- vs. K-selected already at the start of the experiment, our results still indicate that the association of these strategies with the corresponding selection regimes was maintained during the 5-year experimental period
