    SYNTHESIS AND ANTICANCER SCREENING OF TRIAZINE ANALOGUES

    Objective: The study was aimed to investigate the cytotoxic effect of S-5H-[1,2,4]-triazino (5,6-b) indol-3-yl-3,4-phenylethane-thioate derivatives as epidermal growth factor Receptor (EGFR) inhibitors. Methods: In the present study 14 novel triazine analogues were synthesized and characterized using different spectroscopic techniques such as FT-IR, NMR and Mass Spectroscopy. The anticancer activity was performed using MCF-7 (breast cancer) and K-562 (leukaemia) cell lines. Further, molecular docking was carried out using Vlife Molecular Docking Software (MDS) on crystal structure of epidermal growth factor receptor (EGFR) to identify the binding mode of interaction with an active site. Results: Compounds MA-7, MA-8, MA-12, MA-13 and MA-14 show potent activity against cancer cell lines in the range of<10 to 84.4 µg/ml. Further molecular docking on EGFR also supports that there is a strong correlation between in silico and in vitro biological activity. The results of this study may be further useful for lead optimization process. Conclusion: The results of this study indicates that the synthesized triazine analogues can give a potential lead as an anticancer agent

    DEVELOPMENT AND VALIDATION OF UV SPECTROSCOPIC METHOD FOR ESTIMATION OF ABACAVIR IN TABLET DOSAGE FORM

    Objective: To develop and validate simple, rapid, linear, accurate, precise and economical UV Spectroscopic method for estimation of Abacavir in tablet dosage form. Methods: The drug is freely soluble in analytical grade methanol. The drug was identified in terms of solubility studies and on the basis of melting point done on the melting point apparatus of Equiptronics. It showed absorption maxima were determined in analytical grade methanol. The drug obeyed the Beer’s law and showed good correlation of concentration with absorption, which reflect in linearity. The UV spectroscopic method was developed for estimation of Abacavir in tablet dosage form and also validated as per ICH guidelines. Results: The drug is freely soluble in analytical grade methanol, slightly soluble in water and practically insoluble in ethanol. So, the analytical grade methanol is used as a diluent in method. The melting point of Abacavir was found to be 164-165 ˚C (uncorrected). It showed absorption maxima 256 nm in analytical grade methanol. On the basis of absorption spectrum the working concentration was set on 15µg/ml (PPM). The linearity was observed between 5-25 μg/ml (PPM). The results of analysis were validated by recovery studies. The recovery was found to be 98.75, 101 and 99.17% for three levels respectively. The % RSD for precision was found to be 0.32% and for Ruggedness is 0.46% Conclusion: A simple, rapid, linear, accurate, precise and economical UV Spectroscopic method has been developed for estimation of Abacavir in tablet dosage form. The method could be considered for the determination of Abacavir in quality control laboratories

    Onychomycosis: dermatophytes to yeasts: an experience in and around Mumbai, Maharashtra, India

    Background: Onychomycosis is a fungal nail infection having wide range of prevalence in different geographical regions. It becomes imperative to know prevalent causative agent in local area to improve quality of life of patients. Present study was aimed to study epidemiological, clinical and laboratory aspects of onychomycosisMethods: Study was carried out prospectively at a tertiary care teaching hospital Nail scrapings of 630 clinically suspected cases of onychomycosis over a period of 5 years (January 2012 to December 2016) were subjected to KOH examination and culture.Results: Young adults in age group of 21-40 years (67.61%) were predominantly affected with male to female ratio of 1.8:1. Overall prevalence of onychomycosis of the present study is 58.41%. Yeasts were isolated in 47.86%, dermatophytes in 30.71% and non dermatophytic filamentous fungi in 21.43%Conclusions: Present study demonstrates a shift in causative agents from dermatophytes to yeasts.

    Churn Identification and Prediction from a Large-Scale Telecommunication Dataset Using NLP

    The identification of customer churn is a major issue for large telecom businesses. In order to manage the data of current customers as well as acquire and manage new customers, every day, a substantial volume of data gets generated. Therefore, it's crucial to identify the causes of client churn so that the appropriate steps can be taken to lower it. Numerous researchers have already discussed their efforts to combine static and dynamic approaches in order to reduce churn in big data sets, but these systems still have many issues when it comes to actually identifying churn. In this paper, we suggested two methods, the first of which is churn identification and using Natural Language Processing (NLP) methods and machine learning techniques, we make predictions based on a vast telecommunication data set. The NLP process involves data pre-processing, normalization, feature extraction, and feature selection. For feature extraction, we employ unique techniques like TF-IDF, Stanford NLP, and occurrence correlation methods, have been suggested. Throughout the lesson, a machine learning classification algorithm is used for training and testing. Finally, the system employs a variety of cross validation techniques and training and evaluating Machine learning algorithms. The experimental analysis shows the system's efficacy and accuracy

    Impact of wild prey availability on livestock predation by snow leopards

Livestock predation by large carnivores and their retaliatory killing is becoming an economic and conservation concern. A common recommendation for carnivore conservation and for reducing predation on livestock is to increase wild prey populations based on the assumption that the carnivores will consume this alternative food. Livestock predation, however, could either reduce or intensify with increases in wild prey depending on prey choice and trends in carnivore abundance. We show that the extent of livestock predation by the endangered snow leopard Panthera uncia intensifies with increases in the density of wild ungulate prey, and subsequently stabilizes. We found that snow leopard density, estimated at seven sites, was a positive linear function of the density of wild ungulates—the preferred prey—and showed no discernible relationship with livestock density. We also found that modelled livestock predation increased with livestock density. Our results suggest that snow leopard conservation would benefit from an increase in wild ungulates, but that would intensify the problem of livestock predation for pastoralists. The potential benefits of increased wild prey abundance in reducing livestock predation can be overwhelmed by a resultant increase in snow leopard populations. Snow leopard conservation efforts aimed at facilitating increases in wild prey must be accompanied by greater assistance for better livestock protection and offsetting the economic damage caused by carnivores.Publisher PDFPeer reviewe

    FAST DISPERSING TABLETS REVIEW

    ABSTRACT In this investigation fast dissolving tablets were prepared using different Superdisintegrants like crospovidone, croscarmellose sodium and sodium starch glycolate by direct compression method. FDTs were evaluated for physicochemical properties like thickness, uniformity of weight, content uniformity ,hadness, friability, wetting time ,dispersion time in vitro disintegration time and in vitro dissolution. Wetting time of formulations containing Croscarmellose sodium was least and tablets showed fastest disintegration. The drug release from FDTs increased with increasing concentration of superdisintegrants and was found to be highest with formulations containing Croscarmellose sodium. The tablet disintegrated within 16 to 45 seconds. Almost 96% of drug was released from the forumaltion within 16 min. The release of drug from FDTs was found to follow non-Fickian diffusion kinetics.Stability studies of the tablets shows non significant change

    Tubular cell and keratinocyte single-cell transcriptomics applied to lupus nephritis reveal type I IFN and fibrosis relevant pathways

The molecular and cellular processes that lead to renal damage and to the heterogeneity of lupus nephritis (LN) are not well understood. We applied single-cell RNA sequencing (scRNA-seq) to renal biopsies from patients with LN and evaluated skin biopsies as a potential source of diagnostic and prognostic markers of renal disease. Type I interferon (IFN)-response signatures in tubular cells and keratinocytes distinguished patients with LN from healthy control subjects. Moreover, a high IFN-response signature and fibrotic signature in tubular cells were each associated with failure to respond to treatment. Analysis of tubular cells from patients with proliferative, membranous and mixed LN indicated pathways relevant to inflammation and fibrosis, which offer insight into their histologic differences. In summary, we applied scRNA-seq to LN to deconstruct its heterogeneity and identify novel targets for personalized approaches to therapy
